    A novel spontaneous model of epithelial-mesenchymal transition (EMT) using a primary prostate cancer derived cell line demonstrating distinct stem-like characteristics

    Cells acquire the invasive and migratory properties necessary for the invasion-metastasis cascade and the establishment of aggressive, metastatic disease by reactivating a latent embryonic programme: epithelial-to-mesenchymal transition (EMT). Herein, we report the development of a new, spontaneous model of EMT which involves four phenotypically distinct clones derived from a primary tumour-derived human prostate cancer cell line (OPCT-1), and its use to explore relationships between EMT and the generation of cancer stem cells (CSCs) in prostate cancer. Expression of epithelial (E-cadherin) and mesenchymal markers (vimentin, fibronectin) revealed that two of the four clones were incapable of spontaneously activating EMT, whereas the others contained large populations of EMT-derived, vimentin-positive cells having spindle-like morphology. One of the two EMT-positive clones exhibited aggressive and stem cell-like characteristics, whereas the other was non-aggressive and showed no stem cell phenotype. One of the two EMT-negative clones exhibited aggressive stem cell-like properties, whereas the other was the least aggressive of all clones. These findings demonstrate the existence of distinct, aggressive CSC-like populations in prostate cancer, but, importantly, that not all cells having a potential for EMT exhibit stem cell-like properties.     Global patient outcomes after elective surgery: prospective cohort study in 27 low-, middle- and high-income countries.

    BACKGROUND: As global initiatives increase patient access to surgical treatments, there remains a need to understand the adverse effects of surgery and define appropriate levels of perioperative care. METHODS: We designed a prospective international 7-day cohort study of outcomes following elective adult inpatient surgery in 27 countries. The primary outcome was in-hospital complications. Secondary outcomes were death following a complication (failure to rescue) and death in hospital. Process measures were admission to critical care immediately after surgery or to treat a complication and duration of hospital stay. A single definition of critical care was used for all countries. RESULTS: A total of 474 hospitals in 19 high-, 7 middle- and 1 low-income country were included in the primary analysis. Data included 44 814 patients with a median hospital stay of 4 (range 2-7) days. A total of 7508 patients (16.8%) developed one or more postoperative complication and 207 died (0.5%). The overall mortality among patients who developed complications was 2.8%. Mortality following complications ranged from 2.4% for pulmonary embolism to 43.9% for cardiac arrest. A total of 4360 (9.7%) patients were admitted to a critical care unit as routine immediately after surgery, of whom 2198 (50.4%) developed a complication, with 105 (2.4%) deaths. A total of 1233 patients (16.4%) were admitted to a critical care unit to treat complications, with 119 (9.7%) deaths. Despite lower baseline risk, outcomes were similar in low- and middle-income compared with high-income countries. CONCLUSIONS: Poor patient outcomes are common after inpatient surgery. Global initiatives to increase access to surgical treatments should also address the need for safe perioperative care. STUDY REGISTRATION: ISRCTN5181700

    Growth response and apparent nutrient digestibility of weaner rabbits fed with diets containing bamboo leaf meal

    A ten-week feeding trial was conducted to determine the growth response and apparent nutrient digestibility of weaner rabbits fed with diets containing bamboo leaf meal (BLM). Eighty weaner rabbits were allotted into four dietary treatment groups of twenty rabbits each. The dietary treatment groups consisted of four rations containing 0 (control), 10, 20 and 30% BLM respectively. Rabbits fed diet containing 10% BLM had the highest (P<0.05) final live weight, weight gain and apparent ether extract digestibility. Lowest (P<0.05) final live weight, weight gain, apparent crude protein and ash digestibility were noticed with rabbits fed with diets containing 20% or 30% BLM. Rabbits fed with diet containing 30% BLM recorded the highest (P<0.05) feed intake and lowest (P<0.05) NDF digestibility. It was concluded that inclusion of 10% BLM in diets for weaner rabbits supported improved growth and nutrient digestibility.Key words: Bamboo leaf meal, digestibility, performance, rabbi

    Socio-Economics and Marketing of Chewing Stick in Ogbomoso Metropolis of Oyo State, Nigeria

    The study assessed trade in chewing stick in Ogbomoso metropolis of Oyo state with a view to identifying and describing the socio-economic characteristics of the chewing stick wholesalers and retailers, identifying and describing the channels of distribution, determiniming the cost and returns structure in chewing stick trade as well as identifying the major problems facing trade in chewing sticks. Purposive random sampling was employed for the selection of one hundred (100) marketers who were interviewed with semi-structured questionnaires to obtain data on their socio-demographic characteristics, years of experience in the business, costs and revenues. Data generated were analyzed using simple descriptive statistical techniques such as frequency distributions, percentages, cross tabulations, marketing margin and correlation analysis. The result shows that 38.2% of the chewing stick Marketers falls within age of 31-46years and 16-30 (35.3%) with females constituting as much as 60%. A greater part (80%) of the respondents was married, with majority (52%) of them having a household size of 1-5 persons. More than half (55.4%) of the respondents have primary school leaving certificates while 35.3% have secondary education. The Marketers have trading experience ranging between 1 to 5 years (52.9%), 6 to 10 years (35.55%) and 11 to 15 years (11.8%). The study concludes that chewing stick trade is a major means of livelihood in Ogbomoso metropolis, providing income for the sustenance of persons engaged in it and their dependants. As a Non-Timber Forest Product collected from the wild; urgent steps need to be taken for its domestication and cultivation to avoid untimely extinction of the source of livelihood of these respondents and their dependants.Keywords: Non-Timber Forest Products, Chewing stick, Marketing, Channels of distribution, Gender

    Interactions of amino acids with aluminum octacarboxyphthalocyanine hydroxide. Experimental and DFT studies

    The influence of albumin and amino acids (l-serine, glycine, l-histidine, l-tryptophan, l-cysteine) on the properties of aluminum octacarboxyphthalocyanine hydroxide (Al(OH)PcOC) was investigated in a phosphate buffer (pH 8.0). Particular attention was paid to the spectroscopic properties and photostability of Al(OH)PcOC. The effect of albumin or amino acids on the photodegradation of Al(OH)PcOC was examined in water using red light: 685 nm and daylight irradiation. Analysis of kinetic curves indicated that interaction with those molecules increases the photostability of Al(OH)PcOC. The molecular structure of Al(OH)PcOC complexes (in vacuum and in water) with axially or equatorially coordinated amino acids was studied by the B3LYP/6-31G* method, and the effects on molecular structure and electronic absorption spectrum were investigated on the basis of the density functional theory. The calculation results revealed that axial coordination significantly reduces the non-planarity of the phthalocyanine ring, and, thus, alters the electronic structure. On the other hand, hydrogen bonding of phthalocyanine side COOH groups with amino acids, in equatorial complexes, does not change the structure within the center of the phthalocyanine, and causes only a slight increase in UV–vis bands intensity, which is in perfect agreement with experimental data.
