    Challenges of Loss to Follow-up in Tuberculosis Research.

    In studies evaluating methods for diagnosing tuberculosis (TB), follow-up to verify the presence or absence of active TB is crucial and high dropout rates may significantly affect the validity of the results. In a study assessing the diagnostic performance of the QuantiFERON®-TB Gold In-Tube test in TB suspect children in Tanzania, factors influencing patient adherence to attend follow-up examinations and reasons for not attending were examined. In 160 children who attended and 102 children who did not attend scheduled 2-month follow-up baseline health characteristics, demographic data and risk factors for not attending follow-up were determined. Qualitative interviews were used to understand patient and caretakers reasons for not returning for scheduled follow-up. Being treated for active tb in the dots program (OR: 4.14; 95% CI:1.99-8.62;p-value<0.001) and receiving money for the bus fare (OR:129; 95% CI 16->100;P-value<0.001) were positive predictors for attending follow-up at 2 months, and 21/85(25%) of children not attending scheduled follow-up had died. Interviews revealed that limited financial resources, i.e. lack of money for transportation and poor communication, were related to non-adherence. Patients lost to follow-up is a potential problem for TB research. Receiving money for transportation to the hospital and communication is crucial for adherence to follow-up conducted at a study facility. Strategies to ensure follow-up should be part of any study protocol

    A Dual Inhibitory Mechanism Sufficient to Maintain Cell-Cycle-Restricted CENP-A Assembly

    Chromatin featuring the H3 variant CENP-A at the centromere is critical for its mitotic function and epigenetic maintenance. Assembly of centromeric chromatin is restricted to G1 phase through inhibitory action of Cdk1/2 kinases in other phases of the cell cycle. Here, we identify the two key targets sufficient to maintain cell-cycle control of CENP-A assembly. We uncovered a single phosphorylation site in the licensing factor M18BP1 and a cyclin A binding site in the CENP-A chaperone, HJURP, that mediated specific inhibitory phosphorylation. Simultaneous expression of mutant proteins lacking these residues results in complete uncoupling from the cell cycle. Consequently, CENP-A assembly is fully recapitulated under high Cdk activities, indistinguishable from G1 assembly. We find that Cdk-mediated inhibition is exerted by sequestering active factors away from the centromere. Finally, we show that displacement of M18BP1 from the centromere is critical for the assembly mechanism of CENP-A

    The ROHP-PAZ Polarimetric Radio Occultation research dataset and its applications

    Trabajo presentado al 7th International Workshop on Occultations for Probing Atmosphere and Climate y al 9th Workshop of the International Radio Occultation Working Group (OPAC-IROWG), celebrados del 8 al 14 de septiembre de 2022 en Leibnitz, Austria.After more than 4 years on orbit, the Radio Occultations and Heavy Precipitation aboard PAZ satellite (ROHP-PAZ) experiment has already demonstrated the ability of polarimetric radio occultations (PRO) to detect precipitation. In fact, PRO have shown potential not only in rain detection, but also in precipitation characterization and in sensing the associated vertical cloud structures. PAZ PRO Δϕ observable profiles were made available in 2020 trough the ICE servers (https://paz.ice.csic.es), (https://genesis.jpl.nasa.gov). and more recently through the JPL A new re-processing of the PRO observations is being carried out with the aim to make it public during the second half of 2022. In addition to a better treatment of the rainy observations, the new re-processed profiles will come with an extensive collocation dataset that will allow the users to address scientific studies much more easily. These will take into account the limb-sounding geometry of the observations, performing the collocations directly into the RO rays obtained through a ray-tracer. These collocations include observations like the 30-minute geostationary 10.8 µm brightness temperature, GPM IMERG surface precipitation, microwave brightness temperatures from the numerous overpasses by the satellites in the GPM constellation, radar reflectivities from the GPM core satellite and the NEXRAD ground based weather radars, among others. Furthermore, the collocation algorithms are designed so that more external observations can be easily included. In addition to the exact collocations as described above, external databases are also checked so that coincidences with Tropical Cyclones, Mesoscale Convective Systems and other relevant precipitating systems are identified nearby PAZ observations. In this presentation, we will show a brief overview of the re-processing of the ROHP-PAZ data, with emphasis in the differences between the Δϕ profiles obtained from UCAR’s CDAAC excess phases and from those obtained from JPL excess phases. After that, examples of the coincident datasets will be presented. Results will include statistics gathered from the differentiation of different precipitation regimes (e.g. stratiform vs convective), identification and validation of cloud top height determination, and comparison with other relevant parameters obtained from the collocated observations.The ROHP-PAZ project is part of the Grant RTI2018-099008-B-C22 funded by the Spanish Ministry of Science and Innovation MCIN/AEI/10.13039/501100011033 and by “ERDF A way of making Europe” of the “European Union”.     Dry reforming of methane over Co–Mo/Al2O3 catalyst under low microwave power irradiation

    In this work, microwave (MW) irradiation was used to activate Co/Al2O3, Mo/Al2O3, and Co-Mo/Al2O3 catalysts for dry reforming of methane (DRM) reactions. Experimental results indicate that single metallic catalysts of either Co or Mo are inactive for DRM under all the tested conditions due to their limited MW-absorbing ability. In contrast, Co-Mo bimetallic catalysts supported by Al2O3 exhibit high catalytic activity due to the formation of a magnetodielectric Co0.82Mo0.18 alloy, which plays the dual role of a good MW acceptor and the provider of active centers for the DRM reaction. The MW power level required to activate such bimetallic catalysts for DRM is significantly dependent on the molar ratio between Co and Mo. The CoMo2 catalyst (with a molar ratio of 2.0 Co to 1.0 Mo) supported on Al2O3 exhibits the best catalytic performance, converting 80% CH4 and 93% CO2 to syngas at a ratio of H2/CO of 0.80 at the total volumetric hourly space velocity (VHSV) of 10 L g-1 h-1 and MW power of 200 W. As compared to the reported C-based catalysts, the Co-Mo/Al2O3 catalyst delivers more favorable stability over 16 time-on-stream (TOS) by virtue of its intrinsic ability to absorb MW without the inclusion of auxiliary MW acceptors

    Near-infrared study of the stellar population of Sh2-152

    Context: The discovery of new massive star clusters and massive stellar populations in previously known clusters in our Galaxy by means of infrared studies has changed our view of the Milky Way from an inactive to an active star-forming machine. Within this scenario, we present a near-infrared spectrophotometric study of the stellar content of the compact H II region Sh2-152. Aims: We aim to determine the distance, extinction, age, and mass of Sh2-152, using for the first time near-infrared stellar classification for several sources in the region. Methods: Using our near-infrared (J, H, and Ks) photometry and the colour-magnitude diagram for the cluster field, we selected 13 bright stars, candidate members of the reddened cluster's main sequence, for H- and K- spectroscopy and spectral classification. This near-infrared information was complemented with an optical spectrum of the ionizing central star to confirm its spectral nature. Results: From the 13 spectroscopically observed stars, 5 were classified as B-type, 3 as G-type, 2 were young stellar objects (YSOs), and 3 remained unclassified (because of the poor data quality). The cluster's extinction varies from A_{Ks}=0.5 to 2.6 magnitudes (A_V=4.5 to 24 magnitudes) and the distance is estimated to be 3.21\pm0.21 kpc. The age of the cluster is younger than 9.4 Myr and the lower limit to the total mass of the cluster is (2.45\pm0.79)\cdot10^3 M_{Sun}. We compare the number of ionizing photons emitted from the OB-type stars with the Lyman continuum photons derived from the radio observations and conclude that both quantities are consistent for the central region of Sh2-152.     Therapeutic efficacy of sulphadoxine-pyrimethamine and chloroquine for the treatment of uncomplicated malaria in pregnancy in Burkina Faso

    BACKGROUND: A reduction in the therapeutic efficacy of chloroquine (CQ) and sulphadoxine-pyrimethamine (SP) has recently been observed in Burkina Faso. As these two drugs are used in pregnancy, their efficacy in pregnant women was studied to directly assess the level of drug resistance in this specific population, rather than to extrapolate results of studies conducted in children < 5 years of age. METHODS: During the malaria transmission season of 2003 in Ouagadougou, the clinical efficacy of SP and CQ, using the WHO 28-day protocol, was assessed in primigravidae and secundigravidae presenting with uncomplicated malaria. RESULTS: PCR-corrected results by day 28 showed that among 62 women treated with SP, eight (12.9%) experienced late parasitological failure, but no clinical failures. Among 60 women treated with CQ, the overall failure rate was 46.7% including 1.7% early treatment failures, 5% late clinical failures and 40% late parasitological failures. SP induced a haemoglobin gain of 0.3 g/dL by day 14 and 0.9 g/dL by day 28. Treatment responses were independent of gravidity, gestational age and prior antenatal care visits. CONCLUSION: While CQ should no longer be used, the efficacy of SP is still compatible with use for intermittent preventive treatment (IPT) in pregnancy. However, given the possible spread of resistance, the drug should be restricted in its use

    Comprehensive Analysis of 5-Aminolevulinic Acid Dehydrogenase (ALAD) Variants and Renal Cell Carcinoma Risk among Individuals Exposed to Lead

    BACKGROUND: Epidemiologic studies are reporting associations between lead exposure and human cancers. A polymorphism in the 5-aminolevulinic acid dehydratase (ALAD) gene affects lead toxicokinetics and may modify the adverse effects of lead. METHODS: The objective of this study was to evaluate single-nucleotide polymorphisms (SNPs) tagging the ALAD region among renal cancer cases and controls to determine whether genetic variation alters the relationship between lead and renal cancer. Occupational exposure to lead and risk of cancer was examined in a case-control study of renal cell carcinoma (RCC). Comprehensive analysis of variation across the ALAD gene was assessed using a tagging SNP approach among 987 cases and 1298 controls. Occupational lead exposure was estimated using questionnaire-based exposure assessment and expert review. Odds ratios (OR) and 95% confidence intervals (CI) were calculated using logistic regression. RESULTS: The adjusted risk associated with the ALAD variant rs8177796(CT/TT) was increased (OR = 1.35, 95%CI = 1.05-1.73, p-value = 0.02) when compared to the major allele, regardless of lead exposure. Joint effects of lead and ALAD rs2761016 suggest an increased RCC risk for the homozygous wild-type and heterozygous alleles ((GG)OR = 2.68, 95%CI = 1.17-6.12, p = 0.01; (GA)OR = 1.79, 95%CI = 1.06-3.04 with an interaction approaching significance (p(int) = 0.06). No significant modification in RCC risk was observed for the functional variant rs1800435(K68N). Haplotype analysis identified a region associated with risk supporting tagging SNP results. CONCLUSION: A common genetic variation in ALAD may alter the risk of RCC overall, and among individuals occupationally exposed to lead. Further work in larger exposed populations is warranted to determine if ALAD modifies RCC risk associated with lead exposure
