Conserved and species-specific molecular denominators in mammalian skeletal muscle aging
Aging is a complex phenomenon involving functional decline in multiple physiological systems. We undertook a comparative analysis of skeletal muscle from four different species, i.e. mice, rats, rhesus monkeys, and humans, at three different representative stages during their lifespan (young, middle, and old) to identify pathways that modulate function and healthspan. Gene expression profiling and computational analysis revealed that pathway complexity increases from mice to humans, and as mammals age, there is predominantly an upregulation of pathways in all species. Two downregulated pathways, the electron transport chain and oxidative phosphorylation, were common among all four species in response to aging. Quantitative PCR, biochemical analysis, mitochondrial DNA measurements, and electron microscopy revealed a conserved age-dependent decrease in mitochondrial content, and a reduction in oxidative phosphorylation complexes in monkeys and humans. Western blot analysis of key proteins in mitochondrial biogenesis discovered that (i) an imbalance toward mitochondrial fusion occurs in aged skeletal muscle and (ii) mitophagy is not overtly affected, presumably leading to the observed accumulation of abnormally large, damaged mitochondria with age. Select transcript expression analysis uncovered that the skeletal inflammatory profile differentially increases with age, but is most pronounced in humans, while increased oxidative stress (as assessed by protein carbonyl adducts and 4-hydroxynonenal) is common among all species. Expression studies also found that there is unique dysregulation of the nutrient sensing pathways among the different species with age. The identification of conserved pathways indicates common molecular mechanisms intrinsic to health and lifespan, whereas the recognition of species-specific pathways emphasizes the importance of human studies for devising optimal therapeutic modalities to slow the aging process
Much Ado About Leptoquarks: A Comprehensive Analysis
We examine the phenomenological implications of a 200 GeV leptoquark in light
of the recent excess of events at HERA. Given the relative predictions of
events rates in e^+p versus e^-p, we demonstrate that classes of leptoquarks
may be excluded, including those contained in E_6 GUT models. It is shown that
future studies with polarized beams at HERA could reveal the chirality of the
leptoquark fermionic coupling and that given sufficient luminosity in each
e^\pm_{L,R} channel the leptoquark quantum numbers could be determined. The
implications of 200-220 GeV leptoquarks at the Tevatron are examined. While
present Tevatron data most likely excludes vector leptoquarks and leptogluons
in this mass region, it does allow for scalar leptoquarks. We find that while
leptoquarks have little influence on Drell-Yan production, further studies at
the Main Injector are possible in the single production channel. We investigate
precision electroweak measurements as well as the process e^+e^-\to q\bar q at
LEP II and find they provide no further restrictions on these leptoquark
models. We then ascertain that cross section and polarization asymmetry
measurements at the NLC provide the only direct mechanism to determine the
leptoquark's electroweak quantum numbers. The single production of leptoquarks
in \gamma e collisions by both the backscattered laser and Weisacker-Williams
techniques at the NLC is also discussed. Finally, we demonstrate that we can
obtain successful coupling constant unification in models with leptoquarks,
both with or without supersymmetry. The supersymmetric case requires the GUT
group to be larger than SU(5) such as flipped SU(5)\times U(1)_X.Comment: Corrected single production cross section at Tevatron, updated atomic

Overexpression of CYB5R3 and NQO1, Two NAD\u3csup\u3e+\u3c/sup\u3e-Producing Enzymes, Mimics Aspects of Caloric Restriction
Calorie restriction (CR) is one of the most robust means to improve health and survival in model organisms. CR imposes a metabolic program that leads to increased stress resistance and delayed onset of chronic diseases, including cancer. In rodents, CR induces the upregulation of two NADH‐dehydrogenases, namely NAD(P)H:quinone oxidoreductase 1 (Nqo1) and cytochrome b5 reductase 3 (Cyb5r3), which provide electrons for energy metabolism. It has been proposed that this upregulation may be responsible for some of the beneficial effects of CR, and defects in their activity are linked to aging and several age‐associated diseases. However, it is unclear whether changes in metabolic homeostasis solely through upregulation of these NADH‐dehydrogenases have a positive impact on health and survival. We generated a mouse that overexpresses both metabolic enzymes leading to phenotypes that resemble aspects of CR including a modest increase in lifespan, greater physical performance, a decrease in chronic inflammation, and, importantly, protection against carcinogenesis, one of the main hallmarks of CR. Furthermore, these animals showed an enhancement of metabolic flexibility and a significant upregulation of the NAD+/sirtuin pathway. The results highlight the importance of these NAD+ producers for the promotion of health and extended lifespan
Search for SUSY with R-Parity Violation at gamma-proton and gamma-electron colliders
We present an outlook for possible R-parity violating SUSY search at
gamma-proton and gamma-electron colliders. Single production of third
generation slepton/sneutrino through the lambda'-ijk couplings at a gamma-p
collider is investigated and compared with the results of the resonant sfermion
productions at the existing colliders. Also single sneutrino production at a
future gamma-electron collider through the lambda(ijk) couplings is disussed.Comment: 7 pages, 3 figures, enlarged version, a section adde

Metformin improves healthspan and lifespan in mice
Metformin is a drug commonly prescribed to treat patients with type 2 diabetes. Here we show that long-term treatment with metformin (0.1% w/w in diet) starting at middle age extends healthspan and lifespan in male mice, while a higher dose (1% w/w) was toxic. Treatment with metformin mimics some of the benefits of calorie restriction, such as improved physical performance, increased insulin sensitivity, and reduced LDL and cholesterol levels without a decrease in caloric intake. At a molecular level, metformin increases AMP-activated protein kinase activity and increases antioxidant protection, resulting in reductions in both oxidative damage accumulation and chronic inflammation. Our results indicate that these actions may contribute to the beneficial effects of metformin on healthspan and lifespan. These findings are in agreement with current epidemiological data and raise the possibility of metformin-based interventions to promote healthy aging
Search for the standard model Higgs boson in the H to ZZ to 2l 2nu channel in pp collisions at sqrt(s) = 7 TeV
A search for the standard model Higgs boson in the H to ZZ to 2l 2nu decay
channel, where l = e or mu, in pp collisions at a center-of-mass energy of 7
TeV is presented. The data were collected at the LHC, with the CMS detector,
and correspond to an integrated luminosity of 4.6 inverse femtobarns. No
significant excess is observed above the background expectation, and upper
limits are set on the Higgs boson production cross section. The presence of the
standard model Higgs boson with a mass in the 270-440 GeV range is excluded at
95% confidence level.Comment: Submitted to JHE
Clinico-pathological characteristics and outcomes of patients with early-onset colorectal cancer
[Background]: The rising incidence of colorectal cancer (CRC) among young patients is alarming. We aim to characterize the clinico-pathological features and outcomes of patients with early-onset CRC (EOCRC).
[Methods]: We included all of the patients with pathologically confirmed diagnosis of CRC at Hospital Universitario La Paz from October 2016 to September 2020. EOCRC age cut-off was 50 years. All statistical analyses were carried out using SPSS v.25.
[Results]: A total of 1152 patients were diagnosed with CRC, fifty-nine (5,1%) of them were After a median follow-up of 24 months, 279 patients have died. Median overall survival (OS) was not reached in either group (p = 0,06). Three-year OS was 80% (95%CI: 73-87) and 67 (95%CI: 65-69) in the younger and older group, respectively. In patients with localized disease that underwent surgery or other antineoplastic treatment ( n = 856), 159 events for disease-free survival (DFS) were observed. Median DFS was
[Conclusions]: Patients with EOCRC are diagnosed at a more advanced stage and display distinct biological features (more prevalence of dMMR and WT tumors among others). Studies focusing on screening in this population and deeper molecular profiling are needed
