From autism to eating disorders and more: the role of oxytocin in neuropsychiatric disorders

    Get PDF
Oxytocin (oxy) is a pituitary neuropeptide hormone synthesized from the paraventricular and supraoptic nuclei within the hypothalamus. Like other neuropeptides, oxy can modulate a wide range of neurotransmitter and neuromodulator activities. Additionally, through the neurohypophysis, oxy is secreted into the systemic circulation to act as a hormone, thereby influencing several body functions. Oxy plays a pivotal role in parturition, milk let-down and maternal behavior and has been demonstrated to be important in the formation of pair bonding between mother and infants as well as in mating pairs. Furthermore, oxy has been proven to play a key role in the regulation of several behaviors associated with neuropsychiatric disorders, including social interactions, social memory response to social stimuli, decision-making in the context of social interactions, feeding behavior, emotional reactivity, etc. An increasing body of evidence suggests that deregulations of the oxytocinergic system might be involved in the pathophysiology of certain neuropsychiatric disorders such as autism, eating disorders, schizophrenia, mood, and anxiety disorders. The potential use of oxy in these mental health disorders is attracting growing interest since numerous beneficial properties are ascribed to this neuropeptide. The present manuscript will review the existing findings on the role played by oxy in a variety of distinct physiological and behavioral functions (Figure 1) and on its role and impact in different psychiatric disorders. The aim of this review is to highlight the need of further investigations on this target that might contribute to the development of novel more efficacious therapies. Aims: Activation of PPARγ by pioglitazone (PIO) has shown some efficacy in attenuating addictive-like responses in laboratory animals. The ability of PIO to alter the effects of opioids in humans has not been characterized in a controlled laboratory setting. The proposed investigation sought to examine the effects of PIO on the subjective, analgesic, physiological and cognitive effects of oxycodone (OXY). Methods: During this investigation, nondependent prescription opioid abusers (N = 17 completers) were maintained for 2-3 weeks on ascending daily doses of PIO (0 mg, 15 mg, 45 mg) prior to completing a laboratory session assessing the aforementioned effects of OXY [using a within-session cumulative dosing procedure (0, 10, and 20 mg, cumulative dose = 30 mg)]. Results: OXY produced typical mu opioid agonist effects: miosis, decreased pain perception, and decreased respiratory rate. OXY also produced dose-dependent increases in positive subjective responses. Yet, ratings such as: drug "liking," "high," and "good drug effect," were not significantly altered as a function of PIO maintenance dose. Discussion: These data suggest that PIO may not be useful for reducing the abuse liability of OXY. These data were obtained with a sample of nondependent opioid users and therefore may not be applicable to dependent populations or to other opioids. Although PIO failed to alter the abuse liability of OXY, the interaction between glia and opioid receptors is not well understood so the possibility remains that medications that interact with glia in other ways may show more promise

A novel method of pure oxy-fuel circulating fluidized bed combustion with zero recirculation flue gas : experimental validation

Applying oxy-fuel combustion requires more advanced combustion control methods to avoid inadmissible high flam temperature. In fluidized beds and pulverized unites, enhanced heat transfer and recirculation flue gas are used. On other hand, higher oxygen concentration has pluses viz. better heat transfer, higher efficiency, compact setup and lower installation and operating costs. In pulverized power unites, pure oxy-fuel combustion is used with 100% O2 in the oxidant. In contrast, the highest experimental O2 % in oxy-fuel circulating fluidized bed (CFB) combustor is 70%. To the best of authors’ knowledge, there is no single CFB power plant operating under pure oxygen condition. In this work, we are aiming to use pure oxygen for oxy-CFB combustion, with new temperature controlling method for CFBs depending on combustion staging by fuel staging rather than using RFG. Fuel staging allows controlling combustion and varying SR. At the first stage, the used oxidant is 100% O2, and fuel is fed to achieve over SR (λ>1), where the excess oxidant absorbs heat and does not take a part in the reaction. The products of the first stage are reach of O2 and subsequently it is used as an oxidant for the second stage. For validation, a series of experiments are conducted using mini-CFB, and an oxidant of 100% O2 concentration is used with three SR ratios λ=1.25, 2.0, and 3.0. The resulted average temperatures along the riser for biomass are 1031°C, 950°C, and 798°C; and for coal 1129 °C, 1051 °C, and 961 °C respectively. The controlling of AFT with pure oxy-fuel combustion eliminates the recycled flue gas (RFG) in oxy-fuel CFB combustion and flue gas recirculation section; this simplifies the power plants’ design, fabrication and its installing-operating costs. Familiarising this concept can accelerate adapting oxy-fuel combustion in CFB power plant for Carbon Capturing and Sequestration (CCS). This contribution can commence and commercialise the third generation of oxy-fuel CFB combustion with zero recycled flue gas. Finally, the concept of controlling AFT by SR (λ) is validated experimentally

In this paper, the thermodynamic advantage of integrating liquid air power generation (LAPG) process and binary cycle waste heat recovery technology to a standalone pressurized oxy-coal combustion supercritical steam power generation cycle is investigated through modeling and simulation using Aspen Plus® simulation software version 8.4. The study shows that the integration of LAPG process and the use of binary cycle heat engine which convert waste heat from compressor exhaust to electricity, in a standalone pressurized oxy-coal combustion supercritical steam power generation cycle improves the thermodynamic efficiency of the pressurized oxy-coal process. The analysis indicates that such integration can give about 12–15% increase in thermodynamic efficiency when compared with a standalone pressurized oxy-coal process with or without CO2 capture. It was also found that in a pressurized oxy-coal process, it is better to pump the liquid oxygen from the cryogenic ASU to a very high pressure prior to vapourization in the cryogenic ASU main heat exchanger and subsequently expand the gaseous oxygen to the required combustor pressure than either compressing the atmospheric gaseous oxygen produced from the cryogenic ASU directly to the combustor pressure or pumping the liquid oxygen to the combustor pressure prior to vapourization in the cryogenic ASU main heat exchanger. The power generated from the compressor heat in the flue gas purification, carbon capture and compression unit using binary cycle heat engine was also found to offset about 65% of the power consumed in the flue gas cleaning and compression process. The work presented here shows that there is a synergistic and thermodynamic advantage of utilizing the nitrogen-rich stream from the cryogenic ASU of an oxy-fuel power generation process for power generation instead of discarding it as a waste stream

Aryltrioxorhenium (ArReO_3) has been demonstrated to show rapid oxy-functionalization upon reaction with O-atom donors, YO, to selectively generate the corresponding phenols in near quantitative yields. (18)^O-Labeling experiments show that the oxygen in the products is exclusively from YO. DFT studies reveal a 10.7 kcal/mol barrier (Ar = Ph) for oxy-functionalization with H_2O_2 via a Baeyer-Villiger type mechanism involving nudeophilic attack of the aryl group on an electrophilic oxygen of YO coordinated to rhenium

Oxy-functionalization of nucleophilic rhenium(I) metal carbon bonds catalyzed by selenium(IV)

We report that SeO_2 catalyzes the facile oxy-functionalization of (CO)_5Re(I)-Me^(δ−) with IO_4− to generate methanol. Mechanistic studies and DFT calculations reveal that catalysis involves methyl group transfer from Re to the electrophilic Se center followed by oxidation and subsequent reductive functionalization of the resulting CH_3Se(VI) species. Furthermore, (CO)_3Re(I)(Bpy)-R (R = ethyl, n-propyl, and aryl) complexes show analogous transfer to SeO_2 to generate the primary alcohols. This represents a new strategy for the oxy-functionalization of M−R^(δ−) polarized bonds
