    Acute effects of nicotine on visual search tasks in young adult smokers

    Rationale Nicotine is known to improve performance on tests involving sustained attention and recent research suggests that nicotine may also improve performance on tests involving the strategic allocation of attention and working memory. Objectives We used measures of accuracy and response latency combined with eye-tracking techniques to examine the effects of nicotine on visual search tasks. Methods In experiment 1 smokers and non-smokers performed pop-out and serial search tasks. In experiment 2, we used a within-subject design and a more demanding search task for multiple targets. In both studies, 2-h abstinent smokers were asked to smoke one of their own cigarettes between baseline and tests. Results In experiment 1, pop-out search times were faster after nicotine, without a loss in accuracy. Similar effects were observed for serial searches, but these were significant only at a trend level. In experiment 2, nicotine facilitated a strategic change in eye movements resulting in a higher proportion of fixations on target letters. If the cigarette was smoked on the first trial (when the task was novel), nicotine additionally reduced the total number of fixations and refixations on all letters in the display. Conclusions Nicotine improves visual search performance by speeding up search time and enabling a better focus of attention on task relevant items. This appears to reflect more efficient inhibition of eye movements towards task irrelevant stimuli, and better active maintenance of task goals. When the task is novel, and therefore more difficult, nicotine lessens the need to refixate previously seen letters, suggesting an improvement in working memory

    The antisaccade task as an index of sustained goal activation in working memory: modulation by nicotine

    The antisaccade task provides a laboratory analogue of situations in which execution of the correct behavioural response requires the suppression of a more prepotent or habitual response. Errors (failures to inhibit a reflexive prosaccade towards a sudden onset target) are significantly increased in patients with damage to the dorsolateral prefrontal cortex and patients with schizophrenia. Recent models of antisaccade performance suggest that errors are more likely to occur when the intention to initiate an antisaccade is insufficiently activated within working memory. Nicotine has been shown to enhance specific working memory processes in healthy adults. MATERIALS AND METHODS: We explored the effect of nicotine on antisaccade performance in a large sample (N = 44) of young adult smokers. Minimally abstinent participants attended two test sessions and were asked to smoke one of their own cigarettes between baseline and retest during one session only. RESULTS AND CONCLUSION: Nicotine reduced antisaccade errors and correct antisaccade latencies if delivered before optimum performance levels are achieved, suggesting that nicotine supports the activation of intentions in working memory during task performance. The implications of this research for current theoretical accounts of antisaccade performance, and for interpreting the increased rate of antisaccade errors found in some psychiatric patient groups are discussed

Uurimistöös on kasutatud erinevaid eksperimentaalseid katseparadigmasid ja katsestiimuleid ning nii käitumuslikke- kui ka ajukuvamismeetodeid. Esimesed kolm empiirilist uurimust käsitlevad liikumisinformatsiooni töötlust, mis on evolutsiooni käigus kujunenud üheks olulisemaks baasprotsessiks nägemistajus. Esmalt huvitas meid, kuidas avastatakse liikuva objekti suunamuutusi, kui samal ajal toimub ka taustal liikumine (Uurimus I). Nägemistaju uurijad on pikka aega arvanud, et liikumist arvutatakse alati mõne välise objekti või tausta suhtes. Meie uurimistulemused ei kinnitanud taolise suhtelise liikumise printsiibi paikapidavust ning toetavad pigem seisukohta, et eesmärkobjekti liikumisinformatsiooni töötlus on automaatne protsess, mis tuvastab silma põhjas toimuvaid nihkeid, ja taustal toimuv seda eriti ei mõjuta. Teise uurimuse tulemused (Uurimus II) näitasid, et nägemissüsteem töötleb väga edukalt ka seda liikumisinformatsiooni, millele vaatleja teadlikult tähelepanu ei pööra. See tähendab, et samal ajal, kui inimene on mõne tähelepanu hõlmava tegevusega ametis, suudab tema aju taustal toimuvaid sündmusi automaatselt registreerida. Igapäevaselt on inimese nägemisväljas alati palju erinevaid objekte, millel on erinevad omadused, mistõttu järgmiseks huvitas meid (Uurimus III), kuidas ühe tunnuse (antud juhul värvimuutuse) töötlemist mõjutab mõne teise tunnusega toimuv (antud juhul liikumiskiiruse) muutus. Näitasime, et objekti liikumine parandas sama objekti värvimuutuse avastamist, mis viitab, et nende kahe omaduse töötlemine ajus ei ole päris eraldiseisev protsess. Samuti tähendab taoline tulemus, et hoolimata ühele tunnusele keskendumisest ei suuda inimene ignoreerida teist tähelepanu tõmbavat tunnust (liikumine), mis viitab taas kord automaatsetele töötlusprotsessidele. Neljas uurimus keskendus emotsionaalsete näoväljenduste töötlusele, kuna need kannavad keskkonnas hakkamasaamiseks vajalikke sotsiaalseid signaale, mistõttu on alust arvata, et nende töötlus on kujunenud suuresti automaatseks protsessiks. Näitasime, et emotsiooni väljendavaid nägusid avastati kiiremini ja kergemini kui neutraalse ilmega nägusid ning et vihane nägu tõmbas rohkem tähelepanu kui rõõmus (Uurimus IV). Väitekirja viimane osa puudutab visuaalset lahknevusnegatiivsust (ingl Visual Mismatch Negativity ehk vMMN), mis näitab aju võimet avastada automaatselt erinevusi enda loodud mudelist ümbritseva keskkonna kohta. Selle automaatse erinevuse avastamise mehhanismi uurimisse andsid oma panuse nii Uurimus II kui Uurimus IV, mis mõlemad pakuvad välja tõendusi vMMN tekkimise kohta eri tingimustel ja katseparadigmades ning ka vajalikke metodoloogilisi täiendusi. Uurimus V on esimene kogu siiani ilmunud temaatilist teadustööd hõlmav ülevaateartikkel ja metaanalüüs visuaalsest lahknevusnegatiivsusest psühhiaatriliste ja neuroloogiliste haiguste korral, mis panustab oluliselt visuaalse lahknevusnegatiivsuse valdkonna arengusse.The research presented and discussed in the thesis is an experimental exploration of processes in visual perception, which all display a considerable amount of automaticity. These processes are targeted from different angles using different experimental paradigms and stimuli, and by measuring both behavioural and brain responses. In the first three empirical studies, the focus is on motion detection that is regarded one of the most basic processes shaped by evolution. Study I investigated how motion information of an object is processed in the presence of background motion. Although it is widely believed that no motion can be perceived without establishing a frame of reference with other objects or motion on the background, our results found no support for relative motion principle. This finding speaks in favour of a simple and automatic process of detecting motion, which is largely insensitive to the surrounding context. Study II shows that the visual system is built to automatically process motion information that is outside of our attentional focus. This means that even if we are concentrating on some task, our brain constantly monitors the surrounding environment. Study III addressed the question of what happens when multiple stimulus qualities (motion and colour) are present and varied, which is the everyday reality of our visual input. We showed that velocity facilitated the detection of colour changes, which suggests that processing motion and colour is not entirely isolated. These results also indicate that it is hard to ignore motion information, and processing it is rather automatically initiated. The fourth empirical study focusses on another example of visual input that is processed in a rather automatic way and carries high survival value – emotional expressions. In Study IV, participants detected emotional facial expressions faster and more easily compared with neutral facial expressions, with a tendency towards more automatic attention to angry faces. In addition, we investigated the emergence of visual mismatch negativity (vMMN) that is one of the most objective and efficient methods for analysing automatic processes in the brain. Study II and Study IV proposed several methodological gains for registering this automatic change-detection mechanism. Study V is an important contribution to the vMMN research field as it is the first comprehensive review and meta-analysis of the vMMN studies in psychiatric and neurological disorders

    The Elaborated Intrusion Theory of Desire: A 10-year retrospective and implications for addiction treatments

    Ten years after the publication of Elaborated Intrusion (EI) Theory, there is now substantial research into its key predictions. The distinction between intrusive thoughts, which are driven by automatic processes, and their elaboration, involving controlled processing, is well established. Desires for both addictive substances and other desired targets are typically marked by imagery, especially when they are intense. Attention training strategies such as body scanning reduce intrusive thoughts, while concurrent tasks that introduce competing sensory information interfere with elaboration, especially if they compete for the same limited-capacity working memory resources. EI Theory has spawned new assessment instruments that are performing strongly and offer the ability to more clearly delineate craving from correlated processes. It has also inspired new approaches to treatment. In particular, training people to use vivid sensory imagery for functional goals holds promise as an intervention for substance misuse, since it is likely to both sustain motivation and moderate craving

    Adolescent brain cognitive development (ABCD) study: Overview of substance use assessment methods.

    One of the objectives of the Adolescent Brain Cognitive Development (ABCD) Study (https://abcdstudy.org/) is to establish a national longitudinal cohort of 9 and 10 year olds that will be followed for 10 years in order to prospectively study the risk and protective factors influencing substance use and its consequences, examine the impact of substance use on neurocognitive, health and psychosocial outcomes, and to understand the relationship between substance use and psychopathology. This article provides an overview of the ABCD Study Substance Use Workgroup, provides the goals for the workgroup, rationale for the substance use battery, and includes details on the substance use module methods and measurement tools used during baseline, 6-month and 1-year follow-up assessment time-points. Prospective, longitudinal assessment of these substance use domains over a period of ten years in a nationwide sample of youth presents an unprecedented opportunity to further understand the timing and interactive relationships between substance use and neurocognitive, health, and psychopathology outcomes in youth living in the United States

    Interaction of the phencyclidine model of schizophrenia and nicotine on total and categorized ultrasonic vocalizations in rats

    Patients with schizophrenia smoke cigarettes at a higher rate than the general population. We hypothesized that a factor in this comorbidity is sensitivity to the reinforcing and reinforcement- enhancement effects of nicotine. Phencyclidine (PCP) was used to model behavioral changes resembling negative symptoms of schizophrenia in rats. USVs in rats have been used to measure emotional states, with 50 kHz USVs indicating positive states and 22 kHz indicating negative. Total and categorized numbers of 22 and 50 kHz ultrasonic vocalizations (USVs) and USVs during a visual stimulus (e.g. a potential measure of reinforcement-enhancement) were examined in rats following .injection ofh PCP (2.0 mg/kg), and/or nicotine (0.2 or 0.4 mg/kg) daily for 7 days. PCP was then discontinued and all rats received nicotine (0.2 mg/kg and 0.4 mg/kg) and PCP (2.0 mg/kg) on 3 challenge days. PCP acutely decreased 50 kHz vocalizations while repeated nicotine potentiated rates of vocalizations, with similar patterns during light presentations. Rats in the PCP and nicotine combination groups made more 50 kHz vocalizations compared to control groups on challenge days. We conclude that PCP may produce a reward deficit that is shown by decreased 50 kHz USVs, and behaviors post-PCP exposure may best model the comorbidity between schizophrenia and nicotine

    The authors argue that human desire involves conscious cognition that has strong affective connotation and is potentially involved in the determination of appetitive behavior rather than being epiphenomenal to it. Intrusive thoughts about appetitive targets are triggered automatically by external or physiological cues and by cognitive associates. When intrusions elicit significant pleasure or relief, cognitive elaboration usually ensues. Elaboration competes with concurrent cognitive tasks through retrieval of target-related information and its retention in working memory. Sensory images are especially important products of intrusion and elaboration because they simulate the sensory and emotional qualities of target acquisition. Desire images are momentarily rewarding but amplify awareness of somatic and emotional deficits. Effects of desires on behavior are moderated by competing incentives, target availability, and skills. The theory provides a coherent account of existing data and suggests new directions for research and treatment
