    Integration of biomolecular logic principles with electronic transducers on a chip

    Boolean operations applied in biology and integrated with electronic transducers allow the development of a new class of digital biosensors for the detection of multiple input signals simultaneously and in real-time. With the help of Boolean functions (AND, OR, etc.), an electrical output signal will be directly delivered, representing a ”1” or “0” binary notation, corresponding to a “true” or “false” statement, respectively. Such digital biosensors have the future potential to create medical devices and systems for intelligent or smart diagnostics. The present thesis describes the realization of different enzyme-based biomolecular logic gates combined with electronic transducers for the possible application in medicine or food industry. In a first concept, a so called BioLogicChip is developed combining a “sense-act-treat” function integrated on one chip. The present system exemplarily mimics an “artificial pancreas” designed as a closed-loop drug-release system. A glucose sensor is constructed as enzyme-based AND logic gate, a temperature-depending hydrogel imitates the actuator function switching ON and OFF with its shrinking or swelling property, and an additional insulin sensor is developed to monitor and control the release of the drug (here: insulin) from the actuator. In this study, the results of the individual components such as the amperometric glucose sensor, the temperature-dependent hydrogel and the amperometric insulin sensor are presented, which are necessary to create such BioLogicChip. Moreover, a digital adrenaline biosensor is developed to proof the catheter position during adrenal vein sampling. The sensor consists of an oxygen electrode modified by a bi-enzyme system with the enzymes laccase and pyrroloquinoline quinone-dependent glucose dehydrogenase (PQQ-GDH) to realize substrate-recycling principle to detect low adrenaline concentrations (in the nanomolar concentration range). The sensor`s behavior at different pH values and at different temperatures is studied. Measurements in Ringer`s solution are performed. In addition, the sensitivity of the biosensor to other catecholamines such as noradrenaline, dopamine and dobutamine is investigated. Furthermore, the adrenaline biosensor is successfully examined in human blood plasma. Finally, “proof-of-principle” experiments have been performed by combining the adrenaline biosensor with Boolean operations to get a rapid qualitative statement of the presence or absence of adrenaline, thus validating the correct position of the catheter in a YES/NO form. This adrenaline biosensor is further miniaturized as a thin-film platinum adrenaline biosensor. Here, the bioelectrocatalytical measurement principle is applied by immobilization of the enzyme PQQ-GDH to detect adrenaline in the nanomolar concentration range, too. The measurement conditions such as pH value, glucose concentration in the analyte solution and temperature are optimized with regard to a high sensitivity and low detection limit. Also, this sensor has been verified towards other catecholamines (noradrenaline, dopamine and dobutamine). The platinum thin-film adrenaline biosensor is successfully applied in blood plasma for the detection of different spiked adrenaline concentrations. Furthermore, the developed adrenalin biosensor is able to detect the concentration difference between adrenal blood and peripheral blood. In contrast to the above-mentioned amperometric biosensor examples for biomolecular gates, also a field-effect-based platform is given attention in this thesis. The field-effect electrolyte-insulator-semiconductor (EIS) sensor consists of a layer structure of Al/p-Si/SiO2/Ta2O5 and is used to create an acetoin biosensor for the first time to control different fermentation processes. The sensor chip is modified by the enzyme acetoin reductase from B. clausii DSM 8716T for the catalytical reaction of (R)-acetoin to (R,R)-butanediol and meso-butanediol, respectively, in the presence of NADH. The linear measurement range, the optimal immobilization strategy (cross-linking by using glutaraldehyde and adsorptive binding) as well as the optimal working pH value and long-term stability are investigated by means of constant-capacitance measurements. Finally, the acetoin sensor was successfully applied in wine probes to detect different spiked acetoin concentrations.     Potential use of electronic noses, electronic tongues and biosensors, as multisensor systems for spoilage examination in foods

    Development and use of reliable and precise detecting systems in the food supply chain must be taken into account to ensure the maximum level of food safety and quality for consumers. Spoilage is a challenging concern in food safety considerations as it is a threat to public health and is seriously considered in food hygiene issues accordingly. Although some procedures and detection methods are already available for the determination ofspoilage in food products, these traditional methods have some limitations and drawbacks as they are time-consuming,labour intensive and relatively expensive. Therefore, there is an urgent need for the development of rapid, reliable, precise and non-expensive systems to be used in the food supply and production chain as monitoring devices to detect metabolic alterations in foodstuff. Attention to instrumental detection systems such as electronic noses, electronic tongues and biosensors coupled with chemometric approaches has greatly increased because they have been demonstrated as a promising alternative for the purpose of detecting and monitoring food spoilage. This paper mainly focuses on the recent developments and the application of such multisensor systems in the food industry. Furthermore, the most traditionally methods for food spoilage detection are introduced in this context as well. The challenges and future trends of the potential use of the systems are also discussed. Based on the published literature, encouraging reports demonstrate that such systems are indeed the most promising candidates for the detection and monitoring of spoilage microorganisms in different foodstuff

Dentro de este campo, el concepto de lengua electrĂłnica (ETs o e-tongues) ha crecido rĂĄpidamente en los Ășltimos años debido a su gran potencial. Estos dispositivos se basan en sensores electroquĂ­micos combinados con anĂĄlisis de datos multivariantes. De acuerdo con la IUPAC (UniĂłn Internacional de QuĂ­mica Pura y Aplicada), una lengua electrĂłnica es un sistema multisensor, que consiste en un nĂșmero de sensores de baja selectividad y utiliza procedimientos matemĂĄticos avanzados para el procesamiento de señales basados en el reconocimiento de patrones (PARC) y/o anĂĄlisis multivariante [redes neuronales artificiales (RNA), anĂĄlisis de componentes principales (PCA), etc.]. Por lo tanto, las ETs son sistemas holĂ­sticos que proporcionan informaciĂłn global y cualitativa acerca de la muestra en lugar de datos cuantitativos acerca de compuestos especĂ­ficos. Sin embargo, si la matriz de datos obtenida por estos sistemas se analiza con herramientas de procesamiento quimiomĂ©trico adecuadas, se podrĂ­a extraer informaciĂłn descriptiva o predictiva de parĂĄmetros especĂ­ficos.     Valorization of Food Processing By-Products as Smart Food Packaging Materials and Its Application

    Traditional food packaging systems cannot provide any information related to the food quality during storage to consumers. Recently, the renewable resources have been considered as starting materials for making biodegradable packaging film. A variety of food processing by-products have been utilized, either alone or in mixtures, to produce packaging films with proper properties. It shows high possibility for smart biodegradable filmmaking as well as is applicable in the food industry. In order to monitor the food quality and to reduce the food loss and waste, a new packaging technology has been increasingly developed. Smart packaging refers to packaging systems which can monitor, detect, and inform about the qualities of food in real time. Indicator is the most commonly used device, which can communicate through direct visual change, especially in color. Natural extract and synthetic color are usually added into smart packaging films. However, synthetic dyes may be harmful to the consumers’ health. Thus, the use of natural extract has been increased. Smart packaging films can be applied to various types of food products in order to monitor the food quality during transportation and storage. Thus, smart packaging could be used as a nondestructive tool to detect the food quality

    Fundamentals and application of voltammetric electronic tongues in quantitative analysis

Through the application of chemometrics, it is possible to infer underlying relationships between the measured analytical signals and the chemical properties of the samples, both for descriptive and predictive purposes, otherwise impossible to decipher. Research in voltammetric ETs during the last two decades has demonstrated the benefits derived from the use of sensor arrays with complementary response, together with advanced data treatment methods to enhance their overall performance. In this direction, the different approaches followed when developing voltammetric ETs and some relevant applications in quantitative analysis are reviewed herein

    Instrumentation and control of anaerobic digestion processes: a review and some research challenges

    The final publication is available at Springer via http://dx.doi.org/10.1007/s11157-015-9382-6[EN] To enhance energy production from methane or resource recovery from digestate, anaerobic digestion processes require advanced instrumentation and control tools. Over the years, research on these topics has evolved and followed the main fields of application of anaerobic digestion processes: from municipal sewage sludge to liquid mainly industrial then municipal organic fraction of solid waste and agricultural residues. Time constants of the processes have also changed with respect to the treated waste from minutes or hours to weeks or months. Since fast closed loop control is needed for short time constant processes, human operator is now included in the loop when taking decisions to optimize anaerobic digestion plants dealing with complex solid waste over a long retention time. Control objectives have also moved from the regulation of key variables measured online to the prediction of overall process perfor- mance based on global off-line measurements to optimize the feeding of the processes. Additionally, the need for more accurate prediction of methane production and organic matter biodegradation has impacted the complexity of instrumentation and should include a more detailed characterization of the waste (e.g., biochemical fractions like proteins, lipids and carbohydrates)andtheirbioaccessibility andbiodegradability characteristics. However, even if in the literature several methodologies have been developed to determine biodegradability based on organic matter characterization, only a few papers deal with bioaccessibility assessment.     Sensores: De los biosensores a la nariz electrĂłnica

    The recent advances in sensor devices have allowed the developing of new applications in many technological fields. This review describes the current state-of-the-art of this sensor technology, placing special emphasis on the food applications. The design, technology and sensing mechanism of each type of sensor are analysed. A description of the main characteristics of the electronic nose and electronic tongue (taste sensors) is also given. Finally, the applications of some statistical procedures in sensor systems are described briefly.Los recientes avances en los sistemas de sensores han permitido el desarrollo de nuevas aplicaciones en muchos campos tecnológicos. Este artículo de revisión describe el estado actual de esta nueva tecnología, con especial énfasis en las aplicaciones alimentarias. El diseño, la tecnología y el mecanismo sensorial de cada tipo de sensor son analizados en el artículo. También se describen las principales características de la nariz y la lengua electrónica (sensores de sabor). Finalmente, se describe brevemente el uso de algunos procedimientos estadísticos en sistemas de sensores.Peer reviewe
