    Games and Roleplaying in the Classroom

    The Library Ambassador Program hires approximately 20 undergraduate students each fall and provides them with two semesters of information literacy and research skills instruction before deploying them across campus to help students with their research. As part of their training, instructors use various game-play strategies in the classroom including: a card game designed to teach players about databases and how they function, a card game intended to teach players how to evaluate sources of information, and a roleplaying activity meant to prepare ambassadors for experiences in helping students. Many emotions are involved in the action of gameplay such as competitiveness, satisfaction, and excitement. The card games and roleplaying activities we play in class harness these emotions to create a fun and engaging way to develop research skills. Games in this context also provide opportunities for collaborative learning as students work together to problem-solve and to learn new skills

    Wastewater and Mixed Microbial Consortia: a metastudy analysis of Optimal Microbial Fuel Cell configuration

    Microbial Fuel Cells (MFCs) are an area of increasing research for use as an alternative energy source, due to their ability to produce electricity while simultaneously treating organic waste. This meta-study determines the optimal MFC configuration for electricity production, through consideration of the biocatalyst and substrate used. This study focuses primarily on comparing the use of mixed microbial consortia to pure strains of biocatalyst, and the use of waste water in contrast to simple substrates such as; acetate, glucose, and lactate. The use of algae as a substrate, and as a biocatalyst, is also investigated. In this study, only single and dual chamber MFCs are compared, and power density standardised to anode surface area (mW/m2) is used as a metric to facilitate the comparison of different experimental setups. This meta-study shows that dual chamber MFCs, using simple substrates, when catalysed by mixed culture biocatalysts, produce greater power densities, than algae, and complex substrates, with average power densities of 280, 70 and 30 (mW/m2) observed respectively. In single chamber MFC configurations, mixed culture biocatalysts have been observed to yield approximately double the power output of pure culture biocatalysts

    Voltage-gated potassium channels (Kv) are important regulators of membrane potential in vascular smooth muscle cells, which is integral to controlling intracellular Ca2+ concentration and regulating vascular tone. Previous work indicates that Kv channels can be modulated by receptor-driven alterations of cyclic AMP-dependent protein kinase (PKA) activity. Here, we demonstrate that Kv channel activity is maintained by tonic activity of PKA. Whole-cell recording was used to assess the effect of manipulating PKA signalling on Kv and ATP-dependent K+ channels of rat mesenteric artery smooth muscle cells. Application of PKA inhibitors, KT5720 or H89, caused a significant inhibition of Kv currents. Tonic PKA-mediated activation of Kv appears maximal as application of isoprenaline (a β-adrenoceptor agonist) or dibutyryl-cAMP failed to enhance Kv currents. We also show that this modulation of Kv by PKA can be reversed by protein phosphatase 2B/calcineurin (PP2B). PKA-dependent inhibition of Kv by KT5720 can be abrogated by pre-treatment with the PP2B inhibitor cyclosporin A, or inclusion of a PP2B auto-inhibitory peptide in the pipette solution. Finally, we demonstrate that tonic PKA-mediated modulation of Kv requires intact caveolae. Pre-treatment of the cells with methyl-β-cyclodextrin to deplete cellular cholesterol, or adding caveolin-scaffolding domain peptide to the pipette solution to disrupt caveolae-dependent signalling each attenuated PKA-mediated modulation of the Kv current. These findings highlight a novel, caveolae-dependent, tonic modulatory role of PKA on Kv channels providing new insight into mechanisms and the potential for pharmacological manipulation of vascular tone

    Hip fracture evaluation with alternatives of total hip arthroplasty versus hemiarthroplasty (HEALTH): protocol for a multicentre randomised trial

    Introduction: Hip fractures are a leading cause of mortality and disability worldwide, and the number of hip fractures is expected to rise to over 6 million per year by 2050. The optimal approach for the surgical management of displaced femoral neck fractures remains unknown. Current evidence suggests the use of arthroplasty; however, there is lack of evidence regarding whether patients with displaced femoral neck fractures experience better outcomes with total hip arthroplasty (THA) or hemiarthroplasty (HA). The HEALTH trial compares outcomes following THA versus HA in patients 50 years of age or older with displaced femoral neck fractures. Methods and analysis: HEALTH is a multicentre, randomised controlled trial where 1434 patients, 50 years of age or older, with displaced femoral neck fractures from international sites are randomised to receive either THA or HA. Exclusion criteria include associated major injuries of the lower extremity, hip infection(s) and a history of frank dementia. The primary outcome is unplanned secondary procedures and the secondary outcomes include functional outcomes, patient quality of life, mortality and hiprelated complications—both within 2 years of the initial surgery. We are using minimisation to ensure balance between intervention groups for the following factors: age, prefracture living, prefracture functional status, American Society for Anesthesiologists (ASA) Class and centre number. Data analysts and the HEALTH Steering Committee are blinded to the surgical allocation throughout the trial. Outcome analysis will be performed using a χ2 test (or Fisher’s exact test) and Cox proportional hazards modelling estimate. All results will be presented with 95% CIs. Ethics and dissemination: The HEALTH trial has received local and McMaster University Research Ethics Board (REB) approval (REB#: 06-151). Results: Outcomes from the primary manuscript will be disseminated through publications in academic journals and presentations at relevant orthopaedic conferences. We will communicate trial results to all participating sites. Participating sites will communicate results with patients who have indicated an interest in knowing the results.
