Withdrawal to the shadows: dark social media as opportunity structures for extremism
Dark social media has been described as a home base for extremists and a breeding ground for dark participation. Beyond the description of single cases, it often remains unclear what exactly is meant by dark social media and which opportunity structures for extremism emerge on these applications. The current paper contributes to filling this gap. We present a theoretical framework conceptualizing dark social media as opportunity structures shaped by (a) regulation on the macro-level; (b) different genres and types of (dark) social media as influence factors on the meso level; and (c) individual attitudes, salient norms, and technological affordances on the micro-level. The results of a platform analysis and a scoping review identified meaningful differences between dark social media of different types. Particularly social counter-media and fringe communities positioned themselves as "safe havens" for dark participation, indicating a high tolerance for accordant content. This makes them a fertile ground for those spreading extremist worldviews, consuming such content, or engaging in dark participation. Context-bound alternative social media were comparable to mainstream social media but oriented towards different legal spaces and were more intertwined with governments in China and Russia. Private-first channels such as Instant messengers were rooted in private communication. Yet, particularly Telegram also included far-reaching public communication formats and optimal opportunities for the convergence of mass, group, and interpersonal communication. Overall, we show that a closer examination of different types and genres of social media provides a more nuanced understanding of shifting opportunity structures for extremism in the digital realm
Occupant requirements in residential buildings â an empirical study and a theoretical model
Occupant needs with regard to residential buildings are not well known due to a lack of representative scientific studies. To improve the lack of data, a large scale study was carried out using a Post Occupancy Evaluation of 1,416 building occupants. Several criteria describing the needs of occupants were evaluated with regard to their subjective level of relevance. Additionally, we investigated the degree to which deficiencies subjectively exist, and the degree to which occupants were able to accept them. From the data obtained, a hierarchy of criteria was created. It was found that building occupants ranked the physiological needs of air quality and thermal comfort the highest. Health hazards such as mould and contaminated building materials were unacceptable for occupants, while other deficiencies were more likely to be tolerated. Occupant satisfaction was also investigated. We found that most occupants can be classified as satisfied, although some differences do exist between different populations. To explain the relationship between the constructs of what we call relevance, acceptance, deficiency and satisfaction, we then created an explanatory model. Using correlation and regression analysis, the validity of the model was then confirmed by applying the collected data. The results of the study are both relevant in shaping further research and in providing guidance on how to maximize tenant satisfaction in real estate management

Extremist:innen greifen zunehmend auf dunkle sozialen Medien zurĂŒck. Der Begriff der dunklen sozialen Medien umfasst verschiedene Typen alternativer Sozialer Medien (soziale Kontermedien wie Gab, kontextgebundene alternative Soziale Medien wie VKontakte, Fringe Communities wie 4Chan), ebenso wie verschiedene Typen dunkler KanĂ€le (ursprĂŒnglich private KanĂ€le wie Telegram und SeparĂ©e-KanĂ€le wie geschloĂene Facebook-Gruppen). Das vorliegende Gutachten beleuchtet die Gelegenheitsstrukturen fĂŒr Extremismus und ExtremismusprĂ€vention, die sich durch die Verlagerung hin zu dunklen Sozialen Medien ergeben. HierfĂŒr werden in einem theoretischen Rahmenmodel Einflussfaktoren auf drei Ebenen verknĂŒpft: (1) Regulierung (etwa durch das NetzDG) auf der gesellschaftlichen Makro-Ebene. (2) Verschiedene Genres und Typen (dunkler) sozialer Medien auf der Meso-Ebene einzelner Angebote. (3) Einstellungen, Normen und technische Affordanzen als Motivatoren menschlichen Verhaltens im Sinne der Theorie des geplanten Verhaltens (Ajzen und Fishbein, 1977) auf der Mikro-Ebene. Basierend auf diesem Rahmenmodel werden die Gelegenheitsstrukturen fĂŒr Extremismus und ExtremismusprĂ€vention mit Hilfe zweier Studien untersucht: (1) Einer detaillierten Plattformanalyse dunkler und etablierter Sozialer Medien (N = 19 Plattformen). (2) Eine Literaturanalyse (> âscoping reviewâ) des Forschungsstandes zu (dunklen) Sozialen Medien im Kontext von Extremismus und ExtremismusprĂ€vention (N = 142 Texte). Die Ergebnisse der Platformanalyse ermöglichen nuancierte Einblicke in die Gelegenheitsstrukturen, die sich durch unterschiedliche Typen und Genres (dunkler) Sozialer Medien ergeben. Das Scoping Review bietet einen Ăberblick ĂŒber die Entwicklung des Forschungsfeldes und die typischen Untersuchungsmethoden, die eingesetzt werden. Auf der Grundlage der erhobenen Daten werden Forschungsdesiderata und Implikationen fĂŒr die ExtremismusprĂ€vention diskutiert

Effect of Experimental Thyrotoxicosis onto Blood Coagulation: A Proteomics Study
Background: Hyperthyroidism is known to induce a hypercoagulable state. It stimulates plasma levels of procoagulative factors and reduces fibrinolytic activity. So far most of the data have been derived from patients with endogenous hyperthyroidism with a wide variability in the underlying pathogenesis and severity of the disease. Objectives: In this study we experimentally induced thyrotoxicosis in healthy volunteers to explore the effects of thyroxine excess on the plasma proteome. Using a shotgun proteomics approach, the abundance of plasma proteins was monitored before, during and after thyrotoxicosis. Methods: Sixteen healthy male subjects were sampled at baseline, 4 and 8 weeks under 250 ”g/day thyroxine p.o., as well as 4 and 8 weeks after stopping the application. Plasma proteins were analyzed after depletion of 6 high-abundance proteins (MARS6) by LC-ESI-MS/MS mass spectrometry. Mass spectrometric raw data were processed using a label-free, intensity-based workflow. Subsequently, the linear dependence between protein abundances and fT4 levels were calculated using a Pearson correlation. Results: All subjects developed biochemical thyrotoxicosis, and this effect was reversed within the first 4 weeks of follow-up. None of the volunteers noticed any subjective symptoms. Levels of 10 proteins involved in the coagulation cascade specifically correlated with fT4, supporting an influence of thyroid hormone levels on blood coagulation even at nonpathological levels. Conclusions: The results suggest that experimental thyrotoxicosis exerts selective and specific thyroxine-induced effects on coagulation markers. Our study design allows assessment of thyroid hormone effects on plasma protein levels without secondary effects of other diseases or therapies
Extensive alterations of the whole-blood transcriptome are associated with body mass index: results of an mRNA profiling study involving two large population-based cohorts
Background: Obesity, defined as pathologically increased body mass index (BMI),is strongly related to an increased risk for numerous common cardiovascular and metabolic diseases. It is particularly associated with insulin resistance, hyperglycemia, and systemic oxidative stress and represents the most important risk factor for type 2 diabetes (T2D). However, the pathophysiological mechanisms underlying these associations are still not completely understood. Therefore, in order to identify potentially disease-relevant BMI-associated gene expression signatures, a transcriptome-wide association study (TWAS) on BMI was performed. Methods: Whole-blood mRNA levels determined by array-based transcriptional profiling were correlated with BMI in two large independent population-based cohort studies (KORA F4 and SHIP-TREND) comprising a total of 1977 individuals. Results: Extensive alterations of the whole-blood transcriptome were associated with BMI: More than 3500 transcripts exhibited significant positive or negative BMI-correlation. Three major whole-blood gene expression signatures associated with increased BMI were identified. The three signatures suggested: i) a ratio shift from mature erythrocytes towards reticulocytes, ii) decreased expression of several genes essentially involved in the transmission and amplification of the insulin signal, and iii) reduced expression of several key genes involved in the defence against reactive oxygen species (ROS). Conclusions: Whereas the first signature confirms published results, the other two provide possible mechanistic explanations for well-known epidemiological findings under conditions of increased BMI, namely attenuated insulin signaling and increased oxidative stress. The putatively causative BMI-dependent down-regulation of the expression of numerous genes on the mRNA level represents a novel finding. BMI-associated negative transcriptional regulation of insulin signaling and oxidative stress management provide new insights into the pathogenesis of metabolic syndrome and T2D
Genome-Wide Interaction Analyses of Serum Calcium on Ventricular Repolarization Time in 125 393 Participants
Background: Ventricular repolarization time (ECG QT and JT intervals) is associated with malignant arrhythmia. Genome-wide association studies have identified 230 independent loci for QT and JT; however, 50% of their heritability remains unexplained. Previous work supports a causal effect of lower serum calcium concentrations on longer ventricular repolarization time. We hypothesized calcium interactions with QT and JT variant associations could explain a proportion of the missing heritability. Methods and Results: We performed genome-wide calcium interaction analyses for QT and JT intervals. Participants were stratified by their calcium level relative to the study distribution (top or bottom 20%). We performed a 2-stage analysis (genome-wide discovery [N=62 532] and replication [N=59 861] of lead variants) and a single-stage genome-wide meta-analysis (N=122 393, [European ancestry N=117 581, African ancestry N=4812]). We also calculated 2-degrees of freedom joint main and interaction and 1-degree of freedom interaction P values. In 2-stage and single-stage analyses, 50 and 98 independent loci, respectively, were associated with either QT or JT intervals (2-degrees of freedom joint main and interaction P value <5x10(-8)). No lead variant had a significant interaction result after correcting for multiple testing and sensitivity analyses provided similar findings. Two loci in the single-stage meta-analysis were not reported previously (SPPL2B and RFX6). Conclusions: We have found limited support for an interaction effect of serum calcium on QT and JT variant associations despite sample sizes with suitable power to detect relevant effects. Therefore, such effects are unlikely to explain a meaningful proportion of the heritability of QT and JT, and factors including rare variation and other environmental interactions need to be considered
Genome-wide association analyses identify 143 risk variants and putative regulatory mechanisms for type 2 diabetes
Type 2 diabetes (T2D) is a very common disease in humans. Here we conduct a meta-analysis of genome-wide association studies (GWAS) with ~16 million genetic variants in 62,892 T2D cases and 596,424 controls of European ancestry. We identify 139 common and 4 rare variants associated with T2D, 42 of which (39 common and 3 rare variants) are independent of the known variants. Integration of the gene expression data from blood (n = 14,115 and 2765) with the GWAS results identifies 33 putative functional genes for T2D, 3 of which were targeted by approved drugs. A further integration of DNA methylation (n = 1980) and epigenomic annotation data highlight 3 genes (CAMK1D, TP53INP1, and ATP5G1) with plausible regulatory mechanisms, whereby a genetic variant exerts an effect on T2D through epigenetic regulation of gene expression. Our study uncovers additional loci, proposes putative genetic regulatory mechanisms for T2D, and provides evidence of purifying selection for T2D-associated variants
Exome-chip meta-analysis identifies novel loci associated with cardiac conduction, including ADAMTS6.
BACKGROUND: Genome-wide association studies conducted on QRS duration, an electrocardiographic measurement associated with heart failure and sudden cardiac death, have led to novel biological insights into cardiac function. However, the variants identified fall predominantly in non-coding regions and their underlying mechanisms remain unclear. RESULTS: Here, we identify putative functional coding variation associated with changes in the QRS interval duration by combining Illumina HumanExome BeadChip genotype data from 77,898 participants of European ancestry and 7695 of African descent in our discovery cohort, followed by replication in 111,874Â individuals of European ancestry from the UK Biobank and deCODE cohorts. We identify ten novel loci, seven within coding regions, including ADAMTS6, significantly associated with QRS duration in gene-based analyses. ADAMTS6 encodes a secreted metalloprotease of currently unknown function. In vitro validation analysis shows that the QRS-associated variants lead to impaired ADAMTS6 secretion and loss-of function analysis in mice demonstrates a previously unappreciated role for ADAMTS6 in connexin 43 gap junction expression, which is essential for myocardial conduction. CONCLUSIONS: Our approach identifies novel coding and non-coding variants underlying ventricular depolarization and provides a possible mechanism for the ADAMTS6-associated conduction changes.BH
