    The Geology of the Davis Ranch Oil Pool, Wabaunsee County, Kansas

    The discovery of the Davis Ranch oil pool in Wabaunsee County, Kansas, in April 1949 started one of the most important leasing and wildcat drilling programs in Kansas in recent years. In the 21 months since the completion of the discovery well, 65 holes have been drilled in Wabaunsee County alone. Of this total 38 may be classed as wildcat or semiwildcat wells, three of which have opened additional new pools. In view of the increased activity in Wabaunsee County and the Kansas part of the Forest City basin, a report showing our interpretations of the stratigraphy and structural relations of the Davis Ranch pool has been considered appropriate. All interpretations must be considered tentative until further information is received by the Survey. On February 1, 1951, the Davis Ranch pool had 18 producing oil wells. Six dry holes have been drilled and one oil well, the Aladdin and Bakke No. 1 Schutter in the SW SW SW sec. 34, T. 13 S., R. 10 E. was abandoned in December 1950. This well was the only producing well in the Davis Ranch East pool. The structure shown by the surface rocks is a north-northeast trending anticline with a closure of more than 20 feet. The Davis Ranch anticline is located on the northeast-trending subsurface Alma anticline which extends northward into Pottawatomie County and southward into Lyon County. The pre-Pennsylvanian structure is marked by a reverse fault which is not noticeable in the surface structure. Seventeen wells produce from the Viola dolomite and one well from the "Hunton" dolomite in the Davis Ranch pool. The thickness of the oil-saturated porous zone in the Viola ranges from a featheredge to 20 feet. Initial productivity tests indicated little or no water present in the producing zone

    Black hole head-on collisions and gravitational waves with fixed mesh-refinement and dynamic singularity excision

    We present long-term-stable and convergent evolutions of head-on black hole collisions and extraction of gravitational waves generated during the merger and subsequent ring-down. The new ingredients in this work are the use of fixed mesh-refinement and dynamical singularity excision techniques. We are able to carry out head-on collisions with large initial separations and demonstrate that our excision infrastructure is capable of accommodating the motion of the individual black holes across the computational domain as well as their their merger. We extract gravitational waves from these simulations using the Zerilli-Moncrief formalism and find the ring-down radiation to be, as expected, dominated by the l=2, m=0 quasi-normal mode.     Meta-analysis of genome-wide association studies from the CHARGE consortium identifies common variants associated with carotid intima media thickness and plaque

    Carotid intima media thickness (cIMT) and plaque determined by ultrasonography are established measures of subclinical atherosclerosis that each predicts future cardiovascular disease events. We conducted a meta-analysis of genome-wide association data in 31,211 participants of European ancestry from nine large studies in the setting of the Cohorts for Heart and Aging Research in Genomic Epidemiology (CHARGE) Consortium. We then sought additional evidence to support our findings among 11,273 individuals using data from seven additional studies. In the combined meta-analysis, we identified three genomic regions associated with common carotid intima media thickness and two different regions associated with the presence of carotid plaque (P < 5 × 10 -8). The associated SNPs mapped in or near genes related to cellular signaling, lipid metabolism and blood pressure homeostasis, and two of the regions were associated with coronary artery disease (P < 0.006) in the Coronary Artery Disease Genome-Wide Replication and Meta-Analysis (CARDIoGRAM) consortium. Our findings may provide new insight into pathways leading to subclinical atherosclerosis and subsequent cardiovascular events

    Endomembrane targeting of human OAS1 p46 augments antiviral activity

    Many host RNA sensors are positioned in the cytosol to detect viral RNA during infection. However, most positive-strand RNA viruses replicate within a modified organelle co-opted from intracellular membranes of the endomembrane system, which shields viral products from cellular innate immune sensors. Targeting innate RNA sensors to the endomembrane system may enhance their ability to sense RNA generated by viruses that use these compartments for replication. Here, we reveal that an isoform of oligoadenylate synthetase 1, OAS1 p46, is prenylated and targeted to the endomembrane system. Membrane localization of OAS1 p46 confers enhanced access to viral replication sites and results in increased antiviral activity against a subset of RNA viruses including flaviviruses, picornaviruses, and SARS-CoV-2. Finally, our human genetic analysis shows that the OAS1 splice-site SNP responsible for production of the OAS1 p46 isoform correlates with protection from severe COVID-19. This study highlights the importance of endomembrane targeting for the antiviral specificity of OAS1 and suggests that early control of SARS-CoV-2 replication through OAS1 p46 is an important determinant of COVID-19 severity

    A Functional Polymorphism of Toll-Like Receptor 4 Gene Increases Risk of Gastric Carcinoma and Its Precursors

    BACKGROUND AND AIMS: TLR4 is a cell-surface signaling receptor involved in the recognition and host response to Helicobacter pylori. The TLR4+896A>G polymorphism linked with impaired reactivity to bacterial lipopolysaccharide may play a role in gastric carcinogenesis. METHODS: We assessed associations with premalignant gastric changes in 149 relatives of gastric cancer patients, including 45 with hypochlorhydria and gastric atrophy. We also genotyped 2 independent Caucasian population-based case-control studies of upper gastrointestinal tract cancer, initially in 312 noncardia gastric carcinoma cases and 419 controls and then in 184 noncardia gastric carcinomas, 123 cardia carcinomas, 159 esophageal cancers, and 211 frequency-matched controls. Odds ratios were computed from logistic models and adjusted for potential confounding factors. RESULTS: TLR4+896G carriers had an 11-fold (95% confidence interval [CI], 2.5-48) increased odds ratio (OR) for hypochlorhydria; the polymorphism was unassociated with gastric acid output in the absence of H pylori infection. Carriers also had significantly more severe gastric atrophy and inflammation. Seventeen percent of gastric carcinoma patients in the initial study and 15% of the noncardia gastric carcinoma patients in the replication study had 1 or 2 TLR4 variant alleles vs 8% of both control populations (combined OR = 2.3; 95% CI = 1.6-3.4). In contrast, prevalence of TLR4+896G was not significantly increased in esophageal squamous cell (2%, OR = 0.2) or adenocarcinoma (9%, OR = 1.4) or gastric cardia carcinoma (11%, OR = 1.4). CONCLUSIONS: Our data suggest that the TLR4+896A>G polymorphism is a risk factor for noncardia gastric carcinoma and its precursors. The findings underscore the role of the host innate immune response in outcome of H pylori infection
