    Hybrid Petri net model of a traffic intersection in an urban network

    Control in urban traffic networks constitutes an important and challenging research topic nowadays. In the literature, a lot of work can be found devoted to improving the performance of the traffic flow in such systems, by means of controlling the red-to-green switching times of traffic signals. Different techniques have been proposed and commercially implemented, ranging from heuristic methods to model-based optimization. However, given the complexity of the dynamics and the scale of urban traffic networks, there is still a lot of scope for improvement. In this work, a new hybrid model for the traffic behavior at an intersection is introduced. It captures important aspects of the flow dynamics in urban networks. It is shown how this model can be used in order to obtain control strategies that improve the flow of traffic at intersections, leading to the future possibility of controlling several connected intersections in a distributed way

    Arabidopsis thaliana: A model host plant to study plant-pathogen interaction using Chilean field isolates of Botrytis cinerea

Botrytis is a constant threat to crops because the fungus infects a wide range of host species, both native and cultivated. Furthermore, Botrytis persists on plant debris in and on the soil. Some of the most serious diseases caused by Botrytis include gray mold on vegetables and fruits, such as grapes and strawberries. Botrytis also causes secondary soft rot of fruits and vegetables during storage, transit and at the market. In many plant-pathogen interactions, resistance often is associated with the deposition of callose, accumulation of autofluorescent compounds, the synthesis and accumulation of salicylic acid as well as pathogenesis-related proteins. Arabidopsis thaliana has been used as a plant model to study plant-pathogen interaction. The genome of Arabidopsis has been completely sequenced and this plant serves as a good genetic and molecular model. In this study, we demonstrate that Chilean field isolates infect Arabidopsis thaliana and that Arabidopsis subsequently activates several defense response mechanisms associated with a hypersensitive response. Furthermore, we propose that Arabidopsis may be used as a model host species to analyze the diversity associated with infectivity among populations of Botrytis cinerea field isolates

In order to keep Europe as a world leader in the global maritime economy, an effective integrated/interoperable, sustainable maritime surveillance system and situational awareness are needed. A significant number of unlawful maritime activities, such as illegal immigration, drugs trafficking, smuggling, piracy and terrorism involve mainly small boats, because small boats are faster and more difficult to detect using conventional means. Hence, it is very important to find out the feasibility of using SAR Satellite images for small boat detection. Since 2008 the EC-JRC has carried out a number of SAR Small Boat detection experiments to assess the feasibility of using Spaceborne SAR for Small Boat detection. This report presents the results and conclusions of the Spaceborne SAR Small Boat detection campaign in inland sea waters and in coastal waters carried out by the EC-JRC in the South of Spain (Sancti Petri-La Barossa, El Rompido, Punta Umbria, Cadiz and Isla Cristina) and in Portugal (Sagres and Cascais) in December 2010. The results of the experiment show that under suitable conditions of sea state, wind speed and incidence angle it is possible to detect small boats in spaceborne SAR imagery in inland waters, Coastal waters and open sea. Further research is needed to study possible correlations between the empirical probability of detection of small boats in spaceborne SAR and the main parameters involved, such as sea state, wind speed, incidence angle, among others.JRC.G.4-Maritime affair

    A Rapid and Efficient Method for Purifying High Quality Total RNA from Peaches (Prunus persica) for Functional Genomics Analyses

Optimized protocols for RNA isolation are necessary to further advance studies in this model species such that functional genomics analyses may be performed. Here we present an optimized protocol to rapidly and efficiently purify high quality total RNA from peach fruits (Prunus persica). Isolating high-quality RNA from fruit tissue is often difficult due to large quantities of polysaccharides and polyphenolic compounds that accumulate in this tissue and co-purify with the RNA. Here we demonstrate that a modified version of the method used to isolate RNA from pine trees and the woody plant Cinnamomun tenuipilum is ideal for isolating high quality RNA from the fruits of Prunus persica. This RNA may be used for many functional genomic based experiments such as RT-PCR and the construction of large-insert cDNA libraries

    Larval food quantity affects development time, survival and adult biological traits that influence the vectorial capacity of Anopheles darlingi under laboratory conditions

    BACKGROUND: The incidence of malaria in the Amazon is seasonal and mosquito vectorial capacity parameters, including abundance and longevity, depend on quantitative and qualitative aspects of the larval diet. Anopheles darlingi is a major malaria vector in the Amazon, representing >95% of total Anopheles population present in the Porto Velho region. Despite its importance in the transmission of the Plasmodium parasite, knowledge of the larval biology and ecology is limited. Studies regarding aspects of adult population ecology are more common than studies on larval ecology. However, in order develop effective control strategies and laboratory breeding conditions for this species, more data on the factors affecting vector biology is needed. The aim of the present study is to assess the effects of larval food quantity on the vectorial capacity of An. darling under laboratory conditions. METHODS: Anopheles darlingi was maintained at 28°C, 80% humidity and exposed to a daily photoperiod of 12 h. Larvae were divided into three experimental groups that were fed either a low, medium, or high food supply (based on the food amounts consumed by other species of culicids). Each experiment was replicated for six times. A cohort of adults were also exposed to each type of diet and assessed for several biological characteristics (e.g. longevity, bite frequency and survivorship), which were used to estimate the vectorial capacity of each experimental group. RESULTS: The group supplied with higher food amounts observed a reduction in development time while larval survival increased. In addition to enhanced longevity, increasing larval food quantity was positively correlated with increasing frequency of bites, longer blood meal duration and wing length, resulting in greater vectorial capacity. However, females had greater longevity than males despite having smaller wings. CONCLUSIONS: Overall, several larval and adult biological traits were significantly affected by larval food availability. Greater larval food supply led to enhance larval and production and larger mosquitoes with longer longevity and higher biting frequency.     RNA-seq analysis of salt-stressed versus non salt-stressed transcriptomes of chenopodium quinoa landrace R49

    Quinoa (Chenopodium quinoa Willd.), a model halophytic crop species, was used to shed light on salt tolerance mechanisms at the transcriptomic level. An RNA-sequencing analysis of genotype R49 at an early vegetative stage was performed by Illumina paired-ends method comparing high salinity and control conditions in a time-course pot experiment. Genome-wide transcriptional salt-induced changes and expression profiling of relevant salt-responsive genes in plants treated or not with 300 mM NaCl were analyzed after 1 h and 5 days. We obtained up to 49 million pairs of short reads with an average length of 101 bp, identifying a total of 2416 differentially expressed genes (DEGs) based on the treatment and time of sampling. In salt-treated vs. control plants, the total number of up-regulated and down-regulated genes was 945 and 1471, respectively. The number of DEGs was higher at 5 days than at 1 h after salt treatment, as reflected in the number of transcription factors, which increased with time. We report a strong transcriptional reprogramming of genes involved in biological processes like oxidation-reduction, response to stress and response to abscisic acid (ABA), and cell wall organization. Transcript analyses by real-time RT-qPCR supported the RNA-seq results and shed light on the contribution of roots and shoots to the overall transcriptional response. In addition, it revealed a time-dependent response in the expression of the analyzed DEGs, including a quick (within 1 h) response for some genes, suggesting a \u201cstress-anticipatory preparedness\u201d in this highly salt-tolerant genotype
