Pretende-se ainda apresentar um projeto de implementação do Balcão Único de Atendimento (BUA) na Câmara Municipal de Moimenta da Beira, no sentido de adequar os seus serviços ao atual paradigma competitivo, que exige da Administração Pública uma postura moderna e dinamizadora que incremente a melhoria constante dos seus serviços, de forma a responder às exigências de eficiência, eficácia, transparência e qualidade dos seus clientes

    As ilhas, os arquivos e a história : o caso dos Açores

    Nos Açores, a força da geografia define o carácter da história, que evidencia expressões bem diferenciadas. Por um lado, as ilhas agem como meio de aproximação dos continentes, equivalendo a um sinónimo de universalidade, que resulta de um privilegiado posicionamento no Atlântico Norte, movido pelo determinismo do mar e pelas condições da navegação à vela. Por outro lado, as ilhas figuram como factor de cristalização de comportamentos, correspondendo a um sinónimo de isolamento, motivado pelo afastamento do mundo e pela descontinuidade territorial interna. Nestas circunstâncias, os planos insulares de pesquisa histórica primam naturalmente pela pluralidade dos objectivos. Assim, demonstram uma participação muito activa na construção do mundo atlântico, que obriga à integração das investigações açorianas nas categorias mais universais do saber, mas aconselham também à realização de estudos de incidência local, conducentes à individualização de idiossincracias, que derivam da divisão do arquipélago em nove parcelas muito desiguais. Em 1979, por altura da publicação do seu livro O Arquipélago dos Açores no Século XVII: aspectos sócio-económicos (1575-1675), Maria Olímpia da Rocha Gil reconhece precisamente a indispensabilidade do desenvolvimento da investigação histórica açoriana de acordo com duas linhas ao mesmo tempo dissemelhantes e convergentes: “... em primeiro, aquela que nos leva a considerar que a história do arquipélago se integra no longo processo da história do Atlântico; em segundo lugar, a que se orienta para o estudo da evolução histórica local tendo em conta as características que lhe são próprias”. No entanto, desde tempos quase imemoriais, diversos cronistas e historiadores evidenciam um entendimento muito semelhante, que certifica a complexidade dos estudos históricos insulares. A comprová-lo, atentemos nas Saudades da Terra do Doutor Gaspar Frutuoso, redigidas logo no termo do século XVI, que relevam de uma assentada as especificidades locais, as correlações com os demais arquipélagos da Macaronésia e o envolvimento nas dinâmicas do Atlântico.     T-Cell Memory Responses Elicited by Yellow Fever Vaccine are Targeted to Overlapping Epitopes Containing Multiple HLA-I and -II Binding Motifs

    The yellow fever vaccines (YF-17D-204 and 17DD) are considered to be among the safest vaccines and the presence of neutralizing antibodies is correlated with protection, although other immune effector mechanisms are known to be involved. T-cell responses are known to play an important role modulating antibody production and the killing of infected cells. However, little is known about the repertoire of T-cell responses elicited by the YF-17DD vaccine in humans. In this report, a library of 653 partially overlapping 15-mer peptides covering the envelope (Env) and nonstructural (NS) proteins 1 to 5 of the vaccine was utilized to perform a comprehensive analysis of the virus-specific CD4+ and CD8+ T-cell responses. The T-cell responses were screened ex-vivo by IFN-γ ELISPOT assays using blood samples from 220 YF-17DD vaccinees collected two months to four years after immunization. Each peptide was tested in 75 to 208 separate individuals of the cohort. The screening identified sixteen immunodominant antigens that elicited activation of circulating memory T-cells in 10% to 33% of the individuals. Biochemical in-vitro binding assays and immunogenetic and immunogenicity studies indicated that each of the sixteen immunogenic 15-mer peptides contained two or more partially overlapping epitopes that could bind with high affinity to molecules of different HLAs. The prevalence of the immunogenicity of a peptide in the cohort was correlated with the diversity of HLA-II alleles that they could bind. These findings suggest that overlapping of HLA binding motifs within a peptide enhances its T-cell immunogenicity and the prevalence of the response in the population. In summary, the results suggests that in addition to factors of the innate immunity, "promiscuous" T-cell antigens might contribute to the high efficacy of the yellow fever vaccines. © 2013 de Melo et al

    Synthesis and Structural Studies of Er3+ Containing Lead Cadmium Fluoroborate Glasses and Glass-Ceramics

    A partir de resultados de experimentos de fusão e choque térmico, o domínio vítreo foi identificado no sistema PbF2-CdF2-B2O 3. Vidros puros e dopados com Er3+ foram preparados e vitrocerâmicas foram obtidas a partir de tratamentos térmicos adequados. A fase cristalina foi identificada como beta-PbF2. Estudos estruturais foram efetuados por difração e absorção de raios X e o papel desempenhado pelos átomos de Pb2+ e Cd2+ na estrutura vítrea foi estabelecido a partir destas técnicas. Os resultados sugerem que, com a cristalização, os íons Er3+ são segregados na fase cristalina.The vitreous domain was established in the PbF2-CdF2-B2O 3 system from melting and quenching experiments. Er3+ containing glasses were prepared and glass ceramics were obtained by selected heat-treatments. Lead fluoride was identified (beta-PbF2) as the crystalline phase. Structural studies were performed in some glassy and partially crystallized samples by means of X-ray Diffraction (XRD) and Extended X-ray Absorption Fine Structure (EXAFS) measurements. The role of Cd2+ and Pb2+ atoms on the glass network formation and also on the crystallization behavior was put forward by these techniques.     Local order around rare earth ions during the devitrification of oxyfluoride glasses.

    International audienceErbium L(3)-edge extended x-ray absorption fine structure (EXAFS) measurements were performed on rare earth doped fluorosilicate and fluoroborate glasses and glass ceramics. The well known nucleating effects of erbium ions for the crystallization of cubic lead fluoride (based on x-ray diffraction measurements) and the fact that the rare earth ions are present in the crystalline phase (as indicated by Er(3+) emission spectra) seem in contradiction with the present EXAFS analysis, which indicates a lack of medium range structural ordering around the Er(3+) ions and suggests that the lead fluoride crystallization does not occur in the nearest neighbor distance of the rare earth ion. Molecular dynamics simulations of the devitrification process of a lead fluoride glass doped with Er(3+) ions were performed, and results indicate that Er(3+) ions lower the devitrification temperature of PbF(2), in good agreement with the experimental results. The genuine role of Er(3+) ions in the devitrification process of PbF(2) has been investigated. Although Er(3+) ions could indeed act as seeds for crystallization, as experiments suggest, molecular dynamics simulation results corroborate the experimental EXAFS observation that the devitrification does not occur at its nearest neighbor distance

    Immunosensor for the Diagnostics of Autoimmune Hemolytic Anemia (AIHA) Based on Immobilization of a Monoclonal Antibody on a Layer of Silk Fibroin

    European Regional Development Fund (ERDF)The diagnostics of the autoimmune hemolytic anemia (AIHA), a rare disease caused by autoantibody-induced hemolysis, is still prone to false positives for it is based on visual observation in the so-called Direct Coombs test. In this study, we developed a specific IgG hemolysis immunosensor produced with layer-by-layer (LbL) films containing a monoclonal antibody against human immunoglobulin (mAbIMUG) deposited along with a layer of silk fibroin (SF) derived from Bombyx mori cocoons. Adsorption of mAbIMUG on a SF layer was confirmed by the fluorescence emission band at 326 nm. Immunosensors were prepared with LbL films deposited on interdigitated gold electrodes for impedance spectroscopy and on screen printed carbon electrodes for electrochemical measurements. When the SF/mAbIMUGLbL film was exposed to healthy red blood cells (RBCs), no cell binding was observed by the optical microscopy images. In addition, no major changes were observed in the signals of the square wave voltammogram and in the impedance spectra. In contrast, the electrochemical signal was significantly increased and the dielectric loss curve shifted for the sensing units containing RBCs with the antibody attached on the surface (“sick cells”). Furthermore, cell attachment was so strong that optical images still showed covered electrodes even after washing in PBS buffer. The detection with two distinct methods seems promising for an effective diagnosis of AIHA

    Biopolymer-based membranes associated with osteogenic growth peptide for guided bone regeneration

    Barrier membranes for guided bone regeneration (GBR) mainly promote mechanical maintenance of bone defect space and induce osteopromotion. Additionally, biopolymer-based membranes may provide greater bioactivity and biocompatibility due to their similarity to extracellular matrix (ECM).In this study, biopolymers-based membranes from bacterial cellulose (BC) and collagen (COL) associated with osteogenic growth peptide (OGP(10–14)) were evaluated to determine in vitro osteoinductive potential in early osteogenesis; moreover, histological study was performed to evaluate the BC–COL OGP(10–14) membranes on bone healing after GBR in noncritical defects in rat femur. The results showed that the BC–COL and BC–COL OGP(10–14) membranes promoted cell proliferation and alkaline phosphatase activity in osteoblastic cell cultures. However, ECMmineralization was similar between cultures grown on BC OGP(10–14) and BC–COL OGP(10–14) membranes. In vivo results showed that all the membranes tested, including the peptide-free BC membrane, promoted better bone regeneration than control group. Furthermore, the BC–COL OGP(10–14) membranes induced higher radiographic density in the repaired bone than the other groups at 1, 4 and 16 weeks. Histomorpho-metric analyses revealed that the BC–COL OGP(10–14) induced higher percentage of bone tissue in the repaired area at 2 and 4 weeks than others membranes. In general, these biopolymer-based membranes might be potential candidates for bone regeneration applications
