    C-Pen Exam Reader and Reading in Basic Education : Will it be a good partnership?

    There is an increasing number of students in basic education schools with difficulties in reading, and consequently, in writing. Our exploratory study aims to understand if the C Pen Exam Reader (C-Pen) helps in the learning process of students with language and writing difficulties. The methodology chosen was interviews with elementary school teachers, no special education teachers, and a speech therapist, as well as the support of CRTICee of the Amato Lusitano School Grouping. Thus, it was possible to assess the added value for students, not only in terms of their autonomy, since it is a reading pen, but also their self-esteem and inclusion in the school environment. It was also possible to clarify which points to improve in the use of this instrument to maximize its effectiveness and, thus, improve the quality of life of these students, such as prosody (accentuation, intonation, duration, rhythm of words and phrases).info:eu-repo/semantics/publishedVersio

    Insights on Ultrafiltration-Based Separation for the Purification and Quantification of Methotrexate in Nanocarriers

    The evaluation of encapsulation efficiency is a regulatory requirement for the characterization of drug delivery systems. However, the difficulties in efficiently separating nanomedicines from the free drug may compromise the achievement of accurate determinations. Herein, ultrafiltration was exploited as a separative strategy towards the evaluation of methotrexate (MTX) encapsulation efficiency in nanostructured lipid carriers and polymeric nanoparticles. The effect of experimental conditions such as pH and the amount of surfactant present in the ultrafiltration media was addressed aiming at the selection of suitable conditions for the effective purification of nanocarriers. MTX-loaded nanoparticles were then submitted to ultrafiltration and the portions remaining in the upper compartment of the filtering device and in the ultrafiltrate were collected and analyzed by HPLC-UV using a reversed-phase (C18) monolithic column. A short centrifugation time (5 min) was suitable for establishing the amount of encapsulated MTX in nanostructured lipid carriers, based on the assumption that the free MTX concentration was the same in the upper compartment and in the ultrafiltrate. The defined conditions allowed the efficient separation of nanocarriers from the free drug, with recoveries of >85% even when nanoparticles were present in cell culture media and in pig skin surrogate from permeation assays.info:eu-repo/semantics/publishedVersio

    Differential mtDNA damage patterns in a transgenic mouse model of Machado-Joseph disease (MJD/SCA3)

    Mitochondrial dysfunction has been associated with late onset neurodegenerative disorders, among which is Machado-Joseph disease (MJD/SCA3). In a previous study, using a transgenic mouse model of MJD, we reported a decrease in mitochondrial DNA (mtDNA) copy number and an accumulation of the 3876-bp deletion with age and with phenotype development. We extended this study by analyzing the pattern of mtDNA depletion and the accumulation of the 3876-bp deletion in 12 older transgenic (TG) and 4 wild-type (wt) animals, and by investigating the accumulation of somatic mutations in the D-loop region in 76 mice (42 TG and 34 wt). mtDNA damage was studied in TG and wt mice at different ages and tissues (blood, pontine nuclei, and hippocampus). Results for older mice demonstrate an accumulation of the mtDNA 3867-bp deletion with age, which was more pronounced in TG animals. Furthermore, the tendency for mtDNA copy number decrease with age, in all analyzed tissues of TG and wt animals, was also confirmed. No point mutations were detected in the D-loop, neither in TG nor wt animals, in any of the tissues analyzed. Due to the absence of mtDNA somatic mutations, we can suggest that mtDNA point mutation accumulation cannot be used to monitor the development and progression of the phenotype in this mouse model and likely in any MJD mice model.     Features and performance of edible films, obtained from whey protein isolate formulated with antimicrobial compounds

    The goal of this research effort was to assess the efficacy of edible films produced fromwhey protein isolate (WPI) and glycerol, including incorporation of lactic acid (LA) and propionic acid (PRO), chitooligosaccharideswith nominal MW of 3 kDa (COS) and natamycin (NA) as antimicrobial agents. Their features were evaluated in vitro via agar diffusion and viable cell counting, against spoilage microflora often found contaminating cheese surfaces. The effect of incorporating the aforementioned compounds upon thickness, moisture content (MC), solubility (S), density (ρs),water activity (aw) and water vapor permeability (WVP), as well as upon tensile and optical properties of those films were also evaluated. Films formulated with LA, PRO or COS exhibited antimicrobial activity against all microorganisms tested, yet the viable cell count assay was more sensitive and reproducible. COS was the most active against Gram-negative bacteria, whereas LA was the most active against Gram-positive ones. NA was not active against bacteria, but displayed the strongest effect against yeasts. Incorporation of said antimicrobial compounds did not significantly (pN0.05) affect film thickness, yet it significantly (pb0.05) reduced tensile strength (TS). Incorporation of LA and NA in particular did not significantly (pb0.05) affect MC, S, ρs, WVP, elongation at break (EB) and Young's modulus (YM) values; however, a statistically significant increase (pb0.05) of MC, S andWVP, together with a statistically significant decrease (pb0.05) of ρs were attained upon incorporation of PRO or COS.     Developmental Regulation of Transcription in Touriga Nacional Berries under Deficit Irrigation

    Grapevine (Vitis vinifera L.) is one of the most economically important crops worldwide, especially due to the economic relevance of wine production. Abiotic stress, such as drought, may contribute to low yield, shifts in quality, and important economic loss. The predicted climate change phenomena point to warmer and dryer Mediterranean environmental conditions; as such, it is paramount to study the effects of abiotic stress on grapevine performance. Deficit irrigation systems are applied to optimize water use efficiency without compromising berry quality. In this research, the effect of two deficit irrigation strategies, sustained deficit irrigation (SDI) and regulated deficit irrigation (RDI), in the grape berry were assessed. The effects of different levels of drought were monitored in Touriga Nacional at key stages of berry development (pea size, véraison, and full maturation) through RNA-Seq transcriptome analysis and by specific differentially expressed genes (DEGs) monitoring through RT-qPCR. Handy datasets were obtained by bioinformatics analysis of raw RNA-Seq results. The dominant proportion of transcripts was mostly regulated by development, with véraison showing more upregulated transcripts. Results showed that primary metabolism is the functional category more severely affected under water stress. Almost all DEGs selected for RT-qPCR were significantly upregulated in full maturation and showed the highest variability at véraison and the lowest gene expression values in the pea size stageinfo:eu-repo/semantics/publishedVersio

    Effect of whey protein purity and glycerol content upon physical properties of edible films manufactured therefrom

    This manuscript describes the detailed characterization of edible films made from two different protein products e whey protein isolate (WPI) and whey protein concentrate (WPC), added with three levels of glycerol (Gly) e i.e. 40, 50 and 60%(w/w). The molecular structure, as well as barrier, tensile, thermal,surface and optical properties of said films were determined, in attempts to provide a better understanding of the effects of proteinaceous purity and Gly content of the feedstock. WPI films exhibited statistically lower (p < 0.05) moisture content (MC), film solubility (S), water activity, water vapor permeability (WVP), oxygen and carbon dioxide permeabilities (O2P and CO2P, respectively) and color change values, as well as statistically higher (p < 0.05) density, surface hydrophobicity, mechanical resistance, elasticity, extensibility and transparency values than their WPC counterparts, for the same content of Gly. These results are consistent with data from thermal and FTIR analyses. Furthermore,a significant increase (p < 0.05) was observed in MC, S, WVP, O2P, CO2P, weight loss and extensibility of both protein films when the Gly content increased; whereas a significant decrease (p < 0.05) was observed in thermal features, as well as in mechanical resistance and elasticity e thus leading to weaker films.     Relação entre a prática de exercícios físicos e saúde mental em estudantes

    A saúde mental refere-se a um bem estar no qual o indivíduo desenvolve suas habilidades pessoais, consegue lidar com os estresses da vida, trabalha de forma produtiva e encontra-se apto a dar sua contribuição para sua comunidade, sendo o exercício físico um possível aliado para manter esse estado de bem estar.&nbsp; Nessa perspectiva, a presente análise trata-se de uma mini revisão integrativa de literatura que teve como objetivo verificar a existência de relação entre a prática de exercícios físicos e a manutenção da saúde mental nos estudantes. &nbsp;Foram utilizados cinco artigos das bases de dados Scientific Eletronic Library Online (SciELO) e Google Acadêmico, encontrados por meio dos descritores “saúde mental”, “exercício físico” e “estudantes”. Constatou-se, através dos cinco artigos, que o exercício físico é, realmente, essencial para manter a saúde mental e evitar a instalação de doenças mentais e outras patologias psicológicas nos estudantes universitários. Dessa forma, observou-se que diversas modalidades de exercícios físicos reduzem o estresse, alivia a pressão diária, melhora o sono, habilidades de memória, pensamento e percepção, além de melhorar a função física desses jovens. Sendo assim, foi possível constatar que todos os autores evidenciaram que existe a relação entre a prática de exercícios físicos e a manutenção da saúde mental nos estudantes.&nbsp; Portanto, conclui-se que a prática desses exercícios deve ser considerada, uma vez que, há resultados significativos no desempenho e saúde mental dos estudantes universitários, no entanto, não deve ser descartado o uso de medicamentos, caso necessário, para que esses resultados sejam potencializados
