AbstractOxidative stress and endoplasmic reticulum (ER) stress have been associated with Alzheimer's disease (AD) progression. In this study we analyzed whether oxidative stress involving changes in Nrf2 and ER stress may constitute early events in AD pathogenesis by using human peripheral blood cells and an AD transgenic mouse model at different disease stages. Increased oxidative stress and increased phosphorylated Nrf2 (p(Ser40)Nrf2) were observed in human peripheral blood mononuclear cells (PBMCs) isolated from individuals with mild cognitive impairment (MCI). Moreover, we observed impaired ER Ca2+ homeostasis and increased ER stress markers in PBMCs from MCI individuals and mild AD patients. Evidence of early oxidative stress defense mechanisms in AD was substantiated by increased p(Ser40)Nrf2 in 3month-old 3xTg-AD male mice PBMCs, and also with increased nuclear Nrf2 levels in brain cortex. However, SOD1 protein levels were decreased in human MCI PBMCs and in 3xTg-AD mice brain cortex; the latter further correlated with reduced SOD1 mRNA levels. Increased ER stress was also detected in the brain cortex of young female and old male 3xTg-AD mice. We demonstrate oxidative stress and early Nrf2 activation in AD human and mouse models, which fails to regulate some of its targets, leading to repressed expression of antioxidant defenses (e.g., SOD-1), and extending to ER stress. Results suggest markers of prodromal AD linked to oxidative stress associated with Nrf2 activation and ER stress that may be followed in human peripheral blood mononuclear cells

    Get PDF
The aim of this study is to investigate (i) the effect of live and recorded verbal encouragement on muscle activity and ankle force; (ii) the effect of communication/extroversion on the variables; (iii) the reliability intra and inter examiners of the variables. Twenty healthy-youngers were assessed by surface electromyography of tibialis anterior and ankle flexion force by an ergometer twice, with one week apart. No difference was found between ankle force (p = 0.373) and root mean square values (RMS) (p = 0.207) for any of the conditions assessed on day 1 nor between examiners 1 and 2 for both live and recorded conditions in RMS (p = 0.207) and force (p = 0.373). Between the 1st and 7th days, there were no differences for any of the conditions on RMS (main effect “Day” p = 0.261, “condition” p = 0.568, interaction p = 0.936) or force (main effect“Day” p = 0.889, “condition” p = 0.781, interaction p = 0.961). Intraclass correlation coefficients (ICCs) for the ankle force were, for without verbal encouragement (ICC2, k = 0.880), live verbal encouragement of examiner 1 (ICC2, k = 0.870), and recorded verbal encouragement of examiner 1 (ICC2, k = 0.920). RMS without verbal encouragement condition (ICC2, k = 0.860), live verbal encouragement of examiner 1 (ICC2, k = 0.930) and recorded verbal encouragement of examiner 1 (ICC2, k = 0.920). Reproducibility between the two examiner’s live encouragements for ankle force (ICC3, k = 0.981) and RMS (ICC3, k = 0.920). There was no effect of the presence or type of the augmented feedback in RMS and ankle force. We conclude that verbal encouragement does not influence ankle torque or muscle activity and there is good to excellent intra and inter rater reliability for subjects’ performance regardless of verbal encouragement modality. In addition, we observed that psychological traits Communication and Emotional stability does not affect the subjects’ strength performance at the ankle

    European Red List of Trees

    Get PDF
The European Red List is a review of the status of European species according to IUCN regional Red Listing guidelines. It identifies those species that are threatened with extinction at the regional level – in order that appropriate conservation action can be taken to improve their status. This publication summarises results for all Europe’s native species of tree (454 species), of which 265 species (over 58%) are endemic to continental Europe, with 56% (252 species) endemic to the 28 EU Member States. Of these, 168 (42%) of the species are threatened with extinction at the European level, however, for 57 species (nearly 13%) there was insufficient information to assign a conservation status, and are therefore classified as Data Deficient, and in need of further research. The main threat to tree species in Europe has been identified as invasive or problematic species, impacting 38% of tree species, followed by deforestation and wood harvesting, and urban development (both affecting 20% of tree species). For threatened species, livestock farming, land abandonment, changes in forest and woodland management, and other ecosystem modifications such as fire are the major threats, impacting the survival of trees.Peer reviewe

Association of HFE common mutations with Parkinson's disease, Alzheimer's disease and mild cognitive impairment in a Portuguese cohort

    Get PDF
Background: Pathological brain iron deposition has been implicated as a source of neurotoxic reactive oxygen species in Alzheimer (AD) and Parkinson diseases (PD). Iron metabolism is associated with the gene hemochromatosis (HFE Human genome nomenclature committee ID: 4886), and mutations in HFE are a cause of the iron mismetabolism disease, hemochromatosis. Several reports have tested the association of HFE variants with neurodegenerative diseases, such as AD and PD with conflicting results. Methods: Genotypes were analysed for the two most common variants of HFE in a series of 130 AD, 55 Mild Cognitive Impairment (MCI) and 132 PD patients. Additionally, a series of 115 healthy age-matched controls was also screened. Results: A statistically significant association was found in the PD group when compared to controls, showing that the presence of the C282Y variant allele may confer higher risk for developing the disease. Conclusion: Taken together these results suggest that the common variants in HFE may be a risk factor for PD, but not for AD in the Portuguese population

White matter hyperintensities in progranulin-associated frontotemporal dementia: A longitudinal GENFI study

    Get PDF
Frontotemporal dementia (FTD) is a heterogeneous group of neurodegenerative disorders with both sporadic and genetic forms. Mutations in the progranulin gene (GRN) are a common cause of genetic FTD, causing either a behavioural presentation or, less commonly, language impairment. Presence on T2-weighted images of white matter hyperintensities (WMH) has been previously shown to be more commonly associated with GRN mutations rather than other forms of FTD. The aim of the current study was to investigate the longitudinal change in WMH and the associations of WMH burden with grey matter (GM) loss, markers of neurodegeneration and cognitive function in GRN mutation carriers. 336 participants in the Genetic FTD Initiative (GENFI) study were included in the analysis: 101 presymptomatic and 32 symptomatic GRN mutation carriers, as well as 203 mutation-negative controls. 39 presymptomatic and 12 symptomatic carriers, and 73 controls also had longitudinal data available. Participants underwent MR imaging acquisition including isotropic 1 mm T1-weighted and T2-weighted sequences. WMH were automatically segmented and locally subdivided to enable a more detailed representation of the pathology distribution. Log-transformed WMH volumes were investigated in terms of their global and regional associations with imaging measures (grey matter volumes), biomarker concentrations (plasma neurofilament light chain, NfL, and glial fibrillary acidic protein, GFAP), genetic status (TMEM106B risk genotype) and cognition (tests of executive function). Analyses revealed that WMH load was higher in both symptomatic and presymptomatic groups compared with controls and this load increased over time. In particular, lesions were seen periventricularly in frontal and occipital lobes, progressing to medial layers over time. However, there was variability in the WMH load across GRN mutation carriers – in the symptomatic group 25.0% had none/mild load, 37.5% had medium and 37.5% had a severe load – a diffe

Apathy in presymptomatic genetic frontotemporal dementia predicts cognitive decline and is driven by structural brain changes

    Get PDF
Introduction: Apathy adversely affects prognosis and survival of patients with frontotemporal dementia (FTD). We test whether apathy develops in presymptomatic genetic FTD, and is associated with cognitive decline and brain atrophy. Methods: Presymptomatic carriers of MAPT, GRN or C9orf72 mutations (N = 304), and relatives without mutations (N = 296) underwent clinical assessments and MRI at baseline, and annually for 2 years. Longitudinal changes in apathy, cognition, gray matter volumes, and their relationships were analyzed with latent growth curve modeling. Results: Apathy severity increased over time in presymptomatic carriers, but not in non-carriers. In presymptomatic carriers, baseline apathy predicted cognitive decline over two years, but not vice versa. Apathy progression was associated with baseline low gray matter volume in frontal and cingulate regions. Discussion: Apathy is an early marker of FTD-related changes and predicts a subsequent subclinical deterioration of cognition before dementia onset. Apathy may be a modifiable factor in those at risk of FTD
