    Optimization of in vitro multiplication for exotic banana (Musa spp.) in Pakistan

    The present attempt aimed to optimize micropropagation protocols supplemented with different concentrations and combinations of benzylaminopurine (BAP) (0, 2, 4, 6 mg L-1) and indole acetic acid (IAA) (0.5 and 1.0 mg L-1). Exotic banana (Musa spp) genotypes GCTCV-215 (AAA), ‘Yangambi’ Yangambi Km-5 (AAA) and FHIA-23 (AAAA) were used in research work. Experiments were conducted at Plant Tissue Culture Laboratory of Nuclear Institute of Agriculture (NIA), Tando Jam. Data collected for in vitro shoot consists of the following parameters: days for bud initiation, rate of shoot proliferation (%), number of multiple shoots, shoot length (cm) and fresh mass of shoot (g). Significant (p≤ 0.05) variations were observed for varieties, treatments and varieties x treatment for all the parameters. Synergistic effects of BAP and IAA were observed in GCTCV-215 and Yangambi Km-5. Out of various treatments, best concentration for multiple shoot in short period of time for GCTCV-215 and Yangambi Km-5 was found in 4.0 mg /l BAP + 0.5 mg L-1 IAA. Maximum fresh mass of shoot observed at same concentration and combination of BAP and IAA and for shoot length combination of 4.0 mg L-1 BAP with 1.0 mg L-1 IAA was found to be most suitable for GCTCV-215 and Yangambi Km-5. FHIA-23, show better performance in MS medium supplemented with only BAP at concentration 4.0 mg/L-1. After development of root, in vitro plantlets were shifted from growth room to green house in polythene bags containing garden soil and humus mixture in ratio (1:1).Keywords: Micropropagation efficiency, exotic musa genotype, growth regulators

    Monitoring of particulate matter concentrations at high altitude ecosystems of Pakistan and China

    Particulate matter exhibits different behavior with altitude. A comparative analysis was carried out to monitor PM1, PM2.5, PM4, PM10 and PMTotal at elevations above 3000 m in both China and Pakistan. Real time monitoring of PM was carried out at both sites using a DustTrak DRX (model 8533, TSI Inc.) for 24 hours each. In Pakistan, the average value of PMTotal was 415 ± 16 μg/m3 while in China the value was considerably lower i.e. 110 ± 57 μg/m3. The 24-hour mean values recorded were well above the WHO recommended limit of 25 μg/m3. These results indicate that, even at sites some distance from anthropogenic sources, PM concentrations still pose a health risk

    Changes in particulate matter concentrations at different altitudinal levels with environmental dynamics

    Ambient air quality is defined not only by the source strength but a variety of meteorological parameters as well. In the current study, ambient concentrations of PM along with temperature and relative humidity levels were monitored at seven different locations of Pakistan. A DustTrak DRX (Model 8533, TSI Inc.) was employed for twenty four hours real time monitoring of particulate matter at the selected sites. A considerable variation was observed in the 24 hour trend of particulate matter (PM) at different locations owing to variation in meteorological conditions due to different altitudes and seasons, and natural and anthropogenic sources in the vicinity. The highest average concentrations of PM2.5 (407μg/m3 were observed at highest elevation (Makra Peak, Shogran, 3089 m) while lowest averages (102 μg/m3) were obtained at the seaside (Hawks Bay, Karachi, 0 m).On the other hand PMTotal fraction exhibited highest levels at site B (506 μg/m3) and lowest at Site A (121 μg/m3).Correlation factors were determined for PM and meteorological parameters at each location. More research needs to be conducted to have a comprehensive knowledge about the physical parameters controlling particulate dispersal at different altitudes within the country

    Seasonal variation of particulate matter in the ambient conditions of Khanspur, Pakistan

    Concentrations of particulate matter tend to vary with shifting seasons. Levels of particulate matter were monitored during the summer and winter season in Khanspur, a high altitude tourist resort in Pakistan. A DustTrak DRX (Model 8533, TSI Inc.) and Kestrel 4500 Pocket Weather Tracker (Nielsen- Kellerman) were installed at selected site in Khanspur and run for 24 hours. During summer the 24- hour average concentrations of PM1, PM2.5, PM4, PM10 and PMTotal were 96 ± 26.42, 106± 29.02, 118± 33.3, 163± 52.5 and 209 ± 79.5 μg/m3 while these were considerably lower during the winter season for the same size fractions (62 ± 48.6, 63± 49.3, 63 ± 49.5, 65.33 ± 50.06 and 66.96 ± 50.78μg/m3). A one way ANOVA was applied on the obtained data and it was concluded that seasons have a substantial impact upon PM concentrations. Moreover, this study provides evidence that seasonal variation of particulate matter is influenced by meteorological parameters

    Echocardiographic AV-interval optimization in patients with reduced left ventricular function

    BACKGROUND: Ritter's method is a tool used to optimize AV delay in DDD pacemaker patients with normal left ventricular function only. The goal of our study was to evaluate Ritter's method in AV delay-interval optimization in patients with reduced left ventricular function. METHODS: Patients with implanted DDD pacemakers and AVB III° were assigned to one of two groups according to ejection fraction (EF): Group 1 (EF > 35%) and Group 2 (EF < 35%). AV delay optimization was performed by means of radionuclide ventriculography (RNV) and application of Ritter's method. RESULTS: For each of the patients examined, we succeeded in defining an optimal AV interval by means of both RNV and Ritter's method. The optimal AV delay determined by RNV correlated well with the delay found by Ritter's method, especially among those patients with reduced EF. The intra-class correlation coefficient was 0.8965 in Group 1 and 0.9228 in Group 2. The optimal AV interval in Group 1 was 190 ± 28.5 ms, and 180 ± 35 ms in Group 2. CONCLUSION: Ritter's method is also effective for optimization of AV intervals among patients with reduced left ventricular function (EF < 35%). The results obtained by RNV correlate well with those from Ritter's method. Individual programming of the AV interval is fundamentally essential in all cases

    Towards informed and multi-faceted wildlife trade interventions

    International trade in wildlife is a key threat to biodiversity conservation. CITES, the Convention on International Trade in Endangered Species of Wild Fauna and Flora, is the primary mechanism for controlling international wildlife trade and seeks to ensure it is sustainable, relying on trade bans and controls. However, there has been little comprehensive review of the effectiveness of CITES. Here, we review typical and atypical approaches taken to regulate wildlife trade in CITES and assert that it boasts few successes. We attribute this to: non-compliance, an over reliance on regulation, lack of knowledge of listed species, ignorance of the reality of market forces, and influence among CITES actors. To more effectively manage trade we argue that interventions need to go beyond regulation and should be multi-faceted, reflecting the complexity of wildlife trade. To inform such interventions we assert an intensive research effort is needed and we outline six key research areas: (1) factors undermining wildlife trade governance at the national level, (2) determining sustainable harvest rates for CITES species, (3) gaining the buy-in of local communities in implementing CITES, (4) supply and demand based market interventions, (5) means of quantifying illicit trade, and (6) political processes and influence within CITES

    Early start and stop of biologics: has the time come?

    Despite considerable advances in the management of rheumatoid arthritis, results are still not satisfactory for all patients. The treatment goal in rheumatoid arthritis is remission, and there currently are numerous conventional and biological medications available to reach this aim. There are also different treatment strategies but with only limited comparative evidence about their efficacies. More patients now achieve remission while on treatment, but it remains elusive in the majority of patients. Treatment-free remission, the ultimate goal of therapy, is only achieved in very few patients; even when this happens, it is most likely due to the natural course of the disease rather than to any specific therapies. Modern treatment is based on the initiation of aggressive therapy as soon as the diagnosis is established, and on modifying or intensifying therapy guided by frequent assessment of disease activity. In this commentary we will discuss the current treatment paradigm as well as the possibility of an induction-maintenance regimen with biological disease-modifying antirheumatic drugs in early rheumatoid arthriti
