    Comparison of circular orbit and Fourier power series ephemeris representations for backup use by the upper atmosphere research satellite onboard computer

    The Upper Atmosphere Research Satellite (UARS) is a three-axis stabilized Earth-pointing spacecraft in a low-Earth orbit. The UARS onboard computer (OBC) uses a Fourier Power Series (FPS) ephemeris representation that includes 42 position and 42 velocity coefficients per axis, with position residuals at 10-minute intervals. New coefficients and 32 hours of residuals are uploaded daily. This study evaluated two backup methods that permit the OBC to compute an approximate spacecraft ephemeris in the event that new ephemeris data cannot be uplinked for several days: (1) extending the use of the FPS coefficients previously uplinked, and (2) switching to a simple circular orbit approximation designed and tested (but not implemented) for LANDSAT-D. The FPS method provides greater accuracy during the backup period and does not require additional ground operational procedures for generating and uplinking an additional ephemeris table. The tradeoff is that the high accuracy of the FPS will be degraded slightly by adopting the longer fit period necessary to obtain backup accuracy for an extended period of time. The results for UARS show that extended use of the FPS is superior to the circular orbit approximation for short-term ephemeris backup

The pricing rule we propose is an alternative for the wellknown no-arbitrage pricing on markets without frictions.The price of an asset equals the signed Choquet integral of its discounted payo with respect to a concave signed capacity.We show that this pricing rule is consistent with equilibrium.     Exhaustive mutagenesis of six secondary active-site residues in Escherichia coli chorismate mutase shows the importance of hydrophobic side chains and a helix N-capping position for stability and catalysis

    Secondary active-site residues in enzymes, including hydrophobic amino acids, may contribute to catalysis through critical interactions that position the reacting molecule, organize hydrogen-bonding residues, and define the electrostatic environment of the active site. To ascertain the tolerance of an important model enzyme to mutation of active-site residues that do not directly hydrogen bond with the reacting molecule, all 19 possible amino acid substitutions were investigated in six positions of the engineered chorismate mutase domain of the Escherichia coli chorismate mutase-prephenate dehydratase. The six secondary active-site residues were selected to clarify results of a previous test of computational enzyme design procedures. Five of the positions encode hydrophobic side chains in the wild-type enzyme, and one forms a helix N-capping interaction as well as a salt bridge with a catalytically essential residue. Each mutant was evaluated for its ability to complement an auxotrophic chorismate mutase deletion strain. Kinetic parameters and thermal stabilities were measured for variants with in vivo activity. Altogether, we find that the enzyme tolerated 34% of the 114 possible substitutions, with a few mutations leading to increases in the catalytic efficiency of the enzyme. The results show the importance of secondary amino acid residues in determining enzymatic activity, and they point to strengths and weaknesses in current computational enzyme design procedures

    Effects of microflora composition in the phyllosphere on biological regulation of grapevine fungal diseases

    Aqueous fermentation extracts of composted microbiologically active substrates reduced significantly the infestation of grapevine leaves with Plasmopara viticola under growth chamber conditions. Enhancement of efficacy was available through the enrichment of the extracts with nutrients - like sucrose and brewer's yeast - or with a natural surfactant (methylized cellulose). Studies on the microbial composition of the phyllosphere showed that the quantity of the total number of colony forming units, of yeasts/filametous fungi, of enterobacteria, of pseudomonads and of aerobic bacilli depends on the specific amendments to the extracts and on the microclimatic conditions. They ace responsible for the differences in efficiency of the various extracts against P. viticola. Appropriate investigations on the phyllosphere in a field experiment resulted in a different composition of the microflora, though the same combinations of extract amendments were used. This is caused by the varying environmental conditions. In spite of the changed microflora, the disease suppressing effects against Urcinula necator were preserved

    Aldehyde dehydrogenase and HSP90 co-localize in human glioblastoma biopsy cells.

    The concept of a stem cell subpopulation as understood from normal epithelial tissue or bone marrow function has been extended to our understanding of cancer tissue and is now the target of treatment efforts specifically directed to this subpopulation. In glioblastoma, as well as in other cancers, increased expression of aldehyde dehydrogenase (ALDH) has been found localized within a minority sub-population of tumor cells which demonstrate stem cell properties. A separate body of research associated increased expression of heat-shock protein-90 (HSP90) with stem cell attributes. We present here results from our initial immunohistochemistry study of human glioblastoma biopsy tissue where both ALDH and HSP90 tended to be co-expressed in high amounts in the same minority of cells. Since 12% of all cells in the six biopsies studied were ALDH positive and 17% were HSP90 positive, by chance alone 2% would have been expected to be positive for both. In fact 7% of all cells simultaneously expressed both markers-a significant difference (p = 0.037). That two previously identified proteins associated with stem cell attributes tend to be co-expressed in the same individual glioblastoma cells might have clinical utility. Disulfiram, used to treat alcoholism for half-a century now, is a potent ALDH inhibitor and the old anti-viral drug ritonavir inhibits HSP90. These should be explored for the potential to retard aspects of glioblastoma stem cells' function subserved by ALDH and HSP90

    Annexin A2 in Virus Infection

    Viral life cycles consist of three main phases: (1) attachment and entry, (2) genome replication and expression, and (3) assembly, maturation, and egress. Each of these steps is intrinsically reliant on host cell factors and processes including cellular receptors, genetic replication machinery, endocytosis and exocytosis, and protein expression. Annexin A2 (AnxA2) is a membrane-associated protein with a wide range of intracellular functions and a recurrent host factor in a variety of viral infections. Spatially, AnxA2 is found in the nucleus and cytoplasm, vesicle-bound, and on the inner and outer leaflet of the plasma membrane. Structurally, AnxA2 exists as a monomer or in complex with S100A10 to form the AnxA2/S100A10 heterotetramer (A2t). Both AnxA2 and A2t have been implicated in a vast array of cellular functions such as endocytosis, exocytosis, membrane domain organization, and translational regulation through RNA binding. Accordingly, many discoveries have been made involving AnxA2 in viral pathogenesis, however, the reported work addressing AnxA2 in virology is highly compartmentalized. Therefore, the purpose of this mini review is to provide information regarding the role of AnxA2 in the lifecycle of multiple epithelial cell-targeting viruses to highlight recurrent themes, identify discrepancies, and reveal potential avenues for future research

    Autoselection of Cytoplasmic Yeast Virus Like Elements Encoding Toxin/Antitoxin Systems Involves a Nuclear Barrier for Immunity Gene Expression

    Cytoplasmic virus like elements (VLEs) from Kluyveromyces lactis (Kl), Pichia acaciae (Pa) and Debaryomyces robertsiae (Dr) are extremely A/T-rich (>75%) and encode toxic anticodon nucleases (ACNases) along with specific immunity proteins. Here we show that nuclear, not cytoplasmic expression of either immunity gene (PaORF4, KlORF3 or DrORF5) results in transcript fragmentation and is insufficient to establish immunity to the cognate ACNase. Since rapid amplification of 3' ends (RACE) as well as linker ligation of immunity transcripts expressed in the nucleus revealed polyadenylation to occur along with fragmentation, ORF-internal poly(A) site cleavage due to the high A/T content is likely to prevent functional expression of the immunity genes. Consistently, lowering the A/T content of PaORF4 to 55% and KlORF3 to 46% by gene synthesis entirely prevented transcript cleavage and permitted functional nuclear expression leading to full immunity against the respective ACNase toxin. Consistent with a specific adaptation of the immunity proteins to the cognate ACNases, cross-immunity to non-cognate ACNases is neither conferred by PaOrf4 nor KlOrf3. Thus, the high A/T content of cytoplasmic VLEs minimizes the potential of functional nuclear recruitment of VLE encoded genes, in particular those involved in autoselection of the VLEs via a toxin/antitoxin principle

    Learning to Generate Ambiguous Sequences

    In this paper, we experiment with methods for obtaining binary sequences with a random probability mass function and with low autocorrelation and use it to generate ambiguous outcomes. Outputs from a neural network are mixed and shuffled, resulting in binary sequences whose probability mass function is non-convergent, constantly moving and changing. Empirical comparison with algorithms that generate ambiguity shows that the sequences generated by the proposed method have a significantly lower serial dependence. Therefore, the method is useful in scenarios where observes can see and record the outcome of each draw sequentially, by hindering the ability to make useful statistical inferences
