    Motivational factors influencing farming practices in northern Ghana

    Socio-economic factors that influence the adoption of management practices and technologies by farmers have received wide attention in the adoption literature, but the effects of socio-psychological farmer features such as perceptions and motivations have been analysed to a lesser extent. Using farm household survey data from three regions in northern Ghana, this study explores farmers’ motivations and perceived adoption impediments for three sustainable intensification practices (SIPs): improved maize varieties, cropping system strategies, and combined SIPs (i.e. improved maize and cropping system strategies), and the effect of motivational factors on decisions to adopt SIPs. First, explorative factor analysis (EFA) was used in identifying factors of motivations and impediments for adoption of SIPs. Then, a multinomial logit model was used to analyze the effect of socio-economic farm characteristics and motivational factors on farmers’ decisions to adopt SIPs. EFA identified three motivational factors: personal satisfaction, eco-diversity and eco-efficiency, which differed in importance between the three regions. Across these regions, higher scores for aspects of personal satisfaction were associated with lower interest in improved maize varieties compared to cropping system strategies, while the opposite was true for eco-efficiency which was related to a stronger preference for improved maize varieties. Uncertainty, absence of social support, and resource constraints were identified as impediment factors. The logit model demonstrated that extension services seemed to support the use of improved maize varieties more than the implementation of cropping system strategies. We conclude that motivational factors significantly influence farmer adoption decisions regarding sustainable intensification practices and should be considered systematically in combination with socio-economic farm features and external drivers to inform on-farm innovation processes and supporting policies.</p

    bRing: An observatory dedicated to monitoring the Ξ²\beta Pictoris b Hill sphere transit

    Aims. We describe the design and first light observations from the Ξ²\beta Pictoris b Ring ("bRing") project. The primary goal is to detect photometric variability from the young star Ξ²\beta Pictoris due to circumplanetary material surrounding the directly imaged young extrasolar gas giant planet \bpb. Methods. Over a nine month period centred on September 2017, the Hill sphere of the planet will cross in front of the star, providing a unique opportunity to directly probe the circumplanetary environment of a directly imaged planet through photometric and spectroscopic variations. We have built and installed the first of two bRing monitoring stations (one in South Africa and the other in Australia) that will measure the flux of Ξ²\beta Pictoris, with a photometric precision of 0.5%0.5\% over 5 minutes. Each station uses two wide field cameras to cover the declination of the star at all elevations. Detection of photometric fluctuations will trigger spectroscopic observations with large aperture telescopes in order to determine the gas and dust composition in a system at the end of the planet-forming era. Results. The first three months of operation demonstrate that bRing can obtain better than 0.5\% photometry on Ξ²\beta Pictoris in five minutes and is sensitive to nightly trends enabling the detection of any transiting material within the Hill sphere of the exoplanet

    MutLΞ± heterodimers modify the molecular phenotype of Friedreich ataxia

    This article has been made available through the Brunel Open Access Publishing Fund.Background: Friedreich ataxia (FRDA), the most common autosomal recessive ataxia disorder, is caused by a dynamic GAA repeat expansion mutation within intron 1 of FXN gene, resulting in down-regulation of frataxin expression. Studies of cell and mouse models have revealed a role for the mismatch repair (MMR) MutS-heterodimer complexes and the PMS2 component of the MutLΞ± complex in the dynamics of intergenerational and somatic GAA repeat expansions: MSH2, MSH3 and MSH6 promote GAA repeat expansions, while PMS2 inhibits GAA repeat expansions. Methodology/Principal Findings: To determine the potential role of the other component of the MutLΞ± complex, MLH1, in GAA repeat instability in FRDA, we have analyzed intergenerational and somatic GAA repeat expansions from FXN transgenic mice that have been crossed with Mlh1 deficient mice. We find that loss of Mlh1 activity reduces both intergenerational and somatic GAA repeat expansions. However, we also find that loss of either Mlh1 or Pms2 reduces FXN transcription, suggesting different mechanisms of action for Mlh1 and Pms2 on GAA repeat expansion dynamics and regulation of FXN transcription. Conclusions/Significance: Both MutLΞ± components, PMS2 and MLH1, have now been shown to modify the molecular phenotype of FRDA. We propose that upregulation of MLH1 or PMS2 could be potential FRDA therapeutic approaches to increase FXN transcription.     Brain architecture in the terrestrial hermit crab Coenobita clypeatus (Anomura, Coenobitidae), a crustacean with a good aerial sense of smell

    <p>Abstract</p> <p>Background</p> <p>During the evolutionary radiation of Crustacea, several lineages in this taxon convergently succeeded in meeting the physiological challenges connected to establishing a fully terrestrial life style. These physiological adaptations include the need for sensory organs of terrestrial species to function in air rather than in water. Previous behavioral and neuroethological studies have provided solid evidence that the land hermit crabs (Coenobitidae, Anomura) are a group of crustaceans that have evolved a good sense of aerial olfaction during the conquest of land. We wanted to study the central olfactory processing areas in the brains of these organisms and to that end analyzed the brain of <it>Coenobita clypeatus </it>(Herbst, 1791; Anomura, Coenobitidae), a fully terrestrial tropical hermit crab, by immunohistochemistry against synaptic proteins, serotonin, FMRFamide-related peptides, and glutamine synthetase.</p> <p>Results</p> <p>The primary olfactory centers in this species dominate the brain and are composed of many elongate olfactory glomeruli. The secondary olfactory centers that receive an input from olfactory projection neurons are almost equally large as the olfactory lobes and are organized into parallel neuropil lamellae. The architecture of the optic neuropils and those areas associated with antenna two suggest that <it>C. clypeatus </it>has visual and mechanosensory skills that are comparable to those of marine Crustacea.</p> <p>Conclusion</p> <p>In parallel to previous behavioral findings of a good sense of aerial olfaction in C. clypeatus, our results indicate that in fact their central olfactory pathway is most prominent, indicating that olfaction is a major sensory modality that these brains process. Interestingly, the secondary olfactory neuropils of insects, the mushroom bodies, also display a layered structure (vertical and medial lobes), superficially similar to the lamellae in the secondary olfactory centers of <it>C. clypeatus</it>. More detailed analyses with additional markers will be necessary to explore the question if these similarities have evolved convergently with the establishment of superb aerial olfactory abilities or if this design goes back to a shared principle in the common ancestor of Crustacea and Hexapoda.</p

    Scallop swimming kinematics and muscle performance: modelling the effects of "within-animal" variation in temperature sensitivity

    Escape behaviour was investigated in Queen scallops (Aequipecten opercularis) acclimated to 5, 10 or 15 degrees C and tested at their acclimation temperature. Scallops are active molluscs, able to escape from predators by jet-propelled swimming using a striated muscle working in opposition to an elastic hinge ligament. The first cycle of the escape response was recorded using high-speed video ( 250 Hz) and whole-animal velocity and acceleration determined. Muscle shortening velocity, force and power output were calculated using measurements of valve movement and jet area, and a simple biomechanical model. The average shortening speed of the adductor muscle had a Q(10) of 2.04, significantly reducing the duration of the jetting phase of the cycle with increased temperature. Muscle lengthening velocity and the overall duration of the clap cycle were changed little over the range 5 - 15 degrees C, as these parameters were controlled by the relatively temperature-insensitive, hinge ligament. Improvements in the average power output of the adductor muscle over the first clap cycle ( 222 vs. 139 W kg(-1) wet mass at 15 and 5 degrees C respectively) were not translated into proportional increases in overall swimming velocity, which was only 32% higher at 15 degrees C ( 0.37m s(-1)) than 5 degrees C (0.28 m s(-1))

    Distinct Properties of Hexameric but Functionally Conserved Mycobacterium tuberculosis Transcription-Repair Coupling Factor

    Transcription coupled nucleotide excision repair (TC-NER) is involved in correcting UV-induced damage and other road-blocks encountered in the transcribed strand. Mutation frequency decline (Mfd) is a transcription repair coupling factor, involved in repair of template strand during transcription. Mfd from M. tuberculosis (MtbMfd) is 1234 amino-acids long harboring characteristic modules for different activities. Mtbmfd complemented Escherichia coli mfd (Ecomfd) deficient strain, enhanced survival of UV irradiated cells and increased the road-block repression in vivo. The protein exhibited ATPase activity, which was stimulated ∼1.5-fold in the presence of DNA. While the C-terminal domain (CTD) comprising amino acids 630 to 1234 showed ∼2-fold elevated ATPase activity than MtbMfd, the N-terminal domain (NTD) containing the first 433 amino acid residues was able to bind ATP but deficient in hydrolysis. Overexpression of NTD of MtbMfd led to growth defect and hypersensitivity to UV light. Deletion of 184 amino acids from the C-terminal end of MtbMfd (MfdΞ”C) increased the ATPase activity by ∼10-fold and correspondingly exhibited efficient translocation along DNA as compared to the MtbMfd and CTD. Surprisingly, MtbMfd was found to be distributed in monomer and hexamer forms both in vivo and in vitro and the monomer showed increased susceptibility to proteases compared to the hexamer. MfdΞ”C, on the other hand, was predominantly monomeric in solution implicating the extreme C-terminal region in oligomerization of the protein. Thus, although the MtbMfd resembles EcoMfd in many of its reaction characteristics, some of its hitherto unknown distinct properties hint at its species specific role in mycobacteria during transcription-coupled repair

This emergent form of creative work has found an outlet in a wide variety of different domains ranging from community social history, to cookbooks, to the classroom. It is the latter domain that provides the focus for this paper, specifically the online classroom at the tertiary level...Early feedback from students suggests that listening to and telling \u27true stories\u27 was a compelling and emotionally-engaging experience, providing an opportunity for \u27transformative reflection\u27 (Lambert 2000). By including multimedia, learners were able to build upon the fundamentals, presenting content in an easy-to-absorb and compelling way. In terms of team assignments students learned to become more effective actors in collaborative work environments
