    Stability aspects of the traveling salesman problem based on k-best solutions

    AbstractThis paper discusses stability analysis for the Traveling Salesman Problem (TSP). For a traveling salesman tour which is known to be optimal with respect to a given instance (length vector) we are interested in determining the stability region, i.e. the set of all length vectors for which the tour is optimal. The following three subsets of the stability region are of special interest: 1.(1) tolerances, i.e. the maximum perturbations of single edges;2.(2) tolerance regions which are subsets of the stability region that can be constructed from the tolerances; and3.(3) the largest ball contained in the stability region centered at the given length vector (the corresponding radius is known as the stability radius). It is well known that the problems of determining tolerances and the stability radius for the TSP are NP-hard so that in general it is not possible to obtain the above-mentioned three subsets without spending a lot of computation time. The question addressed in this paper is the following: assume that not only an optimal tour is known, but also a set of k shortest tours (k ⩾2) is given. Then to which extent does this allow us to determine the three subsets in polynomial time? It will be shown in this paper that having k-best solutions can give the desired information only partially. More precisely, it will be shown that only some of the tolerances can be determined exactly and for the other ones as well as for the stability radius only lower and/or upper bounds can be derived. Since the amount of information that can be derived from the set of k-best solutions is dependent on both the value of k as well as on the specific length vector, we present numerical experiments on instances from the TSPLIB library to analyze the effectiveness of our approach

    Fabrication of corrugated Ge-doped silica fibers

    We present a method of fabricating Ge-doped SiO2 fibers with corrugations around their full circumference for a desired length in the longitudinal direction. The procedure comprises three steps: hydrogenation of Ge-doped SiO2 fibers to increase photosensitivity, recording of Bragg gratings with ultraviolet light to achieve modulation of refractive index, and chemical etching. Finite-length, radially corrugated fibers may be used as couplers. Corrugated tapered fibers are used as high energy throughput probes in scanning near-field optical microscopy

    Selective laser melting of a high precision turbomachinery application in IN718 alloy

    The paper describes the manufacture of an outlet guide vane (OGV) component, in IN718 alloy, used in jet engines by Selective Laser Melting (SLM). The OGV component is a static part in the last stage of the compressor and is characterised as a series of airfoils or vanes secured by two flanged rings. The part tolerances at the leading and trailing edge require a high dimensional precision of +/-0.072 m whilst the profile tolerances are slightly more generous. The current challenge to manufacture a prototype OGV in IN718 alloy from a wrought stock involves a lengthy machining process in a hard-to-machine alloy.     Thymoquinone Inhibits Bone Metastasis of Breast Cancer Cells Through Abrogation of the CXCR4 Signaling Axis

    Overexpression of chemokine receptor type 4 (CXCR4) has been found to be associated with increased cell proliferation, metastasis and also act as an indicator of poor prognosis in patients with breast cancer. Therefore, new agents that can abrogate CXCR4 expression have potential against breast cancer metastasis. In this study, we examined the potential effect of thymoquinone (TQ), derived from the seeds of Nigella sativa, on the expression and regulation of CXCR4 in breast cancer cells. TQ was found to inhibit the expression of CXCR4 in MDA-MB-231 triple negative breast cancer (TNBC) cells in a dose- and time-dependent manner. It was noted that suppression of CXCR4 by TQ was possibly transcriptionally regulated, as treatment with this drug caused down-regulation of nuclear factor kappa-light-chain-enhancer of activated B cells (NF-kB) activation and suppression of NF-kB binding to the CXCR4 promoter. Pretreatment with a proteasome inhibitor and/or lysosomal stabilization did not affect TQ induced suppression of CXCR4. Down-regulation of CXCR4 was further correlated with the inhibition of CXCL12-mediated migration and invasion of MDA-MB-231 cells. Interestingly, it was observed that the deletion of p65 could reverse the observed antiinvasive/ anti-migratory effects of TQ in breast cancer cells. TQ also dose-dependently inhibited MDA-MB-231 tumor growth and tumor vascularity in a chick chorioallantoic membrane assay model. We also observed TQ (2 and 4 mg/kg) treatment significantly suppressed multiple lung, brain, and bone metastases in a dose-dependent manner in a metastasis breast cancer mouse model. Interestingly, H&E and immunohistochemical analysis of bone isolated from TQ treated mice indicated a reduction in number of osteolytic lesions and the expression of metastatic biomarkers. In conclusion, the results indicate that TQ primarily exerts its anti-metastatic effects by down-regulation of NF-kB regulated CXCR4 expression and thus has potential for the treatment of breast cancer

    Identification of Hepatic Niche Harboring Human Acute Lymphoblastic Leukemic Cells via the SDF-1/CXCR4 Axis

    In acute lymphoblastic leukemia (ALL) patients, the bone marrow niche is widely known to be an important element of treatment response and relapse. Furthermore, a characteristic liver pathology observed in ALL patients implies that the hepatic microenvironment provides an extramedullary niche for leukemic cells. However, it remains unclear whether the liver actually provides a specific niche. The mechanism underlying this pathology is also poorly understood. Here, to answer these questions, we reconstituted the histopathology of leukemic liver by using patients-derived primary ALL cells into NOD/SCID/Yc null mice. The liver pathology in this model was similar to that observed in the patients. By using this model, we clearly demonstrated that bile duct epithelial cells form a hepatic niche that supports infiltration and proliferation of ALL cells in the liver. Furthermore, we showed that functions of the niche are maintained by the SDF-1/CXCR4 axis, proposing a novel therapeutic approach targeting the extramedullary niche by inhibition of the SDF-1/CXCR4 axis. In conclusion, we demonstrated that the liver dissemination of leukemia is not due to nonselective infiltration, but rather systematic invasion and proliferation of leukemic cells in hepatic niche. Although the contribution of SDF-1/CXCR4 axis is reported in some cancer cells or leukemic niches such as bone marrow, we demonstrated that this axis works even in the extramedullary niche of leukemic cells. Our findings form the basis for therapeutic approaches that target the extramedullary niche by inhibiting the SDF-1/CXCR4 axis
