    Expression of cyclin B1 after induction of senescence and cell death in non-small cell lung carcinoma A549 cells

    The purpose of this study was to evaluate the level of mitotic cyclin B1 in the context of senescence and cell death in A549 non-small cell lung carcinoma cells. This was performed through analysis of the cell cycle, the percentage of SA-β-galactosidase-positive, as well as TUNEL-positive cells. Morphological alterations were studied using a transmission electron microscope. Changes in  the intracellular level and the presence of cyclin B1 in the nucleus and cytoplasm areas were detected by flow cytometry and confocal fluorescence microscopy, respectively. In the cells exposed to various concentrations of doxorubicin, different kinds of cell death and senescent phenotype were observed. Alterations in the cell cycle and increased polyploidy may be indicative of mitotic catastrophe execution. Changes in cyclin B1 may also be strictly related to its different regulation at mitotic catastrophe and senescence programs

    Markery stosowane w onkologii stanowią niejednorodną grupę czynników umożliwiających diagnozę lub ocenęinwazyjności nowotworu. Wyróżnia się trzy grupy markerów prognostycznych, wśród których ważne miejsce zajmująmarkery biologii nowotworu. Czynniki prognostyczne należące do tej grupy obrazują między innymi zdolność komórekdo przerzutowania oraz odpowiedzi na zastosowaną terapię. Do markerów biologii nowotworu zaliczane są białkaregulujące progresję cyklu komórkowego, m.in. cykliny. Cykliny są kluczowymi regulatorami cyklu komórkowego,których stężenie jest zależne od fazy cyklu. Zmiany ekspresji cykliny (głównie cyklin typu A, B, D oraz E) wpływają naaktywność proliferacyjną komórek i mają związek z rozwojem różnych nowotworów. Sugeruje się, że badanie profiluekspresji poszczególnych cyklin w komórkach nowotworowych może dostarczać cennych informacji prognostycznychoraz diagnostycznych. Praca ta przedstawia obecny stan wiedzy na temat potencjalnego wykorzystania cyklinw diagnostyce chorób nowotworowych

    Treatment of patients with primary cutaneous lymphomas – real-life data

    Background. Primary cutaneous lymphomas (PCL) comprise a heterogeneous group of neoplasms of mature lymphocytes with skin tropism. Although, by definition, these lymphomas are restricted to the skin at the time of diagnosis, during the course of the disease it may involve also lymph nodes and visceral organs. A close cooperation between a dermatologist and oncologist is required to ensure proper treatment. We present in a real-life data on treatment of patients with PCL between dermatology and oncology department. Material and methods. 104 patients were registered in a joined database of Oncology Department of Oncology Centre in Bydgoszcz and Dermatology Department of Medical University in Toruń between 2007 and 2017. Due to different clinical and prognostic features data from MF/SS (44 patients), non-MF/SS CTCLs and CBCLs were presented separately. Results. Median overall survival for patients with MF/SS was 76.7 months. Median follow-up time was 5 years.Background.     Oxidative stress plays an important role in the etiopathogenesis of acquired vitiligo. In patients with this disorder, impaied performance of the antioxidant system leads to the accumulation of reactive oxygen species in their skin. Under physiological conditions, they play the role of mediators and regulators of many cellular processes. Their excess, however, leads to the destruction of structural and functional elements of dye cells. Previous studies clearly indicate the share of free radicals in the progression of lesions. The use of antioxidant defense elements, including antioxidants, may prove to be an important element of effective therapy of this disease

    W niniejszej pracy przedstawiono własne, jednoośrodkowe doświadczenia dotyczące stosowania interferonu w pierwszej linii leczenia systemowego ziarniniaka grzybiastego. Badaniem objęto grupę chorych leczonych w Centrum Onkologii w Bydgoszczy, u których po wyczerpaniu możliwości leczenia miejscowego oraz w stopniu zaawansowania Ib–III podjęto terapię systemową. Maksymalny czas prowadzenia terapii sięgał 3 lat. Interferon alfa jest leczeniem dobrze tolerowanym, skutkującym poprawąjakości życia u 80% chorych. Lek ten powoduje znaczną bądź całkowitą remisję trwającą do kilkunastu miesięcy. Terapia ziarniniaka grzybiastego jest leczeniem choroby przewlekłej, co wpływa na zasady prowadzenia terapii.This paper reports our own experience on interferon-alpha as the first-line systemic treatment of mycosis fungoides in the group of patients with advanced stages of disease after failure of skin-directedtherapies. Therapy resulted in improved quality of life and long-lasting, high response rate up to 80%. Given chronic character of MF special account must be taken not only to efficacy of the therapy, but also the safety

    SARS-CoV-2 infects an <I>in vitro</I> model of the human developing pancreas through endocytosis

    Recent studies showed that SARS-CoV-2 can infect adult human pancreas and trigger pancreatic damage. Here, using human fetal pancreas samples and 3D differentiation of human pluripotent cells into pancreatic endocrine cells, we determined that SARS-CoV-2 receptors ACE2, TMPRSS2, and NRP1 are expressed in precursors of insulin-producing pancreatic β-cells, rendering them permissive to SARS-CoV-2 infection. We also show that SARS-CoV-2 enters and undergoes efficient replication in human multipotent pancreatic and endocrine progenitors in vitro. Moreover, we investigated mechanisms by which SARS-CoV-2 enters pancreatic cells, and found that ACE2 mediates the entry, while NRP1 and TMPRSS2 do not. Surprisingly, we found that in pancreatic progenitors, SARS-CoV-2 enters cells via cathepsin-dependent endocytosis, which is a different route than in respiratory tract. Therefore, pancreatic spheroids might serve as a model to study candidate drugs for endocytosis-mediated viral entry inhibition and to investigate whether SARS-CoV-2 infection may affect pancreas development, possibly causing lifelong health consequences

    Measurement of the top quark forward-backward production asymmetry and the anomalous chromoelectric and chromomagnetic moments in pp collisions at √s = 13 TeV

    Abstract The parton-level top quark (t) forward-backward asymmetry and the anomalous chromoelectric (d̂ t) and chromomagnetic (μ̂ t) moments have been measured using LHC pp collisions at a center-of-mass energy of 13 TeV, collected in the CMS detector in a data sample corresponding to an integrated luminosity of 35.9 fb−1. The linearized variable AFB(1) is used to approximate the asymmetry. Candidate t t ¯ events decaying to a muon or electron and jets in final states with low and high Lorentz boosts are selected and reconstructed using a fit of the kinematic distributions of the decay products to those expected for t t ¯ final states. The values found for the parameters are AFB(1)=0.048−0.087+0.095(stat)−0.029+0.020(syst),μ̂t=−0.024−0.009+0.013(stat)−0.011+0.016(syst), and a limit is placed on the magnitude of | d̂ t| &lt; 0.03 at 95% confidence level.     MUSiC : a model-unspecific search for new physics in proton-proton collisions at root s=13TeV

    Results of the Model Unspecific Search in CMS (MUSiC), using proton-proton collision data recorded at the LHC at a centre-of-mass energy of 13 TeV, corresponding to an integrated luminosity of 35.9 fb(-1), are presented. The MUSiC analysis searches for anomalies that could be signatures of physics beyond the standard model. The analysis is based on the comparison of observed data with the standard model prediction, as determined from simulation, in several hundred final states and multiple kinematic distributions. Events containing at least one electron or muon are classified based on their final state topology, and an automated search algorithm surveys the observed data for deviations from the prediction. The sensitivity of the search is validated using multiple methods. No significant deviations from the predictions have been observed. For a wide range of final state topologies, agreement is found between the data and the standard model simulation. This analysis complements dedicated search analyses by significantly expanding the range of final states covered using a model independent approach with the largest data set to date to probe phase space regions beyond the reach of previous general searches.Peer reviewe

    Search for new particles in events with energetic jets and large missing transverse momentum in proton-proton collisions at root s=13 TeV

    A search is presented for new particles produced at the LHC in proton-proton collisions at root s = 13 TeV, using events with energetic jets and large missing transverse momentum. The analysis is based on a data sample corresponding to an integrated luminosity of 101 fb(-1), collected in 2017-2018 with the CMS detector. Machine learning techniques are used to define separate categories for events with narrow jets from initial-state radiation and events with large-radius jets consistent with a hadronic decay of a W or Z boson. A statistical combination is made with an earlier search based on a data sample of 36 fb(-1), collected in 2016. No significant excess of events is observed with respect to the standard model background expectation determined from control samples in data. The results are interpreted in terms of limits on the branching fraction of an invisible decay of the Higgs boson, as well as constraints on simplified models of dark matter, on first-generation scalar leptoquarks decaying to quarks and neutrinos, and on models with large extra dimensions. Several of the new limits, specifically for spin-1 dark matter mediators, pseudoscalar mediators, colored mediators, and leptoquarks, are the most restrictive to date.Peer reviewe
