The autophagic response to Staphylococcus aureus provides an intracellular niche in neutrophils
Staphylococcus aureus is a major human pathogen causing multiple pathologies, from cutaneous lesions to life-threatening sepsis. Although neutrophils contribute to immunity against S. aureus, multiple lines of evidence suggest that these phagocytes can provide an intracellular niche for staphylococcal dissemination. However, the mechanism of neutrophil subversion by intracellular S. aureus remains unknown. Targeting of intracellular pathogens by macroautophagy/autophagy is recognized as an important component of host innate immunity, but whether autophagy is beneficial or detrimental to S. aureus-infected hosts remains controversial. Here, using larval zebrafish, we showed that the autophagy marker Lc3 rapidly decorates S. aureus following engulfment by macrophages and neutrophils. Upon phagocytosis by neutrophils, Lc3-positive, non-acidified spacious phagosomes are formed. This response is dependent on phagocyte NADPH oxidase as both cyba/p22phox knockdown and diphenyleneiodonium (DPI) treatment inhibited Lc3 decoration of phagosomes. Importantly, NADPH oxidase inhibition diverted neutrophil S. aureus processing into tight acidified vesicles, which resulted in increased host resistance to the infection. Some intracellular bacteria within neutrophils were also tagged by Sqstm1/p62-GFP fusion protein and loss of Sqstm1 impaired host defense. Together, we have shown that intracellular handling of S. aureus by neutrophils is best explained by Lc3-associated phagocytosis (LAP), which appears to provide an intracellular niche for bacterial pathogenesis, while the selective autophagy receptor Sqstm1 is host-protective. The antagonistic roles of LAP and Sqstm1-mediated pathways in S. aureus-infected neutrophils may explain the conflicting reports relating to anti-staphylococcal autophagy and provide new insights for therapeutic strategies against antimicrobial-resistant Staphylococci
The alpha-particle in nuclear matter
Among the light nuclear clusters the alpha-particle is by far the strongest
bound system and therefore expected to play a significant role in the dynamics
of nuclei and the phases of nuclear matter. To systematically study the
properties of the alpha-particle we have derived an effective four-body
equation of the Alt-Grassberger-Sandhas (AGS) type that includes the dominant
medium effects, i.e. self energy corrections and Pauli-blocking in a consistent
way. The equation is solved utilizing the energy dependent pole expansion for
the sub system amplitudes. We find that the Mott transition of an
alpha-particle at rest differs from that expected from perturbation theory and

Altered precipitation and root herbivory affect the productivity and composition of a mesic grassland
Background Climate change models predict changes in the amount, frequency and seasonality of precipitation events, all of which have the potential to affect the structure and function of grassland ecosystems. While previous studies have examined plant or herbivore responses to these perturbations, few have examined their interactions; even fewer have included belowground herbivores. Given the ecological, economic and biodiversity value of grasslands, and their importance globally for carbon storage and agriculture, this is an important knowledge gap. To address this, we conducted a precipitation manipulation experiment in a former mesic pasture grassland comprising a mixture of C-4 grasses and C-3 grasses and forbs, in southeast Australia. Rainfall treatments included a control [ambient], reduced amount [50% ambient] and reduced frequency [ambient rainfall withheld for three weeks, then applied as a single deluge event] manipulations, to simulate predicted changes in both the size and frequency of future rainfall events. In addition, half of all experimental plots were inoculated with adult root herbivores (Scarabaeidae beetles). Results We found strong seasonal dependence in plant community responses to both rainfall and root herbivore treatments. The largest effects were seen in the cool season with lower productivity, cover and diversity in rainfall-manipulated plots, while root herbivore inoculation increased the relative abundance of C-3, compared to C-4, plants. Conclusions This study highlights the importance of considering not only the seasonality of plant responses to altered rainfall, but also the important role of interactions between abiotic and biotic drivers of vegetation change when evaluating ecosystem-level responses to future shifts in climatic conditions.This work was partially supported by a Higher Degree Research Scholarship from the Hawkesbury Institute for the Environment at Western Sydney University. A log analysis study of 10 years of ebook consumption in academic library collections
Even though libraries have been offering eBooks for more than a decade, very little is known about eBook access and consumption in academic library collections. This paper addresses this gap with a log analysis study of eBook access at the library of the University of Waikato. This in-depth analysis covers a period spanning 10 years of eBook use at this university. We draw conclusions about the use of eBooks at this institution and compare the results with other published studies of eBook usage at tertiary institutes
Longitudinal changes in wellbeing amongst breastfeeding women in Australia and New Zealand during the COVID-19 pandemic
The COVID-19 pandemic has impacted new mothersâ wellbeing and breastfeeding experience. Women have experienced changes in birth and postnatal care and restricted access to their support network. It is unclear how these impacts may have changed over time with shifting rates of infection and policies restricting movement and access to services in Australia and New Zealand. This study investigated the longitudinal effect of the COVID-19 pandemic on breastfeeding and maternal wellbeing in Australia and New Zealand. Mothers (nâ=â246) completed an online survey every 4 weeks for 6 months that examined feeding methods, maternal mental wellbeing, worries, challenges, and positive experiences during the pandemic. Mothers maintained high full breastfeeding rates at 4 months (81%) which decreased to 37% at 6 months. Perceived low milk supply contributed to the earlier cessation of full breastfeeding. Poor infant sleep was associated with stress, perinatal anxiety, mental wellbeing, and breastfeeding status. Although mothers initially reported that lockdowns helped with family bonding and less pressure, prolonged lockdowns appeared to have adverse effects on access to social networks and extended family support.
Conclusion: The results highlight the changing dynamic of the pandemic and the need for adaptable perinatal services which allow mothers access to in-person services and their support network even in lockdowns. Similarly, access to continuous education and clinical care remains critical for women experiencing concerns about their milk supply, infant sleep, and their own wellbeing
Photodisintegration of three- and four- nucleon systems
Three- and four-nucleon photodisintegration processes are quite efficiently
treated by means of effective two-body integral equations in momentum space. We
recall some aspects of their derivation, present previous and most recent
results obtained within this framework, and discuss general features, trends
and effects observed in these investigations: At low energies final-state
interaction plays an important role. Even more pronounced is the effect of
meson exchange currents. A considerable potential dependence shows up in the
low-energy peak region. The different peak heights are found to be closely
correlated with the corresponding binding energies. Above the peak region only
the difference between potentials with or without p-wave contributions remains
relevant. In the differential cross sections the electric quadrupole
contributions have to be taken into account. The remarkable agreement between
theory and experiment in - radiative capture is achieved only when
incorporating this contribution, together with most of the above-mentioned
effects. In the final part of this report we briefly review also methods
developed, and results achieved in three- and four- nucleon
electrodisintegration. We, in particular, compare them with a recent access to
this problem, based on the construction of nucleon-nucleus potentials via

The role of macrophages in Staphylococcus aureus infection
Staphylococcus aureus is a member of the human commensal microflora that exists, apparently benignly, at multiple sites on the host. However, as an opportunist pathogen it can also cause a range of serious diseases. This requires an ability to circumvent the innate immune system to establish an infection. Professional phagocytes, primarily macrophages and neutrophils, are key innate immune cells which interact with S. aureus, acting as gatekeepers to contain and resolve infection. Recent studies have highlighted the important roles of macrophages during S. aureus infections, using a wide array of killing mechanisms. In defense, S. aureus has evolved multiple strategies to survive within, manipulate and escape from macrophages, allowing them to not only subvert but also exploit this key element of our immune system. Macrophage-S. aureus interactions are multifaceted and have direct roles in infection outcome. In depth understanding of these host-pathogen interactions may be useful for future therapeutic developments. This review examines macrophage interactions with S. aureus throughout all stages of infection, with special emphasis on mechanisms that determine infection outcome
Three-Nucleon Photodisintegration of 3He
The three-nucleon photodisintegration of 3He has been calculated in the whole
phase space using consistent Faddeev equations for the three-nucleon bound and
scattering states. Modern nucleon-nucleon and 3N forces have been applied as
well as different approaches to nuclear currents. Phase space regions are
localized where 3N force effects are especially large. In addition
semi-exclusive cross sections for 3He(gamma,N) have been predicted which carry
interesting peak structures. Finally some data for the exclusive 3N breakup
process of 3He and its total breakup cross section have been compared to

The effects of aerobic and resistance exercise on state anxiety and cognitive function
Background: There is a paucity of data directly comparing the effects of aerobic versus resistance exercise within a single cohort. Objective: The study aimed to investigate the effects of an acute bout of aerobic and resistance exercise on state anxiety and cognitive function. Equipment and methods: Participants completed three laboratory visits randomized between aerobic exercise (cycling at a self-selected intensity), resistance exercise (three sets; 10 repetitions; six exercises), and a control condition (seated rest). State anxiety and cognitive function were assessed before and after each condition. Results: Reductions in state anxiety that approached significance were seen immediately following aerobic exercise yet were non-significant in the resistance and control conditions. Cognitive function significantly improved following aerobic exercise yet this was not evident in the resistance or control conditions. Conclusion: Acute aerobic exercise at a self-selected intensity may be an effective way of reducing state anxiety and improving cognitive function, when compared to seated rest or resistance training
Systematic behaviour of 3He/4He in Earthâs continental mantle
Helium isotopes are unrivalled tracers of the origins of melts in the Earthâs convecting mantle but their role in determining melt contributions from the shallower and rigid lithospheric mantle is more ambiguous. We have acquired new 3He/4He data for olivine and pyroxene separates from 47 well-characterised mantle xenoliths from global on- and off-craton settings. When combined with existing data they demonstrate a new systematic relationship between fluid-hosted 3He/4He and major and trace element composition of host minerals and whole rock. We show that a significant proportion (>70âŻ%) of mantle peridotites from continental off-craton settings with depleted major element compositions (e.g., olivine Mg# â„ 89.5) have 3He/4He in the range of modern-day mid-ocean ridge basalt (MORB) source mantle (7â9 Ra) and we propose that they represent underplated melt residues, which initially formed in the convecting upper mantle. Furthermore, we observe that off-craton mantle xenoliths with signatures often attributed to enrichment by melts or fluids from âancientâ subducted oceanic lithosphere have lower 3He/4He (<7 Ra). Modest correlations between 3He/4He and whole rock incompatible trace element signatures commonly used as proxies for metasomatism by small-fraction carbonatite and silicate melts or C-O-H fluids characterise lithospheric mantle with 3He/4He ranging from 5 to 8 Ra.
