    Evaluation of a Smart Fork to Decelerate Eating Rate.

    Overweight is associated with a range of negative health consequences, such as type 2 diabetes, cardiovascular disease, gastrointestinal disorders, and premature mortality.1 One means to combat overweight is through encouraging people to eat more slowly.2 People who eat quickly tend to consume more3, 4 and 5 and have a higher body mass index,6, 7, 8 and 9 whereas people who eat more slowly feel sated earlier and eat less.10, 11, 12 and 13. Unfortunately, eating rate is difficult to modify, because of its highly automatic nature.14 In clinical settings, researchers have had some success changing behavior by using devices that deliver feedback in real time.15, 16 and 17 However, existing technologies are either too cumbersome18 or not engaging enough19 for use in daily life contexts. Training people to eat more slowly in everyday eating contexts, therefore, requires creative and engaging solutions. This article presents a qualitative evaluation of the feasibility of a smart fork to decelerate eating rate in daily life contexts. Furthermore, we outline the planned research to test the efficacy of this device in both laboratory and community settings<br/

    The Hand of Cercopithecoides williamsi (Mammalia, Primates): Earliest Evidence for Thumb Reduction among Colobine Monkeys

    Thumb reduction is among the most important features distinguishing the African and Asian colobines from each other and from other Old World monkeys. In this study we demonstrate that the partial skeleton KNM-ER 4420 from Koobi Fora, Kenya, dated to 1.9 Ma and assigned to the Plio-Pleistocene colobine species Cercopithecoides williamsi, shows marked reduction of its first metacarpal relative to the medial metacarpals. Thus, KNM-ER 4420 is the first documented occurrence of cercopithecid pollical reduction in the fossil record. In the size of its first metacarpal relative to the medial metacarpals, C. williamsi is similar to extant African colobines, but different from cercopithecines, extant Asian colobines and the Late Miocene colobines Microcolobus and Mesopithecus. This feature clearly links the genus Cercopithecoides with the extant African colobine clade and makes it the first definitive African colobine in the fossil record. The postcranial adaptations to terrestriality in Cercopithecoides are most likely secondary, while ancestral colobinans (and colobines) were arboreal. Finally, the absence of any evidence for pollical reduction in Mesopithecus implies either independent thumb reduction in African and Asian colobines or multiple colobine dispersal events out of Africa.     Staphylococcus aureus sigma B-dependent emergence of small-colony variants and biofilm production following exposure to Pseudomonas aeruginosa 4-hydroxy-2-heptylquinoline-N-oxide

    <p>Abstract</p> <p>Background</p> <p><it>Staphylococcus aureus </it>and <it>Pseudomonas aeruginosa </it>are often found together in the airways of cystic fibrosis (CF) patients. It was previously shown that the <it>P. aeruginosa </it>exoproduct 4-hydroxy-2-heptylquinoline-<it>N-</it>oxide (HQNO) suppresses the growth of <it>S. aureus </it>and provokes the emergence of small-colony variants (SCVs). The presence of <it>S. aureus </it>SCVs as well as biofilms have both been associated with chronic infections in CF.</p> <p>Results</p> <p>We demonstrated that HQNO stimulates <it>S. aureus </it>to form a biofilm in association with the formation of SCVs. The emergence of SCVs and biofilm production under HQNO exposure was shown to be dependent on the activity of the stress- and colonization-related alternative sigma factor B (SigB). Analysis of gene expression revealed that exposure of a prototypical <it>S. aureus </it>strain to HQNO activates SigB, which was leading to an increase in the expression of the fibronectin-binding protein A and the biofilm-associated <it>sarA </it>genes. Conversely, the quorum sensing accessory gene regulator (<it>agr</it>) system and the α-hemolysin gene were repressed by HQNO. Experiments using culture supernatants from <it>P. aeruginosa </it>PAO1 and a double chamber co-culture model confirmed that <it>P. aeruginosa </it>stimulates biofilm formation and activates SigB in a <it>S. aureus </it>strain isolated from a CF patient. Furthermore, the supernatant from <it>P. aeruginosa </it>mutants unable to produce HQNO induced the production of biofilms by <it>S. aureus </it>to a lesser extent than the wild-type strain only in a <it>S. aureus </it>SigB-functional background.</p> <p>Conclusions</p> <p>These results suggest that <it>S. aureus </it>responds to HQNO from <it>P. aeruginosa </it>by forming SCVs and biofilms through SigB activation, a phenomenon that may contribute to the establishment of chronic infections in CF patients.</p

    The effect of real-time vibrotactile feedback delivered through an augmented fork on eating rate, satiation, and food intake.

    Eating rate is a basic determinant of appetite regulation, as people who eat more slowly feel sated earlier and eat less. Without assistance, eating rate is difficult to modify due to its automatic nature. In the current study, participants used an augmented fork that aimed to decelerate their rate of eating. A total of 114 participants were randomly assigned to the Feedback Condition (FC), in which they received vibrotactile feedback from their fork when eating too fast (i.e., taking more than one bite per 10 s), or a Non-Feedback Condition (NFC). Participants in the FC took fewer bites per minute than did those in the NFC. Participants in the FC also had a higher success ratio, indicating that they had significantly more bites outside the designated time interval of 10 s than did participants in the NFC. A slower eating rate, however, did not lead to a significant reduction in the amount of food consumed or level of satiation. These findings indicate that real-time vibrotactile feedback delivered through an augmented fork is capable of reducing eating rate, but there is no evidence from this study that this reduction in eating rate is translated into an increase in satiation or reduction in food consumption. Overall, this study shows that real-time vibrotactile feedback may be a viable tool in interventions that aim to reduce eating rate. The long-term effectiveness of this form of feedback on satiation and food consumption, however, awaits further investigation

    Early Pleistocene large mammals from Maka’amitalu, Hadar, lower Awash Valley, Ethiopia

    The Early Pleistocene was a critical time period in the evolution of eastern African mammal faunas, but fossil assemblages sampling this interval are poorly known from Ethiopia's Afar Depression. Field work by the Hadar Research Project in the Busidima Formation exposures (similar to 2.7-0.8 Ma) of Hadar in the lower Awash Valley, resulted in the recovery of an early Homo maxilla (A.L. 666-1) with associated stone tools and fauna from the Maka'amitalu basin in the 1990s. These assemblages are dated to similar to 2.35 Ma by the Bouroukie Tuff 3 (BKT-3). Continued work by the Hadar Research Project over the last two decades has greatly expanded the faunal collection. Here, we provide a comprehensive account of the Maka'amitalu large mammals (Artiodactyla, Carnivora, Perissodactyla, Primates, and Proboscidea) and discuss their paleoecological and biochronological significance. The size of the Maka'amitalu assemblage is small compared to those from the Hadar Formation (3.45-2.95 Ma) and Ledi-Geraru (2.8-2.6 Ma) but includes at least 20 taxa. Bovids, suids, and Theropithecus are common in terms of both species richness and abundance, whereas carnivorans, equids, and megaherbivores are rare. While the taxonomic composition of the Maka'amitalu fauna indicates significant species turnover from the Hadar Formation and Ledi-Geraru deposits, turnover seems to have occurred at a constant rate through time as taxonomic dissimilarity between adjacent fossil assemblages is strongly predicted by their age difference. A similar pattern characterizes functional ecological turnover, with only subtle changes in dietary proportions, body size proportions, and bovid abundances across the composite lower Awash sequence. Biochronological comparisons with other sites in eastern Africa suggest that the taxa recovered from the Maka'amitalu are broadly consistent with the reported age of the BKT-3 tuff.     Multiplicity of northern bright O-type stars with optical long baseline interferometry

    The study of the multiplicity of massive stars gives hints on their formation processes and their evolutionary paths, which are still not fully understood. Large separation binaries (>50 milliseconds of arc, mas) can be probed by adaptive-optics-assisted direct imaging and sparse aperture masking, while close binaries can be resolved by photometry and spectroscopy. However, optical long baseline interferometry is mandatory to establish the multiplicity of Galactic massive stars at the separation gap between 1 and 50 mas. In this paper, we aim to demonstrate the capability of the new interferometric instrument MIRC-X, located at the CHARA Array, to study the multiplicity of O-type stars and therefore probe the full range of separation for more than 120 massive stars (H<7.5 mag). We initiated a pilot survey of bright O-type stars (H<6.5mag) observable with MIRC-X. We observed 29 O-type stars, including two systems in average atmospheric conditions around a magnitude of H=7.5 mag. We systematically reduced the obtained data with the public reduction pipeline of the instrument. We analyzed the reduced data using the dedicated python software CANDID to detect companions. Out of these 29 systems, we resolved 19 companions in 17 different systems with angular separations between ~0.5 and 50 mas. This results in a multiplicity fraction fm=17/29=0.59+/-0.09, and an average number of companions fc=19/29=0.66+/-0.13. Those results are in agreement with the results of the SMASH+ survey in the Southern Hemisphere. Thirteen of these companions have been resolved for the first time, including the companion responsible for the nonthermal emission in Cyg OB2-5 A and the confirmation of the candidate companion of HD 47129 suggested by SMASH+.     High-content siRNA screening of the kinome identifies kinases involved in Alzheimer's disease-related tau hyperphosphorylation

    <p>Abstract</p> <p>Background</p> <p>Neurofibrillary tangles (NFT), a cardinal neuropathological feature of Alzheimer's disease (AD) that is highly correlated with synaptic loss and dementia severity, appear to be partly attributable to increased phosphorylation of the microtubule stabilizing protein tau at certain AD-related residues. Identifying the kinases involved in the pathologic phosphorylation of tau may provide targets at which to aim new AD-modifying treatments.</p> <p>Results</p> <p>We report results from a screen of 572 kinases in the human genome for effects on tau hyperphosphorylation using a loss of function, high-throughput RNAi approach. We confirm effects of three kinases from this screen, the eukaryotic translation initiation factor 2 α kinase 2 (EIF2AK2), the dual-specificity tyrosine-(Y)-phosphorylation regulated kinase 1A (DYRK1A), and the A-kinase anchor protein 13 (AKAP13) on tau phosphorylation at the 12E8 epitope (serine 262/serine 356). We provide evidence that EIF2AK2 effects may result from effects on tau protein expression, whereas DYRK1A and AKAP13 are likely more specifically involved in tau phosphorylation pathways.</p> <p>Conclusions</p> <p>These findings identify novel kinases that phosphorylate tau protein and provide a valuable reference data set describing the kinases involved in phosphorylating tau at an AD-relevant epitope.</p

    A Transient, Neuron-Wide Form of CREB-Mediated Long-Term Facilitation Can Be Stabilized at Specific Synapses by Local Protein Synthesis

    AbstractIn a culture system where a bifurcated Aplysia sensory neuron makes synapses with two motor neurons, repeated application of serotonin (5-HT) to one synapse produces a CREB-mediated, synapse-specific, long-term facilitation, which can be captured at the opposite synapse by a single pulse of 5-HT. Repeated pulses of 5-HT applied to the cell body of the sensory neuron produce a CREB-dependent, cell-wide facilitation, which, unlike synapse-specific facilitation, is not associated with growth and does not persist beyond 48 hr. Persistent facilitation and synapse-specific growth can be induced by a single pulse of 5-HT applied to a peripheral synapse. Thus, the short-term process initiated by a single pulse of 5-HT serves not only to produce transient facilitation, but also to mark and stabilize any synapse of the neuron for long-term facilitation by means of a covalent mark and rapamycin-sensitive local protein synthesis

    High-Resolution Melting System to Perform Multilocus Sequence Typing of Campylobacter jejuni

    Multi-locus sequence typing (MLST) has emerged as the state-of-the-art method for resolving bacterial population genetics but it is expensive and time consuming. We evaluated the potential of high resolution melting (HRM) to identify known MLST alleles of Campylobacter jejuni at reduced cost and time. Each MLST locus was amplified in two or three sub fragments, which were analyzed by HRM. The approach was investigated using 47 C. jejuni isolates, previously characterized by classical MLST, representing isolates from diverse environmental, animal and clinical sources and including the six most prevalent sequence types (ST) and the most frequent alleles. HRM was then applied to a validation set of 84 additional C. jejuni isolates from chickens; 92% of the alleles were resolved in 35 hours of laboratory time and the cost of reagents per isolate was 20comparedwith20 compared with 100 for sequence-based typing. HRM has the potential to complement sequence-based methods for resolving SNPs and to facilitate a wide range of genotyping studies
