    Timing the senses and sensing the time: Individual differences in subjective duration

    Although the experimental investigation of the perception of time dates back to around 1864 (Lejeune & Wearden, 2009) the reasons for variation between individuals are still poorly understood. Advancing in this area has been identified as one of the most important issues for the modern investigation of time perception (Hancock & Block, 2012). Although various individual differences in time perception have been identified, these are usually based on comparisons between different groups depending on characteristics such as age or gender; clinical conditions such as schizophrenia or autism; or induced differences such as temperature or pharmacology. In this thesis we seek to investigate whether, and how, intrinsic individual differences in sensory processing within the general population influence individual timing behaviour. This is accomplished over four experiments. The first seeks a relationship between the EEG alpha rhythm, which is strongly associated with visual and audio-visual temporal integration, and behaviour on four timing tasks; while trends are in the expected directions no significant relationship emerges. The second responds to this null result by seeking a relationship between audio-visual integration and estimated, sub-second, durations in a purely behavioural paradigm, in this case the results show a significant association. The final two experiments are concerned with individual differences in interoceptive accuracy (sensitivity to ones own bodily signals), and how these influence the effect of arousal on time judgement. The first experiment, using supra-second durations, finds no effects, but the second, using shorter durations (under 1200ms) and addressing some methodological concerns, does find a significant moderation, by interoception, of the effect of emotional arousal upon time. We conclude that this provides substantial evidence that exteroceptive and interoceptive primary sensory processes play a role in individual variation in timing tasks, a finding that provides many further avenues of investigation

    Individual differences in sensory integration predict differences in time perception and individual levels of schizotypy

    To interact functionally with our environment, our perception must locate events in time, including discerning whether sensory events are simultaneous. The Temporal Binding Window (TBW; the time window within which two stimuli tend to be integrated into one event) has been shown to relate to individual differences in perception, including schizotypy, but the relationship with subjective estimates of duration is unclear. We compare individual TBWs with individual differences in the filled duration illusion, exploiting differences in perception between empty and filled durations (the latter typically being perceived as longer). Schizotypy has been related to both these measures and is included to explore a potential link between these tasks and enduring perceptual differences. Results suggest that individuals with a narrower TBW make longer estimates for empty durations and demonstrate less variability in both conditions. Exploratory analysis of schizotypy data suggests a relationship with the TBW but is inconclusive regarding time perception

    CrowdQC+—A Quality-Control for Crowdsourced Air-Temperature Observations Enabling World-Wide Urban Climate Applications

    In recent years, the collection and utilisation of crowdsourced data has gained attention in atmospheric sciences and citizen weather stations (CWS), i.e., privately-owned weather stations whose owners share their data publicly via the internet, have become increasingly popular. This is particularly the case for cities, where traditional measurement networks are sparse. Rigorous quality control (QC) of CWS data is essential prior to any application. In this study, we present the QC package “CrowdQC+,” which identifies and removes faulty air-temperature (ta) data from crowdsourced CWS data sets, i.e., data from several tens to thousands of CWS. The package is a further development of the existing package “CrowdQC.” While QC levels and functionalities of the predecessor are kept, CrowdQC+ extends it to increase QC performance, enhance applicability, and increase user-friendliness. Firstly, two new QC levels are introduced. The first implements a spatial QC that mainly addresses radiation errors, the second a temporal correction of the data regarding sensor-response time. Secondly, new functionalities aim at making the package more flexible to apply to data sets of different lengths and sizes, enabling also near-real time application. Thirdly, additional helper functions increase user-friendliness of the package. As its predecessor, CrowdQC+ does not require reference meteorological data. The performance of the new package is tested with two 1-year data sets of CWS data from hundreds of “Netatmo” CWS in the cities of Amsterdam, Netherlands, and Toulouse, France. Quality-controlled data are compared with data from networks of professionally-operated weather stations (PRWS). Results show that the new package effectively removes faulty data from both data sets, leading to lower deviations between CWS and PRWS compared to its predecessor. It is further shown that CrowdQC+ leads to robust results for CWS networks of different sizes/densities. Further development of the package could include testing the suitability of CrowdQC+ for other variables than ta, such as air pressure or specific humidity, testing it on data sets from other background climates such as tropical or desert cities, and to incorporate added filter functionalities for further improvement.     Serological responses in humans to the smallpox vaccine LC16m8

    In response to potential bioterrorism with smallpox, members of the Japanese Self-Defense Forces were vaccinated with vaccinia virus (VACV) strain LC16m8, an attenuated smallpox vaccine derived from VACV strain Lister. The serological response induced by LC16m8 to four virion-surface proteins and the intracellular mature virus (IMV) and extracellular enveloped virus (EEV) was investigated. LC16m8 induced antibody response against the IMV protein A27 and the EEV protein A56. LC16m8 also induced IMV-neutralizing antibodies, but unlike the VACV strain Lister, did not induce either EEV-neutralizing antibody or antibody to EEV protein B5, except after revaccination. Given that B5 is the only target for EEV-neutralizing antibody and that neutralization of both IMV and EEV give optimal protection against orthopoxvirus challenge, these data suggest that immunity induced by LC16m8 might be less potent than that deriving from strain Lister. This potential disadvantage should be balanced against the advantage of the greater safety of LC16m8

    Inhibition of CCL3 abrogated precursor cell fusion and bone erosions in human osteoclast cultures and murine collagen-induced arthritis

    Objective Macrophage inflammatory protein 1-alpha (CCL3) is a chemokine that regulates macrophage trafficking to the inflamed joint. The agonistic effect of CCL3 on osteolytic lesions in patients with multiple myeloma is recognized; however, its role in skeletal damage during inflammatory arthritis has not been established. The aim of the study was to explore the role of osteoclast-associated CCL3 upon bone resorption, and to test its pharmacological blockade for protecting against bone pathology during inflammatory arthritis. Methods CCL3 production was studied during osteoclast differentiation from osteoclast precursor cells: human CD14-positive mononuclear cells. Mice with CIA were treated with an anti-CCL3 antibody. The effect of CCL3 blockade through mAb was studied through osteoclast number, cytokine production and bone resorption on ivory disks, and in vivo through CIA progression (clinical score, paw diameter, synovial inflammation and bone damage). Results Over time, CCL3 increased in parallel with the number of osteoclasts in culture. Anti-CCL3 treatment achieved a concentration-dependent inhibition of osteoclast fusion and reduced pit formation on ivory disks (P â©˝ 0.05). In CIA, anti-CCL3 treatment reduced joint damage and significantly decreased multinucleated tartrate-resistant acid phosphatase-positive osteoclasts and erosions in the wrists (P < 0.05) and elbows (P < 0.05), while also reducing joint erosions in the hind (P < 0.01) and fore paws (P < 0.01) as confirmed by X-ray. Conclusion Inhibition of osteoclast-associated CCL3 reduced osteoclast formation and function whilst attenuating arthritis-associated bone loss and controlling development of erosion in murine joints, thus uncoupling bone damage from inflammation. Our findings may help future innovations for the diagnosis and treatment of inflammatory arthritis

    Discovery and Synthesis of Caracolamide A, an Ion Channel Modulating Dichlorovinylidene Containing Phenethylamide from a Panamanian Marine Cyanobacterium cf. Symploca Species

    A recent untargeted metabolomics investigation into the chemical profile of 10 organic extracts from cf. Symploca spp. revealed several interesting chemical leads for further natural product drug discovery. Subsequent targetdirected isolation efforts with one of these, a Panamanian marine cyanobacterium cf. Symploca sp., yielded a phenethylamide metabolite that terminates in a relatively rare gemdichlorovinylidene moiety, caracolamide A (1), along with a known isotactic polymethoxy-1-alkene (2). Detailed NMR and HRESIMS analyses were used to determine the structures of these molecules, and compound 1 was confirmed by a threestep synthesis. Pure compound 1 was shown to have in vitro calcium influx and calcium channel oscillation modulatory activity when tested as low as 10 pM using cultured murine cortical neurons, but was not cytotoxic to NCI-H460 human non-small-cell lung cancer cells in vitro (IC50 > 10 ÎĽM).A recent untargeted metabolomics investigation into the chemical profile of 10 organic extracts from cf.     Mortuary Caves and the Dammar Trade in the Towuti-Routa Region, Sulawesi, in an Island Southeast Asian context

    Archaeological evidence from survey and cave excavation in the Towuti-Routa region of Sulawesi suggests the following sequence of late Holocene cultural change. Settled communities whose subsistence included an agricultural component had established themselves by the early centuries a.d. and began the use of caves for mortuary purposes. Extended inhumations are the oldest attested mortuary practice, overlapping in time with secondary burials in large earthenware jars dated to around a.d. 1000. The third, ethnohistorically described practice involved the surface disposal of the deceased, including the use of imported martavans for the elite, between approximately a.d. 1500 and 1900. This sequence of mortuary practices has not been documented elsewhere in Island Southeast Asia, although each practice has multiple parallels. The Towuti-Routa dammar trade, which was at its peak at the time of European contact, can perhaps account for the quantity of exotic items imported to the region but not the specifics of the mortuary practices.The Indonesian government’s permission for the research was granted by the Bureau of Research and Technology (Visa 278/SIP/FRP/SM/VII/2013).     Replication and shedding kinetics of infectious hematopoietic necrosis virus in juvenile rainbow trout

    Viral replication and shedding are key components of transmission and fitness, the kinetics of which are heavily dependent on virus, host, and environmental factors. To date, no studies have quantified the shedding kinetics of infectious hematopoietic necrosis virus (IHNV) in rainbow trout (Oncorhynchus mykiss), or how they are associated with replication, making it difficult to ascertain the transmission dynamics of this pathogen of high agricultural and conservation importance. Here, the replication and shedding kinetics of two M genogroup IHNV genotypes were examined in their naturally co-evolved rainbow trout host. Within host virus replication began rapidly, approaching maximum values by day 3 post-infection, after which viral load was maintained or gradually dropped through day 7. Host innate immune response measured as stimulation of Mx-1 gene expression generally followed within host viral loads. Shedding also began very quickly and peaked within 2 days, defining a generally uniform early peak period of shedding from 1 to 4 days after exposure to virus. This was followed by a post-peak period where shedding declined, such that the majority of fish were no longer shedding by day 12 post-infection. Despite similar kinetics, the average shedding rate over the course of infection was significantly lower in mixed compared to single genotype infections, suggesting a competition effect, however, this did not significantly impact the total amount of virus shed. The data also indicated that the duration of shedding, rather than peak amount of virus shed, was correlated with fish mortality. Generally, the majority of virus produced during infection appeared to be shed into the environment rather than maintained in the host, although there was more retention of within host virus during the post-peak period. Viral virulence was correlated with shedding, such that the more virulent of the two genotypes shed more total virus. This fundamental understanding of IHNV shedding kinetics and variation at the individual fish level could assist with management decisions about how to respond to disease outbreaks when they occur.     Measurement of in vitro cardiac deformation by means of 3D digital image correlation and ultrasound 2D speckle-tracking echocardiography

    Ultrasound-based 2D speckle-tracking echocardiography (US-2D-STE) is increasingly used to assess the functionality of the heart. In particular, the analysis of cardiac strain plays an important role in the identification of several cardiovascular diseases. However, this imaging technique presents some limitations associated with its operating principle that result in low accuracy and reproducibility of the measurement. In this study, an experimental framework for multimodal strain imaging in an in vitro porcine heart was developed. Specifically, the aim of this work was to analyse displacement and strain in the heart by means of 3D digital image correlation (3D-DIC) and US-2D-STE. Over a single cardiac cycle, displacement values obtained from the two techniques were in strong correlation, although systematically larger displacements were observed with 3D-DIC. Notwithstanding an absolute comparison of the strain measurements was not possible to achieve between the two methods, maximum principal strain directions computed with 3D-DIC were consistent with the longitudinal and circumferential strain distribution measured with US-2D-STE. 3D-DIC confirmed its high repeatability in quantifying displacement and strain over multiple cardiac cycles, unlike US-2D-STE which is affected by accumulated errors over time (i.e. drift). To conclude, this study demonstrates the potential of 3D-DIC to perform dynamic measurement of displacement and strain during heart deformations and supports future applications of this method in ex vivo beating heart platforms, which replicate more fully the complex contraction of the heart

    The World Urban Database and Access Portal Tools (WUDAPT) is a community initiative to collect worldwide data on urban form (i.e., morphology, materials) and function (i.e., use and metabolism). This is achieved through crowdsourcing, which we define here as the collection of data by a bounded crowd, composed of students. In this process, training data for the classification of urban structures into Local Climate Zones (LCZ) are obtained, which are, like most volunteered geographic information initiatives, of unknown quality. In this study, we investigated the quality of 94 crowdsourced training datasets for ten cities, generated by 119 students from six universities. The results showed large discrepancies and the resulting LCZ maps were mostly of poor to moderate quality. This was due to general difficulties in the human interpretation of the (urban) landscape and in the understanding of the LCZ scheme. However, the quality of the LCZ maps improved with the number of training data revisions. As evidence for the wisdom of the crowd, improvements of up to 20% in overall accuracy were found when multiple training datasets were used together to create a single LCZ map. This improvement was greatest for small training datasets, saturating at about ten to fifteen sets
