Quality assurance of the solar UV network in the Antarctic
Measuring ultraviolet radiation in the Antarctic region, where weather conditions are extremely challenging, is a demanding task. Proper quality control of the measurements and quality assurance of the data, which are the basis of all scientific use of data, has to be especially well planned and executed. In this paper we show the importance of proper quality assurance and describe the methods used to successfully operate the NILU-UV multichannel radiometers of the Antarctic network stations at Ushuaia, 54S, and Marambio, 64S. According to our experience, even though multichannel instruments are supposed to be rather stable as a function of time, severe drifts can occur in the sensitivity of the channels under these harsh conditions. During 2000–2003 the biggest drifts were 35%, both at Ushuaia and Marambio, with the sensitivity of the channels dropping at different rates. Without proper corrections in the data, this would have seriously affected the calculated UV dose rates. As part of the quality assurance of the network a traveling reference NILU-UV, which was found to be stable, was used to transfer the desired irradiance scale to the site NILU-UV data. Relative lamp tests were used to monitor the stability of the instruments. Each site NILU-UV was scaled channel by channel to the traveling reference by performing solar comparisons. The method of scaling each channel separately was found to be successful, even though the differences between the raw data of the site NILU-UV and the reference instruments were, before the data correction, as much as 40%. After the correction, the mean ratios of erythemally weighted UV dose rates measured during the solar comparisons in 2000–2003 between the reference NILU-UV and the site NILU-UV were 1.007 ± 0.011 and 1.012 ± 0.012 for Ushuaia and Marambio, respectively, when the solar zenith angle varied up to 80. These results make possible the scientific use of NILU-UV data measured simultaneously at quite different locations, e.g., the Antarctic and Arctic, and the method presented is also practicable for other multichannel radiometer networks.S, and Marambio, 64S. MIDTAL (Microarrays for the Detection of Toxic Algae)
Microalgae in marine and brackish waters of Europe regularly cause harmful effects, considered from the human perspective, in that they cause economic damage to fisheries and tourism. Cyanobacteria cause similar problems in freshwaters. These episodes encompass a broad range of phenomena collectively referred to as harmful algal blooms (HABs). For adequate management of these phenomena, monitoring of microalgae is required. However, present day monitoring is time consuming and based on morphology as determined by light microscopy, which may be insufficient to give definitive species and toxin attribution. In the European Union (EU) FP7 project MIDTAL (microarrays for the detection of toxic algae), we will first target rapid species identification using rRNA genes. The variable regions of the rRNA genes can be used for probe design to recognize species or even strains. Second, a toxin based microarray will be developed that includes antibody reactions to specific toxins produced by these microalgae because even when cell numbers are low, toxins can be present and can accumulate in the shellfish. Microarrays are the state of the art technology in molecular biology for the processing of bulk samples for detection of target RNA/DNA sequence. Existing rRNA probes and antibodies for toxic algal species/strains and their toxins will be adapted and optimized for microarray use. The purpose of MIDTAL is to support the common fisheries policy and to aid the national monitoring agencies by providing new rapid tools for the identification of toxic algae and their toxins so they can comply with EU directive 91/1491/CEE to monitor for toxic algae, and reduce the need for the mouse bioassay

Fatal poisoning in drug addicts in the Nordic countries in 2017
This study is the seventh report on fatal poisonings among drug addicts in the Nordic countries. In this report, we analyse data from the five Nordic countries: Denmark, Finland, Iceland, Norway and Sweden. Data on gender, number of deaths, places of deaths, age, main intoxicants and substances detected in blood were recorded to obtain national and comparable Nordic data, and to allow comparison with earlier studies conducted in 1984, 1991, 1997, 2002, 2007 and 2012. The death rate (number of deaths per 100,000 inhabitants) was highest in Iceland (6.58) followed closely by Sweden (6.46) and then lowest in Denmark (4.29). The death rate increased in Finland (5.84), Iceland and Sweden and decreased in Denmark compared to earlier studies. The death rate in Norway, which has decreased since 2002, has stabilised around 5.7 as of 2017. Women accounted for 7-23% of the fatal poisonings. The percentage was lowest in Iceland and highest in Finland and Norway. The age range was 14-70 years. The median age (41 years) was highest in Denmark and Norway. The other countries had a median age between 33 and 35 years. Opioids were the main cause of death. Methadone remained the main intoxicant in Denmark, while heroin/morphine was still the main intoxicant in Norway, as was buprenorphine in Finland. However, the picture has changed in Sweden compared to 2012, where heroin/morphine caused most deaths in 2017. Sweden also experienced the highest number of deaths from fentanyl analogues (67 deaths) and buprenorphine (61 deaths). Deaths from fentanyl analogues also occurred in Denmark, Finland and Norway, but to a smaller extent. Over the years, the proportion of opioid deaths has decreased in all countries except Sweden, which has experienced an increase. This decline has been replaced by deaths from CNS stimulants like cocaine, amphetamine and methylenedioxymethamphetamine (MDMA). Cocaine deaths have occurred in all countries but most frequently in Denmark. MDMA deaths have increased in all countries but mostly in Finland. Poly-drug use was widespread, as seen in the earlier studies. The median number of detected drugs per case varied from 4-6. Heroin/morphine, methadone, buprenorphine, cocaine, amphetamine, methamphetamine, MDMA, tetrahydrocannabinol (THC) and benzodiazepines were frequently detected. Pregabalin and gabapentin were detected in all countries, especially pregabalin, which was detected in 42% of the Finnish cases. TMFunction: database for functional residues in membrane proteins
We have developed the database TMFunction, which is a collection of more than 2900 experimentally observed functional residues in membrane proteins. Each entry includes the numerical values for the parameters IC50 (measure of the effectiveness of a compound in inhibiting biological function), Vmax (maximal velocity of transport), relative activity of mutants with respect to wild-type protein, binding affinity, dissociation constant, etc., which are important for understanding the sequence–structure–function relationship of membrane proteins. In addition, we have provided information about name and source of the protein, Uniprot and Protein Data Bank codes, mutational and literature information. Furthermore, TMFunction is linked to related databases and other resources. We have set up a web interface with different search and display options so that users have the ability to get the data in several ways. Loss of fshr prevents testicular maturation in Atlantic Salmon (Salmo salar L.).
Early puberty poses a significant challenge for male Atlantic salmon in aquaculture due to its negative impact on growth and welfare. The regulation of puberty in vertebrates involves 2 key reproductive hormones: follicle-stimulating hormone (FSH) and luteinizing hormone (LH) and their gonadal receptors. In male mice lacking FSH receptor, testes size is reduced, but fertility is maintained, while medaka and zebrafish with a disrupted fshr gene exhibit near normal testis size and fertility. In these fishes both Fsh and Lh are present during puberty and Lh may rescue fertility, while in salmonid fish only Fsh is present in the circulation during puberty. Using CRISPR-Cas9, we produced crispants with a high prevalence of fshr mutations at the target site, which remained fertile, although more than half showed a testis development deviating from wild-type (wt) males
Hypertension and Smoking Are Associated With Reduced Regional Left Ventricular Function in Asymptomatic Individuals The Multi-Ethnic Study of Atherosclerosis
ObjectivesThis study sought to test the hypothesis that reduced regional left ventricular (LV) function is associated with traditional risk factors including hypertension, hypercholesterolemia, and smoking in asymptomatic individuals.BackgroundCoronary artery disease is the main etiology of congestive heart failure in the U.S. and Europe. However, the relationship between risk factors for coronary artery disease and decreased myocardial function has not been studied systematically in asymptomatic individuals.MethodsThe Multi-Ethnic Study of Atherosclerosis (MESA) is a cohort study designed to investigate the nature of atherosclerosis in asymptomatic individuals. A total of 1,184 participants (45 to 84 years old) underwent tagged cardiac magnetic resonance imaging. Regional LV function was quantified by analyzing peak systolic circumferential strain (Ecc) in regions corresponding to the left anterior descending (LAD), circumflex (LCX), and right coronary (RCA) territories. The association between risk factors and strains was studied using multiple linear regression.ResultsHigher diastolic blood pressure (DBP) was associated with lower Ecc (p ≤ 0.002). The Ecc’s in the LAD territory of participants with DBP <80, 80 to 84, 85 to 89, and ≥90 mm Hg were −15.6%, −14.8%, −14.2%, and −13.7%, respectively (p < 0.001). Similar results were documented in other territories and after multivariable analysis. Smokers had lower Ecc in the LAD and RCA regions compared with nonsmokers. Furthermore, dose response relationship between cigarette consumption measured in pack-years and regional LV dysfunction by Ecc was noted (p ≤ 0.01 in LAD and RCA territories). Finally, combined diastolic hypertension and smoking was associated with a greater reduction of regional LV function.ConclusionsHigher diastolic blood pressure and smoking are associated with decreased regional LV function in asymptomatic individuals

Field testing for toxic algae with a microarray: initial results from the MIDTAL project
One of the key tasks in the project MIDTAL (MIcroarrays for the Detection of Toxic ALgae) is to demonstrate the applicability of microarrays to monitor harmful algae across a broad range of ecological niches and toxic species responsible for harmful algal events. Water samples are collected from a series of sites used in national phytoplankton and biotoxin monitoring programmes across Europe. The samples are filtered; the rRNA is extracted, labelled with a fluorescent dye and applied to a microarray chip. The signal intensity from >120 probes previously spotted on the chip is measured and analysed. Preliminary results comparing microarray signal intensities with actual field counts are presented
