    Genomic diversity of Oenococcus oeni from different winemaking regions of Portugal

    Oenococcus oeni is an alcohol-tolerant, acidophilic lactic acid bacterium that plays an important role in the elaboration of wine. It is often added as a starter culture to carry out malolactic conversion. Given the economic importance of this reaction, the taxonomic structure of this species has been studied in detail. In the present work, phenotypic and molecular approaches were used to identify 121 lactic acid bacteria strains isolated from the wines of three winemaking regions of Portugal. The strains were differentiated at the genomic level by M13-PCR fingerprinting. Twenty-seven genomic clusters represented by two or more isolates and 21 single-member clusters, based on an 85% similarity level, were recognized by hierarchic numerical analysis. M13-PCR fingerprinting patterns revealed a high level of intraspecific genomic diversity in O. oeni. Moreover, this diversity could be partitioned according to the geographical origin of the isolates. Thus, M13-PCR fingerprint analysis may be an appropriate methodology to study the O. oeni ecology of wine during malolactic fermentation as well as to trace new malolactic starter cultures and evaluate their dominance over the native microbiota

    Effects of increasing doses of condensed tannins extract from Cistus ladanifer L. on In Vitro ruminal fermentation and biohydrogenation

    Research Areas: Agriculture ; Veterinary Sciences ; ZoologyCistus ladanifer (rockrose) is a perennial shrub quite abundant in the Mediterranean region, and it is a rich source in secondary compounds such as condensed tannins (CTs). Condensed tannins from C. ladanifer were able to change the ruminal biohydrogenation (BH), increasing the t11–18:1 and c9,t11–18:2 production. However, the adequate conditions of the C. ladanifer CTs used to optimize the production of t11–18:1 and c9,t11–18:2 is not yet known. Thus, we tested the effect of increasing the doses of C. ladanifer CT extract (0, 25, 50, 75 and 100 g/kg dry matter (DM)) on in vitro rumen BH. Five in vitro batch incubations replicates were conducted using an oil supplemented high-concentrate substrate, incubated for 24 h with 6 mL of buffered ruminal fluid. Volatile fatty acids (VFAs) and long chain fatty acids (FA) were analyzed at 0 h and 24 h, and BH of c9–18:1, c9, c12–18:2 and c9, c12, c15–18:3, and BH products yield were computed. Increasing doses of C. ladanifer CTs led to a moderate linear decrease (p < 0.001) of the VFA production (a reduction of 27% with the highest dose compared to control). The disappearance of c9–18:1 and c9,c12–18:2 as well as the production of t11–18:1 and c9, t11:18:2 was not affected by increasing doses of C. ladanifer CTs, and only the disappearance of c9, c12, c15–18:3 suffered a mild linear decrease (a reduction of 24% with the highest dose compared to control). Nevertheless, increasing the C. ladanifer CT dose led to a strong depression of microbial odd and branched fatty acids and of dimethyl acetals production (less than 65% with the highest dose compared to control), which indicates that microbial growth was more inhibited than fermentative and biohydrogenation activities, in a possible adaptative response of microbial population to stress induced to CTs and polyunsaturated fatty acids. The ability of C. ladanifer to modulate the ruminal BH was not verified in the current in vitro experimental conditions, emphasizing the inconsistent BH response to CTs and highlighting the need to continue seeking the optimal conditions for using CTs to improve the fatty acid profile of ruminant fat.info:eu-repo/semantics/publishedVersio

    Anti-inflammatory and immunoregulatory action of sesquiterpene lactones

    Sesquiterpene lactones (SL), characterized by their high prevalence in the Asteraceae family, are one of the major groups of secondary metabolites found in plants. Researchers from distinct research fields, including pharmacology, medicine, and agriculture, are interested in their biological potential. With new SL discovered in the last years, new biological activities have been tested, different action mechanisms (synergistic and/or antagonistic effects), as well as molecular structure–activity relationships described. The review identifies the main sesquiterpene lactones with interconnections between immune responses and anti-inflammatory actions, within different cellular models as well in in vivo studies. Bioaccessibility and bioavailability, as well as molecular structure–activity relationships are addressed. Additionally, plant metabolic engineering, and the impact of sesquiterpene lactone extraction methodologies are presented, with the perspective of biological activity enhancement. Sesquiterpene lactones derivatives are also addressed.     Fatores de risco para mortalidade em idosos

    OBJECTIVE: The risk factors for diseases and premature deaths are important in drawing up preventive measures. This study had the aim of analyzing the risk factors for death among elderly people. METHODS: This study was carried out among participants in the SABE (Health, Wellbeing and Aging) study, in the city of São Paulo in 2000. Interviews were conducted with 2,143 elderly people (60 years old or over), using a standardized questionnaire for the SABE study. The sample was obtained from census tracts, in two stages, with replacements and with probability proportional to the population, and with supplementation of the sample of people aged 75 years or over. The final data were weighted so that they could be expanded. Between the two data collection times, there were 38 deaths which comprised study sample. Logistic regression was utilized for the data analysis, with a significance level of 5%. RESULTS: The risk factors found were: locomotion difficulty, advanced age, male gender, self-assessment of health as "bad" and difficulty in going to the bathroom, OR=3.15; 2.93; 2.90; 2.69 and 2.51, respectively. CONCLUSIONS: The results may contribute towards the adoption of preventive measures for elderly people, with the aim of diminishing the expected number of fatal outcomes.OBJETIVO: Os fatores de risco para as doenças e mortes prematuras são importantes para a elaboração de condutas preventivas. O objetivo do trabalho foi analisar os fatores de risco para óbito entre idosos. MÉTODOS: O estudo foi realizado no Município de São Paulo, em participantes do estudo SABE (Saúde, Bem-estar e Envelhecimento), em 2000. Foram entrevistados 2.143 indivíduos com 60 anos ou mais, utilizando-se o questionário padronizado do estudo SABE. A amostra foi obtida de setores censitários em dois estágios, com reposição e com probabilidade proporcional à população, com complementação da amostra de pessoas de 75 anos ou mais. Os dados finais foram ponderados para poderem ser expandidos. Entre os dois períodos de coleta de dados ocorreram 38 óbitos que compuseram a amostra do estudo. Para a análise dos dados, utilizou-se regressão logística, com nível de significância de 5%. RESULTADOS: Os fatores de risco encontrados foram: dificuldade de locomoção, idade avançada, gênero masculino, auto-avaliação de saúde como "má" e dificuldade para ir ao banheiro, OR=3,15; 2,93; 2,90; 2,69 e 2,51, respectivamente. CONCLUSÕES: Os resultados podem contribuir para a adoção de medidas preventivas para os idosos, visando à diminuição de desfechos fatais antecipados

    Análise da prevalência e fatores associados à incontinência urinária entre idosos do Município de São Paulo, Brasil: Estudo SABE (Saúde, Bem-Estar e Envelhecimento)

    To investigate the prevalence of urinary incontinence among elderly people living in São Paulo, Brazil and their associated risk factors. The Pan-American Health Organization and World Health Organization coordinated a multicenter study named Health, Wellbeing and Aging (SABE Study) in elderly people (over 60 years old) living in seven countries in Latin America and the Caribbean. In Brazil, the study was carried out in São Paulo in the year 2000. The total Brazilian sample included 2,143 people. The prevalence of self reported urinary incontinence was 11.8% among men and 26.2% for women. It was verified that among those reporting urinary incontinence, 37% also reported stroke and 34% depression. It was found that the greater the dependence that the elderly people presented, the greater the prevalence of urinary incontinence. The associated factors found were depression (odds ratio = 2.49), female (2.42), advanced age (2.35), important functional limitation (2.01). Urinary incontinence is a highly prevalent symptom among the elderly population of the municipality of São Paulo, especially among women. The adoption of preventive measures can reduce the negative effects of urinary incontinence.Investigar a prevalência de incontinência urinária entre idosos de São Paulo, Brasil, e fatores associados e de risco. A Organização Pan-Americana da Saúde e a Organização Mundial da Saúde coordenaram estudo multicêntrico denominado Saúde, Bem-Estar e Envelhecimento (Estudo SABE) em pessoas idosas (60 anos ou mais) que vivem em sete países da América Latina e Caribe. No Brasil, o estudo populacional foi realizado no Município de São Paulo no ano 2000. A amostra total brasileira foi de 2.143 pessoas. A prevalência da incontinência urinária auto-referida foi de 11,8% entre homens e 26,2% entre mulheres. Verificou-se que entre aqueles que relataram incontinência urinária, 37% também relataram acidente vascular cerebral e 34%, depressão. Observou-se que quanto maior a dependência do idoso, maior era a prevalência de incontinência urinária. Os fatores associados encontrados foram depressão (OR = 2,49), sexo feminino (2,42), idade avançada (2,35), limitação funcional (2,01). Incontinência urinária é um sintoma altamente prevalente entre a população idosa do Município de São Paulo, especialmente entre as mulheres. A adoção de medidas preventivas pode reduzir os efeitos negativos da incontinência urinária

    Insight into the role of facultative bacteria stimulated by micro-aeration in continuous bioreactors converting LCFA to methane

    Conversion of unsaturated long chain fatty acids (LCFA) to methane in continuous bioreactors is not fully understood. Palmitate (C16:0) often accumulates during oleate (C18:1) biodegradation in methanogenic bioreactors, and the reason why this happens and which microorganisms catalyze this reaction remains unknown. Facultative anaerobic bacteria are frequently found in continuous reactors operated at high LCFA loads, but their function is unclear. To get more insight on the role of these bacteria, LCFA conversion was studied under microaerophilic conditions. For that, we compared bioreactors treating oleate-based wastewater (organic loading rates of 1 and 3 kg COD m-3 d-1), operated under different redox conditions (strictly anaerobic-AnR, -350 mV; microaerophilic-MaR, -250 mV). At the higher load, palmitate accumulated 7 times more in the MaR, where facultative anaerobes were more abundant, and only the biomass from this reactor could recover the methanogenic activity after a transient inhibition. In a second experiment, the abundance of facultative anaerobic bacteria, particularly Pseudomonas spp. (from which two strains were isolated), was strongly correlated (p<0.05) with palmitate-to-total LCFA percentage in the biofilm formed in a continuous plug flow reactor fed with very high loads of oleate. This work strongly suggests that micro-aeration stimulates the development of facultative bacteria that are critical for achieving LCFA conversion to methane in continuous bioreactors.     Novel insights for permeant lead structures through in vitro skin diffusion assays of Prunus lusitanica L., the Portugal Laurel

    As a contribution for the generation of libraries in which a natural product (NP) is used as the guiding structure, this work sought to investigate molecular features of triterpenes as deliver leads to cross the stratum corneum at a significant rate. Seeking a bioguided investigation of the dermocosmetic lead-like potential of triterpenes in Prunus lusitanica L., various extracts were obtained by two different methods (Soxhlet extractor and Accelerated Solvent Extraction-ASE) and analyzed by GC–MS and NMR. In vitro assays were conducted to quantify the friedelin 1 and crude plant extract permeation through a membrane of polydimethylsiloxane (PDMS), as well as their skin penetration enhancement capacity using two model molecules, caffeine 19 and ibuprofen 20. Friedelin 1 was identified as the major component (16–77%, GC) with isolated yield of 51% w/w (94%, GC) from Soxhlet residue (1.7% p/p) of the dried aerial parts of the plant harvested when in early flowering stage. Friedelin 1 promoted the penetration of the lipophilic molecule 20, however, it did not influence the permeation of the hydrophilic permeant 20. On the other hand, the crude extract acted as a retardant of the penetration of both substances. Molecular characteristics for the applicability of P. lusitanica L. in the development of dermocosmetics, as well as a new potential use for friedelin 1 in particular, are demonstrated. Probable mechanisms for chemical penetration enhancement using triterpenes as models for transdermal administration are herein discussed

    Pittosporum Undulatum and Hedychium Gardnerianum: nutritive value and secondary metabolites on cattle reproductive performances

    Hedychium gardnerianum (HG) and Pittospporum undulatum (PU) are invasive plants all over the world, being in the Azores supplied to cattle on periods of shortage food. As these plants produce secondary metabolites, including a diverse range of phytochemicals compounds, the aim of the presente study is to identify how these metabolites can be related to animal’s reproductive performances. For such purpose, plants were harvested on winter, compounds extracted by method of decoction and analysed by combination of liquid chromatography and mass spectrometry as well as highperformance liquid chromatography. For nutritive evaluations, Van Soest and Weende methodologies were used. In HG quercetin-3, 4'-di-O-betaglucopyranoside, myricetin rhamnoside, quercetin rhamnoside, and gibberellin A1 and A8 were identified, while for PU were found cafeic acid derivatives, including dicaffeoylquinic acid and caffeoylquinic acid. In nutritional terms, these plants can be considered as poor, presenting percentages of dry matter (DM%) of 16.34% and 40.39%, respectively for HG and PU. Values for ash 10.4%, crude protein (CP) 7.75%, neutral detergent fiber (NDF) 64.5, acid detergent fiber (ADF) 34.69%, acid detergent lignin (ADL) 3.47% and ether extract (EE) 2.03% were found for HG. For PU values were ash 6.64%, CP 6.11%, NDF 43.84%, acid ADF 35.57%, ADL 3.56% and EE 2.71%. This study clearly indicated that, besides their low nutritive values, these plants can be used to feed ruminants, especially when pasture lacks. Nevertheless, as some compounds, namely the caffeoylquinic and dicaffeoylquinic acids, are known to be associated to physiological reproductive mechanisms, one could speculate that these compounds can be directly or indirectly associated to reproductive performances in bovine fed with these plants

    Gestão curricular como prestação de serviço educativo

    Procede-se neste estudo à análise dos processos de gestão do currículo na escola, tais como descritos e avaliados nos 289 relatórios de avaliação externa de escolas da região norte de Portugal, produzidos entre os anos de 2011 e 2017. Os dados indiciam graves fragilidades ao nível da gestão do currículo levada a cabo pelas escolas, permitindo a inferência de que o currículo não é ainda um projeto que as escolas concebam, a partir do currículo nacional. Os índices de insucesso escolar, a indisciplina nas aulas, as dificuldades com as famílias e o desconforto dos professores comprovam que a rigidez curricular e organizacional está na mais completa contradição com a acentuada heterogeneidade discente, tornando um desenvolvimento curricular fundado no paradigma racional-tecnológico. Face à complexidade acrescida da escola para todos, o currículo nacional precisa de ser, ao nível da escola, reconcetualizado e reconstruído, para se tornar acessível aos alunos e para que estes reconheçam a sua relevância. Contextualizar, integrar, adequar e diferenciar o currículo constituem o conjunto de decisões curriculares a serem assumidas pelos professores, individual e coletivamente. Não obstante, os dados indicam que, no essencial, impera a ditadura do currículo prescrito e que a gestão curricular de escola é, ainda incipiente. Os maiores avanços ter-se-ão produzido no âmbito das medidas de recuperação e apoio aos alunos com atrasos e/ou dificuldades de aprendizagem. As escolas parecem não ter consciencializado ainda que o currículo real que desenvolvem é um dos principais fatores desse insucesso e dessas dificuldades e atrasos. E, por isso, lançam mão de uma estratégia remediativa quando podiam minimizar o fenómeno através duma estratégia preventiva: conceber um currículo real contextualizado, adequado e articulado.info:eu-repo/semantics/publishedVersio
