    Collagen-an important fish allergen for improved diagnosis

    Background Fish collagen is widely used in medicine, cosmetics, and the food industry. However, its clinical relevance as an allergen is not fully appreciated. This is likely due to collagen insolubility in neutral aqueous solutions, leading to low abundance in commercially available in vitro and skin prick tests for fish allergy. Objective To investigate the relevance of fish collagen as an allergen in a large patient population (n = 101). Methods Acid-soluble collagen type I was extracted from muscle and skin of Atlantic salmon, barramundi, and yellowfin tuna. IgE binding to collagen was analyzed by ELISA for 101 fish-allergic patients. Collagen-sensitized patients' sera were tested for IgE binding to parvalbumin from the same fish species. IgE cross-linking was analyzed by rat basophil leukemia assay and basophil activation test. Protein identities were confirmed by mass spectrometry. Results Purified fish collagen contained type I α1 and α2 chains and their multimers. Twenty-one of 101 patients (21%) were sensitized to collagen. Eight collagen-sensitized patients demonstrated absence of parvalbumin-specific IgE to some fish species. Collagen induced functional IgE cross-linking, as shown by rat basophil leukemia assay performed using 6 patients' sera, and basophil activation test using fresh blood from 1 patient. Collagen type I α chains from barramundi and Atlantic salmon were registered at www.allergen.org as Lat c 6 and Sal s 6, respectively. Conclusions IgE sensitization and IgE cross-linking capacity of fish collagen were demonstrated in fish-allergic patients. Inclusion of relevant collagen allergens in routine diagnosis is indicated to improve the capacity to accurately diagnose fish allergy

    The Light Dark Matter eXperiment (LDMX) is an electron-beam fixed-target experiment designed to achieve comprehensive model independent sensitivity to dark matter particles in the sub-GeV mass region. An upgrade to the LCLS-II accelerator will increase the beam energy available to LDMX from 4 to 8 GeV. Using detailed GEANT4-based simulations, we investigate the effect of the increased beam energy on the capabilities to separate signal and background, and demonstrate that the veto methodology developed for 4 GeV successfully rejects photon-induced backgrounds for at least 2Ă—10142\times10^{14} electrons on target at 8 GeV.Comment: 28 pages, 20 figures; corrected author lis

    Why political ontology must be experimentalized, On ecoshowhomes as devices of participation

    This paper contributes to debates about the ontological turn and its implications for democracy by proposing an experimental understanding of political ontology. It discusses why the shift from epistemology to ontology in STS has proved inconclusive for the study of politics and democracy: the politics of non-humans has been assumed to operate on a different level from that of politics and democracy understood as institutional and public forms. I distinguish between three different understandings of political ontology: theoretical, empirical and experimental. Each of these implies a different approach to the problem that non-humans pose for democracy. Theoretical ontology proposes to solve it by conceptual means, while empirical ontology renders it manageable by assuming a problematic analytic separation between constituting and constituted ontology. This paper makes the case for the third approach, experimental ontology, by analysing an empirical site, that of the ecoshowhome. In this setting, material entities are deliberately invested with moral and political capacities. As such, ecoshowhomes help to clarify two main features of experimental political ontology: 1) ontological work is here not so much relocated from theory to empirical practice, but distributed among actors and entities involved in them, and 2) normative variability does not just pertain to the enactment of things, but can be conceived of as internal to political objects. From these two features of experimental ontology something follows for democracy as an ontological problem. This problem does not dissolve in empirical settings, but these settings make possible its articulation by experimental means

    A deep learning system accurately classifies primary and metastatic cancers using passenger mutation patterns.

    In cancer, the primary tumour's organ of origin and histopathology are the strongest determinants of its clinical behaviour, but in 3% of cases a patient presents with a metastatic tumour and no obvious primary. Here, as part of the ICGC/TCGA Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium, we train a deep learning classifier to predict cancer type based on patterns of somatic passenger mutations detected in whole genome sequencing (WGS) of 2606 tumours representing 24 common cancer types produced by the PCAWG Consortium. Our classifier achieves an accuracy of 91% on held-out tumor samples and 88% and 83% respectively on independent primary and metastatic samples, roughly double the accuracy of trained pathologists when presented with a metastatic tumour without knowledge of the primary. Surprisingly, adding information on driver mutations reduced accuracy. Our results have clinical applicability, underscore how patterns of somatic passenger mutations encode the state of the cell of origin, and can inform future strategies to detect the source of circulating tumour DNA

    Foodservice Composting Crowds out Consumer Food Waste Reduction Behavior in a Dining Experiment

    Pressure mounts to address food waste, which deprives hungry people of needed nutrition, depletes resources used to produce food, and accounts for substantial greenhouse gas emissions during production, distribution and disposal. Composting, and other food waste recycling technologies that divert food waste from landfills, mitigate the environmental damages of food waste disposal and grow in popularity. We explore whether consumer knowledge that the environmental damage created by their food waste will be mitigated undermines personal food waste reduction behavior. Subjects in a dining situation are randomly assigned whether or not they receive information about the negative effects of landfilling food waste and whether they are told that uneaten food from the study will be composted or landfilled. We find that providing information about the negative effects of food waste in landfills significantly reduces both the propensity to create any food waste and the total amount of solid food waste created when compared to control subjects. However, if subjects are also informed that food waste from the study will be composted, the propensity to create food waste and the amount of solid food waste generated is similar to control situation which features neither a reduction nor a recycling policy. This suggests a crowding out effect or informational rebound effect in which promoting policies that mitigate the environmental damages of food waste may unintentionally undermine policies meant to encourage individual consumer food waste reduction. We discuss key policy implications as well as several limitations of our experimental setting and analysis

    Food waste declined more in rural Chinese households with livestock

    © 2020 Elsevier Ltd The ongoing evolution of food waste trends and the intensification of livestock systems in developing economies play a critical role in shaping global sustainability. We explore the linkage between food waste and livestock systems at the household level and how this connection changed in China during the 1990s and 2000s when market liberalization was followed by policies that led to intensification of livestock production. We find the amount of food waste in all rural households declined significantly over this period. Households with livestock, which often use uneaten food for animal feed, created about 75% more food waste than other rural households at the beginning of this period, but experienced significantly greater reductions in food waste and significantly greater increases in animal protein consumption over the study period. We postulate that intensified livestock production led to less uneaten food being used as animal feed and, in response, led to more efficient household consumption including less discarded food. We reject the separability of household livestock production and consumption decisions prior to the onset of livestock intensification, but cannot not reject separability thereafter. We end by discussing the implications of livestock intensification for overall food system sustainability

    Household Food Waste: Multivariate Regression and Principal Components Analyses of Awareness and Attitudes among U.S. Consumers

    <div><p>We estimate models of consumer food waste awareness and attitudes using responses from a national survey of U.S. residents. Our models are interpreted through the lens of several theories that describe how pro-social behaviors relate to awareness, attitudes and opinions. Our analysis of patterns among respondents’ food waste attitudes yields a model with three principal components: one that represents perceived practical benefits households may lose if food waste were reduced, one that represents the guilt associated with food waste, and one that represents whether households feel they could be doing more to reduce food waste. We find our respondents express significant agreement that some perceived practical benefits are ascribed to throwing away uneaten food, e.g., nearly 70% of respondents agree that throwing away food after the package date has passed reduces the odds of foodborne illness, while nearly 60% agree that some food waste is necessary to ensure meals taste fresh. We identify that these attitudinal responses significantly load onto a single principal component that may represent a key attitudinal construct useful for policy guidance. Further, multivariate regression analysis reveals a significant positive association between the strength of this component and household income, suggesting that higher income households most strongly agree with statements that link throwing away uneaten food to perceived private benefits.</p></div

    Some issues in the ethics of food waste

    © 2020 Elsevier Inc. It is estimated that nearly one-third of food produced on the planet never meets its intended purpose of human nourishment. This represents a substantial stock of resources available for reallocation. Any potential reallocation of resources raises ethical issues – who should sacrifice (change current behaviors), who should benefit, and what methods are appropriate to induce the behavioral change required to invoke the reallocation? In this brief article, we will discuss several topics in the food waste literature that, in our opinion, raise ethical issues that warrant further thought and consideration. These include the emphasis on food donation as a means to reduce food waste, the emergence of markets for food with cosmetic imperfection (i.e., “ugly food”), the appropriateness of guilt appeals to motivate reductions in wasted food, and the ethical tensions in choosing dates on food labels
