    Increased levels of NETosis biomarkers in high-grade serous ovarian cancer patients’ biofluids: Potential role in disease diagnosis and management

    Introduction: High-grade serous ovarian cancer (HGSOC) is the second most frequent gynecological malignancy but the most lethal, partially due to the spread of the disease through the peritoneal cavity. Recent evidence has shown that, apart from their role in immune defense through phagocytosis and degranulation, neutrophils are able to participate in cancer progression through the release of neutrophil extracellular traps (NETs) in a process called NETosis. NETs are composed of DNA, histones, calprotectin, myeloperoxidase (MPO) and elastase and the NETosis process has been proposed as a pre-requisite for the establishment of omental metastases in early stages of HGSOC. Nevertheless, its role in advanced stages remains to be elucidated. Therefore, our principal aim is to characterize a NETosis biomarker profile in biofluids from patients with advanced HGSOC and control women. Methods: Specifically, five biomarkers of NETosis (cell-free DNA (cfDNA), nucleosomes, citrullinated histone 3 (citH3), calprotectin and MPO) were quantified in plasma and peritoneal fluid (PF) samples from patients (n=45) and control women (n=40). Results: Our results showed that HGSOC patients presented a higher concentration of cfDNA, citH3 and calprotectin in plasma and of all five NETosis biomarkers in PF than control women. Moreover, these biomarkers showed a strong ability to differentiate the two clinical groups. Interestingly, neoadjuvant treatment (NT) seemed to reduce NETosis biomarkers mainly systemically (plasma) compared to the tumor environment (PF). Discussion: In conclusion, NETosis biomarkers are present in the tumor environment of patients with advanced HGSOC, which might contribute to the progression of the disease. Besides, plasma cfDNA and calprotectin could represent minimally invasive surrogate biomarkers for HGSOC. Finally, NT modifies NETosis biomarkers levels mainly at the systemic level

    A Text Mining and Statistical Approach for Assessment of Pedagogical Impact of Students’ Evaluation of Teaching and Learning Outcome in Education

    Technology-enhanced learning (TEL) is now at the heart of teaching and learning process in many higher education institutions (HEIs). Today, educators are faced with the challenges of pedagogically specifying what tools, methods, and technologies are used to support the teachers and students, and to help maintain/sustain a continuous education and practices. This study shows that there is an opportunity in the use of (educational) datasets derived about the teaching and learning processes to provide insights for fostering the education process. To this effect, it analyzed the students’ evaluation of teaching (SET) dataset ( n=471968n=471968 ) collected within a higher education setting to determine prominent factors that influences the students’ performance or the way (TEL-based) education is being delivered, including its didactical impact and implications for practice. Theoretically, the study employed a mixed methodology grounded on integration of the Data-structure approach and Descriptive decision theory to study the rationality behind the students’ evaluation of the teaching and performance. This was done through the Textual data quantification (qualitative) and Statistical (quantitative) analysis. Qualitatively, the study applied the Educational Process and Data Mining (EPDM) model (a text mining method) to extract the different sentiments and emotional valence expressed by the students in the SET, and how those characteristically differ based on the period and type of evaluation they have completed (between 2019 to 2021). For the quantitative analysis, the study used a multivariate analysis of covariance (MANCOVA) and multiple pairwise comparisons post-hoc tests to analyze the quantified information (average sentiment and emotional valence) extracted from the SET data to determine the marginal means of effect the different SET types and evaluation period have on the students’ learning outcomes/perception about the teaching-learning process. In addition, the study empirically discussed and shed light on the implications of the main findings for TEL-based Education, particularly implemented by the HEI during the analyzed periods. The scholastic indicator from the study shows that while the flexible digital models or instructional methods are effective for continuous education, innovative pedagogies, and teaching transformations. It also, on the other hand, serve as an incentive for more robust research that idiosyncratically look into their implications for the students’ learning outcomes and assessment done in this study

    A context for the last Neandertals of interior Iberia: Los Casares cave revisited

    Introduction and objectives Although the Iberian Peninsula is a key area for understanding the Middle to Upper Paleolithic transition and the demise of the Neandertals, valuable evidence for these debates remains scarce and problematic in its interior regions. Sparse data supporting a late Neandertal persistence in the Iberian interior have been recently refuted and hence new evidence is needed to build new models on the timing and causes of Neandertal disappearance in inland Iberia and the whole peninsula. In this study we provide new evidence from Los Casares, a cave located in the highlands of the Spanish Meseta, where a Neandertal-associated Middle Paleolithic site was discovered and first excavated in the 1960's. Our main objective is twofold: (1) provide an updated geoarcheological, paleoenvironmental and chronological framework for this site, and (2) discuss obtained results in the context of the time and nature of the last Neandertal presence in Iberia. Methods We conducted new fieldwork in an interior chamber of Los Casares cave named 'Seno A'. Our methods included micromorphology, sedimentology, radiocarbon dating, Uranium/Thorium dating, palinology, microfaunal analysis, anthracology, phytolith analysis, archeo-zoology and lithic technology. Here we present results on site formation processes, paleoenvironment and the chronological setting of the Neandertal occupation at Los Casares cave-Seno A. Results and discussion The sediment sequence reveals a mostly in situ archeological deposit containing evidence of both Neandertal activity and carnivore action in level c, dated to 44,899 +/- 42,175 calendar years ago. This occupation occurred during a warm and humid interval of Marine Isotopic Stage 3, probably correlating with Greenland Interstadial 11, representing one of the latest occurrences of Neandertals in the Iberian interior. However, overlying layer b records a deterioration of local environments, thus providing a plausible explanation for the abandonment of the site, and perhaps for the total disappearance of Neandertals of the highlands of inland Iberia during subsequent Greenland Stadials 11 or 10, or even Heinrich Stadial 4. Since layer b provided very few signs of human activity and no reliable chronometric results, and given the scarce chronostratigrapic evidence recorded so far for this period in interior Iberia, this can only be taken as a working hypothesis to be tested with future research. Meanwhile, 42,000 calendar years ago remains the most plausible date for the abandonment of interior Iberia by Neandertals, possibly due to climate deterioration. Currently, a later survival of this human species in Iberia is limited to the southern coasts
