    Differences between plasma and cerebrospinal fluid glial fibrillary acidic protein levels across the Alzheimer Disease continuum

    Importance: Glial fibrillary acidic protein (GFAP) is a marker of reactive astrogliosis that increases in the cerebrospinal fluid (CSF) and blood of individuals with Alzheimer disease (AD). However, it is not known whether there are differences in blood GFAP levels across the entire AD continuum and whether its performance is similar to that of CSF GFAP. Objective: To evaluate plasma GFAP levels throughout the entire AD continuum, from preclinical AD to AD dementia, compared with CSF GFAP. Design, Setting, and Participants: This observational, cross-sectional study collected data from July 29, 2014, to January 31, 2020, from 3 centers. The Translational Biomarkers in Aging and Dementia (TRIAD) cohort (Montreal, Canada) included individuals in the entire AD continuum. Results were confirmed in the Alzheimer’s and Families (ALFA+) study (Barcelona, Spain), which included individuals with preclinical AD, and the BioCogBank Paris Lariboisière cohort (Paris, France), which included individuals with symptomatic AD. Main Outcomes and Measures: Plasma and CSF GFAP levels measured with a Simoa assay were the main outcome. Other measurements included levels of CSF amyloid-β 42/40 (Aβ42/40), phosphorylated tau181 (p-tau181), neurofilament light (NfL), Chitinase-3-like protein 1 (YKL40), and soluble triggering receptor expressed on myeloid cells 2 (sTREM2) and levels of plasma p-tau181 and NfL. Results of amyloid positron emission tomography (PET) were available in TRIAD and ALFA+, and results of tau PET were available in TRIAD. Results: A total of 300 TRIAD participants (177 women [59.0%]; mean [SD] age, 64.6 [17.6] years), 384 ALFA+ participants (234 women [60.9%]; mean [SD] age, 61.1 [4.7] years), and 187 BioCogBank Paris Lariboisière participants (116 women [62.0%]; mean [SD] age, 69.9 [9.2] years) were included. Plasma GFAP levels were significantly higher in individuals with preclinical AD in comparison with cognitively unimpaired (CU) Aβ-negative individuals (TRIAD: Aβ-negative mean [SD], 185.1 [93.5] pg/mL, Aβ-positive mean [SD], 285.0 [142.6] pg/mL; ALFA+: Aβ-negative mean [SD], 121.9 [42.4] pg/mL, Aβ-positive mean [SD], 169.9 [78.5] pg/mL). Plasma GFAP levels were also higher among individuals in symptomatic stages of the AD continuum (TRIAD: CU Aβ-positive mean [SD], 285.0 [142.6] pg/mL, mild cognitive impairment [MCI] Aβ-positive mean [SD], 332.5 [153.6] pg/mL; AD mean [SD], 388.1 [152.8] pg/mL vs CU Aβ-negative mean [SD], 185.1 [93.5] pg/mL; Paris: MCI Aβ-positive, mean [SD], 368.6 [158.5] pg/mL; AD dementia, mean [SD], 376.4 [179.6] pg/mL vs CU Aβ-negative mean [SD], 161.2 [67.1] pg/mL). Plasma GFAP magnitude changes were consistently higher than those of CSF GFAP. Plasma GFAP more accurately discriminated Aβ-positive from Aβ-negative individuals than CSF GFAP (area under the curve for plasma GFAP, 0.69-0.86; area under the curve for CSF GFAP, 0.59-0.76). Moreover, plasma GFAP levels were positively associated with tau pathology only among individuals with concomitant Aβ pathology. Conclusions and Relevance: This study suggests that plasma GFAP is a sensitive biomarker for detecting and tracking reactive astrogliosis and Aβ pathology even among individuals in the early stages of AD

    Multi-island single-electron devices from self-assembled colloidal nanocrystal chains

    We report the fabrication of multi-island single-electron devices made by lithographic contacting of self-assembled alkanethiol-coated gold nanocrystals. The advantages of this method, which bridges the dimensional gap between lithographic and NC sizes, are (1) that all tunnel junctions are defined by self-assembly rather than lithography and (2) that the ratio of gate capacitance to total capacitance is high. The rich electronic behavior of a double-island device, measured at 4.2 K, is predicted in detail by combining finite element and Monte Carlo simulations with the standard theory of Coulomb blockade with very few adjustable parameters.Comment: 4 page

    La période allant de 1968 à 1981 correspond à des années cruciales pour la géographie physique en France, entre tradition et innovations. Au plan scientifique et sociétal, la géographie physique est en effet confrontée depuis 1950 à divers changements au plan des concepts et des théories, en particulier la double révolution de la systémique et de la quantification. Dans les années 1970 s’accélère la marche aux grands programmes internationaux, à la globalisation et à l’innovation technique. L’article discute des possibles raisons de fond d’une mutation inachevée de la géographie physique française, moins par manque de moyens matériels que du fait d’effectifs insuffisants pour porter le renouvellement des idées.The period from 1968 to 1981 is a crucial one in France for physical geography, between tradition and innovation. Since 1950, physical geography was confronted, scientifically and socially, to various changes in terms of concepts and theories, especially the dual revolution of systems theory and quantitative geography. In the 1970s there was an acceleration towards large international programs, globalisation and technological innovation. The paper discusses some fundamental reasons for the incomplete transformation of the French physical geography, less from lack of resources than as a result of an insufficient number of scholars to put forward the renewal of ideas

    Spectral classification of the brightest objects in the galactic star forming region W40

    We present high S/N, moderate resolution near-infrared spectra, as well as 10 micron imaging, for the brightest members of the central stellar cluster in the W40 HII region, obtained using the SpeX and MIRSI instruments at NASA's Infrared Telescope Facility. Using these observations combined with archival Spitzer Space Telescope data, we have determined the spectral classifications, extinction, distances, and spectral energy distributions for the brightest members of the cluster. Of the eight objects observed, we identify four main sequence (MS) OB stars, two Herbig Ae/Be stars, and two low-mass young stellar objects. Strong HeI absorption at 1.083 micron in the MS star spectra strongly suggests that at least some of these sources are in fact close binaries. Two out of the four MS stars also show significant infrared excesses typical of circumstellar disks. Extinctions and distances were determined for each MS star by fitting model stellar atmospheres to the SEDs. We estimate a distance to the cluster of between 455 and 535 pc, which agrees well with earlier (but far less precise) distance estimates. We conclude that the late-O star we identify is the dominant source of LyC luminosity needed to power the W40 HII region and is the likely source of the stellar wind that has blown a large (~4 pc) pinched-waist bubble observed in wide field mid-IR images. We also suggest that 3.6 cm radio emission observed from some of the sources in the cluster is likely not due to emission from ultra-compact HII regions, as suggested in other work, due to size constraints based on our derived distance to the cluster. Finally, we also present a discussion of the curious source IRS 3A, which has a very strong mid-IR excess (despite its B3 MS classification) and appears to be embedded in a dusty envelope roughly 2700 AU in size.Comment: Accepted for publication in The Astronomical Journal. 29 pages, 10 figure

    Implication of the lopsided growth for the viscosity of Earth's inner core

    Two main seismic features characterize the Earth's inner core: a North-South polar anisotropy and an East-West asymmetry of P-wave velocity and attenuation. Anisotropy is expected if shear deformation is induced by convective motions. Translation has recently been put forward as an important mode of convection of the inner core. Combined with a simple diffusive grain growth model, this mechanism is able to explain the observed seismic asymmetry, but not the bulk anisotropy. The source of anisotropy has therefore to be sought in the shear motions caused by higher modes of convection. Using a hybrid finite-difference spherical harmonics Navier-Stokes solver, we investigate the interplay between translation and convection in a 3D spherical model with permeable boundary conditions at the inner core boundary. Three parameters act independently: viscosity, internal heating and convection velocity in the outer core. Our numerical simulations show the dominance of pure translation for viscosities of the inner core higher than 102010^{20} Pas. Translation is almost completely hampered by convective motions for viscosities lower than 101810^{18} Pas. Between these values, translation and convection develop, but convective downwellings are restricted to the coldest hemisphere where crystallization occurs. On the opposite side, shear is almost absent, thereby allowing grain growth. We propose that the coexistence of translation and convection observed in our numerical models leads to a seismic asymmetry but localizes deformation only in one hemisphere.Comment: Submitted to PNAS on Februray 10th 2012, rejected on March 19th 2012.     Large-scale rock slope failures in the eastern Pyrenees: identifying a sparse but significant population in paraglacial and parafluvial contexts

Around 30 principal RSFs >0.25 km2 and 20 lesser or uncertain cases have been identified from remote imagery and groundtruthing. Compared with other European mountain ranges, RSF incidence is relatively sparse, displays no obvious regional trend or spatial clustering, and occurs across diverse landscape types, if mainly on metamorphic rocks. A transition is observed from paraglacial RSFs in formerly-glaciated valleys to what are here termed ‘parafluvial’ RSFs, within wholly or mainly fluvial valleys but where slope failure is not directly provoked by or linked to river erosion. RSFs are particularly found in three topographic settings: (i) at cirque and trough-head thresholds (transition zones of elevated instability between cirque and main glaciated trough walls); (ii) near the upper or outer periphery of the ice field, where glacial adaptation of fluvial valleys is incomplete; and (iii) in fluvial valleys beyond glacial limits where incision is locally intense. RSF is absent from the range divide, from within cirques, and from most main valleys. In the montane areas, RSF is strongly associated with vestiges of preglacial summit surfaces, confirming that plateau ridges are less stable than sharpened crests and horns. RSF is contributing significantly to the progressive destruction of this paleic relief. The overall sparsity of RSF indicates insufficient rock mass stresses, including rebound after concentrated bedrock erosion. This may reflect a relatively weak imprint of glacial erosion, including breaching, in a context of relatively low mean rates of neotectonic uplift, possibly signalling overall that eastern Pyrenees landscapes are close to dynamic equilibrium.Peer ReviewedPostprint (author’s final draft
