    Experimental Study of the Abalone Shell on Cataract induced by Oxidative Stress

    目的研究石决明提取物对体外培养的晶状体氧化应激性白内障形成的作用及机制。方法离体培养小鼠晶状体,应用不同浓度的石决明提取物预孵育晶状体24h后,加入1mm过氧化氢,继续培养3小时后恢复正常培养,72小时后观察小鼠晶状体混浊程度,测定晶状体组织培养液中的乳酸脱氢酶(LDH)含量,晶状体组织中还原型谷胱甘肽(GSH)含量及超氧化物歧化酶(SOD)活力。结果石决明提取物在1~2 mg/ml浓度范围内减轻氧化应激造成的晶状体混浊,减少晶状体LDH的释放,提高组织内GSH含量和SOD活力。结论石决明提取物可减轻氧化应激白内障的形成,其作用主要与石决明提取物提高内源性抗氧化系统有关。Objective To study the effect of the abalone shell extract on oxidative stress induced cataract formation and its mechanism in cultured mouse lens in vitro. Methods The cultured mouse lens were pretreated with the abalone shell extract in different concentrations for24 hours,and then 1mm hydrogen peroxide was added and continued incubating for 3 hours,and they were changed to normal culture media.After 72 hours,the opacity of each lens was observed under an anatomical microscope,the content of lactate dehydrogenase( LDH)leakages,the content of the reduced glutathione( GSH) and activity of superoxide dismutase( SOD) in lens tissue were assayed. Results Abalone shell extract in the concentration range of 1 ~ 2 mg / ml reduced the lens opacity caused by oxidative stress,alleviated the release of LDH,and increased GSH content and SOD activity in cultured lens.     Construction of a Recombinant Escherichia Coli DH5α for Dextranase Expression

    以Plf3为模板,PCr扩增得到葡聚糖酶的启动子,并将启动子基因重新连接到切除启动子基因及葡聚糖酶基因的Plf3载体上,构建Plf3-PrO载体.采用PCr的方法扩增朱黄青霉总dnA中的α-1-6-葡聚糖酶基因,并插入到Plf3-PrO载体上,构建Plf3-PrO-dEX质粒,以大肠杆菌dH5α为宿主菌,酶切验证及PCr验证均表明成功构建了Plf3-PrO-dEX质粒,且重组质粒中的α-1-6-葡聚糖酶基因与朱黄青霉中的α-1-6-葡聚糖酶基因有93%的同源性.The promoter of dextranase was PCR-amplified from pLF3 vector and inserted into pLF3 plasmid,from which the genes encoding a promoter and β-glucanase had been removed,yielding PLF3-Pro.The DNA encoding the dextranase was PCR-amplified from the total DNA of Penicillium minioluteum MUCL 38929 and cloned into pLF3-Pro vector.The recombinant plasmid pLF3-Pro-Dex was constructed and checked through PCR,restriction enzyme digestion and DNA sequencing.Results shows that the recombinant plasmid pLF3-Pro-Dex is successfully constructed.科技部科研院所技术开发研究专项资金(NCSTE-2007-JKZX-023

    黄河首曲地区生态旅游开发适宜区识别与分类研究——结合旅游者偏好的分析(英文)

    黄河首曲地区具有突出的生态功能和较高的旅游价值。为保障湿地的完整性保护及尊重当地居民的雪山崇拜,研究首先对不可开发区和可开发区进行了识别分析,认为不管是严格的生态旅游还是一般的生态旅游活动都只能在有效生态旅游区进行,生态旅游的适宜性分析与区划也主要围绕可开发区展开。研究选取距水域湿地距离、草地覆盖情况、森林覆盖情况、海拔高度、坡度和原生态性六个表征地区自然性的标准,采用GIS技术与层次分析法相结合的方法对可开发区生态旅游适宜性进行了分析,层次分析法权重源于旅游者偏好调查,并基于适宜度空间分布格局进行了区域分类。可开发区的生态旅游适宜性可划分为五级,I级地区适宜度最低,最不适合发展严格的生态旅游,V级地区适宜度最高,原生态景观价值突出,生态旅游发展潜力最大。出于协调旅游功能与生态功能的统一以实现地区生态旅游资源可持续利用的目的,黄河首曲地区被划分为五类亚区,且均具有各自特殊的主导服务功能。研究最终根据各亚区的特殊服务功能提出了相应的调控措施。研究结论有助于促进生态旅游的合理性规划及未来可持续发展

    Cloning of a Recombinant Dextranase and Its Expression in Pichia pastoris

    通信作者: [email protected][中文文摘]根据朱黄青霉(Penicillium minioluteumHI-4)α-1,6-葡聚糖酶的基因序列,人工合成α-1,6-葡聚糖酶基因并将其插入毕赤酵母(Pichia pastoris)表达载体pPICZαA,PCR和双酶切验证结果均表明成功构建了重组质粒pPICZαA-Dex.重组质粒经SacⅠ线性化后整合到毕赤酵母X33基因组中进行表达.摇瓶发酵表明,重组毕赤酵母经甲醇诱导120 h产酶活力达到最高值29.2 U/mL,SDS-PAGE结果显示在67 ku附近有明显的条带,与α-1,6-葡聚糖酶理论分子质量相符合.[英文文摘]The α-1,6-dextranase gene was amplified from a synthetic α-1,6-dextranase gene derived from Penicillium minioluteum HI-4 and was inserted into plasmid pPICZαA.The recombinant plasmid named pPICZαA-Dex was identified by PCR and enzyme digestion,and was then electrotransformed into Pichia pastoris X33.The recombinant Pichia pastoris X33 was induced to express the dextranase using methanol.The enzyme activity reached 29.2 U/mL after 120 h of induction.The size of the recombinant α-1,6-dextranase is approximately 67 ku analyzed by SDSPAGE, which is consistent with the theoretical molecular weight.科技部科研院所技术开发研究专项资金(NCSTE-2007-JKEC-023

    一、该项目针对农业生产中食品安全和环境污染问题,开展了S-诱抗素、新奥霉素、壳寡糖、棉铃虫病毒杀虫剂与昆虫病原线虫生物制剂、功能性堆肥及其浸提液工业化生产等的试验和示范,立项准确,针对性强,意义重大。 二、项目研究出S-诱抗素等生物制剂及其生产工艺、工厂化技术;研究开发出昆虫病原线虫活体繁殖技术,实现了工厂化生产;研究开发了两种功能性堆肥及浸提液,提出了“功能性堆肥+秸秆生物反应堆+堆肥浸提液+S-诱抗素等生物制剂”健康、安全设施蔬菜生产模式;在宁夏实现了地上、地下,土壤、作物生物制剂联防技术体系,为低耗、高效、安全、健康农产品生产开辟了新途径。 三、在S-诱抗素、新奥霉素高产菌株的生产工艺,昆虫病原线虫活体繁殖工厂化生产方面取得了新突破;在S-诱抗素、新奥霉素、壳寡糖、棉铃虫病毒杀虫剂、功能性堆肥及其浸提液集成应用控制作物病虫害等方面有创新。研究成果达到了国内先进水平,S-诱抗素、新奥霉素高产菌株的生产工艺研究达到国际领先。 四、项目执行期间,在宁夏15个市县建立核心试验基地14个,示范推广点40个,累计推广面积17万亩,新增效益9600万元。获得发明专利4项,实用新型专利1项,制定地方标准5项,专著1部,发表论文19篇(其中SCI收录6篇)。培训农技人员300人次,农民4700多人次

    JUNO Sensitivity on Proton Decay pνˉK+p\to \bar\nu K^+ Searches

    The Jiangmen Underground Neutrino Observatory (JUNO) is a large liquid scintillator detector designed to explore many topics in fundamental physics. In this paper, the potential on searching for proton decay in pνˉK+p\to \bar\nu K^+ mode with JUNO is investigated.The kaon and its decay particles feature a clear three-fold coincidence signature that results in a high efficiency for identification. Moreover, the excellent energy resolution of JUNO permits to suppress the sizable background caused by other delayed signals. Based on these advantages, the detection efficiency for the proton decay via pνˉK+p\to \bar\nu K^+ is 36.9% with a background level of 0.2 events after 10 years of data taking. The estimated sensitivity based on 200 kton-years exposure is 9.6×10339.6 \times 10^{33} years, competitive with the current best limits on the proton lifetime in this channel
