    Evaluation of Solvent Toxicity of Plant Extract with Antiviral Action in Refrigerated Goat Semen

    Background: Caprine Arthritis Encephalitis Virus have been detected in sperm of breeding goats causing economic losses. In order to control the virus, researches aiming to identify natural extracts with potential antiviral effects are performed. However, aqueous or ethanolic extracts must be diluted in dimethyl sulfoxide (DMSO), which is a substance with unknown effects in sperm quality when present in diluting media. Therefore, this study aimed to evaluate sperm viability of refrigerated caprine semen diluted in media containing DMSO. This was performed to provide data that aid in researches involving the use of this component with natural extracts that may inactivate the caprine lentivirus in sperm.Materials, Methods & Results: The experiment was performed at the Laboratory of Seminal Technology in Embrapa Goats and Sheep in the city of Sobral, Brazil. Sperm viability was assessed in caprine semen refrigerated in two dilution media with crescent concentrations of DMSO. Sperm samples of five goats seronegative for the caprine lentivirus were pooled and diluted in minimal essential medium (MEM) enriched with glucose at 0.01 M added of crescent concentrations of DMSO (0%, 1.5%, 1.75%, 2.0%, 2.25% and 2.5%). The same breeders provided the pool of sperm to test Tris added 2.5% of egg yolk and the same concentrations of DMSO previously mentioned. Treatments were refrigerated at 7°C and evaluated up until four h after DMSO addition. Individual progressive motility (MIP), sperm vigor (V), percentage of spermatozoa reactive to hypoosmotic test (HO) and morphologically normal (NOR) were evaluated. IPM, vigor and NOR remained within normal standards for the caprine species in all treatments test. Percentage results of spermatozoa reactive to hypoosmotic was higher in Tris yolk with values ranging between 34.66% to 46.33%. Sperm vigor was positively correlated (r = 0.85) with IPM in the MEM diluted pool of sperm. In Tris yolk, vigor and hypoosmotic test correlated moderately (r = 0.63, r = 0.54, respectively) with IPM. Tris yolk medium added DMSO presented the highest percentage of reactivity to hypoosmotic test in all treatments when compared to MEM added DMSO.Discussion: The fact that DMSO is easily homogenized in water, ethylic alcohol and most organic solvents favors its use in diluting natural extracts. These components are a possible source of products that inactivate caprine arthritis encephalitis virus in sperm, which is the key to promoting the safe use of genetic material of infected breeders, in addition to commercial use of germplasm. In this study, there was no interference of DMSO in the analyzed parameters when added in a maximum concentration of 2.5% to MEM and Tris yolk, which is in accordance with standard values for goats. In addition, Tris yolk may promote greater protection to the membrane of sperm cells, which was demonstrated by hypoosmotic test. This medium could be ideal to be used in new methodologies that incorporate DMSO. In conclusion, DMSO added to dilution media Tris yolk and MEM did not interfere with the quality of refrigerated caprine sperm, which maintained viability. These results indicate that this substance did not present harmful effects to the genetic material, promoting the use as solvent of extracts from plant compounds with potential anti-viral effect. The information in this study may aid new research performed in this area

    Tromboembolismo pulmonar: dos aspectos epidemiológicos ao tratamento / Pulmonary thromboembolism: from epidemiological aspects to treatment

    Introdução: O tromboembolismo pulmonar se trata de uma doença relativamente comum, potencialmente fatal e que requer diagnóstico rápido e preciso. Apesar das inovações tecnológicas na medicina que permitiram avanços em seu tratamento, ele constitui um paradoxo para a área da saúde, uma vez que à medida que ocorre o progresso medicinal, maior é o número de situações que predispõem ao embolismo. Objetivo: Investigar, interpretar e revisar os achados sobre epidemiologia, fisiopatologia, métodos diagnósticos, profiláticos e de tratamento dessa doença. Metodologia: Utilizou-se a base Medical Literature Analysis and Retrieval System Online (MEDLINE/PubMed), sob os descritores Embolia pulmonar; Tratamento; Tromboembolia venosa, com a seleção de artigos de revisão e artigos originais, seguindo critérios de elegibilidade. Foram encontrados 5866 textos, com seleção inicial de 488, dos quais restaram 23 artigos elegíveis. Resultados: Os artigos, em sua maioria redigidos em língua inglesa, contemplavam também publicações de autores e periódicos de diferentes nacionalidades. A maioria dos artigos abrangiam os anos de 2017 (sete) e 2018 (cinco), e grande parte foi publicada nos últimos cinco anos. Considerações finais: Pelo fato de o tromboembolismo pulmonar possuir diferentes formas clínicas e, assim, manifestar sinais e sintomas inespecíficos e capazes de serem confundidos com outros diagnósticos possíveis, é fundamental a tomada de decisão rápida e eficiente sobre a suspeição e a abordagem clínica desse quadro

    Obesidade: principais distúrbios materno-fetais: Obesity: major maternal-fetal disorders

    Introdução: A obesidade gestacional pode ser considerada tanto pelo aumento do IMC quanto pelo ganho de peso excessivo durante o período de prenhez. Mulheres obesas apresentam fatores de riscos aumentados pré-gestação, no início da gestação, no decorrer do período de gravidez, parto e pós-parto. Também, maiores riscos de morbidade e mortalidades para seu bebê. Assim, o presente estudo teve como objetivo descrever as complicações materno fetais presentes em gestantes obesas. Metodologia: Revisão integrativa de literatura, com levantamento de dados nas bases: PubMed, UpToDate e Scielo. Utilizou-se os descritores: “Ganho de Peso Gestacional”; “Ganho de Peso Materno”; “Gestação de Risco”. Totalizando em uma seleção de 30 artigos. Resultados e Discussão: Dentre as principais complicações materno-fetais que o estudo nos mostrou podemos considerar que mulheres obesas estão mais propensas a desenvolver diabetes, hipertensão, trabalho de parto prolongado e parto cirúrgico, enquanto sua prole tem a chance aumentada de apresentar macrossomia, sofrimento fetal, restrição de crescimento intrauterino e prematuridade. Logo necessitaríamos de mudanças no estilo de vida, investindo em dietas e atividade física, para reduzir os riscos destas pacientes em manter este peso pós-parto ou em aumentar ainda mais em gestações subseqüentes. Conclusão: Assim, o planejamento gestacional faz-se fundamental para todas as mulheres em idade fértil, focando principalmente nas portadoras de obesidade, visando a mudança de estilo de vida previamente a gestação, para que, sejam evitadas as complicações maternas e fetais resultantes da obesidade gestacional

    SARS-CoV-2 introductions and early dynamics of the epidemic in Portugal

    Genomic surveillance of SARS-CoV-2 in Portugal was rapidly implemented by the National Institute of Health in the early stages of the COVID-19 epidemic, in collaboration with more than 50 laboratories distributed nationwide. Methods By applying recent phylodynamic models that allow integration of individual-based travel history, we reconstructed and characterized the spatio-temporal dynamics of SARSCoV-2 introductions and early dissemination in Portugal. Results We detected at least 277 independent SARS-CoV-2 introductions, mostly from European countries (namely the United Kingdom, Spain, France, Italy, and Switzerland), which were consistent with the countries with the highest connectivity with Portugal. Although most introductions were estimated to have occurred during early March 2020, it is likely that SARS-CoV-2 was silently circulating in Portugal throughout February, before the first cases were confirmed. Conclusions Here we conclude that the earlier implementation of measures could have minimized the number of introductions and subsequent virus expansion in Portugal.
