    Effect of rare earth elements on growth and antioxidant metabo- lism in Lemna minor L.

    Lemna minor is frequently used in bioremediation processes to remove nutrients and contaminants from waste water. In this work the response of L. minor to treatments with lanthanum nitrate and with a mix of several light rare earth elements (REE) nitrates was investigated. Preliminary results indicate that L. minor shows an overall good tolerance to the presence of REE in the media. Toxic effects were observed after prolonged exposition to high concentration of REE. An increase in ascorbate and glutathione content as well as in ascorbate peroxidase, dehy- droascorbate reductase and ascorbate free radical (AFR) reductase activity was observed in treated plants

    Staphylococcal enterotoxins and enzymes in presence of essential oils

    Nuestro objetivo fue investigar el efecto de concentraciones subinhibitorias de clavo, canela, y tomillo en el crecimiento de S aureus y en la producción de coagulasa, termonucleasa y enterotoxina. Con ese fi n se agregaron diferentes concentraciones de los aceites esenciales a Caldo Cerebro Corazón. La producción de enterotoxina en presencia y en ausencia de los aceites esenciales se determinó por la técnica de enzimoinmunoensayo (ELISA). Nuestros resultados indican que bajas concentraciones que no afectan el crecimiento, reducen la producción de la producción de las enzimas ensayadas y de enterotoxina El aceite esencial de tomillo es el que presenta mayor actividad pues al 0,04% produce la pérdida de la actividad de ambas enzimas y de la enterotoxicidad Esto es importante cuando consideramos su potencial aplicación en alimentos y en la industria farmacéutica.Our aim was to investigate the effect of subinhibitory concentrations of clove, cinnamon, and thyme essential oils on the growth of Staphylococcus aureus and its production of coagulase, thermonuclease and enterotoxin. Different concentrations of the essential oils were added to Brain Heart Infusion broth. The production of enterotoxin in the presence or absence of essential oils was estimated using the ELISA technique. Our results indicate that a lower concentration did not affect the growth S. aureus but reduced the production of metabolites. Thyme oil presents the most signifi cant effect, inhibiting the activity of both enzymes and the production of enterotoxin with 0,04%. There is a renewed interest in the potential application of these essential oils in the food and pharmaceutical industries to control staphylococcal poisoning

    The Rejection of the Epicurean Ideal of Pleasure in Late Antique Sources: not Only Misunderstandings

    Epicureanism was seen by its opponents, both 'pagan' and Christian, as the philosophy of pleasure and atheism. From the theological point of view, the accusation of atheism was incorrect, since Epicurus and the Epicureansadmitted of the existence of deities, and posited them as models of moral perfection, while denying their interest in human affairs, i.e. providence. This denial aimed at guaranteeing their imperturbability (ataraxia). From the ethical point of view, the ideal of pleasure (hedone), on which I shall concentrate here, was grossly misunderstood or distorted by the opponents of Epicureanism, who generally did not take into consideration the moderation, equilibrium, and serenity that the superior 'catastematic pleasure' (Epicurus's real ideal of pleasure) involved. I shall analyse the attitude of late-antique sources, especially Christian, toward Epicureanism and its ethics. A great many of Usener's and Arrighetti's fragments of Epicurus indeed come from Christian late-antique authors, such as Clement, Origen, Eusebius, Lactantius, and Augustine, but other patristic authors should be added, such as Basil and Gregory of Nyssa. Even if patristic interest in Epicureanism is often critical, and sometimes imprecise or distorted, nevertheless it is tangible. I shall focus on the authors who make the most interesting use of Epicurean sources, particularly with respect to the ethical doctrine: Origen, Dionysius of Alexandria, Lactantius, Ambrose, Jerome and Augustine, Gregory Nyssen, and Nazianzen, the only one who really understood and praised Epicurus's notion of hedone. I shall also argue that the fading away of the availability and use of good sources on Epicureanism, along with the disappearance of the Epicurean school itself, brought about an impoverishment in the understanding of, and hostility to, Epicurus and Epicureanism.O Epicurismo foi visto pelos seus oponentes, tanto pagãos quanto cristãos, como a filosofia do prazer e do ateísmo. Do ponto de vista teológico, a acusação de ateísmo era incorreta, desde que o Epicuro e os epicuristas admitiram a existência de divindades e depositaram neles os ideais de perfeição, enquanto negavam seu interesse nos negócios humanos, isto é, a providência. Esta negação visa garantir sua imperturbabilidade (ataraxia). Do ponto de vista ético, o ideal de prazer (hedone), no qual eu irei me concentrar, foi grosseiramente mal-entendido ou distorcido pelos oponentes do Epicurismo, que geralmente não consideravam a moderação, equilíbrio, e serenidade que o "Prazer Catastemático" superior (verdadeiro ideal de prazer do Epicuro) envolvia. Analisarei a atitude em fontes tardo-antigas, especialmente cristãs, a cerca do Epicuro e sua ética. Grande parte de fragmentos do Epicuro de Usener e Arrighetti na verdade são de autores cristãos tardo-antigos, como Clemente, Orígenes, Eusébio, Lactêncio, e Agostinho, mas outros autores patrísticos deveriam ser adicionados, como Basílio e Gregório de Nyssa. Mesmo se o interesse patrístico no Epicurismo fosse frequentemente de crítica, e às vezes imprecisa ou distorcida, no entanto, é tangível. Focarei nos autores que mais se interessaram no uso de documentos do Epicuro, particularmente a despeito da doutrina ética: Orígenes, Dionísio de Alexandria, Lactêncio, Ambrósio, Jerônimo e Agostinho, Gregório Nyssen, e Nazianzeno, o único que realmente entendeu e elogiou noção de hedone do Epicuro. Também argumentarei acerca do desaparecimento da disponibilidade de uso de boas fontes sobre o Epicuro, juntamente com o desaparecimento da própria escola epicurista, que provocou um empobrecimento na compreensão e na hostilidade do Epicuro e epicurismo

    Perosomus elumbis in piglets: Pathological, radiological and cytogenetic findings

    Perosomus elumbis (PE) is a rare congenital condition characterized by agenesis of the lumbar, sacral and coccygeal vertebrae. Perosomus elumbis has rarely been reported in literature as morphological description of singles or few cases. Here we report the first extensive description of eight cases of PE detected in two consecutive litters from the same parents of Casertana pig breed. In August 2018, eight piglets were investigated for multiple malformations. All malformed animals, but one, died in the first day of life. The survivor piglet died at 23 days of age. Pathological, radiological and cytogenetic examination was performed. Furthermore, a farm epidemiological investigation was carried out to investigate the percentage of piglets born dead or with malformations in 2018. The radiological and pathological exams showed skeletal abnormalities at the spinal cord level and visceral malformations. Cytogenetic investigations showed a normal chromosome arrangement. Finally, epidemiological investigation revealed a low prevalence of malformations in newborn pigs, equal to 0.5% of the total birth rate of the farm. Our findings report the first extensive description of PE cases in pigs and suggest an underestimation of this malformation in veterinary medicine. Our findings also suggest a specific genetic etiological basis as cause of PE in pigs and exclude chromosomal abnormalities. Further studies will be performed to confirm this hypothesis

    Human Dental Pulp Stem Cells Hook into Biocoral Scaffold Forming an Engineered Biocomplex

    The aim of this study was to evaluate the behavior of human Dental Pulp Stem Cells (DPSCs), as well as human osteoblasts, when challenged on a Biocoral scaffold, which is a porous natural hydroxyapatite. For this purpose, human DPSCs were seeded onto a three-dimensional (3D) Biocoral scaffold or on flask surface (control). Either normal or rotative (3D) cultures were performed. Scanning electron microscopic analyses, at 8, 24 and 48 h of culture showed that cells did not adhere on the external surface, but moved into the cavities inside the Biocoral structure. After 7, 15 and 30 days of culture, morphological and molecular analyses suggested that the Biocoral scaffold leads DPSCs to hook into the cavities where these cells quickly start to secrete the extra cellular matrix (ECM) and differentiate into osteoblasts. Control human osteoblasts also moved into the internal cavities where they secreted the ECM. Histological sections revealed a diffuse bone formation inside the Biocoral samples seeded with DPSCs or human osteoblasts, where the original scaffold and the new secreted biomaterial were completely integrated and cells were found within the remaining cavities. In addition, RT-PCR analyses showed a significant increase of osteoblast-related gene expression and, above all, of those genes highly expressed in mineralized tissues, including osteocalcin, OPN and BSP. Furthermore, the effects on the interaction between osteogenesis and angiogenesis were observed and substantiated by ELISA assays. Taken together, our results provide clear evidence that DPSCs differentiated into osteoblasts, forming a biocomplex made of Biocoral, ECM and differentiated cells

    Human mandible bone defect repair by the grafting of dental pulp stem/progenitor cells and collagen sponge biocomplexes

    In this study we used a biocomplex constructed from dental pulp stem/progenitor cells (DPCs) and a collagen sponge scaffold for oro-maxillo-facial (OMF) bone tissue repair in patients requiring extraction of their third molars. The experiments were carried out according to our Internal Ethical Committee Guidelines and written informed consent was obtained from the patients. The patients presented with bilateral bone reabsorption of the alveolar ridge distal to the second molar secondary to impaction of the third molar on the cortical alveolar lamina, producing a defect without walls, of at least 1.5 cm in height. This clinical condition does not permit spontaneous bone repair after extraction of the third molar, and eventually leads to loss also of the adjacent second molar. Maxillary third molars were extracted first for DPC isolation and expansion. The cells were then seeded onto a collagen sponge scaffold and the obtained biocomplex was used to fill in the injury site left by extraction of the mandibular third molars. Three months after autologous DPC grafting, alveolar bone of patients had optimal vertical repair and complete restoration of periodontal tissue back to the second molars, as assessed by clinical probing and X-rays. Histological observations clearly demonstrated the complete regeneration of bone at the injury site. Optimal bone regeneration was evident one year after grafting. This clinical study demonstrates that a DPC/collagen sponge biocomplex can completely restore human mandible bone defects and indicates that this cell population could be used for the repair and/or regeneration of tissues and organs

    Transverse aortic constriction induces gut barrier alterations, microbiota remodeling and systemic inflammation

    Accumulating evidence suggests that modifications of gut function and microbiota composition might play a pivotal role in the pathophysiology of several cardiovascular diseases, including heart failure (HF). In this study we systematically analysed gut microbiota composition, intestinal barrier integrity, intestinal and serum cytokines and serum endotoxin levels in C57BL/6 mice undergoing pressure overload by transverse aortic constriction (TAC) for 1 and 4 weeks. Compared to sham-operated animals, TAC induced prompt and strong weakening of intestinal barrier integrity, long-lasting decrease of colon anti-inflammatory cytokine levels, significant increases of serum levels of bacterial lipopolysaccharide and proinflammatory cytokines. TAC also exerted effects on microbiota composition, inducing significant differences in bacterial genera inside Actinobacteria, Firmicutes, Proteobacteria and TM7 phyla as shown by 16S rDNA sequencing of fecal samples from TAC or sham mice. These results suggest that gut modifications represent an important element to be considered in the development and progression of cardiac dysfunction in response to TAC and support this animal model as a valuable tool to establish the role and mechanisms of gut-heart crosstalk in HF. Evidence arising in this field might identify new treatment options targeting gut integrity and microbiota components to face adverse cardiac events

    Detection and Characterization of CD133+ Cancer Stem Cells in Human Solid Tumours

    Osteosarcoma is the most common primary tumour of bone. Solid tumours are made of heterogeneous cell populations, which display different goals and roles in tumour economy. A rather small cell subset can hold or acquire stem potentials, gaining aggressiveness and increasing expectancy of recurrence. The CD133 antigen is a pentaspan membrane glycoprotein, which has been proposed as a cancer stem cell marker, since it has been previously demonstrated to be capable of identifying a cancer initiating subpopulation in brain, colon, melanoma and other solid tumours. Therefore, our aim was to observe the possible presence of cells expressing the CD133 antigen within solid tumour cell lines of osteosarcoma and, then, understand their biological characteristics and performances.In this study, using SAOS2, MG63 and U2OS, three human sarcoma cell lines isolated from young Caucasian subjects, we were able to identify and characterize, among them, CD133+ cells showing the following features: high proliferation rate, cell cycle detection in a G2\M phase, positivity for Ki-67, and expression of ABCG2 transporters. In addition, at the FACS, we were able to observe the CD133+ cell fraction showing side population profile and forming sphere-clusters in serum-free medium with a high clonogenic efficiency.Taken together, our findings lead to the thought that we can assume that we have identified, for the first time, CD133+ cells within osteosarcoma cell lines, showing many features of cancer stem cells. This can be of rather interest in order to design new therapies against the bone cancer

    Human CD34+/CD90+ ASCs Are Capable of Growing as Sphere Clusters, Producing High Levels of VEGF and Forming Capillaries

    Background: Human adult adipose tissue is an abundant source of mesenchymal stem cells (MSCs). Moreover, it is an easily accessible site producing a considerable amount of stem cells. Methodology/Principal Findings: In this study, we have selected and characterized stem cells within the stromal vascular fraction (SVF) of human adult adipose tissue with the aim of understanding their differentiation capabilities and performance. We have found, within the SVF, different cell populations expressing MSC markers – including CD34, CD90, CD29, CD44, CD105, and CD117 – and endothelial-progenitor-cell markers – including CD34, CD90, CD44, and CD54. Interestingly, CD34+/CD90+ cells formed sphere clusters, when placed in non-adherent growth conditions. Moreover, they showed a high proliferative capability, a telomerase activity that was significantly higher than that found in differentiated cells, and contained a fraction of cells displaying the phenotype of a side population. When cultured in adipogenic medium, CD34+/CD90+ quickly differentiated into adipocytes. In addition, they differentiated into endothelial cells (CD31+/VEGF+/Flk- 1+) and, when placed in methylcellulose, were capable of forming capillary-like structures producing a high level of VEGF, as substantiated with ELISA tests. Conclusions/Significance: Our results demonstrate, for the first time, that CD34+/CD90+ cells of human adipose tissue are capable of forming sphere clusters, when grown in free-floating conditions, and differentiate in endothelial cells that form capillary-like structures in methylcellulose. These cells might be suitable for tissue reconstruction in regenerative medicine, especially when patients need treatments for vascular disease
