    Biomonitoring Study of Deoxynivalenol Exposure in Chinese Inhabitants

    Objective: To investigate the levels of a deoxynivalenol (DON) biomarker in the urine of subjects living in two China provinces with different geographic locations and dietary patterns, and estimate their dietary DON exposures and health risks. Methods: First morning urine samples were collected on three consecutive days from 599 healthy subjects—301 from Henan province and 298 from Sichuan province—to analyze the total DON concentrations (tDON) after β-glucuronidase hydrolysis using a high-performance liquid chromatography tandem mass spectrometry-based method. The consumption of cereal foods in the previous 24 h before each urine collection was recorded using a duplicate diet method. DON exposure levels were estimated based on the urinary tDON concentrations. Results: Total DON were detected in 100% and 92% of the urine samples from Henan and Sichuan, respectively. Mean urinary tDON concentrations were 52.83 ng/mL in Henan subjects and 12.99 ng/mL in Sichuan subjects, respectively. The tDON levels were significantly higher in the urine of Henan subjects than that of the Sichuan subjects (p < 0.001). Urinary tDON levels were significantly different among age groups in both areas (Henan: p < 0.001; Sichuan: p = 0.026) and were highest in adolescents aged 13–17 years, followed by children aged 7–12 years. Based on the DON biomarker and exposure conversion reported by the European Food Safety Authority (EFSA), the mean estimated dietary intakes of DON were 1.82 μg/kg bw/day in Henan subjects and 0.45 μg/kg bw/day in Sichuan subjects. A total of 56% of Henan subjects and 12% of Sichuan subjects were estimated to exceed the PMTDI of 1 μg/kg bw/day. Consistent with urinary tDON levels, the highest estimated dietary DON intakes were also in children and adolescents aged 7–17 years. For all kinds of wheat-based foods except dumplings, the consumptions were significantly higher in Henan than those in Sichuan. The mean consumption of steamed buns was 8.4-fold higher in Henan (70.67 g/d) than that in Sichuan (8.45 g/d). The mean consumption of noodles in Henan (273.91 g/d) was 3.6-fold higher than that in Sichuan (75.87 g/d). Conclusions: The levels of urinary DON biomarker and the estimated dietary DON intakes in Henan province were high and concerning, especially for children and adolescents. The overall exposure level of Sichuan inhabitants was low

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals &lt;1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    Exploring the patterns of food waste generation by tourists in a popular destination

    Tourist food consumption is an important driver of food waste generation within the hotel/restaurant/café (HORECA) sector of popular destinations. Little is however known about the exact magnitude of food wastage by tourists alongside the determinants of their wasteful behaviour. This study has striven to contribute to knowledge with an exploratory survey in Lhasa, a popular destination in China, which set to establish the size of food wastage by tourists and explain the role of various socio-demographic and food consumption-related factors in its occurrence by statistical analysis and multiple linear regression analysis, respectively. The study found that tourists generated circa 15% of the total food waste in the HORECA sector, while the taste preferences and portion size were two major causes. The level of tourist education and personal satisfaction with meals exerted a significant negative impact on food waste generation. To reduce food wastage, policy-makers and HORECA professionals should educate tourists about the detrimental effect of wasted food and increase their satisfaction with meals. The research findings provided insights into achieving sustainability objectives in national and international tourism sectors

    Buddhist art originated with the emergence of Buddhism and has experienced continual innovation and development throughout history. Yingluo jewellery was introduced to China from India, with its form continuously incorporating ancient Chinese aesthetics in its inheritance and development. The designs of Ming Dynasty’s Yingluo jewellery exhibited a more secular and everyday trend. This article centres on the design of Ming Dynasty necklaces, using the Fahai Temple mural and the Yongxuan court statues as representative examples. The article summarises the distinct artistic features of Ming Dynasty necklaces and explores their aesthetics, focusing on three aspects: shape structure, colour matching, and pattern design. Additionally, 3D digital modelling is employed to revitalise and restore the necklaces’ beauty. Technical abbreviations will be explained upon first usage, and the text adheres to standard academic formatting and language conventions. The objective is to enhance national cultural confidence, bridge the gap between cultural heritage, appreciate the allure of traditional art treasures, and revive cultural artifacts. The infusion of rich cultural nuances into contemporary jewellery design, integration of traditional artistic components into modern aesthetics, and merging exceptional Ming Dynasty necklace works with contemporary aesthetic context, facilitates the seamless integration of outstanding culture into contemporary society

    The First Transcriptome Assembly of Yenyuan Stream Salamander (<i>Batrachuperus yenyuanensis</i>) Provides Novel Insights into Its Molecular Evolution

    The Yenyuan stream salamander (Batrachuperus yenyuanensis) has been previously evaluated with regards to phylogeny, population genetics, and hematology, but genomic information is sparse due to the giant genome size of salamanders which contain highly repetitive sequences, thus resulting in the lack of a complete reference genome. This study evaluates the encoding genetic sequences and provides the first transcriptome assembly of Yenyuan stream salamander based on mixed samples from the liver, spermary, muscle and spleen tissues. Using this transcriptome assembly and available encoding sequences from other vertebrates, the gene families, phylogenetic status, and species divergence time were compared or estimated. A total of 13,750 encoding sequences were successfully obtained from the transcriptome assembly of Yenyuan stream salamander, estimated to contain 40.1% of the unigenes represented in tetrapod databases. A total of 88.79% of these genes could be annotated to a biological function by current databases. Through gene family clustering, we found multiple possible isoforms of the Scribble gene&#8212;whose function is related to regeneration&#8212;based on sequence similarity. Meanwhile, we constructed a robust phylogenetic tree based on 56 single-copy orthologues, which indicates that based on phylogenetic position, the Yenyuan stream salamander presents the closest relationship with the Chinese giant salamander (Andrias davidianus) of the investigated vertebrates. Based on the fossil-calibrated phylogeny, we estimated that the lineage divergence between the ancestral Yenyuan stream salamander and the Chinese giant salamander may have occurred during the Cretaceous period (~78.4 million years ago). In conclusion, this study not only provides a candidate gene that is valuable for exploring the remarkable capacity of regeneration in the future, but also gives an interesting insight into the understanding of Yenyuan stream salamander by this first transcriptome assembly

    Characteristics of microscopic pore-throat structure of tight oil reservoirs in Sichuan Basin measured by rate-controlled mercury injection

    Based on the results of rate-controlled mercury-injection experiments, the microscopic pore-throat structure characteristics of tight sandstone in Sha-1 Section and tight limestone in Da’anzhai Section of Sichuan Basin were quantitatively characterized. The results show that the pore radius distribution characteristics of tight oil reservoirs are similar. The main distribution is between 100~190 μm, and the average pore radius is 160 μm. While the distribution of the throat radius of tight sandstone and limestone is quite different, the distribution of the throat of sandstone samples is relatively concentrated, and the distribution of the throat of limestone samples is relatively sparse. There is a good positive correlation between the average throat radius and permeability, but the correlation between fractal dimension and permeability is not obvious. This indicates that the permeability is mainly affected by the radius of the throat. The pore-throat ratio in tight oil reservoirs is relatively large, and the resistance to seepage is greater during development. Therefore, during the development of tight oil, measures should be taken to increase the radius of the throat, reduce the ratio of pore radius to pore-throat radius, and improve the seepage capacity of the reservoir, thereby improving the development of tight oil
