    Determination method of the structure size interval of dynamic similar models for predicting vibration characteristics of the isotropic sandwich plates

    A method is studied for determining the structure size interval of dynamic similar models of the isotropic sandwich plates. Firstly, a comparison between the two theories of plates, the Resineer theory and the Hoff theory, is conducted, including their governing equations and the ANSYS analytic solutions of frequency. The Resineer theory is chosen as the basic theory of this paper finally. Secondly, the scaling laws between the model and prototype of isotropic sandwich plate are established by combining the dimensional analysis and governing analysis. Both complete and incomplete geometric similarity conditions are discussed. Thirdly, the determination method of the structure size interval of the models is proposed. The nature vibration mode keeps the same and the nature frequency and harmonic response keep in proportion with the prototype of the sandwich plate. At last, the flow step of the intervals determination method is given

    Effect of Calcium Chloride on Energy Level and Quality of Tan Sheep Meat during Postmortem Aging

    Our purpose was to clarify the effect of CaCl2 treatment on the energy level and quality of Tan sheep meat during postmortem aging. Tan sheep hind leg meat was injected with 200 mmol/L CaCl2 solution, and aged at 4 ℃ for 0, 2, 4, 6, and 8 days. The protein expression of phosphofructokinase (PFKM) was detected by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Western blot to verify whether the glycolysis pathway could be activated by CaCl2. At the same time, the energy level and meat quality indexes were measured during postmortem aging to analyze the mechanism by which the glycolysis pathway affects the metabolic level and quality of Tan sheep meat. The results showed that the protein expression of PFKM decreased during postmortem aging, indicating activation of the glycolytic pathway. CaCl2 treatment accelerated the glycolysis process after slaughter, thereby promoting the decomposition of glycogen, increasing the production of lactic acid, and resulting in a rapid decrease in pH. Water-holding capacity (WHC), myofibril fragmentation index (MFI) and b* value were higher and shear force and a* were lower in the treatment group than in the control and blank groups. Therefore, CaCl2 could affect the color, WHC and tenderness of Tan sheep meat at different aging stages. By accelerating the decrease of pH and the accumulation of energy metabolites, the glycolytic pathway increased the level of energy metabolism during postmortem aging, while inhibiting the production of oxymyoglobin, activating the activity of calpain, changing the structure of myofibrils and shortening the space between muscle protein molecules, which accelerated the process of myofibril fragmentation and the hydrolysis of myofibrillar protein, and led to a deterioration in meat color and an increase in water loss and meat tenderness

    Postmortem Degradation of Qinchuan Beef Protein by Proteasome and Its Mediated Ubiquitin-Proteasome Pathway

    In this study, the Longissimus dorsi muscle of Qinchuan cattle was injected with the proteasome inhibitor MG-132 immediately postmortem and then stored at 4 ℃. The effect of the ubiquitin-proteasome pathway (UPP) on protein degradation as well as changes in the proteasome activity, ubiquitin content and microstructure of the muscle during postmortem storage was explored in order to provide theoretical support for precise postmortem regulation of beef quality. With the extension of storage time, proteasome activity was lower and the contents of total soluble protein and ubiquitin were higher in the MG-132 group than in the control group. The sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) results showed that the band intensity of total soluble proteins between 40 and 250 kDa was greater in the MG-132 group than in the control group; muscle structure was better preserved in the MG-132 group, and the Z line and the boundary between light and dark bands were clearer than those in the control group. The contents of total soluble protein and ubiquitin showed a significantly positive correlation (P < 0.05). In conclusion, postmortem injection of MG-132 inhibited the proteasome activity and the degradation of ubiquitinated proteins in the UPP in Qinchuan beef, which in turn altered protein degradation and attenuated muscle damage. This suggests that the UPP has a potential role in meat quality formation; the proteasome not only degrades proteins by itself alone to destroy beef myofibrillar structure, but also influences postmortem beef protein degradation through mediating the UPP, ultimately affecting postmortem beef quality

    Using histogram analysis of the intrinsic brain activity mapping to identify essential tremor

    BackgroundEssential tremor (ET) is one of the most common movement disorders. Histogram analysis based on brain intrinsic activity imaging is a promising way to identify ET patients from healthy controls (HCs) and further explore the spontaneous brain activity change mechanisms and build the potential diagnostic biomarker in ET patients.MethodsThe histogram features based on the Resting-state functional magnetic resonance imaging (Rs-fMRI) data were extracted from 133 ET patients and 135 well-matched HCs as the input features. Then, a two-sample t-test, the mutual information, and the least absolute shrinkage and selection operator methods were applied to reduce the feature dimensionality. Support vector machine (SVM), logistic regression (LR), random forest (RF), and k-nearest neighbor (KNN) were used to differentiate ET and HCs, and classification performance of the established models was evaluated by the mean area under the curve (AUC). Moreover, correlation analysis was carried out between the selected histogram features and clinical tremor characteristics.ResultsEach classifier achieved a good classification performance in training and testing sets. The mean accuracy and area under the curve (AUC) of SVM, LR, RF, and KNN in the testing set were 92.62%, 0.948; 92.01%, 0.942; 93.88%, 0.941; and 92.27%, 0.939, respectively. The most power-discriminative features were mainly located in the cerebello-thalamo-motor and non-motor cortical pathways. Correlation analysis showed that there were two histogram features negatively and one positively correlated with tremor severity.ConclusionOur findings demonstrated that the histogram analysis of the amplitude of low-frequency fluctuation (ALFF) images with multiple machine learning algorithms could identify ET patients from HCs and help to understand the spontaneous brain activity pathogenesis mechanisms in ET patients

    Chromosomal-level assembly of the Asian Seabass genome using long sequence reads and multi-layered scaffolding

    We report here the ~670 Mb genome assembly of the Asian seabass (Lates calcarifer), a tropical marine teleost. We used long-read sequencing augmented by transcriptomics, optical and genetic mapping along with shared synteny from closely related fish species to derive a chromosome-level assembly with a contig N50 size over 1 Mb and scaffold N50 size over 25 Mb that span ~90% of the genome. The population structure of L. calcarifer species complex was analyzed by re-sequencing 61 individuals representing various regions across the species' native range. SNP analyses identified high levels of genetic diversity and confirmed earlier indications of a population stratification comprising three clades with signs of admixture apparent in the South-East Asian population. The quality of the Asian seabass genome assembly far exceeds that of any other fish species, and will serve as a new standard for fish genomics

    The Asian arowana (<i>Scleropages formosus</i>) genome provides new insights into the evolution of an early lineage of teleosts

    The Asian arowana (Scleropages formosus), one of the world’s most expensive cultivated ornamental fishes, is an endangered species. It represents an ancient lineage of teleosts: the Osteoglossomorpha. Here, we provide a high-quality chromosome-level reference genome of a female golden-variety arowana using a combination of deep shotgun sequencing and high-resolution linkage mapping. In addition, we have also generated two draft genome assemblies for the red and green varieties. Phylogenomic analysis supports a sister group relationship between Osteoglossomorpha (bonytongues) and Elopomorpha (eels and relatives), with the two clades together forming a sister group of Clupeocephala which includes all the remaining teleosts. The arowana genome retains the full complement of eight Hox clusters unlike the African butterfly fish (Pantodon buchholzi), another bonytongue fish, which possess only five Hox clusters. Differential gene expression among three varieties provides insights into the genetic basis of colour variation. A potential heterogametic sex chromosome is identified in the female arowana karyotype, suggesting that the sex is determined by a ZW/ZZ sex chromosomal system. The high-quality reference genome of the golden arowana and the draft assemblies of the red and green varieties are valuable resources for understanding the biology, adaptation and behaviour of Asian arowanas

    Leukocyte Tyrosine Kinase Functions in Pigment Cell Development

    A fundamental problem in developmental biology concerns how multipotent precursors choose specific fates. Neural crest cells (NCCs) are multipotent, yet the mechanisms driving specific fate choices remain incompletely understood. Sox10 is required for specification of neural cells and melanocytes from NCCs. Like sox10 mutants, zebrafish shady mutants lack iridophores; we have proposed that sox10 and shady are required for iridophore specification from NCCs. We show using diverse approaches that shady encodes zebrafish leukocyte tyrosine kinase (Ltk). Cell transplantation studies show that Ltk acts cell-autonomously within the iridophore lineage. Consistent with this, ltk is expressed in a subset of NCCs, before becoming restricted to the iridophore lineage. Marker analysis reveals a primary defect in iridophore specification in ltk mutants. We saw no evidence for a fate-shift of neural crest cells into other pigment cell fates and some NCCs were subsequently lost by apoptosis. These features are also characteristic of the neural crest cell phenotype in sox10 mutants, leading us to examine iridophores in sox10 mutants. As expected, sox10 mutants largely lacked iridophore markers at late stages. In addition, sox10 mutants unexpectedly showed more ltk-expressing cells than wild-type siblings. These cells remained in a premigratory position and expressed sox10 but not the earliest neural crest markers and may represent multipotent, but partially-restricted, progenitors. In summary, we have discovered a novel signalling pathway in NCC development and demonstrate fate specification of iridophores as the first identified role for Ltk
