    Molecular cloning and characterization of a novel expressed sequence tag (EST) associated with fecundity in goats

    To screen the genes controlling the fecundity traits in goats, a DDRT-PCR technique was applied. We found a new EST which highly expressed in Chinese native prolific goat breed, Haimen goats. There exists a difference of EST expression level between the prolific and non-prolific goat breed, indicating EST might associate to fecundity in goats. A full-length cDNA with 2253 base pairs was obtained by the 3’- and 5’-RACE method based on the EST sequence encoding a protein segment of 201 amino acid residues. Tissue specific distribution and sequence analysis implicated the likely involvement of EST in the regulation of the hormones related to fecundity

    Influence of the dimensions of spheroniser plate protuberances on the production of pellets by extrusion-spheronisation

    This study systematically investigated the influence of the dimensions of square-patterned pyramidal protuberances on a spheronisation plate on pellet yield, size and shape distributions, surface tensile strength and surface morphology was investigated using a 45 wt% microcrystalline cellulose/ water paste. Tests were conducted using four different extrudate diameters (1.0, 1.5, 2.0 and 2.5 mm) generated by screen extrusion and seven plate geometries, including a flat plate as a control, allowing the relative size of extrudate to protuberance feature to be studied. Geometrical analyses of the protuberance shapes provide some insight into the observed differences, with protuberance angle being significant in many cases. Sharper protuberances reduced yield (promoting breakage and attrition) but tended to give narrower size distributions and more uneven pellet surface. Pellet yield for 1 mm extrudates was also subject to losses caused by fragments passing through the 1.0 mm gap between the plate and spheroniser wall. Pellet tensile strength was noticeably greater for 1.0 mm diameter extrudates, which is attributed to the greater extensional strain imparted on the paste during the extrusion step.     Evidence for genetic association of TBX21 and IFNG with systemic lupus erythematosus in a Chinese Han population

    TBX21 recode T-bet which is an important transcription factor that drives the Th1 immune response primarily by promoting expression of the interferon-gamma (IFNG) gene. Recent studies have shown that genetic variants in TBX21 and IFNG are connected with risk of systemic lupus erythematosus (SLE). The aim of the present study was to replicate these genetic associations with SLE in Anhui Chinese population. Genotyping of 3 variants (rs4794067 in TBX21, rs2069705 and rs2069718 in IFNG) was performed. A total of 3732 subjects were included in the final analysis. The study only identified the association of rs2069705 with SLE susceptibility (T vs. C: odds ratio [OR] = 1.12, 95% confidence interval [CI] = 1.00-1.26, P = 0.046). Combined analysis with Hong Kong GWAS showed that the OR for rs2069705 was 1.10 (95% CI: 1.01-1.21, P = 0.027). Further pooled analysis with Korean populations involving 10498 subjects showed a more significant association between rs2069705 and SLE (T vs. C: OR = 1.11, 95%CI = 1.04-1.19, P = 0.002; TT + TC vs. CC: OR = 1.11, 95%CI = 1.02-1.21, P = 0.012; TT vs. TC + CC: OR = 1.28, 95%CI = 1.07-1.54, P = 0.008; TT vs. CC: OR = 1.33, 95%CI = 1.10-1.60, P = 0.003). In addition, we also identified a significant genetic interaction between rs2069705 and rs4794067 in Anhui Chinese population. Our study suggests that IFNG and IFNG-TBX21 interaction are involved in SLE susceptibility.published_or_final_versio

    Development of a Cryogenic Calorimeter for Investigating Beam-Based Heat Load of Superconducting Undulators

    Superconducting undulators provide higher magmatic field to increase the brilliance and photon energy of synchrotron light sources. To quantify the amount of beam-based heat load of storage rings and optimize the design of cryogenic system, Lawrence Berkeley National Laboratory (LBNL) proposed a cryogenic calorimeter to perform the working condition of superconducting undulators. The calorimeter has been developed by Shanghai Institute of Applied Physics (SINAP) and installed on storage ring of Shanghai Synchrotron Radiation Facility (SSRF). Also, online experiments started in September of 2012. This paper describes the cryogenic system and beam-based heat load measurement system. Also, some measurement results are given in the paper

    SiC Nanowires Synthesized by Rapidly Heating a Mixture of SiO and Arc-Discharge Plasma Pretreated Carbon Black

    SiC nanowires have been synthesized at 1,600 °C by using a simple and low-cost method in a high-frequency induction furnace. The commercial SiO powder and the arc-discharge plasma pretreated carbon black were mixed and used as the source materials. The heating-up and reaction time is less than half an hour. It was found that most of the nanowires have core-shell SiC/SiO2nanostructures. The nucleation, precipitation, and growth processes were discussed in terms of the oxide-assisted cluster-solid mechanism

    Engineered polyketide biosynthesis and biocatalysis in Escherichia coli

    Polyketides are important bioactive natural products biosynthesized by bacteria, fungi, and plants. The enzymes that synthesize polyketides are collectively referred to as polyketide synthases (PKSs). Because many of the natural hosts that produce polyketides are difficult to culture or manipulate, establishing a universal heterologous host that is genetically tractable has become an important goal toward the engineered biosynthesis of polyketides and analogues. Here, we summarize the recent progresses in engineering Escherichia coli as a heterologous host for reconstituting PKSs of different types. Our increased understanding of PKS enzymology and structural biology, combined with new tools in protein engineering, metabolic engineering, and synthetic biology, has firmly established E. coli as a powerful host for producing polyketides

    China’s urban methane emissions from municipal wastewater treatment plant

    The increased number and capacity of municipal wastewater treatment plants (WWTPs) in China has driven the emission of methane (CH4). Few studies have focused on quantification of CH4 emissions from municipal WWTPs of different cities and analysis of socioeconomic factors influencing the quantity of emissions. Here we estimated CH4 emissions from WWTPs in China for 229 prefectural‐level cities, based on data from 2,019 working municipal WWTPs. The results show the total CH4 emissions to be 1,169.8 thousand tons (29.2 MtCO2e) in 2014, which is over three times that of the municipal WWTPs in the United States in 2016. Large cities along the east coast regions had larger CH4 emissions in absolute and per capita terms. Correlation analysis shows that cities with higher gross domestic product, household food consumption expenditure, or household consumption expenditure produced more degradable organics in wastewater, thus more CH4 emissions. Measures to control the sources of degradable organics and regulate WWTP processes with less emission factor are key to mitigate CH4 emissions. In addition to aerobic or anaerobic wastewater treatment systems, factors such as wastewater temperature, length of sewer, and the addition of nitrate that influencing emission factor are suggested to be involved in CH4 emission modeling

    The fibroblast mitogen platelet-derived growth factor -BB (PDGF-BB) induces a transient expression of the orphan nuclear receptor NR4A1 (also named Nur77, TR3 or NGFIB). The aim of the present study was to investigate the pathways through which NR4A1 is induced by PDGF-BB and its functional role. We demonstrate that in PDGF-BB stimulated NIH3T3 cells, the MEK1/2 inhibitor CI-1040 strongly represses NR4A1 expression, whereas Erk5 downregulation delays the expression, but does not block it. Moreover, we report that treatment with the NF-κB inhibitor BAY11-7082 suppresses NR4A1 mRNA and protein expression. The majority of NR4A1 in NIH3T3 was found to be localized in the cytoplasm and only a fraction was translocated to the nucleus after continued PDGF-BB treatment. Silencing NR4A1 slightly increased the proliferation rate of NIH3T3 cells; however, it did not affect the chemotactic or survival abilities conferred by PDGF-BB. Moreover, overexpression of NR4A1 promoted anchorage-independent growth of NIH3T3 cells and the glioblastoma cell lines U-105MG and U-251MG. Thus, whereas NR4A1, induced by PDGF-BB, suppresses cell growth on a solid surface, it increases anchorage-independent growth
