    Noncontingent and Response-Contingent Intravenous Ethanol Attenuates the Effect of Naltrexone on Hypothalamic-Pituitary-Adrenal Activity in Rhesus Monkeys

    Background : The mechanism by which the opioid antagonist naltrexone suppresses overconsumption of ethanol is unclear. Oral ethanol consumption in humans increases hypothalamic-pituitary-adrenal (HPA) activity, and recent studies suggest that naltrexone may reduce ethanol consumption by modifying the HPA-stimulating effects of ethanol. The purpose of this study was to measure in rhesus monkeys the effects of ethanol and naltrexone, alone and in combination, on plasma levels of adrenocorticotropin hormone (ACTH). Methods : Nine adult male and female rhesus monkeys with chronic, indwelling intravenous catheters were maintained on tethers that allowed ethanol delivery and blood sampling. In one study, the monkeys received intramuscular injections of saline or 0.32 mg/kg naltrexone followed by noncontingent intravenous bolus infusions of saline or 0.3 to 1.8 g/kg ethanol. In a second study, other monkeys were given intramuscular injections of saline or 0.01 to 0.3 mg/kg naltrexone and subsequently responded on levers to receive intravenous saline or ethanol 0.03 g/kg per injection. Results : Ethanol, delivered either response contingently or noncontingently, did not produce systematic changes in ACTH plasma levels. Naltrexone alone produced increases in plasma ACTH that were attenuated by the subsequent administration of noncontingent or response-contingent ethanol. Naltrexone also produced dose-dependent reductions in intravenous ethanol self-administration. Linear regression analysis indicated that ethanol intake was negatively correlated with the plasma levels of ACTH over time. Conclusions : The route of administration may modulate ethanol's effects on HPA activity. Ethanol may attenuate naltrexone's effect on the HPA axis by impairing HPA axis sensitivity to other stimuli. The negative correlation between ethanol intake and ACTH levels supports the notion that naltrexone's effect of increasing HPA axis activity may be related to its ability to suppress ethanol consumption.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/66078/1/01.ALC.0000121655.48922.C4.pd

    The role of Neuropilin-1 in COVID-19

    Neuropilin-1 (NRP-1), a member of a family of signaling proteins, was shown to serve as an entry factor and potentiate SARS Coronavirus 2 (SARS-CoV-2) infectivity in vitro. This cell surface receptor with its disseminated expression is important in angiogenesis, tumor progression, viral entry, axonal guidance, and immune function. NRP-1 is implicated in several aspects of a SARS-CoV-2 infection including possible spread through the olfactory bulb and into the central nervous system and increased NRP-1 RNA expression in lungs of severe Coronavirus Disease 2019 (COVID-19). Up-regulation of NRP-1 protein in diabetic kidney cells hint at its importance in a population at risk of severe COVID-19. Involvement of NRP-1 in immune function is compelling, given the role of an exaggerated immune response in disease severity and deaths due to COVID-19. NRP-1 has been suggested to be an immune checkpoint of T cell memory. It is unknown whether involvement and up-regulation of NRP-1 in COVID-19 may translate into disease outcome and long-term consequences, including possible immune dysfunction. It is prudent to further research NRP-1 and its possibility of serving as a therapeutic target in SARS-CoV-2 infections. We anticipate that widespread expression, abundance in the respiratory and olfactory epithelium, and the functionalities of NRP-1 factor into the multiple systemic effects of COVID-19 and challenges we face in management of disease and potential long-term sequelae

    Home Modification Outcomes in the Residences of Older People as a Result of Cougar Home Safety Assessment (Version 4.0) Recommendations

    The purpose of this study was to determine if recommendations made as a result of the administration of the Cougar Home Safety Assessment Version 4.0 (CHSA 4.0) were effective in causing older residents to make environmental safety modifications in their homes. Initial data were collected during the administration of the CHSA 4.0 in the homes of 40 older people in four northeastern states. After completing the initial assessments, the researchers provided the participants with recommendations for improving the environmental safety of their homes. Approximately one month later, each home was reassessed with the CHSA 4.0. Overall, improvement in environmental safety was demonstrated in the homes with the greatest increases being in fire safety, emergency phone number placement, and bathroom safety. A t-test demonstrated a significant gain in compliance (t = 7.8, p < .001) by comparing the difference between the mean initial and reassessment home safety scores. A very large effect size (Cohen’s d = 1.7) was also found, indicating a high magnitude of difference between initial and reassessment safety ratings. The assessment proved to be beneficial in increasing the environmental safety of homes

    The dependence of galaxy group star formation rates and metallicities on large scale environment

    We construct a sample of 75,863 star forming galaxies with robust metallicity and star formation rate measurements from the Sloan Digital Sky Survey Data Release 7 (SDSS DR7), from which we select a clean sample of compact group (CG) galaxies. The CGs are defined to be close configurations of at least 4 galaxies that are otherwise apparently isolated. Our selection results in a sample of 112 spectroscopically identified compact group galaxies, which can be further divided into groups that are either embedded within a larger structure, such as a cluster or large group, or truly isolated systems. The compact groups then serve as a probe into the influence of large scale environment on a galaxy's evolution, while keeping the local density fixed at high values. We find that the star formation rates (SFRs) of star forming galaxies in compact groups are significantly different between isolated and embedded systems. Galaxies in isolated systems show significantly enhanced SFR, relative to a control sample matched in mass and redshift, a trend not seen in the embedded systems. Galaxies in isolated systems exhibit a median SFR enhancement at fixed stellar mass of +0.07 \pm 0.03 dex. These dependences on large scale environment are small in magnitude relative to the apparent influence of local scale effects found in previous studies, but the significance of the difference in SFRs between our two samples constrains the effect of large scale environment to be non-zero. We find no significant change in the gas-phase interstellar metallicity for either the isolated or embedded compact group sample relative to their controls. However, simulated samples that include artificial offsets indicate that we are only sensitive to metallicity changes of log O/H >0.13 dex (at 99% confidence), which is considerably larger than the typical metallicity differences seen in previous environmental studies.Comment: Accepted for publication in MNRAS. 16 pages, 9 figure

    Young people’s experiences of COVID-19 messaging at the start of the UK lockdown:Lessons for positive engagement and information sharing

    BACKGROUND: To reduce COVID-19 infection rates during the initial stages of the pandemic, the UK Government mandated a strict period of restriction on freedom of movement or 'lockdown'. For young people, closure of schools and higher education institutions and social distancing rules may have been particularly challenging, coming at a critical time in their lives for social and emotional development. This study explored young people's experiences of the UK Government's initial response to the pandemic and related government messaging.METHODS: This qualitative study combines data from research groups at the University of Southampton, University of Edinburgh and University College London. Thirty-six online focus group discussions (FGDs) were conducted with 150 young people (Southampton: n = 69; FGD = 7; Edinburgh: n = 41; FGD = 5; UCL: n = 40; FGD = 24). Thematic analysis was conducted to explore how young people viewed the government's response and messaging and to develop recommendations for how to best involve young people in addressing similar crises in the future. RESULTS: The abrupt onset of lockdown left young people shocked, confused and feeling ignored by government and media messaging. Despite this, they were motivated to adhere to government advice by the hope that life might soon return to normal. They felt a responsibility to help with the pandemic response, and wanted to be productive with their time, but saw few opportunities to volunteer.CONCLUSIONS: Young people want to be listened to and feel they have a part to play in responding to a national crisis such as the COVID-19 epidemic. To reduce the likelihood of disenfranchising the next generation, Government and the media should focus on developing messaging that reflects young people's values and concerns and to provide opportunities for young people to become involved in responses to future crises

    Galaxy pairs in the Sloan Digital Sky Survey - IV: Interactions trigger AGN

    Galaxy-galaxy interactions are predicted to cause gas inflows leading to enhanced nuclear star formation. In this paper we test the further prediction that the gas inflows lead to enhanced accretion onto the central supermassive black hole, triggering activity in the nucleus. Based on a sample of 11,060 SDSS galaxies with a close companion (rp < 80 kpc, Delta V < 200 km/s), we classify AGN based either on emission line ratios or on spectral classification as a quasar. The AGN fraction in the close pairs sample is compared to a control sample of 110,600 mass- and redshift-matched control galaxies with no nearby companion. We find a clear increase in the AGN fraction in close pairs of galaxies with projected separations < 40 kpc by up to a factor of 2.5 relative to the control sample (although the enhancement depends on the chosen S/N cut of the sample). The increase in AGN fraction is strongest in equal mass galaxy pairings, and weakest in the lower mass component of an unequal mass pairing. The increased AGN fraction at small separations is accompanied by an enhancement in the number of `composite' galaxies whose spectra are the result of photoionization by both AGN and stars. Our results indicate that AGN activity occurs (at least in some cases) well before final coalescence and concurrently with ongoing star formation. Finally, we find a marked increase at small projected separations of the fraction of pairs in which BOTH galaxies harbour AGN. We demonstrate that the fraction of double AGN exceeds the expected random fraction, indicating that some pairs undergo correlated nuclear activity. Taken together with complimentary studies, we favour an interpretation where interactions trigger AGN, but are not the only cause of nuclear activity.Comment: Accepted for publication in MNRA
