    Athirat, Asherah, Ashratu: a reassessment according to the textual sources

    This dissertation was undertaken partially in response to previous studies on the goddess Asherah. These studies have tended to gather together information from the various cultures in which 'Asherah' appears, and the information is generally presented as a portrait of the goddess. This dissertation approaches the problem from a different perspective. The primary issue addressed is: did the goddess 'Asherah' develop in the same way in all the cultures in which she appears? In order to answer this question, this study considers the evidence as contained in the written records of the first two millennia B.C.E.The mythology preserved in the tablets written by Elimelek in ancient Ugarit is the primary source of information on the goddess Athirat. After considering this mythology, it should be possible to examine Athirat's role in other mythologies, and to attempt to distil her essential characteristics and nature.Within the Ugaritic mythology of Elimelek, she appears most active in the 'Palace of Baal' episode in the Baal Cycle. In this culture Athirat appears primarily in relationship to other gods. She is the consort of El, the head of the pantheon. In the Elimelek tablets her title is rbt alrt ym.This title indicates an unspecified relationship with the sea. She is the mother of the gods but does not, however, appear as an amorphous 'mother goddess'. Her role as a mother is limited to divine children and royal children. She appears to be the rabitu, the 'queen mother'. Although Athirat is associated with the head of the pantheon, she maintains a connection with mortal women. This may account for her emblem, which is a spindle.A goddess Asherah may appear in the Old Testament. Certain passages seem to require a goddess interpretation for the word asherah. In other texts asherah designates a cultic object. If Asherah does appear as a goddess in the Old Testament, her characteristics are difficult to discern.She does not, however, appear as the consort of Baal or as a fertility goddess.Ashratu appears in Mesopotamian sources. The information within these texts are our oldest records of the goddess, and point to her Amorite origins. She is the consort of Amurru and she is connected with the steppe.That she had a temple and active cultus is amply attested in the materials.Ashratu is also attested in a Hittite version of a Canaanite myth. She is known from a number of Epigraphic South Arabian inscriptions. These inscriptions may point to a solar nature in that culture.TThe Khirbet el-Qom and Kuntillet ÂŁAjrud inscriptions may refer to a goddess Asherah, but more likely they denote a cultic object. Their interpretation is uncertain since they cannot be explained adequately with our present knowledge of Hebrew grammar.Conclusions are drawn on the basis of the information available from these individual cultures, each within its own context

    IL-1α cleavage by inflammatory caspases of the noncanonical inflammasome controls the senescence-associated secretory phenotype.

    Interleukin-1 alpha (IL-1α) is a powerful cytokine that modulates immunity, and requires canonical cleavage by calpain for full activity. Mature IL-1α is produced after inflammasome activation and during cell senescence, but the protease cleaving IL-1α in these contexts is unknown. We show IL-1α is activated by caspase-5 or caspase-11 cleavage at a conserved site. Caspase-5 drives cleaved IL-1α release after human macrophage inflammasome activation, while IL-1α secretion from murine macrophages only requires caspase-11, with IL-1ÎČ release needing caspase-11 and caspase-1. Importantly, senescent human cells require caspase-5 for the IL-1α-dependent senescence-associated secretory phenotype (SASP) in vitro, while senescent mouse hepatocytes need caspase-11 for the SASP-driven immune surveillance of senescent cells in vivo. Together, we identify IL-1α as a novel substrate of noncanonical inflammatory caspases and finally provide a mechanism for how IL-1α is activated during senescence.     Can Observation Skills of Citizen Scientists Be Estimated Using Species Accumulation Curves?

    Volunteers are increasingly being recruited into citizen science projects to collect observations for scientific studies. An additional goal of these projects is to engage and educate these volunteers. Thus, there are few barriers to participation resulting in volunteer observers with varying ability to complete the project’s tasks. To improve the quality of a citizen science project’s outcomes it would be useful to account for inter-observer variation, and to assess the rarely tested presumption that participating in a citizen science projects results in volunteers becoming better observers. Here we present a method for indexing observer variability based on the data routinely submitted by observers participating in the citizen science project eBird, a broad-scale monitoring project in which observers collect and submit lists of the bird species observed while birding. Our method for indexing observer variability uses species accumulation curves, lines that describe how the total number of species reported increase with increasing time spent in collecting observations. We find that differences in species accumulation curves among observers equates to higher rates of species accumulation, particularly for harder-to-identify species, and reveals increased species accumulation rates with continued participation. We suggest that these properties of our analysis provide a measure of observer skill, and that the potential to derive post-hoc data-derived measurements of participant ability should be more widely explored by analysts of data from citizen science projects. We see the potential for inferential results from analyses of citizen science data to be improved by accounting for observer skill

    Conflict over African land – between small holders and large industrial farmers and between domestic farmers and global agribusinesses – raises key questions about who will make the best use of African land and which farmers do most to decrease poverty and produce more food, industrial inputs, and exports. Zimbabwe has already gone through two major changes in land occupation, and thus provides an important test of what is the 'best' use of the land. Three measures of 'best' use have been cited in Zimbabwe: reward for military victory, poverty reduction, and agricultural production.     Genome-wide association studies (GWAS) have begun to identify the common genetic component to ischaemic stroke (IS). However, IS has considerable phenotypic heterogeneity. Where clinical covariates explain a large fraction of disease risk, covariate informed designs can increase power to detect associations. As prevalence rates in IS are markedly affected by age, and younger onset cases may have higher genetic predisposition, we investigated whether an age-at-onset informed approach could detect novel associations with IS and its subtypes; cardioembolic (CE), large artery atherosclerosis (LAA) and small vessel disease (SVD) in 6,778 cases of European ancestry and 12,095 ancestry-matched controls. Regression analysis to identify SNP associations was performed on posterior liabilities after conditioning on age-at-onset and affection status. We sought further evidence of an association with LAA in 1,881 cases and 50,817 controls, and examined mRNA expression levels of the nearby genes in atherosclerotic carotid artery plaques. Secondly, we performed permutation analyses to evaluate the extent to which age-at-onset informed analysis improves significance for novel loci. We identified a novel association with an MMP12 locus in LAA (rs660599; p = 2.5×10⁻⁷), with independent replication in a second population (p = 0.0048, OR(95% CI) = 1.18(1.05-1.32); meta-analysis p = 2.6×10⁻⁞). The nearby gene, MMP12, was significantly overexpressed in carotid plaques compared to atherosclerosis-free control arteries (p = 1.2×10⁻Âč⁔; fold change = 335.6). Permutation analyses demonstrated improved significance for associations when accounting for age-at-onset in all four stroke phenotypes (p<0.001). Our results show that a covariate-informed design, by adjusting for age-at-onset of stroke, can detect variants not identified by conventional GWAS

    Genetic overlap between diagnostic subtypes of ischemic stroke

    Background and Purpose: Despite moderate heritability, the phenotypic heterogeneity of ischemic stroke has hampered gene discovery, motivating analyses of diagnostic subtypes with reduced sample sizes. We assessed evidence for a shared genetic basis among the 3 major subtypes: large artery atherosclerosis (LAA), cardioembolism, and small vessel disease (SVD), to inform potential cross-subtype analyses. Methods: Analyses used genome-wide summary data for 12 389 ischemic stroke cases (including 2167 LAA, 2405 cardioembolism, and 1854 SVD) and 62 004 controls from the Metastroke consortium. For 4561 cases and 7094 controls, individual-level genotype data were also available. Genetic correlations between subtypes were estimated using linear mixed models and polygenic profile scores. Meta-analysis of a combined LAA-SVD phenotype (4021 cases and 51 976 controls) was performed to identify shared risk alleles.     The first demonstration of laser action in ruby was made in 1960 by T. H. Maiman of Hughes Research Laboratories, USA. Many laboratories worldwide began the search for lasers using different materials, operating at different wavelengths. In the UK, academia, industry and the central laboratories took up the challenge from the earliest days to develop these systems for a broad range of applications. This historical review looks at the contribution the UK has made to the advancement of the technology, the development of systems and components and their exploitation over the last 60 years
