    Some aspects of its post-roman influence on the landscape

    Hadrian's Wall is the longest single stone built ancient monument in the United Kingdom and its influence on the man-made landscape has been greater than that of any other surviving structure. Over the last century and a half archaeological excavation and research have largely pieced together the history and function of the monument as it pertains to the Roman period. Little in-depth research has been undertaken regarding its function in and influence on the landscape from the end of the Roman period to the present day. This study has attempted to understand the role that various factors including: geology, topography, population distribution, building types, farming practices, local history and national politics have had on the survival or destruction of the Wall and the influence that the Wall has had on such things as the place-names of settlements, field names, land forms, and even in the naming of houses, streets and businesses. The Wall is referred to in early 8th century manuscripts, Norman charters as well as medieval documents, charters, maps and estate plans and is described in varying detail by antiquarian sources. The present landscape along the Wall owes its existence to past generations and their close relationship to the Wall of Hadrian

    The flow of the Antarctic circumpolar current over the North Scotia Ridge

    The transports associated with the Subantarctic Front (SAF) and the Polar Front (PF) account for the majority of the volume transport of the Antarctic Circumpolar Current (ACC). After passing through Drake Passage, the SAF and the PF veer northward over the steep topography of the North Scotia Ridge. Interaction of the ACC with the North Scotia Ridge influences the sources of the Malvinas Current. This ridge is a major obstacle to the flow of deep water, with the majority of the deep water passing through the 3100 m deep gap in the ridge known as Shag Rocks Passage. Volume transports associated with these fronts were measured during the North Scotia Ridge Overflow Project, which included the first extensive hydrographic survey of the ridge, carried out in April and May 2003. The total net volume transport northward over the ridge was found to be . The total net transport associated with the SAF was approximately , and the total transport associated with the PF was approximately . Weddell Sea Deep Water was not detected passing through Shag Rocks Passage, contrary to some previous inferences

    UTILISING PROBABILISTIC TECHNIQUES IN THE ASSESSMENT OF EXTREME COASTAL FLOODING FREQUENCY- MAGNITUDE RELATIONSHIPS USING A CASE STUDY FROM SOUTH-WEST ENGLAND

    Full version unavailable due to 3rd party copyright restrictions.Recent events such as the New Orleans floods and the Japanese tsunami of 2011 have highlighted the uncertainty in the quantification of the magnitude of natural hazards. The research undertaken here has focussed on the uncertainty in evaluating storm surge magnitudes based on a range of statistical techniques including the Generalised Extreme Value distribution, Joint Probability and Monte Carlo simulations. To support the evaluation of storm surge frequency magnitude relationships a unique hard copy observed sea level data set, recording hourly observations, was acquired and digitised for Devonport, Plymouth, creating a 40 year data set. In conjunction with Devonport data, Newlyn (1915-2012) tide gauge records were analysed, creating a data set of 2 million data points. The different statistical techniques analysed led to an uncertainty range of 0.4 m for a 1 in 250 year storm surge event, and 0.7 m for a 1 in 1000 storm surge event. This compares to a 0.5 m uncertainty range between the low and high prediction for sea level rise by 2100. The Geographical Information system modelling of the uncertainty indicated that for a 1 in 1000 year event the level uncertainty (0.7 m) led to an increase of 100% of buildings and 50% of total land affect. Within the study area of south-west England there are several critical structures including a nuclear licensed site. Incorporating the uncertainty in storm surge and wave height predictions indicated that the site would be potentially affected today with the combination of a 1 in 1000 year storm surge event coincident with a 1 in 1000 wave. In addition to the evaluation of frequency magnitude relations this study has identified several trends in the data set. Over the data period sea level rise is modelled as an exponential growth (0.0001mm/yr2), indicating the modelled sea level rise of 1.9 mm/yr and 2.2 mm/yr for Newlyn and Devonport, will potentially increase over the next century by a minimum of 0.2 m by 2100.The increase in storm frequency identified as part of this analysis has been equated to the rise in sea level, rather than an increase in the severity of storms, with decadal variations in the observed frequency, potentially linked to the North Atlantic Oscillation. The identification as part of this study of a significant uncertainty in the evaluation of storm surge frequency magnitude relationships has global significance in the evaluation of natural hazards. Guidance on the evaluation of external hazards currently does not adequately consider the effect of uncertainty; an uncertainty of 0.7 m identified within this study could potentially affect in the region of 500 million people worldwide living close to the coast

    SPR-measured dissociation kinetics of PROTAC ternary complexes influence target degradation rate

    Bifunctional degrader molecules, known as proteolysis-targeting chimeras (PROTACs), function by recruiting a target to an E3 ligase, forming a target/PROTAC/ligase ternary complex. Despite the importance of this key intermediate species, no detailed validation of a method to directly determine binding parameters for ternary complex kinetics has been reported, and it remains to be addressed whether tuning the kinetics of PROTAC ternary complexes may be an effective strategy to improve the efficiency of targeted protein degradation. Here, we develop an SPR-based assay to quantify the stability of PROTAC-induced ternary complexes by measuring for the first time the kinetics of their formation and dissociation <i>in vitro</i> using purified proteins. We benchmark our assay using four PROTACs that target the bromodomains (BDs) of bromodomain and extraterminal domain proteins Brd2, Brd3, and Brd4 to the von Hippel–Lindau E3 ligase (VHL). We reveal marked differences in ternary complex off-rates for different PROTACs that exhibit either positive or negative cooperativity for ternary complex formation relative to binary binding. The positively cooperative degrader MZ1 forms comparatively stable and long-lived ternary complexes with either Brd4<sup>BD2</sup> or Brd2<sup>BD2</sup> and VHL. Equivalent complexes with Brd3<sup>BD2</sup> are destabilized due to a single amino acid difference (Glu/Gly swap) present in the bromodomain. We observe that this difference in ternary complex dissociative half-life correlates to a greater initial rate of intracellular degradation of Brd2 and Brd4 relative to Brd3. These findings establish a novel assay to measure the kinetics of PROTAC ternary complexes and elucidate the important kinetic parameters that drive effective target degradation

    Long-term monitoring of Scripps’s Murrelet and Guadalupe Murrelet at San Clemente Island, California: evaluation of baseline data in 2012–2016

    San Clemente Island (SCI) supports one of the smallest Scripps’s Murrelet (Synthliboramphus scrippsi; SCMU) colonies in the world, and perhaps the only colony of Guadalupe Murrelets (S. hypoleucus; GUMU) in California. In 2012–2016, the U.S. Navy sponsored development of a long-term murrelet monitoring program at SCI that utilized nocturnal spotlight surveys, night-lighting at-sea captures, and nest monitoring. Standardized spotlight survey transects were established in nearshore waters off breeding areas at Seal Cove and southeast SCI (SESCI). Baseline mean spotlight counts were 29 ± 15 murrelets (n = 31) at Seal Cove in 2013–2016 and 21 ± 10 murrelets (n = 15) at SESCI in 2014–2016. We banded 201 SCMU captured in congregations at Seal Cove (n = 158) and SESCI (n = 43); 12% of the SCMU from Seal Cove and 7% from SESCI were recaptured ≥1 year after banding. We also banded 21 GUMU at Seal Cove, but none were recaptured. Murrelet nests or eggs were found in 6 shoreline breeding “refuges” at Seal Cove and SESCI that were seldom if ever visited by island foxes (Urocyon littoralis clementae) and feral cats (Felis catus). Incubating SCMU were observed in 4 nest sites, but in 8 other sites only eggs or eggshells were found. Overall hatching success was very low (12%; n = 17 clutches) in 2012–2016, apparently due to intraspecific competition for limited nest crevices at Seal Cove and predation (or possibly abandonment and subsequent egg scavenging) by foxes or black rats (Rattus rattus) at SESCI. Using spotlight survey data, we estimated 115 murrelet pairs (range 79–208) at SCI, including 110 pairs (range 76–199) of SCMU and 5 pairs (range 3–9) of GUMU, although a GUMU nest has not yet been found. Power analyses of Seal Cove spotlight data indicated that surveys conducted over 9 nights per year for 20 years could reliably (power ≥ 0.90) detect minimum population changes of ± 1.7% per annum. Additional efforts are needed to (1) confirm the breeding status of GUMU; (2) investigate alternative methods of rat control to increase hatching success in murrelet breeding refuges; and (3) enhance breeding habitats to reduce intraspecific competition for nest sites and increase the number of monitored nests

    Jet-Induced Emission-Line Nebulosity and Star Formation in the High-Redshift Radio Galaxy 4C41.17

    The high redshift radio galaxy 4C41.17 consists of a powerful radio source in which previous work has shown that there is strong evidence for jet-induced star formation along the radio axis. We argue that nuclear photoionization is not responsible for the excitation of the emission line clouds and we construct a jet-cloud interaction model to explain the major features revealed by the data. The interaction of a high-powered jet with a dense cloud in the halo of 4C41.17 produces shock-excited emission-line nebulosity through ~1000 km/s shocks and induces star formation. The CIII to CIV line ratio and the CIV luminosity emanating from the shock, imply that the pre-shock density in the line-emitting cloud is high enough (~1-10 cm^-3) that shock initiated star formation could proceed on a timescale of order a few x 10^6 yrs, well within the estimated dynamical age of the radio source. Broad (FWHM ~ 100 - 1400 km/s) emission lines are attributed to the disturbance of the gas cloud by a partial bow--shock and narrow emission lines (FWHM ~ 500 - 650 km/s) (in particular CIV) arise in precursor emission in relatively low metallicity gas. The implied baryonic mass ~ 8 \times 10^{10} solar masses of the cloud is high and implies that Milky Way size condensations existed in the environments of forming radio galaxies at a redshift of 3.8. Our interpretation of the data provides a physical basis for the alignment of the radio, emission-line and UV continuum images in some of the highest redshift radio galaxies and the analysis presented here may form a basis for the calculation of densities and cloud masses in other high redshift radio galaxies.Comment: 18 pages, 5 figures; uses astrobib.sty and aaspp4.sty.
