Východiskem práce je analýza vnějšího a vnitřního prostředí, pomocí níž jsou zachyceny a popsány nejdůležitější vlivy, které na ni působí. Na základě výsledků je navržena strategie pro posílení současného postavení společnosti a její další rozvoj.This diploma thesis is focused on the creation of strategy for small company and the increase of its competitiveness. The base of this thesis is the analysis of external and internal environment, where are the most important influences noticed and described. On the basis of the results the strategy is suggested. This strategy leads to strengthening of current company position and its next development.

Táňa Dluhošová, Ph.D. Abstrakt: Diplomová práce si dává za cíl analyzovat odlišné diskurzy o pandemii COVID-19 na Taiwanu v období od propuknutí nemoci na konci roku 2019 až do konce roku 2021. V této době došlo k objevení viru v čínském Wuhanu (Wuhan 武汉), zamlčování závažnosti situace čínskou vládou, rozšíření viru do podoby světové pandemie a k následné reakci států a jejich vlád. Taiwan byl dlouhou dobu dáván za příklad zvládnutí pandemie nulovou tolerancí, zavedením přísných kontrol a opatření, které mnohé světové vlády obdivovaly. Jak však byla pandemie vnímána a prezentována taiwanskými politickými špičkami? Jak se jednotlivé politické strany k pandemii postavily? A jak to vše bylo podáváno taiwanskými novinami? K odpovědi na tyto otázky bude kromě sekundární literatury použita také diskurzivní analýza vybraných čínsky psaných textů (vyšší stovky článků) týkajících se pandemie ze dvou největších taiwanských periodik: 1) noviny Ziyou shibao ( 自 由 時 報 ), které se přiklání k Demokratické pokrokové straně a 2) Lianhe bao (聯合報), které se přiklání k Národní straně, Kuomintangu. Zároveň se práce zaměří na to, jak situaci prezentují v anglickém jazyce...The aim of the thesis is to analyze different discourses about the COVID-19 pandemic in Taiwan in the period from the outbreak of the virus at the end of 2019 to the end of 2021. During this period, the virus was discovered in chinese Wuhan (Wuhan 武汉) and turned into a global pandemic and caused reactions of states and their governments. Taiwan has long been cited as an example of good practice with zero-case policy, introducing strict controls and measures that many world governments have admired. But how was the pandemic perceived and presented by Taiwanese political leaders? How did the individual political parties react to the pandemic? And how was it all communicated by Taiwanese newspapers? To answer these question, the theses - in addition to secondary literature - will utilize discourse analysis of selected Chinese-language newspaper articles related to the pandemic from the two largest Taiwanese periodicals (up to several hundred item): 1) Ziyou shibao (自由 時報) close to the Democratic Progressive Party, and 2) Lianhe bao (聯合 報) leaning towards the National Party, the Kuomintang.     Treatment of Pelvic Ring Fracture Using Spinopelvic Fixation (Types of Fixation, Indications, Functional Results, Mechanical Characteristics)

    Při vysokoenergetickém poranění (dopravní nehoda, pád z výše) může dojít k poranění pánevního kruhu s vertikální nestabilitou a/nebo k poranění křížové kosti se spinopelvickou nestabilitou. Takové nestability ohrožují základní funkce pánevního kruhu a lumbosakrální junkce. Přes takto poraněné segmenty není možný adekvátní přenos zátěže trupu a končetin. Řešením je repozice a stabilizace, kterou docílíme obnovy anatomických poměrů a šetrného a pevného spojení. Spinopelvická disociace provázející v některých případech zlomeninu pánevního kruhu a/nebo sakra je úrazem složitým s obtížným řešením. Četnost výskytu je uváděna v literatuře 2,9- 3,1 %. Jejich počet je na vzestupu při vyšší hustotě a agresivitě dopravního provozu, při velké oblibě nebezpečných lidských aktivit a při zrychlujícím se životním tempu. Většina těchto zlomenin je léčena operačně, neoperační léčení je vyhrazeno pouze pro pacienty, kteří nejsou schopni podstoupit operaci z interních či lokálních příčin. V našem sledovaném souboru jsme zhodnotili skupinu pacientů se zlomeninou pánevního kruhu a/nebo sakra se spinopelvickou disociací, kteří byli léčeni na našem pracovišti v období 2009-2019. V tomto období bylo zde operačně léčeno 222 pacientů se zlomeninou pánevního kruhu. Celkový počet pacientů řešených spinopelvickou fixací byl 15....In the case of a high-energy injury (traffic accident, fall from above), a pelvic ring injury with vertical instability and/or a sacrum injury with spinopelvic instability may occur. Such instabilities threaten the basic functions of the pelvic ring and lumbosacral junction. Adequate transmission of body and limb load is not possible through such injured segments. The solution is some form of reduction and stabilization to achieve the restoration of anatomical conditions and a gentle and firm connection. Spinopelvic dissociation in some cases accompanying pelvic ring fractures and / or damages is a complex injury with a difficult solution. The frequency of occurrence is reported in the literature 2.9-3.1 %. The number of such accidents is on the rise with higher density and aggressiveness of traffic, the great popularity of dangerous human activities and the accelerating pace of life. Most of these fractures are treated surgically, non-surgical treatment is reserved only for patients who are unable to undergo surgery for internal or local reasons. In our study group, we evaluated a group of patients with a pelvic ring fracture and / or sacral fracture with spinopelvic dissociation who were treated at our workplace in the period 2009- 2019.     Mitochondrial and plasma membrane potential of cultured cerebellar neurons during glutamate-induced necrosis, apoptosis, and tolerance.

    A failure of mitochondrial bioenergetics has been shown to be closely associated with the onset of apoptotic and necrotic neuronal injury. Here, we developed an automated computational model that interprets the single-cell fluorescence for tetramethylrhodamine methyl ester (TMRM) as a consequence of changes in either delta psi(m) or delta psi(p), thus allowing for the characterization of responses for populations of single cells and subsequent statistical analysis. Necrotic injury triggered by prolonged glutamate excitation resulted in a rapid monophasic or biphasic loss of delta psi(m) that was closely associated with a loss of delta psi(p) and a rapid decrease in neuronal NADPH and ATP levels. Delayed apoptotic injury, induced by transient glutamate excitation, resulted in a small, reversible decrease in TMRM fluorescence, followed by a sustained hyperpolarization of delta psi(m) as confirmed using the delta psi(p)-sensitive anionic probe DiBAC2(3). This hyperpolarization of delta psi(m) was closely associated with a significant increase in neuronal glucose uptake, NADPH availability, and ATP levels. Statistical analysis of the changes in delta psi(m) or delta psi(p) at a single-cell level revealed two major correlations; those neurons displaying a more pronounced depolarization of delta psi(p) during the initial phase of glutamate excitation entered apoptosis more rapidly, and neurons that displayed a more pronounced hyperpolarization of delta psi(m) after glutamate excitation survived longer. Indeed, those neurons that were tolerant to transient glutamate excitation (18%) showed the most significant increases in delta psi(m). Our results indicate that a hyperpolarization of delta psi(m) is associated with increased glucose uptake, NADPH availability, and survival responses during excitotoxic injury

    Latrepirdine/Dimebon is a small-molecule compound with attributed neurocognitive-enhancing activities, which has recently been tested in clinical trials for the treatment of Alzheimer\u27s and Huntington\u27s disease. Latrepirdine has been suggested to be a neuroprotective agent that increases mitochondrial function, however the molecular mechanisms underlying these activities have remained elusive. We here demonstrate that latrepirdine, at (sub)nanomolar concentrations (0.1 nM), activates the energy sensor AMP-activated protein kinase (AMPK). Treatment of primary neurons with latrepirdine increased intracellular ATP levels and glucose transporter 3 translocation to the plasma membrane. Latrepirdine also increased mitochondrial uptake of the voltage-sensitive probe TMRM. Gene silencing of AMPKα or its upstream kinases, LKB1 and CaMKKβ, inhibited this effect. However, studies using the plasma membrane potential indicator DisBAC2(3) demonstrated that the effects of latrepirdine on TMRM uptake were largely mediated by plasma membrane hyperpolarization, precluding a purely \u27mitochondrial\u27 mechanism of action. In line with a stabilizing effect of latrepirdine on plasma membrane potential, pretreatment with latrepirdine reduced spontaneous Ca(2+) oscillations as well as glutamate-induced Ca(2+) increases in primary neurons, and protected neurons against glutamate toxicity. In conclusion, our experiments demonstrate that latrepirdine is a potent activator of AMPK, and suggest that one of the main pharmacological activities of latrepirdine is a reduction in neuronal excitability
