Mediterranean isolation preconditioning the Earth System for late Miocene climate cooling
© 2019, The Author(s). A global Neogene cooling trend culminated ~7 million years ago with the onset of Greenland glaciation. Increased ocean-atmosphere interaction and low- to high-latitude circulation are thought to be key factors in reorganizing late Miocene global temperature and precipitation patterns, but the drivers of this reorganization have yet to be identified. Here, we present new information about the evolution of the Atlantic-Mediterranean gateway that generated Mediterranean overflow. We use sedimentary and palaeogeographic evidence to constrain the timing and dimensions of this gateway and document the initiation of a saline plume of water within the North Atlantic. Today, this saline jet entrains and transports Eastern North Atlantic water and its dissolved inorganic carbon into the interior of the ocean, contributing to the drawdown of CO 2 and the sensitivity of the ocean to atmospheric changes. We show that during the Miocene this transport emerged simultaneously with gateway restriction and propose that the resulting interaction of ocean-surface and ocean-interior carbon inventories would have greatly enhanced ocean-atmosphere exchange, preconditioning the Earth System for late Miocene cooling

The Gibraltar Corridor:Watergate of the Messinian Salinity Crisis
The existence and evolution of a Messinian salt giant in the Mediterranean Sea has caused much debate in the marine science community. Especially the suggestion that the Mediterranean was a deep desiccated basin during the Messinian Salinity Crisis (MSC, 5.97–5.33 Ma), triggered by a temporal disconnection from the global ocean, made it a well-known crisis beyond the scientific boundaries. Approximately ~50 years after this provocative statement, it remained unknown which Mediterranean–Atlantic seaway delivered the 5–6% of the global ocean's salt into the Mediterranean basin. Here, we review the changes in Mediterranean-Atlantic connectivity throughout the late Miocene in order to locate, date and quantify the missing Messinian gateway that provided the salt water inflow during the MSC. We conclude that all the known pre-MSC gateways through southern Spain and northern Morocco were closed, leaving the “Gibraltar Corridor” at its Messinian configuration as the sole candidate. We consider the possibility of longer and narrower straits existing at depth below the present Gibraltar region, and using strait dynamic theory we calculate its dimensions during the Messinian based on the salinity changes in the Mediterranean. A marine Messinian gateway through the Gibraltar Corridor is in agreement with growing evidence that Atlantic waters reached the Mediterranean Sea during all three stages of the MSC
Genome Biology / Genome and transcriptome analysis of the Mesoamerican common bean and the role of gene duplications in establishing tissue and temporal specialization of genes
Background:
Legumes are the third largest family of angiosperms and the second most important crop class. Legume genomes have been shaped by extensive large-scale gene duplications, including an approximately 58 million year old whole genome duplication shared by most crop legumes.
Results:
We report the genome and the transcription atlas of coding and non-coding genes of a Mesoamerican genotype of common bean (Phaseolus vulgaris L., BAT93). Using a comprehensive phylogenomics analysis, we assessed the past and recent evolution of common bean, and traced the diversification of patterns of gene expression following duplication. We find that successive rounds of gene duplications in legumes have shaped tissue and developmental expression, leading to increased levels of specialization in larger gene families. We also find that many long non-coding RNAs are preferentially expressed in germ-line-related tissues (pods and seeds), suggesting that they play a significant role in fruit development. Our results also suggest that most bean-specific gene family expansions, including resistance gene clusters, predate the split of the Mesoamerican and Andean gene pools.
Conclusions:
The genome and transcriptome data herein generated for a Mesoamerican genotype represent a counterpart to the genomic resources already available for the Andean gene pool. Altogether, this information will allow the genetic dissection of the characters involved in the domestication and adaptation of the crop, and their further implementation in breeding strategies for this important crop
No difference in penetrance between truncating and missense/aberrant splicing pathogenic variants in mlh1 and msh2: A prospective lynch syndrome database study
Background. Lynch syndrome is the most common genetic predisposition for hereditary cancer. Carriers of pathogenic changes in mismatch repair (MMR) genes have an increased risk of developing colorectal (CRC), endometrial, ovarian, urinary tract, prostate, and other cancers, depending on which gene is malfunctioning. In Lynch syndrome, differences in cancer incidence (penetrance) according to the gene involved have led to the stratification of cancer surveillance. By contrast, any differences in penetrance determined by the type of pathogenic variant remain unknown. Objective. To determine cumulative incidences of cancer in carriers of truncating and missense or aberrant splicing pathogenic variants of the MLH1 and MSH2 genes. Methods. Carriers of pathogenic variants of MLH1 (path_MLH1) and MSH2 (path_MSH2) genes filed in the Prospective Lynch Syndrome Database (PLSD) were categorized as truncating or missense/aberrant splicing according to the InSiGHT criteria for pathogenicity. Results. Among 5199 carriers, 1045 had missense or aberrant splicing variants, and 3930 had truncating variants. Prospective observation years for the two groups were 8205 and 34,141 years, respectively, after which there were no significant differences in incidences for cancer overall or for colorectal cancer or endometrial cancers separately. Conclusion. Truncating and missense or aberrant splicing pathogenic variants were associated with similar average cumulative incidences of cancer in carriers of path MLH1 and path_MSH2.Fil: Dominguez Valentin, Mev. Uptake of hysterectomy and bilateral salpingooophorectomy in carriers of pathogenic mismatch repair variants:a Prospective Lynch Syndrome Database report
Purpose: This study aimed to report the uptake of hysterectomy and/or bilateral salpingo-oophorectomy (BSO) to prevent gynaecological cancers (risk-reducing surgery [RRS]) in carriers of pathogenic MMR (path_MMR) variants. Methods: The Prospective Lynch Syndrome Database (PLSD) was used to investigate RRS by a cross-sectional study in 2292 female path_MMR carriers aged 30-69 years. Results: Overall, 144, 79, and 517 carriers underwent risk-reducing hysterectomy, BSO, or both combined, respectively. Two-thirds of procedures before 50 years of age were combined hysterectomy and BSO, and 81% of all procedures included BSO. Risk-reducing hysterectomy was performed before age 50 years in 28%, 25%, 15%, and 9%, and BSO in 26%, 25%, 14% and 13% of path_MLH1, path_MSH2, path_MSH6, and path_PMS2 carriers, respectively. Before 50 years of age, 107 of 188 (57%) BSO and 126 of 204 (62%) hysterectomies were performed in women without any prior cancer, and only 5% (20/392) were performed simultaneously with colorectal cancer (CRC) surgery. Conclusion: Uptake of RRS before 50 years of age was low, and RRS was rarely undertaken in association with surgical treatment of CRC. Uptake of RRS aligned poorly with gene-and age-associated risk estimates for endometrial or ovarian cancer that were published recently from PLSD and did not correspond well with current clinical guidelines. The reasons should be clarified. Decision-making on opting for or against RRS and its timing should be better aligned with predicted risk and mortality for endometrial and ovarian cancer in Lynch syn-drome to improve outcomes.
