Exploring alternative purchasing strategies: just-in-time or just enough?
What are the prevalent purchasing strategies used by manufacturing firms to purchase components that are critical to the quality of their most important products? This research reports the findings from data on purchasing strategies collected from 248 companies. The data indicate that although firms seem to be moving away from a transaction-based purchasing strategy towards partnership relations necessary for successful just-in-time strategies, firms are likely to embrace one of four hybrid purchasing strategies that on a spectrum would fall somewhere between the two pure strategies. These identified strategies offer purchasing managers viable alternatives to moving directly into a just-in-time environment

The beginning of time? Evidence for catastrophic drought in Baringo in the early nineteenth century
New developments in the collection of palaeo-data over the past two decades have transformed our understanding of climate and environmental history in eastern Africa. This article utilises instrumental and proxy evidence of historical lake-level fluctuations from Baringo and Bogoria, along with other Rift Valley lakes, to document the timing and magnitude of hydroclimate variability at decadal to century time scales since 1750. These data allow us to construct a record of past climate variation not only for the Baringo basin proper, but also across a sizable portion of central and northern Kenya. This record is then set alongside historical evidence, from oral histories gathered amongst the peoples of northern Kenya and the Rift Valley and from contemporary observations recorded by travellers through the region, to offer a reinterpretation of human activity and its relationship to environmental history in the nineteenth century. The results reveal strong evidence of a catastrophic drought in the early nineteenth century, the effects of which radically alters our historical understanding of the character of settlement, mobility and identity within the Baringo–Bogoria basin
BLAST05: Power Spectra of Bright Galactic Cirrus at Submillimeter Wavelengths
We report multi-wavelength power spectra of diffuse Galactic dust emission
from BLAST observations at 250, 350, and 500 microns in Galactic Plane fields
in Cygnus X and Aquila. These submillimeter power spectra statistically
quantify the self-similar structure observable over a broad range of scales and
can be used to assess the cirrus noise which limits the detection of faint
point sources. The advent of submillimeter surveys with the Herschel Space
Observatory makes the wavelength dependence a matter of interest. We show that
the observed relative amplitudes of the power spectra can be related through a
spectral energy distribution (SED). Fitting a simple modified black body to
this SED, we find the dust temperature in Cygnus X to be 19.9 +/- 1.3 K and in
the Aquila region 16.9 +/- 0.7 K. Our empirical estimates provide important new
insight into the substantial cirrus noise that will be encountered in

A shared population of epidemic methicillin-resistant Staphylococcus aureus 15 circulates in humans and companion animals.
UNLABELLED: Methicillin-resistant Staphylococcus aureus (MRSA) is a global human health problem causing infections in both hospitals and the community. Companion animals, such as cats, dogs, and horses, are also frequently colonized by MRSA and can become infected. We sequenced the genomes of 46 multilocus sequence type (ST) 22 MRSA isolates from cats and dogs in the United Kingdom and compared these to an extensive population framework of human isolates from the same lineage. Phylogenomic analyses showed that all companion animal isolates were interspersed throughout the epidemic MRSA-15 (EMRSA-15) pandemic clade and clustered with human isolates from the United Kingdom, with human isolates basal to those from companion animals, suggesting a human source for isolates infecting companion animals. A number of isolates from the same veterinary hospital clustered together, suggesting that as in human hospitals, EMRSA-15 isolates are readily transmitted in the veterinary hospital setting. Genome-wide association analysis did not identify any host-specific single nucleotide polymorphisms (SNPs) or virulence factors. However, isolates from companion animals were significantly less likely to harbor a plasmid encoding erythromycin resistance. When this plasmid was present in animal-associated isolates, it was more likely to contain mutations mediating resistance to clindamycin. This finding is consistent with the low levels of erythromycin and high levels of clindamycin used in veterinary medicine in the United Kingdom. This study furthers the "one health" view of infectious diseases that the pathogen pool of human and animal populations are intrinsically linked and provides evidence that antibiotic usage in animal medicine is shaping the population of a major human pathogen. IMPORTANCE: Methicillin-resistant Staphylococcus aureus (MRSA) is major problem in human medicine. Companion animals, such as cats, dogs, and horses, can also become colonized and infected by MRSA. Here, we demonstrate that a shared population of an important and globally disseminated lineage of MRSA can infect both humans and companion animals without undergoing host adaptation. This suggests that companion animals might act as a reservoir for human infections. We also show that the isolates from companion animals have differences in the presence of certain antibiotic resistance genes. This study furthers the "one health" view of infectious diseases by demonstrating that the pool of MRSA isolates in the human and animal populations are shared and highlights how different antibiotic usage patterns between human and veterinary medicine can shape the population of bacterial pathogens.This work was supported by a Medical Research Council Partnership

A Novel Flow Cytometric HTS Assay Reveals Functional Modulators of ATP Binding Cassette Transporter ABCB6
ABCB6 is a member of the adenosine triphosphate (ATP)-binding cassette family of transporter proteins that is increasingly recognized as a relevant physiological and therapeutic target. Evaluation of modulators of ABCB6 activity would pave the way toward a more complete understanding of the significance of this transport process in tumor cell growth, proliferation and therapy-related drug resistance. In addition, this effort would improve our understanding of the function of ABCB6 in normal physiology with respect to heme biosynthesis, and cellular adaptation to metabolic demand and stress responses. To search for modulators of ABCB6, we developed a novel cell-based approach that, in combination with flow cytometric high-throughput screening (HTS), can be used to identify functional modulators of ABCB6. Accumulation of protoporphyrin, a fluorescent molecule, in wild-type ABCB6 expressing K562 cells, forms the basis of the HTS assay. Screening the Prestwick Chemical Library employing the HTS assay identified four compounds, benzethonium chloride, verteporfin, tomatine hydrochloride and piperlongumine, that reduced ABCB6 mediated cellular porphyrin levels. Validation of the identified compounds employing the hemin-agarose affinity chromatography and mitochondrial transport assays demonstrated that three out of the four compounds were capable of inhibiting ABCB6 mediated hemin transport into isolated mitochondria. However, only verteporfin and tomatine hydrochloride inhibited ABCB6’s ability to compete with hemin as an ABCB6 substrate. This assay is therefore sensitive, robust, and suitable for automation in a high-throughput environment as demonstrated by our identification of selective functional modulators of ABCB6. Application of this assay to other libraries of synthetic compounds and natural products is expected to identify novel modulators of ABCB6 activity
The Balloon-Borne Large Aperture Submillimeter Telescope (BLAST) 2005: A 10 deg^2 Survey of Star Formation in Cygnus X
We present Cygnus X in a new multi-wavelength perspective based on an
unbiased BLAST survey at 250, 350, and 500 micron, combined with rich datasets
for this well-studied region. Our primary goal is to investigate the early
stages of high mass star formation. We have detected 184 compact sources in
various stages of evolution across all three BLAST bands. From their
well-constrained spectral energy distributions, we obtain the physical
properties mass, surface density, bolometric luminosity, and dust temperature.
Some of the bright sources reaching 40 K contain well-known compact H II
regions. We relate these to other sources at earlier stages of evolution via
the energetics as deduced from their position in the luminosity-mass (L-M)
diagram. The BLAST spectral coverage, near the peak of the spectral energy
distribution of the dust, reveals fainter sources too cool (~ 10 K) to be seen
by earlier shorter-wavelength surveys like IRAS. We detect thermal emission
from infrared dark clouds and investigate the phenomenon of cold ``starless
cores" more generally. Spitzer images of these cold sources often show stellar
nurseries, but these potential sites for massive star formation are ``starless"
in the sense that to date there is no massive protostar in a vigorous accretion
phase. We discuss evolution in the context of the L-M diagram. Theory raises
some interesting possibilities: some cold massive compact sources might never
form a cluster containing massive stars; and clusters with massive stars might
not have an identifiable compact cold massive precursor.Comment: 42 pages, 31 Figures, 6 table
