    Transcriptional profiling of colicin-induced cell death of Escherichia coli MG1655 identifies potential mechanisms by which bacteriocins promote bacterial diversity

    We report the transcriptional response of Escherichia coli MG1655 to damage induced by colicins E3 and E9, bacteriocins that kill cells through inactivation of the ribosome and degradation of chromosomal DNA, respectively. Colicin E9 strongly induced the LexA-regulated SOS response, while colicin E3 elicited a broad response that included the induction of cold shock genes, symptomatic of translational arrest. Colicin E3 also increased the transcription of cryptic prophage genes and other laterally acquired mobile elements. The transcriptional responses to both these toxins suggest mechanisms that may promote genetic diversity in E. coli populations, pointing to a more general role for colicins in adaptive bacterial physiology than has hitherto been realized

    Dissolved oxygen dynamics during a phytoplankton bloom in the Ross Sea polynya

    The Ross Sea polynya is one of the most productive regions in the Southern Ocean. However, limited access and high spatio-temporal variability of physical and biological processes limit the use of conventional oceanographic methods to measure early season primary productivity. High-resolution observations from two Seagliders provide insights into the timing of a bloom in the southern Ross Sea polynya in December 2010. Changes in chlorophyll and oxygen concentrations are used to assess bloom dynamics. Using a ratio of dissolved oxygen to carbon, net primary production is estimated over the duration of the bloom showing a sensitive balance between net autotrophy and heterotrophy. The two gliders, observing spatially distinct regions during the same period, found net community production rates of -0.9±0.7 and 0.7±0.4 g C m-2 d-1. The difference highlights the spatial variability of biological processes and is probably caused by observing different stages of the bloom. The challenge of obtaining accurate primary productivity estimates highlights the need for increased observational efforts, particularly focusing on subsurface processes not resolved using surface or remote observations. Without an increased observational effort and the involvement of emerging technologies, it will not be possible to determine the seasonal trophic balance of the Ross Sea polynya and quantify the shelf's importance in carbon export

    A Preliminary Evaluation of Deer Behavior and Nontarget Animal Use Associated with the 4-Poster Tickicide Device on Long Island, New York

    The 4-Poster device is a host-applied tick control technology. White-tailed deer (Odocoileus virginianus) are attracted to devices by whole kernel corn, and when feeding, each animal contacts rollers that apply an acaricide to the animal\u27s head and neck. The blacklegged tick (Ixodes scapularis), and lone star tick (Amblyomma americanum), are both nuisance and medical pests on Long Island, and deer are a keystone host for both species. By targeting adult ticks where they are most commonly found, deer application is highly effective for concentrating acaricide in key target areas, compared to conventional lawn spraying of pesticides. However, attracting deer to baited stations may have negative impacts including: 1) potential damage to vegetation near the deployed stations in both natural and residential settings; 2) possible increases in deer road crossings, leading to more vehicle collisions; 3) increased deer survivorship or fecundity due to supplemental feeding or reduced tick pressure; and 4) impacts of feeding stations on nontarget wildlife. We are completing the first year of a multi-year project, and report nontarget wildlife use at 4-Poster devices, and deer home ranges for areas with, and without 4-Poster feeding stations. These data will provide management information concerning the collateral effects of deploying 4-Poster devices to reduce tick abundance

    Spin Waves in Detwinned BaFe2_2As2_2

    Understanding magnetic interactions in the parent compounds of high-temperature superconductors forms the basis for determining their role for the mechanism of superconductivity. For parent compounds of iron pnictide superconductors such as AAFe2_2As2_2 (A=A= Ba, Ca, Sr), although spin excitations have been mapped out throughout the entire Brillouin zone (BZ), measurements were carried out on twinned samples and did not allow for a conclusive determination of the spin dynamics. Here we use inelastic neutron scattering to completely map out spin excitations of \sim100\% detwinned BaFe2_2As2_2. By comparing observed spectra with theoretical calculations, we conclude that the spin excitations can be well described by an itinerant model with important contributions from electronic correlations.Comment: 6 pages, 4 figures, with supplemental materia

    Dark matter annihilation and decay in dwarf spheroidal galaxies: The classical and ultrafaint dSphs

    Dwarf spheroidal (dSph) galaxies are prime targets for present and future gamma-ray telescopes hunting for indirect signals of particle dark matter. The interpretation of the data requires careful assessment of their dark matter content in order to derive robust constraints on candidate relic particles. Here, we use an optimised spherical Jeans analysis to reconstruct the `astrophysical factor' for both annihilating and decaying dark matter in 21 known dSphs. Improvements with respect to previous works are: (i) the use of more flexible luminosity and anisotropy profiles to minimise biases, (ii) the use of weak priors tailored on extensive sets of contamination-free mock data to improve the confidence intervals, (iii) systematic cross-checks of binned and unbinned analyses on mock and real data, and (iv) the use of mock data including stellar contamination to test the impact on reconstructed signals. Our analysis provides updated values for the dark matter content of 8 `classical' and 13 `ultrafaint' dSphs, with the quoted uncertainties directly linked to the sample size; the more flexible parametrisation we use results in changes compared to previous calculations. This translates into our ranking of potentially-brightest and most robust targets---viz., Ursa Minor, Draco, Sculptor---, and of the more promising, but uncertain targets---viz., Ursa Major 2, Coma---for annihilating dark matter. Our analysis of Segue 1 is extremely sensitive to whether we include or exclude a few marginal member stars, making this target one of the most uncertain.     Candidates for asteroid dust trails

    The contribution of different sources to the circumsolar dust cloud (known as the zodiacal cloud) can be deduced from diagnostic observations. We used the Spitzer Space Telescope to observe the diffuse thermal emission of the zodiacal cloud near the ecliptic. Several structures were identified in these observations, including previously known asteroid dust bands, which are thought to have been produced by recent asteroid collisions, and cometary trails. Interestingly, two of the detected dust trails, denoted t1 and t2 here, cannot be linked to any known comet. Trails t1 and t2 represent a much larger integrated brightness than all known cometary trails combined and may therefore be major contributors to the circumsolar dust cloud. We used our Spitzer observations to determine the orbits of these trails and were able to link them to two ("orphan" or type II) trails that were discovered by the Infrared Astronomical Satellite (IRAS) in 1983. The orbits of trails t1 and t2 that we determined by combining the Spitzer and IRAS data have semimajor axes, eccentricities, and inclinations like those of the main-belt asteroids. We therefore propose that trails t1 and t2 were produced by very recent (<~100 kyr old) collisional breakups of small, <~10 km diameter main-belt asteroids

    MultiTurnCleanup: A Benchmark for Multi-Turn Spoken Conversational Transcript Cleanup

However, numerous discontinuity phenomena in spoken conversational transcripts occur across multiple turns, hampering human readability and the performance of downstream NLP tasks. This study addresses these phenomena by proposing an innovative Multi-Turn Cleanup task for spoken conversational transcripts and collecting a new dataset, MultiTurnCleanup1. We design a data labeling schema to collect the high-quality dataset and provide extensive data analysis.     Phantom and clinical evaluation of the effect of full Monte Carlo collimator modelling in post-SIRT yttrium-90 Bremsstrahlung SPECT imaging

    Background: Post-therapy SPECT/CT imaging of Y-90 microspheres delivered to hepatic malignancies is difficult, owing to the continuous, high-energy Bremsstrahlung spectrum emitted by Y-90. This study aimed to evaluate the utility of a commercially available software package (HybridRecon, Hermes Medical Solutions AB) which incorporates full Monte Carlo collimator modelling. Analysis of image quality was performed on both phantom and clinical images in order to ultimately provide a recommendation of an optimum reconstruction for post-therapy Y-90 microsphere SPECT/CT imaging. A 3D-printed anthropomorphic liver phantom was filled with Y-90 with a sphere-to-background ratio of 4:1 and imaged on a GE Discovery 670 SPECT/CT camera. Datasets were reconstructed using ordered-subsets expectation maximization (OSEM) 1-7 iterations in order to identify the optimal OSEM reconstruction (5 iterations, 15 subsets). Quantitative analysis was subsequently carried out on phantom datasets obtained using four reconstruction algorithms: the default OSEM protocol (2 iterations, 10 subsets) and the optimised OSEM protocol, both with and without full Monte Carlo collimator modelling. The quantitative metrics contrast recovery (CR) and background variability (BV) were calculated. The four algorithms were then used to retrospectively reconstruct 10 selective internal radiation therapy (SIRT) patient datasets which were subsequently blind scored for image quality by a consultant radiologist. Results: The optimised OSEM reconstruction (5 iterations, 15 subsets with full MC collimator modelling) increased the CR by 42% (p <0.001) compared to the default OSEM protocol (2 iterations, 10 subsets). The use of full Monte Carlo collimator modelling was shown to further improve CR by 14% (30 mm sphere, CR = 90%, p <0.05). The consultant radiologist had a significant preference for the optimised OSEM over the default OSEM protocol (p <0. 001), with the optimised OSEM being the favoured reconstruction in every one of the 10 clinical cases presented. Conclusions: OSEM (5 iterations, 15 subsets) with full Monte Carlo collimator modelling is quantitatively the optimal image reconstruction for post-SIRT 90Y Bremsstrahlung SPECT/CT imaging. The use of full Monte Carlo collimator modelling for correction of image-degrading effects significantly increases contrast recovery without degrading clinical image quality.Peer reviewe

    Strategy for Expanding Nutrition Professionals’ Competency: A Pilot Case Study in Dissemination and Implementation Science Training

    Dissemination and Implementation (D&I) science trainings are essential to build knowledge among a variety of current and future health professionals. The objective of this study was to pilot-test and assess implementation of a nutrition-specific D&I science training. Participants (students enrolled in nutrition and public health programs) completed pre/post surveys and exit interviews. Descriptive statistics and a qualitative thematic analysis used deductive coding; in which coding and theme development are directed by existing concepts. Initial coding was completed by one researcher and validated by an additional researcher to describe and provide examples of the categories the Kirkpatrick Model and Implementation Outcomes Framework. The evaluation of the training was positively supported through the Kirkpatrick Scale results (mean scores between 6.94 ± 1.7 (Learning) and 7.35 ± 1.9 (Reaction)) and qualitative findings (increased confidence in D&I science and positive feedback on active learning strategies (application-based learning, mentorship, and discussions). Participants (n=8) described the learning activities (case studies, discussions, projects), the structure of the course (flipped classroom, content, learning strategies), the setting (hybrid, online), and mentorship (continuous feedback on assignments) as enabling effective implementation, which reflects with positive Implementation Outcome findings (3.59 ± 1.26, appropriateness score 3.94 ± 0.85, and feasibility score of 4.09 ± 0.67). These findings support positive implementation feasibility and program evaluation. Future studies need to compare changes in knowledge, attitudes, and beliefs among current or future nutrition professionals before and after completing this training

    A Cure for HIV Infection: "Not in My Lifetime" or "Just Around the Corner"?

    With the advent and stunning success of combination antiretroviral therapy (ART) to prolong and improve quality of life for persons with HIV infection, HIV research has been afforded the opportunity to pivot towards studies aimed at finding "a cure." The mere idea that cure of HIV might be possible has energized researchers and the community towards achieving this goal. Funding agencies, both governmental and private, have targeted HIV cure as a high priority; many in the field have responded to these initiatives and the cure research agenda is robust. In this "salon" two editors of Pathogens and Immunity, Michael Lederman and Daniel Douek ask whether curing HIV is a realistic, scalable objective. We start with an overview perspective and have asked a number of prominent HIV researchers to add to the discussion
