    A comparison of three systemic accident analysis methods using 46 SPAD (Signals Passed at Danger) incidents

    During the period 1996-2003 there were five fatal accidents on the UK railway network, three of which were Signals Passed at Danger (SPAD) events (Watford Junction, 1996; Southall, 1997; Ladbroke Grove, 1999). SPAD events vary in severity and whilst most are not fatal there is the potential to cause serious injuries to passengers and train staff and damage to railway infra-structure. This paper investigates how the current system accident analysis tool used within the railway, the Incident Factor Classification System (IFCS) identifies and analyses causal factors of SPAD events. To evaluate the effectiveness IFCS was used to analysis SPAD incident reports (n=46) and the outputs were compared with two systemic accident analysis methods and relevant outputs (the Human Factors Analysis and Classification System – HFACS and Acci-Maps). The initial reporting process proved to hinder all systemic accident analysis methods in the extraction of causal factors. However, once extracted, all system accident analysis methods were successful in categorizing causal factors and demonstrated various outputs to illustrate the findings

    Fast and reliable storage using a 5 bit, nonvolatile photonic memory cell

Available from Optical Society of America via the DOI in this record.Optically storing and addressing data on photonic chips is of particular interest as such capability would eliminate optoelectronic conversion losses in data centers. It would also enable on-chip non-von Neumann photonic computing by allowing multinary data storage with high fidelity. Here, we demonstrate such an optically addressed, multilevel memory capable of storing up to 34 nonvolatile reliable and repeatable levels (over 5 bits) using the phase change material Ge2Sb2Te5 integrated on a photonic waveguide. Crucially, we demonstrate for the first time, to the best of our knowledge, a technique that allows us to program the device with a single pulse regardless of the previous state of the material, providing an order of magnitude improvement over previous demonstrations in terms of both time and energy consumption. We also investigate the influence of write-and-erase pulse parameters on the single-pulse recrystallization, amorphization, and readout error in our multilevel memory, thus tailoring pulse properties for optimum performance.     Modal Ω-Logic: Automata, Neo-Logicism, and Set-Theoretic Realism

    This essay examines the philosophical significance of Ω\Omega-logic in Zermelo-Fraenkel set theory with choice (ZFC). The duality between coalgebra and algebra permits Boolean-valued algebraic models of ZFC to be interpreted as coalgebras. The modal profile of Ω\Omega-logical validity can then be countenanced within a coalgebraic logic, and Ω\Omega-logical validity can be defined via deterministic automata. I argue that the philosophical significance of the foregoing is two-fold. First, because the epistemic and modal profiles of Ω\Omega-logical validity correspond to those of second-order logical consequence, Ω\Omega-logical validity is genuinely logical, and thus vindicates a neo-logicist conception of mathematical truth in the set-theoretic multiverse. Second, the foregoing provides a modal-computational account of the interpretation of mathematical vocabulary, adducing in favor of a realist conception of the cumulative hierarchy of sets

    Entropy Projection Curved Gabor with Random Forest and SVM for Face Recognition

    In this work, we propose a workflow for face recognition under occlusion using the entropy projection from the curved Gabor filter, and create a representative and compact features vector that describes a face. Despite the reduced vector obtained by the entropy projection, it still presents opportunity for further dimensionality reduction. Therefore, we use a Random Forest classifier as an attribute selector, providing a 97% reduction of the original vector while keeping suitable accuracy. A set of experiments using three public image databases: AR Face, Extended Yale B with occlusion and FERET illustrates the proposed methodology, evaluated using the SVM classifier. The results obtained in the experiments show promising results when compared to the available approaches in the literature, obtaining 98.05% accuracy for the complete AR Face, 97.26% for FERET and 81.66% with Yale with 50% occlusion

    Recurrent De Novo NAHR Reciprocal Duplications in the ATAD3 Gene Cluster Cause a Neurogenetic Trait with Perturbed Cholesterol and Mitochondrial Metabolism.

    Recent studies have identified both recessive and dominant forms of mitochondrial disease that result from ATAD3A variants. The recessive form includes subjects with biallelic deletions mediated by non-allelic homologous recombination. We report five unrelated neonates with a lethal metabolic disorder characterized by cardiomyopathy, corneal opacities, encephalopathy, hypotonia, and seizures in whom a monoallelic reciprocal duplication at the ATAD3 locus was identified. Analysis of the breakpoint junction fragment indicated that these 67 kb heterozygous duplications were likely mediated by non-allelic homologous recombination at regions of high sequence identity in ATAD3A exon 11 and ATAD3C exon 7. At the recombinant junction, the duplication allele produces a fusion gene derived from ATAD3A and ATAD3C, the protein product of which lacks key functional residues. Analysis of fibroblasts derived from two affected individuals shows that the fusion gene product is expressed and stable. These cells display perturbed cholesterol and mitochondrial DNA organization similar to that observed for individuals with severe ATAD3A deficiency. We hypothesize that the fusion protein acts through a dominant-negative mechanism to cause this fatal mitochondrial disorder. Our data delineate a molecular diagnosis for this disorder, extend the clinical spectrum associated with structural variation at the ATAD3 locus, and identify a third mutational mechanism for ATAD3 gene cluster variants. These results further affirm structural variant mutagenesis mechanisms in sporadic disease traits, emphasize the importance of copy number analysis in molecular genomic diagnosis, and highlight some of the challenges of detecting and interpreting clinically relevant rare gene rearrangements from next-generation sequencing data

    The ‘SELection End points in Communities of bacTeria’ (SELECT) Method: A Novel Experimental Assay to Facilitate Risk Assessment of Selection for Antimicrobial Resistance in the Environment

A growing body of research demonstrates selection for AMR likely occurs at environmental concentrations of antibiotics. However, no standardized experimental approaches for determining selective concentrations of antimicrobials currently exist, preventing appropriate environmental and human health risk assessment of AMR. Objectives: We aimed to design a rapid, simple, and cost-effective novel experimental assay to determine selective effect concentrations of antibiotics and to generate the largest experimental data set of selective effect concentrations of antibiotics to date. Methods: Previously published methods and data were used to validate the assay, which determines the effect concentration based on reduction of bacterial community (wastewater) growth. Risk quotients for test antibiotics were generated to quantify risk. Results: The assay (SELection End points in Communities of bacTeria, or the SELECT method) was used to rapidly determine selective effect concentrations of antibiotics. These were in good agreement with quantitative polymerase chain reaction effect concentrations determined within the same experimental system. The SELECT method predicted no effect concentrations were minimally affected by changes in the assay temperature, growth media, or microbial community used as the inoculum. The predicted no effect concentrations for antibiotics tested ranged from 0.05μg/L for ciprofloxacin to 1,250μg/L for erythromycin. Discussion: The lack of evidence demonstrating environmental selection for AMR, and of associated human health risks, is a primary reason for the lack of action in the mitigation of release of antibiotics into the aquatic environment. We present a novel method that can reliably and rapidly fill this data gap to enable regulation and subsequent mitigation (where required) to lower the risk of selection for, and human exposure to, AMR in aquatic environments.     Proximity curves for potential-based clustering

    YesThe concept of proximity curve and a new algorithm are proposed for obtaining clusters in a finite set of data points in the finite dimensional Euclidean space. Each point is endowed with a potential constructed by means of a multi-dimensional Cauchy density, contributing to an overall anisotropic potential function. Guided by the steepest descent algorithm, the data points are successively visited and removed one by one, and at each stage the overall potential is updated and the magnitude of its local gradient is calculated. The result is a finite sequence of tuples, the proximity curve, whose pattern is analysed to give rise to a deterministic clustering. The finite set of all such proximity curves in conjunction with a simulation study of their distribution results in a probabilistic clustering represented by a distribution on the set of dendrograms. A two-dimensional synthetic data set is used to illustrate the proposed potential-based clustering idea. It is shown that the results achieved are plausible since both the ‘geographic distribution’ of data points as well as the ‘topographic features’ imposed by the potential function are well reflected in the suggested clustering. Experiments using the Iris data set are conducted for validation purposes on classification and clustering benchmark data. The results are consistent with the proposed theoretical framework and data properties, and open new approaches and applications to consider data processing from different perspectives and interpret data attributes contribution to patterns

    Using item response theory to explore the psychometric properties of extended matching questions examination in undergraduate medical education

    BACKGROUND: As assessment has been shown to direct learning, it is critical that the examinations developed to test clinical competence in medical undergraduates are valid and reliable. The use of extended matching questions (EMQ) has been advocated to overcome some of the criticisms of using multiple-choice questions to test factual and applied knowledge. METHODS: We analysed the results from the Extended Matching Questions Examination taken by 4th year undergraduate medical students in the academic year 2001 to 2002. Rasch analysis was used to examine whether the set of questions used in the examination mapped on to a unidimensional scale, the degree of difficulty of questions within and between the various medical and surgical specialties and the pattern of responses within individual questions to assess the impact of the distractor options. RESULTS: Analysis of a subset of items and of the full examination demonstrated internal construct validity and the absence of bias on the majority of questions. Three main patterns of response selection were identified. CONCLUSION: Modern psychometric methods based upon the work of Rasch provide a useful approach to the calibration and analysis of EMQ undergraduate medical assessments. The approach allows for a formal test of the unidimensionality of the questions and thus the validity of the summed score. Given the metric calibration which follows fit to the model, it also allows for the establishment of items banks to facilitate continuity and equity in exam standards

    Understanding the dynamics of Toll-like Receptor 5 response to flagellin and its regulation by estradiol

    © 2017 The Author(s). Toll-like receptors (TLRs) are major players of the innate immune system. Once activated, they trigger a signalling cascade that leads to NF-ΰ B translocation from the cytoplasm to the nucleus. Single cell analysis shows that NF-ΰ B signalling dynamics are a critical determinant of transcriptional regulation. Moreover, the outcome of innate immune response is also affected by the cross-talk between TLRs and estrogen signalling. Here, we characterized the dynamics of TLR5 signalling, responsible for the recognition of flagellated bacteria, and those changes induced by estradiol in its signalling at the single cell level. TLR5 activation in MCF7 cells induced a single and sustained NF-k B translocation into the nucleus that resulted in high NF-k B transcription activity. The overall magnitude of NF-k B transcription activity was not influenced by the duration of the stimulus. No significant changes are observed in the dynamics of NF-k B translocation to the nucleus when MCF7 cells are incubated with estradiol. However, estradiol significantly decreased NF-k B transcriptional activity while increasing TLR5-mediated AP-1 transcription. The effect of estradiol on transcriptional activity was dependent on the estrogen receptor activated. This fine tuning seems to occur mainly in the nucleus at the transcription level rather than affecting the translocation of the NF-k B transcription factor
