    Fractional Langevin equation

    We investigate fractional Brownian motion with a microscopic random-matrix model and introduce a fractional Langevin equation. We use the latter to study both sub- and superdiffusion of a free particle coupled to a fractal heat bath. We further compare fractional Brownian motion with the fractal time process. The respective mean-square displacements of these two forms of anomalous diffusion exhibit the same power-law behavior. Here we show that their lowest moments are actually all identical, except the second moment of the velocity. This provides a simple criterion which enables to distinguish these two non-Markovian processes.Comment: 4 page

    Novel genetic loci associated with hippocampal volume

    The hippocampal formation is a brain structure integrally involved in episodic memory, spatial navigation, cognition and stress responsiveness. Structural abnormalities in hippocampal volume and shape are found in several common neuropsychiatric disorders. To identify the genetic underpinnings of hippocampal structure here we perform a genome-wide association study (GWAS) of 33,536 individuals and discover six independent loci significantly associated with hippocampal volume, four of them novel. Of the novel loci, three lie within genes (ASTN2, DPP4 and MAST4) and one is found 200 kb upstream of SHH. A hippocampal subfield analysis shows that a locus within the MSRB3 gene shows evidence of a localized effect along the dentate gyrus, subiculum, CA1 and fissure. Further, we show that genetic variants associated with decreased hippocampal volume are also associated with increased risk for Alzheimer's disease (rg =-0.155). Our findings suggest novel biological pathways through which human genetic variation influences hippocampal volume and risk for neuropsychiatric illness

    Risk profiles and one-year outcomes of patients with newly diagnosed atrial fibrillation in India: Insights from the GARFIELD-AF Registry.

    BACKGROUND: The Global Anticoagulant Registry in the FIELD-Atrial Fibrillation (GARFIELD-AF) is an ongoing prospective noninterventional registry, which is providing important information on the baseline characteristics, treatment patterns, and 1-year outcomes in patients with newly diagnosed non-valvular atrial fibrillation (NVAF). This report describes data from Indian patients recruited in this registry. METHODS AND RESULTS: A total of 52,014 patients with newly diagnosed AF were enrolled globally; of these, 1388 patients were recruited from 26 sites within India (2012-2016). In India, the mean age was 65.8 years at diagnosis of NVAF. Hypertension was the most prevalent risk factor for AF, present in 68.5% of patients from India and in 76.3% of patients globally (P < 0.001). Diabetes and coronary artery disease (CAD) were prevalent in 36.2% and 28.1% of patients as compared with global prevalence of 22.2% and 21.6%, respectively (P < 0.001 for both). Antiplatelet therapy was the most common antithrombotic treatment in India. With increasing stroke risk, however, patients were more likely to receive oral anticoagulant therapy [mainly vitamin K antagonist (VKA)], but average international normalized ratio (INR) was lower among Indian patients [median INR value 1.6 (interquartile range {IQR}: 1.3-2.3) versus 2.3 (IQR 1.8-2.8) (P < 0.001)]. Compared with other countries, patients from India had markedly higher rates of all-cause mortality [7.68 per 100 person-years (95% confidence interval 6.32-9.35) vs 4.34 (4.16-4.53), P < 0.0001], while rates of stroke/systemic embolism and major bleeding were lower after 1 year of follow-up. CONCLUSION: Compared to previously published registries from India, the GARFIELD-AF registry describes clinical profiles and outcomes in Indian patients with AF of a different etiology. The registry data show that compared to the rest of the world, Indian AF patients are younger in age and have more diabetes and CAD. Patients with a higher stroke risk are more likely to receive anticoagulation therapy with VKA but are underdosed compared with the global average in the GARFIELD-AF.     Geomorphological characterization, remote sensing monitoring, and modeling of a slow-moving landslide in Alcoy (Southern Spain)

    In the present paper, an active landslide located to the southeast of the urban area of Alcoy (Alicante, Spain) is analysed. This landslide affects the N-340 road, as well as an area in which some industrial buildings dated from the fifteenth century and catalogued as Places of Cultural Interest are placed. This region is characterised by the existence of a steep relief composed of materials from the Miocene to the present, on which many gravitational movements are developed. The displacements have been analysed using high-resolution spotlight mode SAR images captured by the Spanish SAR satellite PAZ (which operates at X-band), which were processed by advanced differential SAR Interferometry (A-DInSAR). The behaviour of the instability has been determined using InSAR-derived time series displacement maps. Additionally, the study has been completed with a detailed geomorphological mapping at 1:2000 scale and an analysis of structural damage carried out in situ. The landslide was modelled in 3D, from a deterministic and probabilistic point of view, confirming the global instability of the area and the relationship of the landslide with rainfall and the construction of a new road. The integration of all this information obtained and its subsequent analysis has made possible to precisely define the extension of the instability, its kinematics, and to identify the main triggering factors.     Pivotal role of curcuminoids on the antimutagenic activity of Curcuma zedoaria extracts

    In an investigation of the mutagenic activity of two extracts from rhizomes of Curcuma zedoaria (the mathanolic, CME, and the aqueous, CAE) and antimutagenic activity against mutagens, either 2-amino-3-methylimidazo (4,5-f) quinoline (IQ) or 4-nitroquinoline-N-oxide (4-NQO), in dosages of 1-50 mu g/plate was assayed by using Salmonella typhimurium TA97, TA98, TA100, and TA102 strains. We found that the two extracts showed no mutagenicity when tested with all the tester strains either with or without the S9 mix. Moreover, the two extracts, particularly CME, presented a greater antimutagenicity than CAE did either in IQ or 4-NQO mutagens. However, the inhibition effect on lipid peroxidation was similar with both extracts. The amount of major antioxidants beta-carotene, ascorbic acid, and total polyphenols present in both CME and CAE were similar between each other. In contrast, the content of cucuminoids (44.3 mg/g extract) in CME were only found in small amounts in CAE (0.09 mg/g extract). Consequently, although both of the extracts showed similar antioxidant effects, the curcuminoids, which seem to be the main active principles from this plant, were suggested to play a pivotal role in antimutagenic activity

    Opportunities for Quantitative Translational Modeling in Oncology

    A 2‐day meeting was held by members of the UK Quantitative Systems Pharmacology Network (http://www.qsp‐uk.net/) in November 2018 on the topic of Translational Challenges in Oncology. Participants from a wide range of backgrounds were invited to discuss current and emerging modeling applications in nonclinical and clinical drug development, and to identify areas for improvement. This resulting perspective explores opportunities for impactful quantitative pharmacology approaches. Four key themes arose from the presentations and discussions that were held, leading to the following recommendations: • Evaluate the predictivity and reproducibility of animal cancer models through precompetitive collaboration. • Apply mechanism of action (MoA) based mechanistic models derived from nonclinical data to clinical trial data. • Apply MoA reflective models across trial data sets to more robustly quantify the natural history of disease and response to differing interventions. • Quantify more robustly the dose and concentration dependence of adverse events through mathematical modelling techniques and modified trial design.</p
