    Death receptor 5 expression is inversely correlated with prostate cancer progression.

    Prostate carcinoma (PCa) is one of the most common cancers in men. Prostate-specific antigen (PSA) has been widely used to predict the outcome of PCa and screening with PSA has resulted in a decline in mortality. However, PSA is not an optimal prognostic tool as its sensitivity may be too low to reduce morbidity and mortality. Consequently, there is a demand for additional robust biomarkers for prostate cancer. Death receptor 5 (DR5) has been implicated in the prognosis of several cancers and it has been previously shown that it is negatively regulated by Yin Yang 1 (YY1) in prostate cancer cell lines. The present study investigated the clinical significance of DR5 expression in a prostate cancer patient cohort and its correlation with YY1 expression. Immunohistochemical analysis of protein expression distribution was performed using tissue microarray constructs from 54 primary PCa and 39 prostatic intraepithelial neoplasia (PIN) specimens. DR5 expression was dramatically reduced as a function of higher tumor grade. By contrast, YY1 expression was elevated in PCa tumors as compared with that in PIN, and was increased with higher tumor grade. DR5 had an inverse correlation with YY1 expression. Bioinformatic analyses corroborated these data. The present findings suggested that DR5 and YY1 expression levels may serve as progression biomarkers for prostate cancer

    An LRP6 mutation (Arg360His) associated with low bone mineral density but not cardiovascular events in a caucasian family

    We present a family with a rare mutation of the LRP6 gene and for the first time provide evidence for its association with low bone mineral density. Introduction: The Wnt pathway plays a critical role in bone homeostasis. Pathogenic variants of the Wnt co-receptor LRP6 have been associated with abnormal skeletal phenotypes or increased risk of cardiovascular events. Patient and methods: Here we report an index premenopausal patient and her family carrying a rare missense LRP6 pathogenic variant (rs141212743; 0.0002 frequency among Europeans). This variant has been previously associated with metabolic syndrome and atherosclerosis, in the presence of normal bone mineral density. However, the LRP6 variant was associated with low bone mineral density in this family, without evidence for association with serum lipid levels or cardiovascular events. Conclusion: Thus, this novel association shows that LRP6 pathogenic variants may be involved in some cases of early-onset osteoporosis, but the predominant effect, either skeletal or cardiovascular, may vary depending on the genetic background or other acquired factors.Funding: Open Access funding provided thanks to the CRUE-CSIC agreement with Springer Natur

    Use of digital teaching materials in Primary classrooms

    El presente artículo se analizan las prácticas docentes en aulas de Educación Primaria en las que se emplean plataformas digitales comerciales o institucionales, así como otros materiales didácticos digitales. Se pretende dar respuesta, principalmente, a las siguientes preguntas: ¿Qué materiales didácticos digitales se utilizan en las aulas de Primaria? ¿Qué tipología de actividades se llevan a cabo con estos recursos? ¿Cómo se organiza el aula y qué metodologías didácticas se emplean? Para el desarrollo de este estudio, se toman como instrumentos de recogida de datos las entrevistas a docentes y las observaciones, de aula y de espacios, efectuadas en tres aulas de Canarias, Valencia y Galicia durante el curso escolar 2018/2019. Los resultados muestran las diferencias y similitudes de los casos analizados, a la vez que presentan una visión general y actual de las prácticas docentes con recursos digitales. This article analyzes the teaching practices in Primary classrooms in which commercial or institutional digital platforms are used, as well as other digital teaching materials. It is intended to answer mainly these following questions: Which digital didactic materials are used? What type of activities are carried out with these resources? How is the classroom organized and what methods are used? For this work, interviews are taken as tools for teachers and observations of classroom and spaces made in classrooms of the Canary Islands, Valencia and Galicia during the school year 2018/2019.     Are you ready for retirement? The influence of values on membership in voluntary organizations in midlife and old age

    Membership in voluntary organizations is associated with individual and social benefits. Due to the negative consequences of the global pandemic on older people, and the governmental challenges posed by population aging, voluntary membership is of great importance to society. To effectively promote volunteering among older people, it is necessary to understand the determinants of voluntary membership. This study analyses the influence of individual values—secular/traditional and survival/self-expression–on voluntary membership among European adults (N = 31,985). Specifically, it examines which values orient two age groups (middle age: 50–64 and old age: 65–79), as well as men and women toward a certain type of association (Social Awareness; Professional and Political; Education and Leisure; Religion). The sample of 31,985 comprises 60% of adults aged 50–64 and 40% aged 65–79; of which 56% are women and 44% men. The empirical estimation considers different levels of data aggregation: individual, national and welfare system, therefore multilevel analysis is used as an analytical strategy. Individual-level variables from the Integrated Values Survey (2005/09, 2010/14, and 2017/20) and national-level variables (Gini Index and Gross Domestic Product Per Capita) from the World Bank and Eurostat are used. The results indicate that traditional and self-expression values promote membership in voluntary organizations in general more than secular and survival values. However, there are differences according to the type of organization. Furthermore, values are found to moderate the effect of age and gender on voluntary membership

    The Apical Submembrane Cytoskeleton Participates in the Organization of the Apical Pole in Epithelial Cells

    In a previous publication (Rodriguez, M.L., M. Brignoni, and P.J.I. Salas. 1994. J. Cell Sci. 107: 3145–3151), we described the existence of a terminal web-like structure in nonbrush border cells, which comprises a specifically apical cytokeratin, presumably cytokeratin 19. In the present study we confirmed the apical distribution of cytokeratin 19 and expanded that observation to other epithelial cells in tissue culture and in vivo. In tissue culture, subconfluent cell stocks under continuous treatment with two different 21-mer phosphorothioate oligodeoxy nucleotides that targeted cytokeratin 19 mRNA enabled us to obtain confluent monolayers with a partial (40–70%) and transitory reduction in this protein. The expression of other cytoskeletal proteins was undisturbed. This downregulation of cytokeratin 19 resulted in (a) decrease in the number of microvilli; (b) disorganization of the apical (but not lateral or basal) filamentous actin and abnormal apical microtubules; and (c) depletion or redistribution of apical membrane proteins as determined by differential apical–basolateral biotinylation. In fact, a subset of detergent-insoluble proteins was not expressed on the cell surface in cells with lower levels of cytokeratin 19. Apical proteins purified in the detergent phase of Triton X-114 (typically integral membrane proteins) and those differentially extracted in Triton X-100 at 37°C or in n-octyl-β-d-glycoside at 4°C (representative of GPIanchored proteins), appeared partially redistributed to the basolateral domain. A transmembrane apical protein, sucrase isomaltase, was found mispolarized in a subpopulation of the cells treated with antisense oligonucleotides, while the basolateral polarity of Na+– K+ATPase was not affected. Both sucrase isomaltase and alkaline phosphatase (a GPI-anchored protein) appeared partially depolarized in A19 treated CACO-2 monolayers as determined by differential biotinylation, affinity purification, and immunoblot. These results suggest that an apical submembrane cytoskeleton of intermediate filaments is expressed in a number of epithelia, including those without a brush border, although it may not be universal. In addition, these data indicate that this structure is involved in the organization of the apical region of the cytoplasm and the apical membrane

    Aspectos tefrológicos de la erupción del volcán Quizapú de 1932 en la región de la laguna Llancanelo, Payenia (Mendoza, Argentina)

    El Volcán Quizapú es parte del Complejo Volcánico Cerro Azul-Descabezado Grande, ubicado en la Provincia de Talca, Chile (36,67°S - 70,77°O, altura máxima: 3788 m s.n.m.). La erupción del 10 de abril de 1932 fue uno de los mayores eventos volcánicos del siglo XX. Tuvo un carácter pliniano y arrojó un volumen de tefras entre 5 y 30 km3 (según diferentes autores), que por efecto de los vientos dominantes del oeste cubrieron gran parte de la región central de Argentina, llegando a la costa atlántica y afectando a otros países del este de Sudamérica. Los efectos climáticos y el impacto en las regiones más proximales del sur de Mendoza, particularmente en el Departamento de Malargüe, fueron muy significativos. El estudio de los eyectos constituye un campo de exploración de gran valor no solamente para conocer las características, alcances y efectos de esa erupción sino también para evaluar aspectos relacionados con la tefrología. En esta contribución se analiza un depósito de tefras en los alrededores de la Laguna Llancanelo, en las cercanías de Malargüe, una de las áreas más afectadas por la erupción. Las determinaciones sedimentológicas, mineralógicas y texturales (petrografía, microscopía electrónica y determinaciones químicas semicuantitativas con EDS) permitieron caracterizar la composición granulométrica, petrográfica y química semicuantitativa de las tefras. Estas características son afines a las de los materiales piroclásticos eyectados por la erupción del volcán Quizapú de 1932 estudiados por otros autores, por lo que se asignan a dicho evento volcánico. Las tefras depositadas en la zona de estudio son de tamaño arena muy fina a mediana con significativa cantidad de fracciones menores a 10 µm. Las trizas son pumíceas, fibrosas, con diferentes conformaciones morfológicas y abundante vesicularidad que favorece el entrampamiento de partículas menores en las vesículas de las mayores. La composición química revela un alto contenido de sílice que alcanza hasta cerca del 70% de los componentes, con alrededor de un 15% de Al y cantidades subordinadas de K, Na, Ca, Zn, Mg, Cu, Fe y Ti. Es notorio el alto contenido de K, asociado a un aumento relativo por desilicación de la tefra con el transcurso del tiempo. También son importantes los contenidos de Fe y Cu, en este último caso posiblemente asociado a transformaciones post-depositacionales por meteorización. La composición de las trizas señala una erupción de tipo pliniano magmática andesítico-dacítica con contenido hidromagmático, intensa fragmentación y aglutinación durante el enfriamiento. Las evidencias de campo demostraron que la erupción afectó severamente la región sepultando y quemando suelos, carbonizando vegetación y cegando sectores marginales de la laguna. Estudios de esta naturaleza aportan a una clasificación sistemática y comparativa de la peligrosidad volcánica, considerando que las tefras resultantes de la erupción del Quizapú contienen hasta un 10% de partículas PM 10 (tamaño <10 µm) que corresponden a los tamaños “respirables” y en consecuencia altamente nocivos para la salud. Desde el enfoque socioeconómico, estos estudios sirven de base para su aplicación a otros eventos eruptivos muy recientes ocurridos en Argentina (volcanes Hudson, Copahue, Chaitén, Llaima, Peteroa y Puyehue-Cordón Caulle), y permiten elaborar metodologías específicas para analizar la peligrosidad de futuros eventos volcánicos

    Screening of Ionic Liquids and Deep Eutectic Solvents for Physical CO2Absorption by Soft-SAFT Using Key Performance Indicators

    RC2-2019-007 PID2019108014RB-C21 SFRH/BD/130965/2017 UID/QUI/50006/2019The efficient screening of solvents for CO2 capture requires a reliable and robust equation of state to characterize and compare their thermophysical behavior for the desired application. In this work, the potentiality of 14 ionic liquids (ILs) and 7 deep eutectic solvents (DESs) for CO2 capture was examined using soft-SAFT as a modeling tool for the screening of these solvents based on key process indicators, namely, cyclic working capacity, enthalpy of desorption, and CO2 diffusion coefficient. Once the models were assessed versus experimental data, soft-SAFT was used as a predictive tool to calculate the thermophysical properties needed for evaluating their performance. Results demonstrate that under the same operating conditions, ILs have a far superior performance than DESs primarily in terms of amount of CO2 captured, being at least two-folds more than that captured using DESs. The screening tool revealed that among all the examined solvents and conditions, [C4 py][NTf2] is the most promising solvent for physical CO2 capture. The collection of the acquired results confirms the reliability of the soft-SAFT EoS as an attractive and valuable screening tool for CO2 capture and process modeling.publishersversionpublishe

    A Multicentre, Randomised, Controlled Trial of a Combined Clinical Treatment for First-Episode Psychosis

    Nurses working at nursing homes are one of the most vulnerable populations for suffering burnout and compassion fatigue. In Spain, the concept of compassion fatigue and psychological flexibility related to stress in geriatric nurses has not been fully explored until now. It is important to analyze their situation in order to design robust coping and management strategies. The aim was to analyze the relationship between burnout, compassion fatigue and psychological flexibility in geriatric nurses in Spain. Participants included 291 nurses from 97 centers in 51 cities across Spain. Psychological flexibility (AAQ-II), burnout (MBI) and compassion fatigue (ProQOL) were evaluated. Responses were recievced from 281 nurses (91% women), with an average of 7.6 years of work experience. The MBI results were average (26.71), and the ProQOL scores were average for compassion fatigue (40.2%) and high for compassion satisfaction (70.3%), whereas for AAQ-II, the mean score was 37.34 (SD 4.21). The correlation was significant and negative for flexibility, burnout and compassion fatigue, and positive for compassion satisfaction. The ANOVA indicated a significant association between all variables (p < 0.05). We can conclude that geriatric nurses suffer from medium levels of burnout and compassion fatigue, together with high levels of psychological flexibility, which appears to act as a stress reliever, supporting compassion satisfaction
