Study of the scientific potential of a three 40 cm Telescopes Interferometer at Dome C
Recent site testing (see:
http://www-luan.unice.fr/Concordiastro/indexantartic.html) has shown that Dome
C in Antarctica might have a high potential for stellar interferometry if some
solutions related to the surface atmospheric layer are found. A demonstrator
interferometer could be envisioned in order to fully qualify the site and
prepare the future development of a large array.
We analyse the performances of a prototype interferometer for Dome C made
with 3 telescopes of 40 cm diameter. It assumes classical Michelson
recombination. The most recent atmospheric and environmental conditions
measured at Dome C are considered (see K. Agabi "First whole atmosphere
night-time seeing measurements at Dome C, Antarctica"). We also study the
possible science reachable with such a demonstrator. Especially we evaluate
that even such small aperture interferometer could allow the detection and low

Dynamics of glycine receptor insertion in the neuronal plasma membrane
The exocytosis site of newly synthesized glycine receptor was defined by means of a morphological assay to characterize its export from the trans-Golgi Network to the plasma membrane. This was achieved by expressing in transfected neurons an alpha1 subunit bearing an N-terminal tag selectively cleavable from outside the cell by thrombin. This was combined with a transient temperature-induced block of exocytic transport that creates a synchronized exocytic wave. Immunofluorescence microscopy analysis of the cell surface appearance of newly synthesized receptor revealed that exocytosis mainly occurred at nonsynaptic sites in the cell body and the initial portion of dendrites. At the time of cell surface insertion, the receptors existed as discrete clusters. Quantitative analysis showed that glycine receptor clusters are stable in size and subsequently appeared in more distal dendritic regions. This localization resulted from diffusion in the plasma membrane and not from exocytosis of transport vesicles directed to dendrites. Kinetic analysis established a direct substrate-product relationship between pools of somatic and dendritic receptors. This indicated that clusters represent intermediates between newly synthesized and synaptic receptors. These results support a diffusion-retention model for the formation of receptor-enriched postsynaptic domains and not that of a vectorial intracellular targeting to synapses
User-friendly imaging algorithms for interferometry
OPTICON currently supports a Joint Research Activity (JRA) dedicated to providing easy to use image reconstruction algorithms for optical/IR interferometric data. This JRA aims to provide state-of-the-art image reconstruction methods with a common interface and comprehensive documentation to the community. These tools will provide the capability to compare the results of using different settings and algorithms in a consistent and unified way. The JRA is also providing tutorials and sample datasets to introduce the principles of image reconstruction and illustrate how to use the software products. AMBER/VLTI high spectral resolution observations of the Br emitting region in HD 98922. A compact disc wind launched from the inner disc region
We analyse the main physical parameters and the circumstellar environment of
the young Herbig Be star HD 98922. We present AMBER/VLTI high spectral
resolution (R =12000) interferometric observations across the Br line,
accompanied by UVES high-resolution spectroscopy and SINFONI-AO assisted
near-infrared integral field spectroscopic data. To interpret our observations,
we develop a magneto-centrifugally driven disc-wind model. Our analysis of the
UVES spectrum shows that HD 98922 is a young (~5x10^5 yr) Herbig Be star
(SpT=B9V), located at a distance of 440(+60-50) pc, with a mass accretion rate
of ~9+/-3x10^(-7) M_sun yr^(-1). SINFONI K-band AO-assisted imaging shows a
spatially resolved circumstellar disc-like region (~140 AU in diameter) with
asymmetric brightness distribution. Our AMBER/VLTI UT observations indicate
that the Br emitting region (radius ~0.31+/-0.04 AU) is smaller than
the continuum emitting region (inner dust radius ~0.7+/-0.2 AU), showing
significant non-zero V-shaped differential phases (i.e. non S-shaped, as
expected for a rotating disc). The value of the continuum-corrected pure
Br line visibility at the longest baseline (89 m) is ~0.8+/-0.1, i.e.
the Br emitting region is partially resolved. Our modelling suggests
that the observed Br line-emitting region mainly originates from a disc
wind with a half opening angle of 30deg, and with a mass-loss rate of ~2x10(-7)
M_sun yr^(-1). The observed V-shaped differential phases are reliably
reproduced by combining a simple asymmetric continuum disc model with our
Br disc-wind model. The Br emission of HD 98922 can be modelled
with a disc wind that is able to approximately reproduce all interferometric
observations if we assume that the intensity distribution of the dust continuum

Cytochemical techniques and energy-filtering transmission electron microscopy applied to the study of parasitic protozoa
The study of parasitic protozoa plays a major role in cell biology, biochemistry and molecular biology. Numerous cytochemical techniques have been developed in order to unequivocally identify the nature of subcellular compartments. Enzyme and immuno-cytochemistry allow the detection of, respectively, enzymatic activity products and antigens in particular sites within the cell. Energy-filtering transmission electron microscopy permits the detection of specific elements within such compartments. These approaches are particularly useful for studies employing antimicrobial agents where cellular compartments may be destroyed or remarkably altered and thus hardly identified by standard methods of observation. In this regard cytochemical and spectroscopic techniques provide valuable data allowing the determination of the mechanisms of action of such compounds
What Role for Angiogenesis in Childhood Acute Lymphoblastic Leukaemia?
The role of angiogenesis in acute leukaemia has been discussed since the cloning of the gene of vascular endothelial growth factor (VEGF) from the acute myelogenous leukemia cell line (HL60) and, thereafter, when the first studies reported increased bone marrow vascularity and elevation of angiogenic cytokines in acute lymphoblastic leukaemia (ALL). VEGF and basic fibroblast growth factor (bFGF) are the major proangiogenic cytokines that have been studied, and evaluation of their prognostic impact in childhood ALL has been reported in several studies, though with controversial results. The antiangiogenic response, contributing to the angiogenic balance, has scarcely been reported. The origin of the factors, their prognostic value, and their relevance as good markers of what really happens in the bone marrow are discussed in this paper. The place of antiangiogenic drugs in ALL has to be defined in the global treatment strategy
A method for detection of structure
Context. In order to understand the evolution of molecular clouds it is
important to identify the departures from self-similarity associated with the
scales of self-gravity and the driving of turbulence.
Aims. A method is described based on structure functions for determining
whether a region of gas, such as a molecular cloud, is fractal or contains
structure with characteristic scale sizes.
Methods. Using artificial data containing structure it is shown that
derivatives of higher order structure functions provide a powerful way to
detect the presence of characteristic scales should any be present and to
estimate the size of such structures. The method is applied to observations of
hot H2 in the Kleinman-Low nebula, north of the Trapezium stars in the Orion
Molecular Cloud, including both brightness and velocity data. The method is
compared with other techniques such as Fourier transform and histogram
techniques.
Results. It is found that the density structure, represented by H2 emission
brightness in the K-band (2-2.5micron), exhibits mean characteristic sizes of
110, 550, 1700 and 2700AU. The velocity data show the presence of structure at
140, 1500 and 3500AU. Compared with other techniques such as Fourier transform
or histogram, the method appears both more sensitive to characteristic scales

Revealing the inner circumstellar disk of the T Tauri star S Coronae Australis N using the VLTI
Copyright © ESO, 2012Aims. We investigate the structure of the circumstellar disk of the T Tauri star S CrA N and test whether the observations agree with the standard picture proposed for Herbig Ae stars.
Methods. Our observations were carried out with the VLTI/AMBER instrument in the H and K bands with the low spectral resolution mode. For the interpretation of our near-infrared AMBER and archival mid-infrared MIDI visibilities, we employed both geometric and temperature-gradient models.
Results. To characterize the disk size, we first fitted geometric models consisting of a stellar point source, a ring-shaped disk, and a halo structure to the visibilities. In the H and K bands, we measured ring-fit radii of 0.73 ± 0.03 mas (corresponding to 0.095 ± 0.018 AU for a distance of 130 pc) and 0.85 ± 0.07 mas (0.111 ± 0.026 AU), respectively. This K-band radius is approximately two times larger than the dust sublimation radius of ≈0.05 AU expected for a dust sublimation temperature of 1500 K and gray dust opacities, but approximately agrees with the prediction of models including backwarming (namely a radius of ≈0.12 AU). The derived temperature-gradient models suggest that the disk is approximately face-on consisting of two disk components with a gap between star and disk. The inner disk component has a temperature close to the dust sublimation temperature and a quite narrow intensity distribution with a radial extension from 0.11 AU to 0.14 AU.
Conclusions. Both our geometric and temperature-gradient models suggest that the T Tauri star S CrA N is surrounded by a circumstellar disk that is truncated at an inner radius of ≈ 0.11 AU.
