    Induced Pluripotent Stem Cells: Advances in the Quest for Genetic Stability during Reprogramming Process.

    Evaluation of the extent and nature of induced pluripotent stem cell (iPSC) genetic instability is important for both basic research and future clinical use. As previously demonstrated regarding embryonic stem cells, such DNA aberrations might affect the differentiation capacity of the cells and increase their tumorigenicity. Here, we first focus on the contribution of multiple DNA damage response pathways during cellular reprogramming. We then discuss the origin and mechanisms responsible for the modification of genetic material in iPSCs (pre-existing variations in somatic cells, mutations induced by reprogramming factors, and mutations induced by culture expansion) and deepen the possible functional consequences of genetic variations in these cells. Lastly, we present some recent improvements of iPSC generation methods aimed at obtaining cells with fewer genetic variations

    Self-Assessment of Adherence to Medication: A Case Study in Campania Region Community-Dwelling Population.

    Objectives. The aim of the study was to assess self-reported medication adherence measure in patients selected during a health education and health promotion focused event held in the Campania region. The study also assessed sociodemographic determinants of adherence. Methods. An interviewer assisted survey was conducted to assess adherence using the Italian version of the 8-item Morisky Medication Adherence Scale (MMAS-8). Participants older than 18 years were interviewed by pharmacists while waiting for free-medical checkup. Results. A total of 312 participants were interviewed during the Health Campus event. A total of 187 (59.9%) had low adherence to medications. Pearson's bivariate correlation showed positive association between the MMAS-8 score and gender, educational level and smoking (P < 0.05). A multivariable analysis showed that the level of education and smoking were independent predictors of adherence. Individuals with an average level of education (odds ratio (OR), 2.21, 95% confidence interval (CI), 1.08-4.52) and nonsmoker (odds ratio (OR) 1.87, 95% confidence interval (CI), 1.04-3.35) were found to be more adherent to medication than those with a lower level of education and smoking. Conclusion. The analysis showed very low prescription adherence levels in the interviewed population. The level of education was a relevant predictor associated with that result

    Stjenovita zajednica mediolitoralnog makrozoobentosa u Tršćanskom zaljevu (sjeverni Jadran) duž hidromorfološkog gradijenta

    Despite the increasing urban and industrial development in coastal areas our knowledge on direct consequences of coastal modifications on benthic communities is still limited. The aim of the present study was to assess the response of rocky macrozoobenthos mediolittoral communities to human-induced hydromorphological pressures. Sampling was carried out by SCUBA diving and snorkeling in June 2008. Ten sites were selected along a gradient of hydromorphological alterations in the southern part of the Gulf of Trieste. Variables used to describe the stressor gradient were: water retention (from normal hydrology in unprotected coast to closed areas with only one opening), substrate composition, texture and rugosity. Despite natural differences between upper and lower mediolittoral subbelts, the present work showed that human-induced alterations of the coastal zone impact biological assemblages. There was a marked difference in biodiversity among sites with pristine conditions and stressed zones, mainly due to evenness of species distribution. Structural complexity of the substrate resulted to be the main factor influencing benthic diversity in the upper mediolittoral subbelt, while in the lower subbelt also the human-induced water retention seemed to play a key role. Anyhow, this response was complex, and the major human-induced alterations considered had different level of pressure within the two subbelts. The current study has a good potential to contribute to existing coastal assessment methods, since the impact of hydromorphological pressures on mediolittoral communities was almost neglected in the past. However, further work is needed to fully explain the impact of main human-induced threats on benthic communitiesUnatoč rastućem urbanom i industrijskom razvoju u obalnim područjima samo nekolicina istraživača je fokusirana na direktne posljedice obalnih modifikacija na bentičke zajednice. Cilj ovog istraživanja je procijeniti odgovore stjenovitih zajednica mediolitoralnog makrozoobentosa na hidromorfološke pritiske. Uzorkovanje je provedeno ronjenjem sa bocama i ronjenjem na dah u lipnju 2008. godine. Izabrano je deset postaja duž hidromorfološkog gradijenta u južnom dijelu Tršćanskog zaljeva. Upotrebljavane varijable za razvoj gradijenta stresora bile su: zadržavanje vode, sastav supstrata, tekstura i naboranost. Unatoč prirodnim razlikama između gornjih i donjih mediolitoralnih podpojaseva, sadašnji rad je pokazao da promjene krajolika povezane sa ljudskim djelovanjem u obalnoj zoni utječu na biološke cjeline. Postoji značajna razlika u bioraznolikosti između postaja s odličnim ekološkim stanjem i ostalih postaja, uglavnom zbog ravnomjerne rasprostranjenosti vrsta. Strukturna složenost supstrata rezultatirala je time da je bila glavni čimbenik koji utječe na raznolikost bentosa u gornjem mediolitoralnom podpojasu, dok je izgleda u donjem podpojasu ili podregiji također cirkulacija vode odigrala ključnu ulogu (CCA analiza). Odgovor bentičke zajednice bio je kompleksan i vjerojatno su također biotički čimbenici (pokrivenost algama, kompeticija) bili uključeni. Ovo istraživanje ima dobar potencijal da doprinese postojećim metodama procjene obale, iako se treba provesti daljnje istraživanje na području utjecaja kojeg imaju hidromorfološke promjene na obalne bentičke zajednice

    Participatory research for sustainable agriculture: the case of the Italian agroecological rice network

    Since the Green Revolution, worldwide agriculture has been characterized by a typical top – down approach. The degree of autonomy, creativity, and responsibility of farmers has been limited by the continuous external inputs of chemicals, machinery, advice, subsidies and knowledge. The issue of sustainability has brought complexity and uncertainty to this mainly linear process of innovation, steering agriculture toward alternative models. Agroecology represents an innovative paradigm of agriculture in which external inputs are minimized, and the assets of the farm are greatly valued. Agroecological production relies on the farmers ’ direct management of resources and on their active engagement in the agricultural knowledge and innovation system. This paper focuses on the experience of a group of farmers, scientists, public officials, and managers of private companies who are experimenting with agroecology in rice production in one of the most intensively farmed, profitable and environmentally sensitive areas of Italy. The partnership regularly comes together to discuss agricultural techniques and results, needs, and paths of innovation; in addition, it stimulates and takes part in research projects, following a participatory process based on co-learning and mutual responsibility. By using ethnographic methods such as direct observations and in-depth interviews, our work may contribute to understanding the role of participatory research in sustainable agriculture and what makes for good participation

    Case report: Cerebral venous thrombosis in a patient with human immunodeficiency virus infection

    La trombosis venosa cerebral (TVC) es una presentación clínica poco común del tromboembolismo venoso caracterizada por cefalea, crisis convulsivas, déficits neurológicos focales y papiledema. El diagnóstico es confirmado con Tomografía axial computarizada (TAC) de cráneo y Resonancia magnética nuclear (RMN) cerebral. La TVC tiene una relación importante con el virus de inmunodeficiencia humana (VIH), ya que los pacientes VIH positivos cursan con hiperviscosidad sanguínea, alteraciones de factores anticoagulantes endógenos y riesgo de sobre infección, entre otros; que predisponen a estados protrombóticos y lesión vascular como lo es la TVC. El tratamiento de la TVC es terapia anticoagulante, por lo general se utiliza heparina no fraccionada o heparina de bajo peso molecular para la fase aguda y anticoagulantes orales como la warfarina para el mantenimiento posterior. Reportamos el primer caso documentado de TVC en un paciente VIH positivo en Colombia.Cerebral venous thrombosis (CVT) is a rare clinical presentation of venous thromboembolism characterized by headache, seizures, neurological deficits and papilledema. The diagnosis is confirmed using computed tomography scan (CT scan) and magnetic resonance imaging (MRI) of the brain. CVT has an important relationship with the human immunodeficiency virus (HIV) given that HIV-positive patients may present with blood hyperviscosity, irregular levels of endogenous anticoagulation factors and risk of sepsis among others, that predispose to prothrombotic states and vascular injury such as CVT. The treatment of CVT is anticoagulant therapy, generally unfractionated heparin or low molecular weight heparin for the early phase and oral anticoagulants such as warfarin for the late phase. This case reports the first documented case of CVT in an HIV positive patient in Colombia

    Microbial Enzyme Biotechnology to Reach Plastic Waste Circularity: Current Status, Problems and Perspectives

    The accumulation of synthetic plastic waste in the environment has become a global concern. Microbial enzymes (purified or as whole-cell biocatalysts) represent emerging biotechnological tools for waste circularity; they can depolymerize materials into reusable building blocks, but their contribution must be considered within the context of present waste management practices. This review reports on the prospective of biotechnological tools for plastic bio-recycling within the framework of plastic waste management in Europe. Available biotechnology tools can support polyethylene terephthalate (PET) recycling. However, PET represents only ≈7% of unrecycled plastic waste. Polyurethanes, the principal unrecycled waste fraction, together with other thermosets and more recalcitrant thermoplastics (e.g., polyolefins) are the next plausible target for enzyme-based depolymerization, even if this process is currently effective only on ideal polyester-based polymers. To extend the contribution of biotechnology to plastic circularity, optimization of collection and sorting systems should be considered to feed chemoenzymatic technologies for the treatment of more recalcitrant and mixed polymers. In addition, new bio-based technologies with a lower environmental impact in comparison with the present approaches should be developed to depolymerize (available or new) plastic materials, that should be designed for the required durability and for being susceptible to the action of enzymes
