    Polymorphism analysis of the CTLA-4 gene in paracoccidioidomycosis patients

    The CTLA-4 protein is expressed in activated T cells and plays an essential role in the immune response through its regulatory effect on T cell activation. Polymorphisms of the CTLA-4 gene have been correlated with autoimmune, neoplastic and infectious illnesses. This work aimed to verify possible associations between single nucleotide polymorphisms (SNPs) in CTLA-4, -318C/T in the promoter and +49A/G in exon 1 and paracoccidioidomycosis (PCM) caused by Paracoccidioides brasiliensis. For this purpose, 66 chronic form PCM patients and 76 healthy controls had their allele, genotype and haplotype frequencies determined. The genetic admixture structure of the patients and controls was evaluated to eliminate ancestral bias. The comparison of frequencies indicated no significant differences between patients and controls that could link the SNPs to PCM. Groups were admixture matched with no difference observed in population ancestry inference, indicating that the absence of association between CTLA-4 polymorphisms and PCM could not be attributed to ancestral bias. This study showed that there was no association between the CTLA-4 SNPs -318 and +49 and the resistance or susceptibility to PCM

    A influência do ultrassom e do treinamento físico sobre os triglicérides séricos e intramusculares em ratos diabéticos experimentais

    O ultrassom terapêutico é visto hoje como um dos recursos mais utilizados na prática da medicina clínica e o exercício físico é consolidado como uma terapêutica eficaz e eficiente em diversos casos, porém ainda pouco investigados em conjunto; por isso, o presente trabalho tem como objetivo analisar a influência do ultrassom e do exercício físico sobre as concentrações de triglicérides sérico e intramusculares (IMTG) em ratos diabéticos experimentais. Foram utilizados ratos Wistar adultos divididos em oitos grupos: Diabéticos Sedentários (DS), Diabéticos Treinados (DT), Diabéticos Sedentários e Ultrassom (DSUs), Diabéticos Treinados e Ultrassom (DTUs), Controle Sedentário (CS), Controle Treinado (CT), Controle Sedentário e Ultrassom (CSUs), Controle Treinado e Ultrassom (CTUs). O protocolo de treinamento constituía de natação cinco dias por semana, 30 minutos, por dia com uma carga máxima equivalente a 8% da massa corporal, durante três semanas. A terapia ultrassônica foi realizada cinco dias por semana, durante duas semanas, com intensidade de 0,2W/cm² e frequência de 1,0MHz. Não houve diferenças significativas nos triglicerídeos séricos e nos músculo Tibial Anterior e Gastrocnêmio. Para o músculo Sóleo as concentrações dos grupos diabéticos foram menores comparados com as dos grupos controles e também entre os grupos DT e DTUs comparado com DS e DSUS, sendo que treinados apresentaram as menores concentrações. O ultrassom pulsado na intensidade proposta não influenciou as concentrações séricas de triglicerídeos nem de IMTG. Porém o exercício físico foi eficaz em reduzir IMTG no músculo Sóleo

    Effectiveness of nano-calcium phosphate paste on sensitivity during and after bleaching: a randomized clinical trial

    The study aimed to evaluate the effectiveness of in-office bleaching and associated tooth sensitivity on application of nano-calcium phosphate paste as desensitizing agent. Bleaching was performed with 35% hydrogen peroxide gel in 40 patients who were randomly divided into placebo and nano-calcium phosphate paste groups. Bleaching efficacy (BE) was evaluated using a value-oriented Vita shade guide. Tooth sensitivity was recorded using a numeric rating scale (0–4) during bleaching and up to 48 h after each session. The primary outcome of absolute risk of tooth sensitivity was compared using the Fisher’s exact test (α = 0.05). The intensity of tooth sensitivity and the efficacy of in-office bleaching were also statistically evaluated. No significant differences in absolute risk and intensity of tooth sensitivity were detected between the groups (p = 1.0 and p = 0.53, respectively). BE was also found to be similar between the groups (p = 0.67). Although the use of a nano-calcium phosphate paste associated with fluoride and potassium nitrate did not influence the whitening outcome, but it also did not reduce bleaching-induced tooth sensitivity.</p

    CTIP2 Expression in Human Head and Neck Squamous Cell Carcinoma Is Linked to Poorly Differentiated Tumor Status

    We have demonstrated earlier that CTIP2 is highly expressed in mouse skin during embryogenesis and in adulthood. CTIP2 mutant mice die at birth with epidermal differentiation defects and a compromised epidermal permeability barrier suggesting its role in skin development and/or homeostasis. CTIP2 has also been suggested to function as tumor suppressor in cells, and several reports have described a link between chromosomal rearrangements of CTIP2 and human T cell acute lymphoblast leukemia (T-ALL). The aim of the present study was to look into the pattern of CTIP2 expression in Head and Neck Squamous Cell Carcinoma (HNSCC).In the present study, we analyzed CTIP2 expression in human HNSCC cell lines by western blotting, in paraffin embedded archival specimens by immunohistochemistry (IHC), and in cDNA samples of human HNSCC by qRT-PCR. Elevated levels of CTIP2 protein was detected in several HNSCC cell lines. CTIP2 staining was mainly detected in the basal layer of the head and neck normal epithelium. CTIP2 expression was found to be significantly elevated in HNSCC (p<0.01), and increase in CTIP2 expression was associated with poorly differentiated tumor status. Nuclear co-localization of CTIP2 protein and cancer stem cell (CSC) marker BMI1 was observed in most, if not all of the cells expressing BMI1 in moderately and poorly differentiated tumors.We report for the first time expression of transcriptional regulator CTIP2 in normal human head and neck epithelia. A statistically significant increase in the expression of CTIP2 was detected in the poorly differentiated samples of the human head and neck tumors. Actual CTIP2, rather than the long form of CTIP2 (CTIP2(L)) was found to be more relevant to the differentiation state of the tumors. Results demonstrated existence of distinct subsets of cancer cells, which express CTIP2 and underscores the use of CTIP2 and BMI1 co-labeling to distinguish tumor initiating cells or cancer stem cells (CSCs) from surrounding cancer cells

    Population analysis of vitamin D receptor polymorphisms and the role of genetic ancestry in an admixed population

    The vitamin D receptor (VDR) is an essential protein related to bone metabolism. Some VDR alleles are differentially distributed among ethnic populations and display variable patterns of linkage disequilibrium (LD). In this study, 200 unrelated Brazilians were genotyped using 21 VDR single nucleotide polymorphisms (SNPs) and 28 ancestry informative markers. The patterns of LD and haplotype distribution were compared among Brazilian and the HapMap populations of African (YRI), European (CEU) and Asian (JPT+CHB) origins. Conditional regression and haplotype-specific analysis were performed using estimates of individual genetic ancestry in Brazilians as a quantitative trait. Similar patterns of LD were observed in the 5′ and 3′ gene regions. However, the frequency distribution of haplotype blocks varied among populations. Conditional regression analysis identified haplotypes associated with European and Amerindian ancestry, but not with the proportion of African ancestry. Individual ancestry estimates were associated with VDR haplotypes. These findings reinforce the need to correct for population stratification when performing genetic association studies in admixed populations

    Envelhecimento por Deformação em Juntas Soldadas de Tubos de Aço API5L X65Q sem Costura

    Resumo Neste trabalho, investigaram-se efeitos do fenômeno de envelhecimento por deformação em juntas obtidas por soldagem a arco elétrico com proteção gasosa (GMAW), de tubos sem costura de aço API5L X65Q. Corpos de prova (CPs) obtidos a partir das juntas soldadas foram submetidos à uma deformação plástica a frio de 3% e posteriormente envelhecidas por 1h a 250°C, de forma a se induzir envelhecimento estático. Buscando avaliar o envelhecimento dinâmico, os CPs foram deformados até 3% e o carregamento foi mantido durante 1h a 250°C. Os CPs envelhecidos foram submetidos a ensaios de tração e amostras representativas foram analisadas ao microscópio óptico e microscópio eletrônico de varredura. Observou-se que o fenômeno modificou a fração volumétrica de constituintes secundários no metal de solda e na ZTA, com aumento na proporção de agregados ferrita-carbonetos. Com relação às propriedades mecânicas, constatou-se que o fenômeno reduziu a razão elástica das juntas soldadas devido a um aumento no limite de resistência, além de aumentar o alongamento total, não prejudicando, portanto, as propriedades mecânicas em tração das juntas soldadas. Ao se comparar o envelhecimento estático com o envelhecimento dinâmico, observou-se que o aumento na capacidade de deformação plástica das juntas soldadas foi maior após envelhecimento estático por deformação

    Use of the Brazilian People’s Pharmacy Program by older adults

    ABSTRACT OBJECTIVE To assess the prevalence and factors associated with the use of the expanded Brazilian People’s Pharmacy Program among older adults and the reasons for not using it. METHODS In this population-based cross-sectional study conducted in the urban area of Pelotas, RS, Southern Brazil, we evaluated 1,305 older adults (aged 60 years or over) who had used medication in the last 15 days. Independent variables were socioeconomic factors, economic status, household income in minimum wages, educational attainment in years of schooling and occupational status. Demographic variables were sex, age, marital status, and self-reported skin color/race. Poisson regression was employed to analyze the factors associated with the use of the program. RESULTS The prevalence of use was 57.0% whilst the prevalence of knowledge of the program was 87.0%. In individuals aged 80 years or over, use of the program was 41.0%. As to the origin of the prescriptions used by older adults, 46.0% were from the Brazilian Unified Health System. The main reasons for not using the program were: difficulty in getting prescriptions, medication shortage, and ignorance about the medications offered and about the program. Higher age, lower income, presence of chronic diseases, and use of four or more medications were associated with use of the program. CONCLUSIONS It is necessary to expand the knowledge and use of the Brazilian People’s Pharmacy Program, especially among older adults, and to improve the dissemination of its list of medications to users and physicians. Thus it will be possible to reduce spending on long-term medications, which are especially important for this population
