    Analysis of accounting data from the explanatory notes to the financial statements for the estimation of the enterprises investments

    The Italian national institute of statistics (Istat) produces estimates on the main economic variables of enterprises, both from a Structural Business Statistics (SBS) and a National Accounts (NA) perspective. The two production processes present similarities and discrepancies. As far the production process of estimates on “acquisitions of fixed assets” during the year, also known as “enterprise investments”, both SBS and NA processes use survey data, but only NA process also uses administrative information. The aim of the present work is to evaluate the content of the information on Investments, at the level of the legal unit, from the administrative source of the Explanatory Notes, beside the administrative sources already used in Istat processes. This work represents an exploratory and study phase to evaluate the usability of this source in the production processes of Investments of both SBS and NA

    Natural deep eutectic solvents as thermostabilizer for Humicola insolens cutinase

    As a new generation of green solvents, deep eutectic solvents (DESs) are considered a promising alternative to current harsh organic solvents and find application in many chemical processing methods such as extraction and synthesis. DESs, normally formed by two or more components via various hydrogen bond interactions, offer high potential as medium for biocatalysis reactions where they can improve efficiency by enhancing substrate solubility and the activity and stability of the enzymes. In the current study, the stabilization of Humicola insolens cutinase (HiC) in natural deep eutectic solvents (NADESs) was assessed. The best hydrogen bond donor among sorbitol, xylitol, erythritol, glycerol and ethylene glycol, and the best acceptor among betaine, choline chloride, choline acetate, choline dihydrogen citrate and tetramethylammonium chloride, were selected, evaluating binding energies and molecular orientations through molecular docking simulations, and finally used to prepare NADES aqueous solutions. The effects of component ratio and NADES concentration on HiC thermostability at 90 degrees C were also investigated. The choline dihydrogen citrate:xylitol, in a 1:1 ratio with a 20 wt% concentration, was selected as the best combination in stabilizing HiC, increasing its half-life three-fold

    Tailoring the specificity of the type C feruloyl esterase FoFaeC from Fusarium oxysporum towards methyl sinapate by rational redesign based on small molecule docking simulations

    The type C feruloyl esterase FoFaeC from Fusarium oxysporum is a newly discovered enzyme with high potential for use in the hydrolysis of lignocellulosic biomass but it shows low activity towards sinapates. In this work, small molecule docking simulations were employed in order to identify important residues for the binding of the four model methyl esters of hydroxycinnamic acids, methyl ferulate/caffeate/sinapate/p-coumarate, to the predicted structure of FoFaeC. Subsequently rational redesign was applied to the enzyme’ active site in order to improve its specificity towards methyl sinapate. A double mutation (F230H/T202V) was considered to provide hydrophobic environment for stabilization of the methoxy substitution on sinapate and a larger binding pocket. Five mutant clones and the wild type were produced in Pichia pastoris and biochemically characterized. All clones showed improved activity, substrate affinity, catalytic efficiency and turnover rate compared to the wild type against methyl sinapate, with clone P13 showing a 5-fold improvement in catalytic efficiency. Although the affinity of all mutant clones was improved against the four model substrates, the catalytic efficiency and turnover rate decreased for the substrates containing a hydroxyl substitution

    The Vallo di Diano Range-Bounding Fault-System (Southern Italy): New Evidence of Recent Activity From High-Resolution Seismic Profiling

    The Vallo di Diano is the largest intermountain basin in the Southern Apennines (Italy). The basin evolution was controlled by the Quaternary activity of a range-bounding, SW-dipping normal fault system located to the east (Vallo di Diano Fault System, VDFS). Geological and oil industry data define the sin-sedimentary activity of the VDFS up to the Middle Pleistocene. However, commercial profiles do not resolve the shallower, eastern portion of the basin, due to strong lateral heterogeneities and unfavourable surface conditions. Therefore, Late Pleistocene-Holocene activity of the VDFS and its seismogenic potential are still uncertain. To better constrain the shallow structure of the basin, we performed four high-resolution seismic surveys, along its eastern side, where slope breccias and fans cover the Mesozoic carbonate bedrock and bury the VDFS. We also investigated some NW-trending flexures affecting Late Pleistocene fans, that we had previously detected and dubitatively ascribed to recent faulting. Seismic data were acquired with a dense wide-aperture geometry. Two high-resolution (HR) NE-trending profiles, about 1.5 km long, were collected using respectively 5 m and 10 m spaced receivers and sources. Two very high-resolution (VHR) NE-trending profiles, 400 and 350 m long, with densely spaced sources (4 m) and receivers (2 m) were also collected. HR profiling was aimed at imaging alluvial fan thickness and morphology of the underlying carbonate bedrock. VHR surveys targeted the flexures and their possible origin. All lines were acquired with a HR vibroseis source, except for the shortest profile, where we used a buffalo-gun, better suited for very near-surface imaging (z < 50 m depth). Seismic imaging consists of reflectivity images obtained by CDP-processing of reflection data complemented by Vp images obtained by multi-scale seismic tomography. The stack sections illuminated the basin down to 0.4-0.5 s TWT and reveal an array of high-angle, generally SW-dipping faults dissecting the bedrock and the alluvial fans. Faulting created accommodation space in the hanging-wall and displaced the different fan generations. Clear reflection truncations in the stack-sections correspond to significant Vp lateral changes in the tomographic images. VHR tomography is well defined along the shortest line down to 40 m depth, where two steps within slope breccias are visible. Moreover, two low-velocity wedges (colluvial packages) are imaged in the near surface (5-20 m depth). These data support recent faulting consistently with surface geomorphic features. We interpret these fault structures as splays of the range bounding master fault. Comparison with commercial reflection profiles nearby reveals a great improvement in seismic imaging achieved by HR surveys, which allow a detailed seismostratigraphic analysis of the basin.UnpublishedSan Francisco (CA), Moscone West, Howard Street3.2.     Demonstrator of Time Services based on European GNSS signals: the H2020 DEMETRA Project

    During 2015-2016, a European Consortium of 15 partners from 8 different countries, developed the DEMETRA (DEMonstrator of EGNSS services based on Time Reference Architecture), a project funded by the European Union in the frame of the Horizon 2020 program. This project aims at developing and experimenting time dissemination services dedicated to specific users like traffic control, energy distribution, finance, telecommunication, and scientific institutions. Nine services have been developed. These services provide time dissemination with accuracy levels from millisecond to the sub-ns, and also additional services like certification, calibration, or integrity. Five of these services are based on the European GNSS. After a development phase (see PTTI 2016 presentation) the full DEMETRA system has been working during six months for demonstration. The paper will report about the experimentation results, showing performances and limits of the proposed time dissemination services, aiming to foster the exploitation of the European GNSS for timing applications

    MEDIATE - Molecular DockIng at homE: Turning collaborative simulations into therapeutic solutions

    IntroductionCollaborative computing has attracted great interest in the possibility of joining the efforts of researchers worldwide. Its relevance has further increased during the pandemic crisis since it allows for the strengthening of scientific collaborations while avoiding physical interactions. Thus, the E4C consortium presents the MEDIATE initiative which invited researchers to contribute via their virtual screening simulations that will be combined with AI-based consensus approaches to provide robust and method-independent predictions. The best compounds will be tested, and the biological results will be shared with the scientific community.Areas coveredIn this paper, the MEDIATE initiative is described. This shares compounds' libraries and protein structures prepared to perform standardized virtual screenings. Preliminary analyses are also reported which provide encouraging results emphasizing the MEDIATE initiative's capacity to identify active compounds.Expert opinionStructure-based virtual screening is well-suited for collaborative projects provided that the participating researchers work on the same input file. Until now, such a strategy was rarely pursued and most initiatives in the field were organized as challenges. The MEDIATE platform is focused on SARS-CoV-2 targets but can be seen as a prototype which can be utilized to perform collaborative virtual screening campaigns in any therapeutic field by sharing the appropriate input files

    Fog computing security: a review of current applications and security solutions

    Fog computing is a new paradigm that extends the Cloud platform model by providing computing resources on the edges of a network. It can be described as a cloud-like platform having similar data, computation, storage and application services, but is fundamentally different in that it is decentralized. In addition, Fog systems are capable of processing large amounts of data locally, operate on-premise, are fully portable, and can be installed on heterogeneous hardware. These features make the Fog platform highly suitable for time and location-sensitive applications. For example, Internet of Things (IoT) devices are required to quickly process a large amount of data. This wide range of functionality driven applications intensifies many security issues regarding data, virtualization, segregation, network, malware and monitoring. This paper surveys existing literature on Fog computing applications to identify common security gaps. Similar technologies like Edge computing, Cloudlets and Micro-data centres have also been included to provide a holistic review process. The majority of Fog applications are motivated by the desire for functionality and end-user requirements, while the security aspects are often ignored or considered as an afterthought. This paper also determines the impact of those security issues and possible solutions, providing future security-relevant directions to those responsible for designing, developing, and maintaining Fog systems

    Risk of Myocardial Infarction in Parents of HIV-infected Individuals: a population-based Cohort Study

    <p>Abstract</p> <p>Background</p> <p>Previous studies have indicated an increased risk of myocardial infarction (MI) in HIV infected individuals especially after start of highly active antiretroviral therapy (HAART). It is however controversial whether the increased risk of atherosclerotic disease is exclusively associated with the HIV disease and HAART or whether life-style related or genetic factors also increase the risk in this population. To establish whether the increased risk of myocardial infarction in HIV patients partly reflects an increased risk of MI in their families, we estimated the relative risk of MI in parents of HIV-infected individuals.</p> <p>Methods</p> <p>From the Danish HIV Cohort Study and the Danish Civil Registration System we identified the parents of all HIV-infected patients born in Denmark after 1952 in whom a Danish born mother was identifiable. For each HIV patient, 4 matched population controls and their parents were identified. Cumulative incidence functions were constructed to illustrate time to first MI of the parents as registered in the Danish National Hospital Registry. Incidence rate ratios (IRR) were estimated by Cox's regression analyses. Due to the confidential type of the analysed data the study was approved by the Danish Data Protection Agency.</p> <p>Results</p> <p>2,269 mothers and 2,022 fathers of HIV patients as well as 9,076 mothers and 8,460 fathers of control subjects were identified. We observed an increased risk of MI in mothers of HIV patients (adjusted IRR, 1.31; 95% CI: 1.08-1.60). The strongest association was seen in case the offspring was infected heterosexually (adjusted IRR, 1.59; 95% CI: 1.07-2.35) or by IV drug abuse (IVD) (adjusted IRR, 1.63; 95% CI: 1.02-2.60). In fathers of HIV patients the risk of MI was only increased if the offspring was infected by IVD (adjusted IRR, 1.42; 95% CI: 1.01-2.00).</p> <p>Conclusion</p> <p>Mothers of HIV-infected patients have an increased risk of MI. We presume that this stems from family related life style risk factors, some of which may also influence the risk of MI in HIV-infected patients.</p

    Human Immunodeficiency Virus Impairs Reverse Cholesterol Transport from Macrophages

    Several steps of HIV-1 replication critically depend on cholesterol. HIV infection is associated with profound changes in lipid and lipoprotein metabolism and an increased risk of coronary artery disease. Whereas numerous studies have investigated the role of anti-HIV drugs in lipodystrophy and dyslipidemia, the effects of HIV infection on cellular cholesterol metabolism remain uncharacterized. Here, we demonstrate that HIV-1 impairs ATP-binding cassette transporter A1 (ABCA1)-dependent cholesterol efflux from human macrophages, a condition previously shown to be highly atherogenic. In HIV-1–infected cells, this effect was mediated by Nef. Transfection of murine macrophages with Nef impaired cholesterol efflux from these cells. At least two mechanisms were found to be responsible for this phenomenon: first, HIV infection and transfection with Nef induced post-transcriptional down-regulation of ABCA1; and second, Nef caused redistribution of ABCA1 to the plasma membrane and inhibited internalization of apolipoprotein A-I. Binding of Nef to ABCA1 was required for down-regulation and redistribution of ABCA1. HIV-infected and Nef-transfected macrophages accumulated substantial amounts of lipids, thus resembling foam cells. The contribution of HIV-infected macrophages to the pathogenesis of atherosclerosis was supported by the presence of HIV-positive foam cells in atherosclerotic plaques of HIV-infected patients. Stimulation of cholesterol efflux from macrophages significantly reduced infectivity of the virions produced by these cells, and this effect correlated with a decreased amount of virion-associated cholesterol, suggesting that impairment of cholesterol efflux is essential to ensure proper cholesterol content in nascent HIV particles. These results reveal a previously unrecognized dysregulation of intracellular lipid metabolism in HIV-infected macrophages and identify Nef and ABCA1 as the key players responsible for this effect. Our findings have implications for pathogenesis of both HIV disease and atherosclerosis, because they reveal the role of cholesterol efflux impairment in HIV infectivity and suggest a possible mechanism by which HIV infection of macrophages may contribute to increased risk of atherosclerosis in HIV-infected patients
