    The endophytic bacterial community of healthy and Xylella-infected olive sapwood

    Endophytic bacteria are of biotechnological and agronomic interest as they promote plant healthiness by producing and secreting plant growth regulators, and antagonizing phytopathogens through the induction of resistance mechanisms, and the supply of nutritional elements. One of the factors that may influence the behavior of olive towards the ‘quick decline syndrome’ is the nature of the endophytic microbial community occurring in sapwood. Objectives of the research was to characterize the bacterial endophytic population occurring into the xylem of healthy and Xylellainfected olive trees by an isolation-dependent approach. Preliminary results indicate that under field conditions, the population level of cultivable endophytic bacteria is highly variable, being mainly affected by the host genotype, host age, and wilting severity. Among the different group are Pseudomonas, Bacillus, Lysinibacillus, Pantoea, Microbacterium, Stenotrophomonas, and Methylobacterium spp. Bacteria of the Methylobacetrium genus occupy the same ecological niche of X. fastidiosa subsp. pauca. It has been reported as potential biocontrol agent of the pathogen, being its population higher in citrus plant showing mild symptoms of variegated chlorosis. Further research is in progress to better characterize the different Methylobacterium strains, using both biochemical and molecular approaches, and to evaluate its activity in reducing the severity of olive quick decline syndrome

    Fluorescence and Morphology of Self-Assembled Nucleobases and Their Diphenylalanine Hybrid Aggregates

    Studies carried out in recent decades have revealed that the ability to self-assemble is a widespread property among biomolecules. Small nucleic acid moieties or very short peptides are able to generate intricate assemblies endowed with remarkable structural and spectroscopic properties. Herein, the structural/spectroscopic characterization of aggregates formed by nucleobases and peptide nucleic acid (PNA)-peptide conjugates are reported. At high concentration, all studied nucleobases form aggregates characterized by previously unreported fluorescence properties. The conjugation of these bases, as PNA derivatives, to the dipeptide Phe-Phe leads to the formation of novel hybrid assemblies, which are characterized by an amyloid-like association of the monomers. Although these compounds share the same basic cross-\u3b2 motif, the nature and number of PNA units have an important impact on both the level of structural order and the intrinsic fluorescence of the self-assembled nanostructure

    Geomorphological processes, forms and features in the surroundings of the Melka Kunture Palaeolithic site, Ethiopia

    The landscape of the surroundings of the Melka Kunture prehistoric site, Upper Awash Basin, Ethiopia, were studied intensively in the last decades. Nonetheless, the area was mainly characterized under a stratigraphic/geological and archaeological point of view. However, a detailed geomorphological map is still lacking. Hence, in this study, we identify, map and visualize geomorphological forms and processes. The morphology of the forms, as well as the related processes, were remotely sensed with available high-resolution airborne and satellite sources and calibrated and validated through extensive field work conducted in 2013 and 2014. Furthermore, we integrated multispectral satellite imagery to classify areas affected by intensive erosion processes and/or anthropic activities. The Main Map at 1:15,000 scale reveals structural landforms as well as intensive water-related degradation processes in the Upper Awash Basin. The recent establishment of Xylella fastidiosa subspecies pauca in the southern Italian region of Apulia threatens agricultural crops and the environment. Olive is an important and widespread ancient crop in Italy and, so far, the most impacted host. The meadow spittlebug Philaenus spumarius (Hemiptera, Aphrophoridae) has been identified as a vector of X. fastidiosa in southern Italy; this species is one of the most common potential vectors in Europe. To generate disease management strategies, data on X. fastidiosa transmission by P. spumarius are necessary. Therefore, we carried out transmission experiments by using field-collected spittlebugs in 2014 and 2015 (5 and 11 collection dates, respectively), and transferring groups of insects immediately on to recipient plants. Various host plant species were tested: olive, oleander, sweet orange, grapevine and the stone fruit rootstock GF677 (Prunus persica × Prunus amygdalus). Xylella fastidiosa was detected in all the host plants after insect plant access except for grapevine; infections to sweet orange and stone fruit were not systemic. In 2015, estimates of insect X. fastidiosa infectivity were obtained; the number of PCR-positive P. spumarius on each plant was positively correlated with the plant infection status. The proportion of P. spumarius infected with X. fastidiosa ranged from 25% to 71% during the entire survey period. The number of X. fastidiosa cells detected in P. spumarius heads ranged from 3.5 × 10 to 4.0 × 102(CFU equivalents), which is lower than that reported for leafhopper vectors in the Americas. These data show that field-collected P. spumarius have high rates of X. fastidiosa infection and are competent vectors

    May selective serotonin reuptake inhibitors (SSRIs) provide some benefit for the treatment of schizophrenia?

    INTRODUCTION: The treatment of some psychopathological dimensions of schizophrenia (e.g. negative and depressive symptoms) is still challenging for the modest efficacy of atypical antipsychotics. Among pharmacological alternatives, augmentative Selective Serotonin Reuptake Inhibitors (SSRIs) to antipsychotics are frequently prescribed in clinical practice to improve negative/depressive symptoms of schizophrenia patients; however, the data about the efficacy of these molecules on negative, depressive and obsessive-compulsive symptoms of schizophrenia are contrasting. AREAS COVERED: Research using the main database sources has been conducted to obtain an overview of the use and efficacy of SSRIs in schizophrenia. EXPERT OPINION: Data are too scanty to draw definitive recommendations. In a preliminary way, it can be said that available data do not show effectiveness of SSRIs on depressive symptoms of schizophrenia. Regarding negative symptoms, studies are contrasting, but paroxetine appears to be the most effective compound among SSRIs. Despite limited data, SSRIs appear to be useful for the treatment of obsessive-compulsive symptoms of schizophrenia, particularly fluvoxamine. Close clinical and pharmacological monitoring is needed in case of concomitant administration of antipsychotics and antidepressants for potential serious side effects and influence on plasma drug dosages

    The ALTCRISS Project On Board the International Space Station

    The Altcriss project aims to perform a long term survey of the radiation environment on board the International Space Station. Measurements are being performed with active and passive devices in different locations and orientations of the Russian segment of the station. The goal is perform a detailed evaluation of the differences in particle fluence and nuclear composition due to different shielding material and attitude of the station. The Sileye-3/Alteino detector is used to identify nuclei up to Iron in the energy range above approximately equal to 60 MeV/n; a number of passive dosimeters (TLDs, CR39) are also placed in the same location of Sileye-3 detector. Polyethylene shielding is periodically interposed in front of the detectors to evaluate the effectiveness of shielding on the nuclear component of the cosmic radiation. The project was submitted to ESA in reply to the AO the Life and Physical Science of 2004 and was begun in December 2005. Dosimeters and data cards are rotated every six months: up to now three launches of dosimeters and data cards have been performed and have been returned with the end expedition 12 and 13

    High mobilization of CD133+/CD34+ cells expressing HIF-1α and SDF-1α in septic abdominal surgical patients

    Background: The control of endothelial progenitor cells (CD133+/CD34+ EPCs) migrating from bone marrow to peripheral blood is not completely understood. Emerging evidence suggests that stromal cell-derived factor-1α (SDF-1α) mediates egression of EPCs from bone marrow, while the hypoxia inducible factor (HIF) transcriptional system regulates SDF-1α expression. Our study aimed to investigate the time course of circulating CD133+/CD34+ EPCs and its correlation with the expression of HIF-1α protein and SDF-1α in postoperative laparoscopic abdominal septic patients. Methods: Postoperative patients were divided in control (C group) and septic group (S group) operated immediately after the diagnosis of sepsis/septic shock. Blood samples were collected at baseline (0), 1, 3 and 7 postoperative days for CD133+/CD34+ EPCs count expressing or not the HIF-1α and SDF-1α analysis. Results: Thirty-two patients in S group and 39 in C group were analyzed. In C group CD133+/CD34+ EPCs count remained stable throughout the study period, increasing on day 7 (173 [0-421] /μl vs baseline: P = 0.04; vs day 1: P = 0.002). In S group CD133+/CD34+ EPCs count levels were higher on day 3 (vs day 1: P = 0.006 and day 7: P = 0.026). HIF-1α expressing CD133+/CD34+ EPCs count decreased on day 1 as compared with the other days in C group (day 0 vs 1: P = 0.003, days 3 and 7 vs 1: P = 0.008), while it was 321 [0-1418] /μl on day 3 (vs day 1; P = 0.004), and 400 [0-587] /μl on day 7 in S group. SDF-1α levels were higher not only on baseline but also on postoperative day 1 in S vs C group (219 [124-337] pg/ml vs 35 [27-325] pg/ml, respectively; P = 0.01). Conclusion: Our results indicate that sepsis in abdominal laparoscopic patients might constitute an additional trigger of the EPCs mobilization as compared with non-septic surgical patients. A larger mobilization of CD133+/CD34+ EPCs, preceded by enhanced plasmatic SDF-1α, occurs in septic surgical patients regardless of HIF-1α expression therein.
