    KM Capability Assessment: A Qualitative Approach

    The knowledge management (KM) literature highlights both the desire of organizations to assess KM capability and the need to create better methodologies and tools to do so. Although some progress has been made in developing valid assessment tools, the topic still remains inadequately explored. Answering a call for the exploration of KM capability maturity assessment across a variety of organizations (Kulkarni & St Louis, 2003), this research uses the Knowledge Management Capability Assessment (KMCA) methodology (Kulkarni & Freeze, 2004) and Freeze & Kulkarni (2005; 2006) as a guiding framework to qualitatively assess the KM capability of the Secretary of the Air Force Financial Management and Comptroller (SAF/FM) organization—a military organization recognized for exceptional KM efforts. The research resulted in rich, contextual findings with regard to the specific KM efforts underway within SAF/FM. Interestingly, the nature of these efforts translated into KM capability levels lower than expected; however, precise areas for improvement were identified

    Copper(ii) complexes of N-propargyl cyclam ligands reveal a range of coordination modes and colours, and unexpected reactivity

    The coordination chemistry of N-functionalised cyclam ligands has a rich history, yet cyclam derivatives with pendant alkynes are largely unexplored. This is despite the significant potential and burgeoning application of N-propargyl cyclams and related compounds in the creation of diversely functionalised cyclam derivatives via copper-catalysed azide–alkyne ‘click’ reactions. Herein we describe single crystal X-ray diffraction and spectroscopic investigations of the coordination chemistry of copper(II) complexes of cyclam derivatives with between 1 and 4 pendant alkynes. The crystal structures of these copper complexes unexpectedly reveal a range of coordination modes, and the surprising occurrence of five unique complexes within a single recrystallisation of the tetra-N-propargyl cyclam ligand. One of these species exhibits weak intramolecular copper-alkyne coordination, and another is formed by a surprising intramolecular copper-mediated hydroalkoxylation reaction with the solvent methanol, transforming one of the pendant alkynes to an enol ether. Multiple functionalisation of the tetra-N-propargyl ligand is demonstrated via a ‘tetra-click’ reaction with benzyl azide, and the copper-binding behaviour of the resulting tetra-triazole ligand is characterised spectroscopically

    A Cohort Study Assessing the Impact of Anki as a Spaced Repetition Tool on Academic Performance in Medical School

    Introduction Anki is an application that capitalizes upon the techniques of spaced repetition and is increasingly utilized by medical students for examination preparation. This study examines the impact of Anki usage in a medical school curriculum on academic performance. Secondary objectives analyzed individual Anki utilization and a qualitative assessment of Anki use. Methods A cohort-control study was conducted at Boonshoft School of Medicine. One hundred thirty first-year medical students were enrolled in an Anki utilization training program from July 2021 to September 2021. Training included educational Anki courses and subsequent survey data collection over Anki usage. Data variables included all course final examinations, the Comprehensive Basic Science Exam (CBSE), individual Anki user statistics, nationally standardized exams scores, and Qualtrics surveys on student perceived ease of use. Results Seventy-eight students reported using Anki for at least one of the exams, and 52 students did not use Anki for any exam. Anki users scored significantly higher across all four exams: Course I (6.4%; p \u3c 0.001); Course II (6.2%; p = 0.002); Course III (7.0%; p = 0.002); and CBSE (12.9%; p = 0.003). Students who reported higher dependency on Anki for studying performed significantly better on the Course I, II, and CBSE exams. Conclusion Anki usage may be associated with an increase in standardized examination scores. This supports Anki as an evidence-based spaced repetition and active retrieval learning modality for medical school standardized examinations. There was little correlation between its specific statistical markers and examination performance. This is pertinent to physicians and medical students alike as the learning and preservation of biomedical knowledge is required for examinations and effective clinical care

    Cost-effective targeting of conservation investments to reduce the northern Gulf of Mexico hypoxic zone

    A seasonally occurring summer hypoxic (low oxygen) zone in the northern Gulf of Mexico is the second largest in the world. Reductions in nutrients from agricultural cropland in its watershed are needed to reduce the hypoxic zone size to the national policy goal of 5,000 km2 (as a 5-y running average) set by the national Gulf of Mexico Task Force’s Action Plan. We develop an integrated assessment model linking the water quality effects of cropland conservation investment decisions on the more than 550 agricultural subwatersheds that deliver nutrients into the Gulf with a hypoxic zone model. We use this integrated assessment model to identify the most cost-effective subwatersheds to target for cropland conservation investments. We consider targeting of the location (which subwatersheds to treat) and the extent of conservation investment to undertake (how much cropland within a subwatershed to treat). We use process models to simulate the dynamics of the effects of cropland conservation investments on nutrient delivery to the Gulf and use an evolutionary algorithm to solve the optimization problem. Model results suggest that by targeting cropland conservation investments to the most cost-effective location and extent of coverage, the Action Plan goal of 5,000 km2 can be achieved at a cost of 2.7billionannually.Alargesetofcost−hypoxiatradeoffsisdeveloped,rangingfromthebaselinetothenontargetedadoptionofthemostaggressivecroplandconservationinvestmentsinallsubwatersheds(estimatedtoreducethehypoxiczonetolessthan3,000km2atacostof2.7 billion annually. A large set of cost-hypoxia tradeoffs is developed, ranging from the baseline to the nontargeted adoption of the most aggressive cropland conservation investments in all subwatersheds (estimated to reduce the hypoxic zone to less than 3,000 km2 at a cost of 5.6 billion annually)

    Synthesis and Evaluation of 1,8-Disubstituted-Cyclam/Naphthalimide Conjugates as Probes for Metal Ions

    Fluorescent molecular probes for metal ions have a raft of potential applications in chemistry and biomedicine. We report the synthesis and photophysical characterisation of 1,8-disubstituted-cyclam/naphthalimide conjugates and their zinc complexes. An efficient synthesis of 1,8-bis-(2-azidoethyl)cyclam has been developed and used to prepare 1,8-disubstituted triazolyl-cyclam systems, in which the pendant group is connected to triazole C4. UV/Vis and fluorescence emission spectra, zinc binding experiments, fluorescence quantum yield and lifetime measurements and pH titrations of the resultant bis-naphthalimide ligand elucidate a complex pattern of photophysical behaviour. Important differences arise from the inclusion of two fluorophores in the one probe and from the variation of triazole substitution pattern (dye at C4 vs. N1). Introducing a second fluorophore greatly extends fluorescence lifetimes, whereas the altered substitution pattern at the cyclam amines exerts a major influence on fluorescence output and metal binding.     High-throughput identification of genotype-specific cancer vulnerabilities in mixtures of barcoded tumor cell lines.

    Hundreds of genetically characterized cell lines are available for the discovery of genotype-specific cancer vulnerabilities. However, screening large numbers of compounds against large numbers of cell lines is currently impractical, and such experiments are often difficult to control. Here we report a method called PRISM that allows pooled screening of mixtures of cancer cell lines by labeling each cell line with 24-nucleotide barcodes. PRISM revealed the expected patterns of cell killing seen in conventional (unpooled) assays. In a screen of 102 cell lines across 8,400 compounds, PRISM led to the identification of BRD-7880 as a potent and highly specific inhibitor of aurora kinases B and C. Cell line pools also efficiently formed tumors as xenografts, and PRISM recapitulated the expected pattern of erlotinib sensitivity in vivo

    Star formation in bright-rimmed clouds and cluster associated with W5 E H{\sc ii} region

    The aim of this paper is to present the results of photometric investigations of the central cluster of the W5 E region as well as a follow-up study of the triggered star formation in and around bright-rimmed clouds (BRCs). We have carried out wide field UBVIcUBVI_c and deep VIcVI_c photometry of the W5 E H{\sc ii} region. A distance of ∼\sim2.1 kpc and a mean age of ∼\sim1.3 Myr have been obtained for the central cluster. The young stellar objects (YSOs) associated with the region are identified on the basis of near-infrared and mid-infrared observations. We confirmed our earlier results that the average age of the YSOs lying on/inside the rim are younger than those lying outside the rim. The global distribution of the YSOs shows an aligned distribution from the ionising source to the BRCs. These facts indicate that a series of radiation driven implosion processes proceeded from near the central ionising source towards the periphery of the W5 E H{\sc ii} region. We found that, in general, the age distributions of the Class II and Class III sources are the same. This result is apparently in contradiction with the conclusion by Bertout, Siess & Cabrit (2007) and Chauhan et al. (2009) that classical T Tauri stars evolve to weak-line T Tauri stars. The initial mass function of the central cluster region in the mass range 0.4≤M/M⊙≤300.4 \le M/M_\odot \le 30 can be represented by Γ=−1.29±0.03\Gamma = -1.29 \pm 0.03. The cumulative mass functions indicate that in the mass range 0.2≤M/M⊙≤0.80.2 \le M/M_\odot \le 0.8, the cluster region and BRC NW have more low mass YSOs in comparison to BRCs 13 and 14.Comment: 43 pages, 17 figures, accepted for publication in MNRA

    Identification and Characterization of Optimal Gene Expression Markers for Detection of Breast Cancer Metastasis

    Sentinel lymph node (SLN) status is highly predictive of overall axillary lymph node involvement in breast cancer. Historically, SLN-positive patients have undergone axillary lymph node dissection in a second surgery. Intraoperative SLN analysis could reduce the cost and complications of a second surgery; however, existing histopathological methods lack standardization and exhibit poor sensitivity. Rapid molecular methods may lead to improved intraoperative diagnosis of SLN metastasis. In this study,we used a genome-wide gene expression analysis of breast and other tissues to identify seven putative markers for detecting breast cancer metastasis. We assessed the utility of these markers for identifying clinically actionable metastases in lymph nodes through reverse transcriptase-polymerase chain reaction analysis of SLNs from 254 breast cancer patients. Polymerase chain reaction signals were compared to pathology on a per-patient basis. The optimal two-gene combination, mammaglobin and cytokeratin 19, detected clinically actionable metastasis in breast SLNs with 90% sensitivity and 94% specificity. Application of stringent criteria for identifying presumptive hematoxylin- and eosin-positive samples increased sensitivity and specificity to 91 and 97%, respectively. This study represents the first comprehensive demonstration of the utility of gene expression markers for detecting clinically actionable breast metastases. An intraoperative molecular assay using these markers has the potential to significantly reduce second surgeries for patients undergoing SLN dissection.
