    Use of a Global Positioning System (GPS) to Manage Extensive Sheep Farming and Pasture Land

    The terrestrial climate is not sufficient to produce enough food to meet the roughage needs of the animals benefiting from the pasture lands because of excessive and early grazing of those areas. Plant growth is adversely affected in pastures that are not uniformly grazed. Tracking animals using the Global Positioning System (GPS) is a very important factor in determining the uniform distribution of grazing animals in a pasture, increasing the utilization rate of the pasture, and saving costs and time. With GPS tracking systems, establishing more effective pasture-use systems by monitoring the feeding regimes of small animals, the status of feed in the pasture, and the grazing behavior of the animals would be possible. The present study aimed to investigate the use of GPS for pasture and herd management in Turkey in addition to using the traditional techniques.In the present study conducted in the village of Köseyusuflu in Yozgat Province in May 2017, 2018, and 2019, grazing benefits that were determined from the pasture containing two Akkaraman sheep herds were recorded using GPS tracking devices. The results suggested that the area covered with vegetation along the sheep’s spring grazing routes varied between 43.6 and 62.9%, the ratio of legumes in the pasture grass in the low grazing areas was between 0.50 and 4.10%, and the grass species were between 12.75 and 44.50%. We determined that the sheep in herd A traveled between 7.6 and 9.9 km, while the sheep in herd B traveled between 4.7 and 5.7 km daily, and the two herds grazed an average of between 122 and 254 daa

    Incidence of fibroblast growth factor receptor 3 gene (FGFR3) A248C, S249C, G372C, and T375C mutations in bladder cancer

    Bladder cancer is the most frequent cancer of the urinary system. Fibroblast growth factor receptors (FGFR) belong to the tyrosine kinase family and have important roles in cell differentiation and proliferation and embryogenesis. FGFR3 is located on chromosome 4p16.3, and missense mutations of FGFR3 are associated with autosomal dominant human skeletal disorders and have some oncogenic effects. We examined the incidence of FGFR3 thanatophoric dysplasia mutations located in exon 7, A248C and S249C, and in exon 10, G372C and T375C, and their correlation with clinical-pathological parameters in bladder carcinoma patients. Fifty-six paraffin-embedded specimens of transitional cell carcinoma of the urinary bladder were included in this study. Analysis of FGFR3 thanatophoric dysplasia mutations located in exon 7, A248C and S249C, and in exon 10, G372C and T375C, was performed by PCR- restriction fragment length polymorphism (RFLP) analysis and DNA sequencing. FGFR3 thanatophoric dysplasia mutations located in exon 7, A248C and S249C, and in exon 10, G372C and T375C, were detected in 33 of the 56 patients (heterozygous mutant). Among the 56 transitional cell carcinomas, missense point mutations were detected in seven of them at codon A248C, 28 of them at codon S249C, and three of them at codon T375C, similar to data from previous reports. When the results of the FGFR3 thanatophoric dysplasia mutations located in exon 7, A248C and S249C and in exon 10, G372C and T375C, were analyzed one by one or as a group, despite the findings of previous research reports, our data suggest that these mutations are detected homogenously regardless of the tumor classification and tumor grade. © FUNPEC-RP

    Restoration of Noradrenergic Function in Parkinson’s Disease Model Mice

    Dysfunction of the central noradrenergic and dopaminergic systems is the primary neurobiological characteristic of Parkinson’s disease (PD). Importantly, neuronal loss in the locus coeruleus (LC) that occurs in early stages of PD may accelerate progressive loss of dopaminergic neurons. Therefore, restoring the activity and function of the deficient noradrenergic system may be an important therapeutic strategy for early PD. In the present study, the lentiviral constructions of transcription factors Phox2a/2b, Hand2 and Gata3, either alone or in combination, were microinjected into the LC region of the PD model VMAT2 Lo mice at 12 and 18 month age. Biochemical analysis showed that microinjection of lentiviral expression cassettes into the LC significantly increased mRNA levels of Phox2a, and Phox2b, which were accompanied by parallel increases of mRNA and proteins of dopamine β-hydroxylase (DBH) and tyrosine hydroxylase (TH) in the LC. Furthermore, there was considerable enhancement of DBH protein levels in the frontal cortex and hippocampus, as well as enhanced TH protein levels in the striatum and substantia nigra. Moreover, these manipulations profoundly increased norepinephrine and dopamine concentrations in the striatum, which was followed by a remarkable improvement of the spatial memory and locomotor behavior. These results reveal that over-expression of these transcription factors in the LC improves noradrenergic and dopaminergic activities and functions in this rodent model of PD. It provides the necessary groundwork for the development of gene therapies of PD, and expands our understanding of the link between the LC-norepinephrine and dopamine systems during the progression of PD

    Estimates for heritability and consumer-validation of a penetrometer method for phenotyping softness of cooked cassava roots

    Although breeders have made significant progress in the genetic improvement of cassava ( Manihot esculenta Crantz) for agronomic traits, lack of information on heritability and limited testing of high-throughput phenotyping methods are major limitations to improving root quality traits, such as softness after cooking, which rank high among Ugandan consumers. The objectives of this study were to determine heritability for softness of cooked cassava roots, and quantify the relationship between penetrometer and consumer testing methods for phenotyping softness of cassava roots. Softness defined as the maximum force (N) needed to penetrate cooked root samples using a penetrometer, was evaluated at four cooking time intervals: 15, 30, 45, and 60 min on 268 cassava genotypes. Estimates of broad-sense heritability (repeatability) ranged from 0.17 to 0.37, with the highest value observed at 45 min of cooking time interval. In the second study involving 135 cassava consumers from Kibaale district in Uganda, penetrometer measurements of cooked roots from six cassava varieties were found to be in strong agreement (r2 = 0.91; P-value = 0.003) with ordinal scores of root softness from consumer testing. These results suggest that: (a) softness of cooked cassava roots is a trait amenable for evaluation and selection; and (b) a penetrometer can readily be used for assessment of cooked root softness. These findings form the basis for operationalising the routine assessment of root softness in cassava breeding trials, an output that will enhance ongoing efforts to breed for desired end-user root quality traits.Les s\ue9lectionneurs ont fait des progr\ue8s consid\ue9rables dans l\u2019am\ue9lioration g\ue9n\ue9tique des caract\ue8res agronomiques du manioc ( Manihot esculenta Crantz).     Understanding cassava varietal preferences through pairwise ranking of gari-eba and fufu prepared by local farmer-processors

    Within communities in Osun and Imo States of Nigeria, farmer‐processors grew and processed a diverse set of improved and landrace cassava varieties into the locally popular foods, gari, eba and fufu. Local and 15 main varieties were grown in a 'mother and baby trials' design in each state. Mother trials with three replications were processed by farmer‐processors renown in their community for their processing skills. Baby trials were managed and processed by other farmer‐processors. The objective was to identify food quality criteria to inform demand‐led breeding to benefit users, especially women, given their key roles in processing. Farmer‐processors evaluated the overall quality of fresh roots and derived food products through pairwise comparisons. Improved varieties had higher fresh and dry root yield. Overall, landraces ranked first for quality of gari and eba, but several improved varieties were also appreciated for good quality. Landraces in Osun had higher gari yield and a higher swelling power compared to improved varieties. Colour (browning), bulk density, swelling power, solubility and water absorption capacity were the criteria most related to food product ranking by farmer‐processors. Evaluation of varieties under farmer‐processors' conditions is crucial for providing guidance to breeders on critical selection criteria

    Baicalin administration attenuates hyperglycemia-induced malformation of cardiovascular system

    In this study, the effects of Baicalin on the hyperglycemia-induced cardiovascular malformation during embryo development were investigated. Using early chick embryos, an optimal concentration of Baicalin (6 μM), was identified which could prevent hyperglycemia-induced cardiovascular malformation of embryos. Hyperglycemia-enhanced cell apoptosis was reduced in embryos and HUVECs in the presence of Baicalin. Hyperglycemia-induced excessive ROS production was inhibited when Baicalin was administered. Analyses of SOD, GSH-Px, MAQE and GABAA suggested Baicalin plays an antioxidant role in chick embryos possibly through suppression of outwardly rectifying Cl(-) in the high-glucose microenvironment. In addition, hyperglycemia-enhanced autophagy fell in the presence of Baicalin, through affecting the ubiquitin of p62 and accelerating autophagy flux. Both Baicalin and Vitamin C could decrease apoptosis, but CQ did not, suggesting autophagy to be a protective function on the cell survival. In mice, Baicalin reduced the elevated blood glucose level caused by streptozotocin (STZ). Taken together, these data suggest that hyperglycemia-induced embryonic cardiovascular malformation can be attenuated by Baicalin administration through suppressing the excessive production of ROS and autophagy. Baicalin could be a potential candidate drug for women suffering from gestational diabetes mellitus

    Giant cervicothoracic extradural arachnoid cyst: case report

    The pathogenesis, etiology, and treatment of the spinal arachnoid cyst have not been well established because of its rarity. A 57-year-old male was presented with spastic quadriparesis predominantly on the left side. His radiological examination showed widening of the cervical spinal canal and left neural foramina due to a cerebrospinal fluid - filled extradural cyst that extended from C2 to T2 level. The cyst was located left anterolaterally, compressing the spinal cord. Through a C4–T2 laminotomy, the cyst was excised totally and the dural defect was repaired. Several features of the reported case, such as cyst size, location, and clinical features make it extremely unusual. The case is discussed in light of the relevant literature

    Solar flare prediction using advanced feature extraction, machine learning and feature selection

    YesNovel machine-learning and feature-selection algorithms have been developed to study: (i) the flare prediction capability of magnetic feature (MF) properties generated by the recently developed Solar Monitor Active Region Tracker (SMART); (ii) SMART's MF properties that are most significantly related to flare occurrence. Spatio-temporal association algorithms are developed to associate MFs with flares from April 1996 to December 2010 in order to differentiate flaring and non-flaring MFs and enable the application of machine learning and feature selection algorithms. A machine-learning algorithm is applied to the associated datasets to determine the flare prediction capability of all 21 SMART MF properties. The prediction performance is assessed using standard forecast verification measures and compared with the prediction measures of one of the industry's standard technologies for flare prediction that is also based on machine learning - Automated Solar Activity Prediction (ASAP). The comparison shows that the combination of SMART MFs with machine learning has the potential to achieve more accurate flare prediction than ASAP. Feature selection algorithms are then applied to determine the MF properties that are most related to flare occurrence. It is found that a reduced set of 6 MF properties can achieve a similar degree of prediction accuracy as the full set of 21 SMART MF properties
