    Se realizó una investigación de Desarrollo Tecnológico, descriptiva, transversal y prospectiva, con el objetivo de evaluar el estado funcional e integral de los adultos mayores pertenecientes al Consultorio Médico No. 262, Grupo Básico de trabajo No.5, Consejo Popular "Capitán San Luís", Policlínico Universitario "Turcios Lima", Municipio Pinar del Río en el período comprendido de julio-diciembre de 2006. El universo está constituido por los 110 ancianos de nuestro consultorio, correspondiéndose con la muestra. Para la recogida de la información se utilizó el análisis de la situación de salud del sector, las historias de salud familiar e individuales, también se aplicó un cuestionario siempre bajo la voluntariedad y aceptación de las personas a participar en nuestra investigación, basado en la Escala Geriátrica de Evaluación Funcional(EGEF), así como en el Índice de Actividades Básicas de la Vida Diaria (ABVD) de Katz y la Escala de Actividades Instrumentadas de la Vida Diaria de Lawton-Brody. Para su análisis estadístico se aplicó el Test de proporción de variables. Los resultados demostraron que el sexo femenino (63.64 %) y las edades de 60-64 años (22.73 %) fueron los que predominaron, constatándose un envejecimiento demográfico. El uso de medicamentos se detectó en el 70 % de los gerontes mientras la movilidad y el equilibrio estaban afectados en el 60.91 % y 56.36 %, respectivamente. La memoria fue el componente de la esfera psicológica más frecuentemente afectado (56.64 %). Las mujeres presentaron mayores dificultades que los hombres en la mayoría de las esferas, y existe en ellas también un predominio de las alteraciones del estado funcional e integral. Los ancianos independientes, activos y satisfechos con la vida que llevan, se presentaron de forma más relevante en el sexo masculino. Palabras clave: evaluación geriátrica, adulto mayor. To record the information an analysis of the health situation in the sector was performed, reviewing medical and familial histories, a questionnaire was applied always under voluntary nature, and the agreement of the persons to participate in the research based on the Scale of the Functional Geriatric Assessment (SFGA); as well as in the Katz's Rate of Basic Activities on Daily Life (RBADL), and the Scale of Lawton-Brody to the Instrumental Daily Activities was arranged. To the statistical analysis the Test of proportional variables was used. Results showed that female sex (63.64%) and ages 60-64 years old (22.73%) prevailed, verifying a demographic aging. The use of medications was detected in the 70% of the aged, while mobility and balance were influenced in 60.91% and 56.36%, respectively. Memory was the most affected component in the psychological sphere (56.64%). Women presented more difficulties than men in the majority of the spheres; prevailing in women disorders of the functional and general condition.     Caracterización clínico-epidemiológica de los ancianos hipertensos en casas de abuelos / Clinical epidemiological characterization of the hypertensive aged in the Elderly Home

    La hipertensión arterial es la enfermedad cardiovascular más extendida universalmente; existen más de mil millones de hipertensos en el mundo. Con el objetivo de profundizar en el conocimiento de los aspectos clínico-epidemiológicos de la hipertensión en los ancianos, se realiza una investigación descriptiva, transversal y analítica. El universo de estudio lo constituyeron 130 ancianos, de ellos 77 hipertensos. Los datos fueron recogidos a través de las historias clínicas individuales, de salud familiar y una encuesta elaborada por el autor; se obtuvieron datos generales de los pacientes, así como variables para su estudio: edad, sexo, raza, factores de riesgo, compromiso de órganos diana. La muestra seleccionada fueron tres conglomerados, las Casas de Abuelos de los municipios Pinar del Río, San Juan y Martínez y Consolación del Sur. Los datos cualitativos se resumieron en frecuencias absolutas y relativas porcentuales. Se construyeron intervalos de confianza, y realizaron pruebas de hipótesis, regresión logística y pruebas no paramétricas. Se pudo apreciar que independientemente de la edad se puede presentar o no hipertensión arterial al menos en la muestra seleccionada, prevaleciendo el sexo femenino. Hubo predominio de hipertensión en la raza negra, solo el daño vascular cerebral y el color de la piel están asociados. Los factores de riesgo más frecuentes fueron el tabaquismo y la dislipidemia. With the purpose of deepening in the knowledge of the clinical-epidemiological hypertension in the elderly a descriptive, cross-sectional and analytic research was carried out. The universe of the study was comprised of 130 aged, out of them 77 hypertensive. Data were collected by means of the individual clinical histories, familial health and a survey designed by the author; obtaining patient's general data, and variables to the study: age, sex, race, risk factors, and influence in target organs. The sample chosen took three conglomerates, the Houses of the Elderly in Pinar del Rio, San Juan y Martinez and Consolacion del Sur. Reviewing qualitative data in absolute and relative frequencies. Confidence intervals, hypothesis testing, logistic regressions and non-parametric tests were made. Showing that regardless of age; hypertension can or cannot be present at least in the sample chosen, prevailing female sex. Observing that hypertension prevailed in black race, and that only vascular brain damage and the color of the skin had association. The most common risk factors were smoking and dyslipemia.     Light induced decoupling of electronic and magnetic properties in manganites

    The strongly correlated material La0.7Sr0.3MnO3 (LSMO) exhibits metal-to-insulator and magnetic transition near room temperature. Although the physical properties of LSMO can be manipulated by strain, chemical doping, temperature, or magnetic field, they often require large external stimuli. To include additional flexibility and tunability, we developed a hybrid optoelectronic heterostructure that uses photocarrier injection from cadmium sulfide (CdS) to an LSMO layer to change its electrical conductivity. LSMO exhibits no significant optical response, however, the CdS/LSMO heterostructures show an enhanced conductivity, with ~ 37 % resistance drop, at the transition temperature under light stimuli. This enhanced conductivity in response to light is comparable to the effect of a 9 T magnetic field in pure LSMO. Surprisingly, the optical and magnetic responses of CdS/LSMO heterostructures are decoupled and exhibit different effects when both stimuli are applied. This unexpected behavior shows that heterostructuring strongly correlated oxides may require a new understanding of the coupling of physical properties across the transitions and provide the means to implement new functionalities

    Ultrafine anaphase bridges, broken DNA and illegitimate recombination induced by a replication fork barrier

    Most DNA double-strand breaks (DSBs) in S- and G2-phase cells are repaired accurately by Rad51-dependent sister chromatid recombination. However, a minority give rise to gross chromosome rearrangements (GCRs), which can result in disease/death. What determines whether a DSB is repaired accurately or inaccurately is currently unclear. We provide evidence that suggests that perturbing replication by a non-programmed protein–DNA replication fork barrier results in the persistence of replication intermediates (most likely regions of unreplicated DNA) into mitosis, which results in anaphase bridge formation and ultimately to DNA breakage. However, unlike previously characterised replication-associated DSBs, these breaks are repaired mainly by Rad51-independent processes such as single-strand annealing, and are therefore prone to generate GCRs. These data highlight how a replication-associated DSB can be predisposed to give rise to genome rearrangements in eukaryotes

    Development of a rapid, sensitive, and field-deployable Razor Ex BioDetection system and quantitative PCR assay for detection of Phymatotrichopsis omnivora using multiple gene targets

    A validated, multigene-based method using real-time quantitative PCR (qPCR) and the Razor Ex BioDetection system was developed for detection of Phymatotrichopsis omnivora. This soilborne fungus causes Phymatotrichopsis root rot of cotton, alfalfa, and other dicot crops in the southwestern United States and northern Mexico, leading to significant crop losses and limiting the range of crops that can be grown in soils where the fungus is established. It is on multiple lists of regulated organisms. Because P. omnivora is difficult to isolate, accurate and sensitive culture-independent diagnostic tools are needed to confirm infections by this fungus. Specific PCR primers and probes were designed based on P. omnivora nucleotide sequences of the genes encoding rRNA internal transcribed spacers, beta-tubulin, and the second-largest subunit of RNA polymerase II (RPB2). PCR products were cloned and sequenced to confirm their identity. All primer sets allowed early detection of P. omnivora in infected but asymptomatic plants. A modified rapid DNA purification method, which facilitates a quick (about 30-min) on-site assay capability for P. omnivora detection, was developed. Combined use of three target genes increased the assay accuracy and broadened the range of detection. To our knowledge, this is the first report of a multigene-based, field-deployable, rapid, and reliable identification method for a fungal plant pathogen and should serve as a model for the development of field-deployable assays of other phytopathogens.Peer reviewedEntomology and Plant PathologyBiochemistry and Molecular Biolog

    Identification of ovule transcripts from the Apospory-Specific Genomic Region (ASGR)-carrier chromosome

    <p>Abstract</p> <p>Background</p> <p>Apomixis, asexual seed production in plants, holds great potential for agriculture as a means to fix hybrid vigor. Apospory is a form of apomixis where the embryo develops from an unreduced egg that is derived from a somatic nucellar cell, the aposporous initial, via mitosis. Understanding the molecular mechanism regulating aposporous initial specification will be a critical step toward elucidation of apomixis and also provide insight into developmental regulation and downstream signaling that results in apomixis. To discover candidate transcripts for regulating aposporous initial specification in <it>P. squamulatum</it>, we compared two transcriptomes derived from microdissected ovules at the stage of aposporous initial formation between the apomictic donor parent, <it>P. squamulatum </it>(accession PS26), and an apomictic derived backcross 8 (BC<sub>8</sub>) line containing only the Apospory-Specific Genomic Region (ASGR)-carrier chromosome from <it>P. squamulatum</it>. Toward this end, two transcriptomes derived from ovules of an apomictic donor parent and its apomictic backcross derivative at the stage of apospory initiation, were sequenced using 454-FLX technology.</p> <p>Results</p> <p>Using 454-FLX technology, we generated 332,567 reads with an average read length of 147 base pairs (bp) for the PS26 ovule transcriptome library and 363,637 reads with an average read length of 142 bp for the BC<sub>8 </sub>ovule transcriptome library. A total of 33,977 contigs from the PS26 ovule transcriptome library and 26,576 contigs from the BC<sub>8 </sub>ovule transcriptome library were assembled using the Multifunctional Inertial Reference Assembly program. Using stringent <it>in silico </it>parameters, 61 transcripts were predicted to map to the ASGR-carrier chromosome, of which 49 transcripts were verified as ASGR-carrier chromosome specific. One of the alien expressed genes could be assigned as tightly linked to the ASGR by screening of apomictic and sexual F<sub>1</sub>s. Only one transcript, which did not map to the ASGR, showed expression primarily in reproductive tissue.</p> <p>Conclusions</p> <p>Our results suggest that a strategy of comparative sequencing of transcriptomes between donor parent and backcross lines containing an alien chromosome of interest can be an efficient method of identifying transcripts derived from an alien chromosome in a chromosome addition line.</p

    HMDB 5.0: the Human Metabolome Database for 2022

    The Human Metabolome Database or HMDB (https://hmdb.ca) has been providing comprehensive reference information about human metabolites and their associated biological, physiological and chemical properties since 2007. Over the past 15 years, the HMDB has grown and evolved significantly to meet the needs of the metabolomics community and respond to continuing changes in internet and computing technology. This year's update, HMDB 5.0, brings a number of important improvements and upgrades to the database. These should make the HMDB more useful and more appealing to a larger cross-section of users. In particular, these improvements include: (i) a significant increase in the number of metabolite entries (from 114 100 to 217 920 compounds); (ii) enhancements to the quality and depth of metabolite descriptions; (iii) the addition of new structure, spectral and pathway visualization tools; (iv) the inclusion of many new and much more accurately predicted spectral data sets, including predicted NMR spectra, more accurately predicted MS spectra, predicted retention indices and predicted collision cross section data and (v) enhancements to the HMDB's search functions to facilitate better compound identification. Many other minor improvements and updates to the content, the interface, and general performance of the HMDB website have also been made. Overall, we believe these upgrades and updates should greatly enhance the HMDB's ease of use and its potential applications not only in human metabolomics but also in exposomics, lipidomics, nutritional science, biochemistry and clinical chemistry.Analytical BioScience

    MAMMALS IN PORTUGAL : A data set of terrestrial, volant, and marine mammal occurrences in P ortugal

    Mammals are threatened worldwide, with 26% of all species being includedin the IUCN threatened categories. This overall pattern is primarily associatedwith habitat loss or degradation, and human persecution for terrestrial mam-mals, and pollution, open net fishing, climate change, and prey depletion formarine mammals. Mammals play a key role in maintaining ecosystems func-tionality and resilience, and therefore information on their distribution is cru-cial to delineate and support conservation actions. MAMMALS INPORTUGAL is a publicly available data set compiling unpublishedgeoreferenced occurrence records of 92 terrestrial, volant, and marine mam-mals in mainland Portugal and archipelagos of the Azores and Madeira thatincludes 105,026 data entries between 1873 and 2021 (72% of the data occur-ring in 2000 and 2021). The methods used to collect the data were: live obser-vations/captures (43%), sign surveys (35%), camera trapping (16%),bioacoustics surveys (4%) and radiotracking, and inquiries that represent lessthan 1% of the records. The data set includes 13 types of records: (1) burrowsjsoil moundsjtunnel, (2) capture, (3) colony, (4) dead animaljhairjskullsjjaws, (5) genetic confirmation, (6) inquiries, (7) observation of live animal (8),observation in shelters, (9) photo trappingjvideo, (10) predators dietjpelletsjpine cones/nuts, (11) scatjtrackjditch, (12) telemetry and (13) vocalizationjecholocation. The spatial uncertainty of most records ranges between 0 and100 m (76%). Rodentia (n=31,573) has the highest number of records followedby Chiroptera (n=18,857), Carnivora (n=18,594), Lagomorpha (n=17,496),Cetartiodactyla (n=11,568) and Eulipotyphla (n=7008). The data setincludes records of species classified by the IUCN as threatened(e.g.,Oryctolagus cuniculus[n=12,159],Monachus monachus[n=1,512],andLynx pardinus[n=197]). We believe that this data set may stimulate thepublication of other European countries data sets that would certainly contrib-ute to ecology and conservation-related research, and therefore assisting onthe development of more accurate and tailored conservation managementstrategies for each species.
